Abstract

The experiment was carried out to characterize the ability of rhizobacteria isolated from the rhizosphere of healthy lowland rice in dissolving phosphate or fixing nitrogen.  The research was conducted from February to June 2011 at Agronomy Laboratory, Agriculture Faculty, Halu Oleo University. The experiment was arranged based on descriptive methods. Overall, the number of isolates characterized were 40 isolates.  The results of the isolate characterization were documentated with specific indicators (eg. the ability to dissolve phosphate in the form of halo/clear zone). The research indicated that most of these isolates had the ability to dissolve phosphate and fix nitrogen.  Isolates PKNW 6, PKMN 7,  PKNS 3, PKNS 9 and PKNW 4 showed better ability in dissolving phosphate and fixing nitrogen than other isolates evaluated. Further studies are still needed for the development of the potential isolates as biological agensts or biofertilizer. Abstract

The objective of this research was to evaluate the effect of seed invigoration on improving seed viability and vigor, and to overcome problems of postharvest physiological dormancy of upland rice seed. The research was conducted from February to June 2011 at Agrotechnology Laboratory, Faculty of Agriculture, Halu Oleo University. The experiment used randomized complete design which consisted of 13 treatments, i.e control (G0), Hidration-Dehidration (G1), Matriconditioning using ground brick (G2), Matriconditioning using ground burned-rice husk (G3), Bacillus CKD061 (G4), P. fluorescens PG01 (G5), Serratia CMN175 (G6), Biomatriconditioning using ground brick + Bacillus CKD061 (G7), Biomatriconditioning using ground brick + P. fluorescens PG01 (G8), Biomatriconditioning using ground brick + Serratia CMN175 (G9), Biomatriconditioning using ground burned-rice husk + Bacillus CKD061 (G10), Biomatriconditioning using ground burned-rice husk + P. fluorescens PG01 (G11), Biomatriconditioning using ground burned-rice husk + Serratia CMN175 (G12). Each treatment was replicated three times, therefore, overall there were 36 experimental units. Data obtained were analyzed using analysis of variance and followed with Duncan’s Multiple Range Test. The research results showed that bio-invigoration treatments integrated with rhizobacteria were effective to overcome the problems of postharvest physiological dormancy when local upland rice seeds were harvested. In addition, this treatment was also able to increase viability and vigour of the seed. Among treatments tested, the use of Bacillus sp. CKD061 or P. fluorescens PG01, either independently or integrated with matriconditioning using ground brick or ground burned- rice husk was more effective in improving viability and vigour of local upland rice seed compared to other treatments.

Abstract

The research aims to determine the effectiveness of various biological agents in increasing the growth of tomato plants. The study used a randomized block design, consisting of 10 treatments, namely without the use of biological agents, Pseudomonas sp. SWRIIA02, Bacillus sp. CKD061, Pseudomonas sp. Ab06, Pseudomonas sp. Bb05, SWRIIA02 + CKD061, Ab06 + CKD061, Bb05 + CKD061, SWRIIA02 + CKD061 and SWRII02 + CKD061 + Bb05 and each treatment was repeated 3 times. The results showed that the treatment of biological agents had a very significant effect on plant height 14 DAP, number of leaves 12 DAP, plant dry weight and significantly affected plant height 28 DAP, 42 DAP, stem diameter 42 DAP, root fresh weight, root dry weight and length root. The biological agent Pseudomonas sp. SWRIIA02, Bb05+CKD061, Bacillus sp. CKD061, SWRIIA02+CKD061+Bb05 were better at increasing the growth of local tomato plants compared to those without biological agents, with the percentage increase in plant dry weight in each of these treatments reaching 62.30%, 58.78%, 55.91% and 51.76% which was not significantly different from other treatments but significantly different from the treatment without biological agents.

Abstract

This study aims to determine the genetic diversity of several local cayenne pepper cultivars from Southeast Sulawesi and to find out which local cayenne pepper cultivars from Southeast Sulawesi show the best growth and yields. This research was carried out at the Agricultural Extension Center (BPP) Pilot Land located in Langkoroni Village, Maligano District, Muna Regency. The research implementation time starts from October 2021 to February 2022. Data analysis using variety fingerprints, if it shows a real and very real influence then continued Duncan multiple range test (DMRT) with the help of statistical product and service solution (SPSS) application facility and calculate genetic diversity value. The results showed that quantitative characters with high diversity were found in the characters of flowering age, harvest age, number of fruits, fruit weight, fruit length and fruit diameter. Somewhat high is found in the character of plant height and leaf area. Slightly low was found in the character of the number of leaves and low was found in the character of the length of the fruit stalk. The qualitative characters observed were uniform because there were no differences in the observed local cayenne pepper cultivars from Southeast Sulawesi. The local cayenne pepper cultivar from Southeast Sulawesi that showed the highest growth and fruit yield was found in the cultivar from Kolaka (145.96 grams/plant), followed by North Buton-2 (125.71 grams/plant), then South Konawe (121.93 grams/plant) compared to other local cayenne pepper cultivars.

Abstract

Rhizobacteria have been used for plant growth enhancement. The research was aimed on  finding  the  best  method  of  preplant  seed  treatment through  seed  invigoration plus rhizobacteria as biofertilizer to increase physiological quality of preplant sorghum   seed. The research was undertaken in Agronomy Laboratory, Agriculture Faculty, Haluoleo University, Kendari Southeast Sulawesi.    Research results showed that seed bio- matriconditioning with rhizobacteria was effective in enhancing seed quality of sorghum. The most effective treatment was the integration of seed matriconditioning using rice hull charcoal  or  ‘serbuk  bata  merah’  with  rhizobacteria  Pseudomonas  fluorescens  PG01  or Bacillus spp.CKD061. The seed treatment increased seedling growth by 120% compared to untreated seed. Based on this experiment, rhizobacteria P.fluorescens PG01 or Bacillus CKD061 integrated with   rice hull charcoal or ‘serbuk bata merah’ seed matriconditioning can be used as biological agents in improving seed quality of sorghum. Key words:   bio-matriconditioning, rhizobacteria, seed quality, sorghum

Abstract

ABSTRACT The research was aimed to analysis the integration of seed invigoration with rhizobacteria to improve the growth of Numbu and Kawali varieties of sorghum (Sorghumbicolor L.). The experiment design was a randomized block design arranged in Split-plot pattern. The main factor was sorghum varieties which consisted of 2 varieties ie. Numbu and Kawali. The sub plot was seed biomatriconditioning integrated with rhizobacteria treatments which consisted of 5 treatments, namely: control, seed biomatriconditioning with Bacillus sp. CKD061 + rice hulls charcoal, seed biomatriconditioning with Bacillus sp. CKD061 + red brick powder, seed biomatriconditioning with Pseudomonas fluorescens PG01 + rice hulls charcoal, and seed biomatriconditioning with P. fluorescens PG01 + red brick powder. Every treatment was replicated 3 times, therefore, overall there were 30 experimental units. Data obtained were analized using analysis of variance and followed by Duncan’s Multiple Range Test. The result of growth analysis showed that the interaction of both factors significantly influenced the leaf area index, plant growth rate, and net assimilation rate. In both varieties, seed biomatriconditioning with Bacillus sp. CKD061 or P. fluorescens PG01 using red brick powder or rice hulls charcoal were effective in increasing leaf area index, plant growth rate, and net assimilation rate of sorghum compared with untreated treatment. Keywords: biomatriconditioning, rhizobacteria, sorghum

Abstract

Viability and Inhibition of Endophytic Bacteria Pseudomonas fluorescens 4RS1 Against Phytophthora palmivora in Flour Formula Pseudomonas fluorescens 4RS1 is an endophytic bacterium that has good inhibition against Phytophthora palmivora, and it has the potential to be developed as a biological agent for black pod rot disease in cocoa. It is necessary to have a formulation that can maintain viability and inhibition and facilitate its use and application, to apply this bacterial agent at the farm level. This study aimed to determine the ability of talc and tapioca flour as the ingredients of formulations that have strong solubility and maintain the viability and inhibition of P. fluorescens 4RS1. The results showed that the use of tapioca flour as the basis for the formulation of P. fluorescens 4RS1 was better than talc because it had high solubility and low precipitation. The addition of nutrients in the form of brown sugar, peptone, and CMC in the tapioca flour formulation was able to maintain the viability and inhibition of P. fluorescens 4RS1 for 12 weeks of storage.

Abstract

Microalgae Chlorella sp. grows in the seas of South-East Sulawesi, Indonesia. However, information on the chemical and pharmaceutical aspects of this species from this region is still limited. Therefore, this research aims to explore the chemical contents and biological activities of Chlorella sp. The sample was collected from the Kali Biru and Nambo waters, Kendari. The microalgae were isolated by the agar plating technique and extracted by ethanol. Chemical content was analyzed by phytochemical screening, Liquid Chromatography-tandem Mass Spectroscopy (LC-MS/MS), Total Phenolics Content (TPC) and Total Flavonoids Contents (TFC). Biological activities evaluation includes antioxidant, toxicity, cytotoxicity and antimicrobial. Antioxidant potency was evaluated by DPPH (2,2-diphenyl-1-picrylhydrazyl) radicals and ABTS (2,2’-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid). Toxicity and cytotoxicity properties were analyzed by BSLT (Brine Shrimp Lethality Test) and MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assays against Breast cancer (MCF-7) cell lines. Antimicrobial potency was tested towards E. coli, S. aureus and C. albicans. The result showed that ethanol extract of Chlorella sp. (EEC) contained phenolics, flavonoids, alkaloids and terpenoids/steroids which were supported by LC-MS/MS data, TPC and TFC with value 150.33 and 33.18 mgGAE.g-1 extract. Seven of ten identified major compounds caused this extract to have antioxidant and anticancer including xanthin, 1,1-Diethyl-3-oxo-2- triazanolate, Azelaic acid, ruspolinone, 6-keto prostaglandin F1a and glycol stearate. In general, the biological activity of EEC is less active than the positive controls for each assay. However, the EEC is more potential to be developed as antioxidant such as sunscreen and anticancer especially breast cancer with IC50 value 151-200 ppm and 100-500 ppm, respectively.

Abstract

Penelitian bertujuan mengetahui pengaruh berbagai kerapatan deleterious rizobakter Pseudomonas aeruginosa A08) sebagai bioherbisida dan pemacu pertumbuhan tanaman kedelai (Glycine max L. Merril). Penelitian dilaksanakan di Desa Jati Bali, Kecamatan Ranomeeto Barat, Kabupaten Konawe Selatan, Provinsi Sulawesi Tenggara. Penelitian secara eksperimen dengan rancangan acak kelompok (RAK), terdiri dari empat taraf yaitu tanpa bakteri (B0), OD 1 (B1), 1,5 (B2) dan 2 (B3), masing-masing dengan empat ulangan. Data dianalisis dengan Anova dan perbedaan antar perlakuan diuji dengan BNJ dengan taraf kepercayaan 95%. Deleterious rizobakteri P. aeruginosa A08 dalam berbagai kepadatan memberi pengaruh yang nyata dalam menekan pertumbuhan gulma dan memacu pertumbuhan tanaman kedelai. Perlakuan terbaik dalam menekan pertumbuhan gulma dipertanaman kedelai diperoleh pada perlakuan B3 (P. aeruginosa A08 OD 2). Sementara Perlakuan terbaik dalam memacu pertumbuhan kedelai diperoleh pada perlakuan B2 (P. aeruginosa A08 OD 1,5) dan B3 (P. aeruginosa A08 OD 2) dibandingkan dengan perlakuan lain dan kontrol.

Abstract

The use of beneficial microbes in increasing plant growth and yield is a very appropriate choice to reduce synthetic chemicals that can cause negative impacts on the environment. The purpose of this study was to evaluate the effectiveness of post-save formulations of Wakatobi indigenous rhizobacteria in increasing the growth and yield of shallots. The study was conducted in Jati Bali Village, Konawe Selatan District, using a Randomized Block Design, consisting of 10 rhizobacterial formulation treatments, a combination of three types of biological agents, namely Pseudomonas sp. LP03, Pseudomonas sp. TWB02 and Pseudomonas sp. TWB11, and three types of formulation materials: ground brick powder, ground burned rice husk powder, and bentonite; one control using NaCl 0.85%. The experiment was repeated three times so that there were 360 treatment units. The results showed that of the three types of rhizobacteria tested, only Pseudomonas sp. TWB02 and Pseudomonas sp. LP03 can display better performance in increasing the growth and yield of shallots. These rhizobacteria are more compatible using the ground burned rice-husk powder formulation. Increased shallot yield (tuber fresh weight) in seed treatment using Pseudomonas sp. TWB02 and Pseudomonas sp. LP03 in the formulation of ground burned rice-husk powder reached 121% and 117% compared to controls. Further research needs to be done on a broader scale and different environmental conditions to see the stability of these biological agents' effects on the growth and yield of shallots.
