Abstract

Three rhizobacteria (Bacillus polymixa  BG25,  Pseudomonas fluorescens  PG01 and Serratia liquefaciens SG01) which inhibited the conidia germination of Colletotrichum capsici  and produced the phytohormone indoleacetic acid (IAA) were selected and evaluated as biocontrol agents for controlling Colletotrichum capsici, the causal agent of hot pepper anthracnose and for improving the growth, yield and seed quality of hot pepper in glasshouse and field conditions. Two cultivars of hot pepper, cv. Lokal Brebes and cv. Tit Super were also used in this experiment. The result of the experiment showed that seed treatments with P. fluorescens PG01 either alone or in combination with B. polymixa BG25 increase yields and seed quality under glasshouse and field conditions. However, the most effective treatment was the combination of both agents. Moreover, biological seed  treatment with P. fluorescens PG01 either alone or in combination with B. polymixa BG25 led to induction of resistance against C. capsici, as a result of increase in peroxidase activity and biosynthesis of phytoalexins that have been considered as resistance mechanisms against plant diseases.

Abstract

Characterization and activities assay of rhizosphere bacteria from ultisol land for plant-growth promoting and biocontrol agents of soil-borne fungus pathogens under in vitro test. Although many studies have been conducted to identify the specific traits of the plant growth-promoting and bioprotecting rhizobacteria (PGPBR), they were limited to studying specific PGPBR isolates from ultisol lands. We selected 273 isolates from bulk soil and plant rhizosphere and examined them for a wide array traits that might inhibit the growth of plant pathogens and increase early cucumber growth in ultisol soil. A subsample of 25 isolates, all positively produce chitinase and sellulose enzymes, 18 positively produce protease and 7 were fluorescens on KB medium under UV lighting. All isolates could produce IAA and be able to solubilize phosphor in vitro test, 10 exhibited low level of nitrogenase activity. Futher test showed that out of 25 isolates, 12 inhibited F. oxysporum, P. capsici, R. solani and S. rolfsii in vitro. All isolates increased seed germination, but only 5 isolates significanty increased early cucumber growth in ultisol soil. The results suggest that rhizobacteria be able to produce extracelluler enzymes, siderophore, ACC deasiminase, and IAA or those which are able to solubilize phosphor in vitro may be potential to be uses as biofertilizer and biological control agents in ultisol land.

Abstract

This research was aimed to examine and get biomatriconditioning technique that increase viability and seed vigor rice (Oryza sativaL.). This research was conducted at Agronomi Unit of the Laboratory Agrotechnology of Haluoleo University, Kendari from January until March 2013. This research used completely randomized design with factorial. The first factor was the variety: Konawe and Inpari 10, the second factor was biomatriconditioningtechnique Control, Bacillus CKD061+Husk Charcoal Powder, Bacillus CKD061+Powder Red Brick. Serratia CMN 175 + Husk Charcoal Powder, Serratia CMN175+Red Brick powder. For the all numbers there were 14 combination treatments, repeated for 3 times, so the total of experimental units were 42. Replication consisted of 5 polybag so there were 210 plants. Data were analyzed using a manner sidik, followed by Test Honestly Significant Difference. The results showed that the best biomatriconditioning for Konawe varieties was Serratia CMN175 + rice husk powder treatment, while for the Inpari varieties 10 Bacillus CKD061 + husk powder for the both of treatment could be able to improve simultaneity grow, vigor index, relative growth speeds, T50, plant height and number of tillers.Keywords: Bacillus CKD061, P. fluorescensPG01, seed of rice, Serratia

Abstract

Aunupe Village, Wolasi District is one of the villages for the development of organic vegetables in Konawe Selatan Regency. The frequent occurrence of high levels of pest and disease attacks is the main reason for the use of synthetic pesticides that are not justified in organic farming systems. On the other hand, in the same location, there is a group of women farmers who are not productive. Both of these problems can be overcome if the farmer women's group produces quality biological agents that can be utilized by horticultural farmers in an environmentally friendly cultivation system. The implementation of the PKM Program in Aunupe Village is very appropriate as a means and media for transferring technology on how to increase the productivity of environmentally friendly vegetable plants by developing organic farming techniques. Control of pests and plant diseases by using biological agents and botanical pesticides is a wise choice in efforts to increase agricultural production with high economic value while maintaining biodiversity to support sustainable agricultural cultivation, especially in the cultivation of vegetable crops as a source of community nutrition.

Abstract

Persistence and Breaking of Local Chilli Seed Dormancy Using Seed Bio-Invigoration Techniques. This study aims to evaluate the persistence of local chilli seed dormancy while evaluating seed bio-invigoration techniques that effectively break the dormancy of local chilli. The study consisted of two series of experiments, namely: the persistence test of local chilli. seed dormancy (Series 1) was observed descriptively using a germination indicator for several weeks to reach a germination percentage ≥ 80%. Testing of seed dormancy breakage by seed bio-invigoration technique (Series 2) using a randomized complete design with 3 replications and 7 treatments namely: no treatment (A0), matriconditioning of husk charcoal powder (SAS) + Bacillus sp. CKD061 (A1), matriconditioning SAS + Pseudomonas sp. TBT214 (A2), red brick powder matriconditioning (SBM) + Bacillus sp. CKD061 (A3), matriconditioning SBM + Pseudomonas sp. TBT214 (A4), matriconditioning SAS + Bacillus sp. CKD061 + Pseudomonas sp. TBT214 (A5), and matriconditioning SBM + Bacillus sp. CKD061 + Pseudomonas sp. TBT214 (A6). The results showed that the persistence of local chilli seed dormant cultivars Konsel 1 and Konsel 2 cultivars were 6 weeks. While the dormancy break test on the Konsel 1 cultivar was 2 weeks at A1 treatment with breaking dormancy of 90.00%. While in Konsel 2 cultivar which is 4 weeks in the A6 treatment with breaking dormancy  86.67% which is significantly different from the control. This study shows that A1 treatment can break dormancy of chilli seeds in Konsel 1 cultivar and A6 treatment in Konsel 2 cultivar.

Abstract

Penggunaan agens hayati merupakan trend perkembangan pertanian saat ini, diantaranya endo-rhizobakteri. Endo-rhizobakteri mampu menghasilkan hormon tumbuh, melarutkan fosfat dan memfiksasi nitrogen, yang berperan dalam perkecambahan dan perkembangan tanaman. Penelitian ini bertujuan untuk meningkatkan vigor benih padi gogo lokal yang diaplikasi konsorsium endo-rhizobakteri. Penelitian dilakukan di Laboratorium Agroteknologi Unit Agronomi Fakultas Pertanian Universitas Halu Oleo, Kendari pada bulan April 2018. Penelitian menggunakan rancangan acak lengkap, dengan 5 perlakuan yaitu: kontrol, isolat Be02 + PKLK, isolat Be02 + Bacillus sp. CKD061, isolat PKLK5 + Bacillus sp. CKD061 dan isolat Be02 + PKLK5 + Bacillus sp. CKD061. Setiap perlakuan diulang 3 kali sehingga diperoleh 15 unit percobaan. Pengamatan dilakukan terhadap daya berkecambah, potensi tumbuh maksimum, indeks vigor, keserempakkan tumbuh, kecepatan tumbuh relatif, T50 dan panjang akar. Data hasil pengamatan dianalisis menggunakan analisis ragam dan dilanjutkan dengan uji DMRTα=0,05. Hasil penelitian konsorsium endo-rhizobakteri mampu meningkatkan viabilitas dan vigor benih padi gogo lokal. Perlakuan konsorsium endo-rhizobakteri isolat Be02 + PKLK5 + Bacillus sp. CKD061 efektif meningkatkan daya berkecambah (25,49%) dan potensi tumbuh maksimum (26,32%) bila dibandingkan dengan kontrol. Perlakuan konsorsium endo-rhizobakteri isolat Be02 + Bacillus sp. CKD061 efektif meningkatkan indeks vigor (36,58%), keserempakkan tumbuh (18,75%), T50 (20,06%) dan panjang akar (42,76%) bila dibandingkan dengan kontrol

Abstract

Penelitian ini bertujuan untuk mengkaji bagaimana pengaruh teknik LEISA terhadap pertumbuhan dan hasil tanaman bawang merah lokal di Kota Baubau, mengkaji bagaimana pengaruh teknik LEISA terhadap pertumbuhan dan hasil tanaman bawang merah lokal di Kota Baubau dan mengkaji perlakuan yang memberi pengaruh terbaik terhadap pertumbuhan dan hasil tanaman bawang merah lokal di Kota Baubau. Penelitian ini dilaksanakan di Laboratorium Lapangan Sekolah Menengah Kejuruan Negeri 5 (SMKN 5) Kecamatan Gonda Baru, Kota Baubau, Sulawesi Tenggara yang berlangsung pada bulan November 2020 sampai Februari 2021. Penelitian ini dilaksanakan menggunakan Rancangan Acak Kelompok Tunggal (RAK) dengan perlakuan Teknik LEISA yang terdiri atas 6 (enam) taraf yaitu P0=Tanpa aplikasi, P1=Pupuk Organik 5 ton ha-1, P2=Pupuk NPK Majemuk 250 kg ha-1, P3= Pupuk Organik 5 ton ha-1 + Pupuk NPK Majemuk 250 kg ha-1, P4= Pupuk Organik 5 ton ha-1 + 1/ 2 Pupuk NPK Majemuk 250 kg ha-1, P5= Pupuk Organik 5 ton ha-1 + 1/ 4 Pupuk NPK Majemuk 250 kg ha-1 .Dengan demikian setiap perlakuan diulang 3 kali, sehingga terdapat 18 unit percobaan. Variabel yang diamati yaitu tinggi tanaman, jumlah daun, jumlah anakan, berat kering, jumlah umbi, berat umbi dan produksi per petak. Data hasil pengamatan dianalisis ragam dan data yang menunjukkan pengaruh nyata atau sangat nyata dilanjutkan dengan Uji Jarak Berganda Duncan (UJBD) pada taraf nyata α = 0,05. Hasil penelitian menunjukkan bahwa LEISA mampu meningkatkan pertumbuhan dan hasil tanaman bawang merah terutama pada variabel jumlah daun, jumlah anakan, bobot kering tanaman, jumlah umbi, berat umbi dan produksi per petak. Perlakuan sistem LEISA terbaik diperoleh pada perlakuan Pupuk Organik 5 ton ha-1 + Pupuk NPK Majemuk 250 kg ha-1 (P3) dengan dibandingkan dengan kontrol.

Abstract

Pengembangan tanaman temu-temuan dan empon-empon organik memiliki prospek yang sangat cerah dan menjanjikan karena terbukti mampu meningkatkan daya tahan tubuh terhadap berbagai penyakit termasuk virus. Kondisi dunia saat ini yang tengah dilanda pandemi coronavirus disease 2019 (covid-19), memerlukan perhatian serius dari semua komponen masyarakat untuk ikut berkontribusi dalam memerangi wabah ini. Salah satunya adalah melalui upaya masyarakat itu sendiri dalam memerangi virus dengan mengkonsumsi makanan yang dapat meningkatkan daya tahan tubuh seperti jenis temu-temuan dan empon-empon, sehingga tubuh tetap sehat dan fit. Sampai saat ini, sebagian besar masyarakat menanam tanaman temu-temuan dan empon-empon terbatas hanya untuk memenuhi kebutuhan pribadi, padahal jenis tanaman ini dapat dikembangkan lebih luas dalam skala komersial. Kegiatan pengabdian kepada masyarakat (Program Kreativitas Masyarakat Internal) ini, dilaksanakan dengan tujuan membantu memberikan pengertian dan pemahaman kepada peserta tentang besarnya manfaat tanaman temu-temuan dan empon-empon sebagai bahan obat-obatan herbal yang dapat meningkatkan daya tahan tubuh terhadap berbagai infeksi penyakit termasuk yang disebabkan oleh virus seperti Covid-19. Sasaran PKMI ini adalah ibu-ibu Kelompok Dasawisma Mepokoaso di Kelurahan Tobimeita Kecamatan Abeli Kota Kendari. Metode kegiatan dilaksanakan dalam bentuk penyuluhan dan bimbingan teknis budidaya tanaman temu-temuan dan empon-empon berbasis teknologi ramah lingkungan. Masyarakat sasaran sangat antusias mengikuti kegiatan ini, mereka menyatakan akan meneruskan kegiatan secara mandiri. Luaran kegiatan PKMI ini adalah peningkatan keberdayaan kelompok tani sasaran, eksistensi dan keberlanjutan usaha agribisnis tanaman temu-temuan dan empon-empon organik yang berimplikasi pada peningkatan pendapatan kelompok tani sasaran, artikel ilmiah yang dipublikasikan pada jurnal nasional (Jurnal Pepadu), dan penerbitan artikel pada media cetak/elektronik.

Abstract

Penelitian ini bertujuan mengetahui pengaruh bahan amelioran yang dapat meningkatkan pertumbuhan dan produksi tanaman cabai rawit. Penelitian ini menggunakan Rancangan Acak Kelompok dengan 4 perlakuan bahan amelioran yaitu tanpa bahan amelioran (B0), biochar kulit buah durian 4 t ha-1(B1), biochar sekam padi 4 t ha-1 (B2), pupuk kandang sapi 4 t ha-1 (B3). Masing-masing perlakuan diulang sebanyak tiga kali. Hasil penelitian menunjukkan bahwa aplikasi biochar dan pupuk kandang sapi sebagai amelioran mampu meningkatkan tinggi tanaman, jumlah daun, luas daun total, indeks luas daun, jumlah buah, berat buah, dan produktivitas tanaman cabai rawit. Peningkatan produktivitas tanaman cabe rawit dengan aplikasi pupuk kandang sapi, biochar kulit buah durian dan biochar sekam padi dosis 4 t ha-1 yaitu berturut-turut masing-masing sebesar 83%, 70%,  dan 64 % dibandingkan dengan tanpa perlakuan.

Abstract

The research aims to determine the interaction effect of the dose of straw mulch and organic plus fertilizer on the growth of soybean (Glycine max L.) on marginal dry land. This research was conducted from August to December 2021 at the research area of the Agriculture Faculty, Halu Oleo University, Kendari, Southeast Sulawesi. The research design used was a Split Plot arranged in a Randomized Block Design. The main plot was straw mulch (M) consisting of three levels, i.e., without straw mulch (M0), 4 tons ha-1 (M1), and 8 tons ha-1(M2); and Sub-plots were plus organic fertilizer (P) consisting of four levels, namely without plus organic fertilizer (P0); 2.5 tons ha-1(P1); 5 tons ha-1, (P2); and 7.5 tons ha-1 (P3). The measured variable is plant height, number of leaves, leaf area, dry weight of root, dry weight of shoot, and . The analysis of data using ANOVA, if The result of significant (Fhit> Ftab) continues with the DMRT test on α=0.05. The results showed that the treatment of Straw mulch 8 tons ha-1 and organic fertilizer 7.5 tons ha-1 resulted in the highest soybean growth.
