Abstract

Objectives: In spontaneous abortion cases without previously known predisposing factors, an increase proinflammatory cytokines, especially interleukin 6 (IL-6), found in local and systemic specimens compared to normal 1st -trimester pregnancies. Therefore, IL-6 is thought to have a role in implantation failure in early pregnancy resulting in spontaneous abortion. However, research on serum IL-6 levels in spontaneous abortion patients is still limited in Indonesia. This study aims to determine the difference in serum IL-6 levels in normal pregnancy 1 st - trimester and spontaneous abortion in hospitals in Padang City; Method: This study is an analytical study with a cross-sectional design using secondary data of 21 blood serum samples from each research group at the Biomedical Laboratory of FK Unand. Samples were collected using a consecutive sampling technique. A serum examination was performed with the ELISA technique. Bivariate data analysis using Mann-Whitney statistical test based on data type and distribution; Result: The results showed that the mean value of serum IL-6 levels was 280,56 ng/L in the normal pregnancy group in the 1st -trimester and the median value of the spontaneous abortion group was 279,78 ng/L. There was no significant difference in serum IL-6 levels in normal pregnancy in the 1st -trimester and spontaneous abortion (p = 0,980); Conclusion: This study concluded that there was no difference in serum IL-6 levels between patients with normal pregnancy in the 1st -trimester and spontaneous abortion in hospitals in Padang City. Abstract

Mesenchymal Stem Cells Wharton's Jelly (MSC-WJ) has become a new hope for Alzheimer's disease due to its capability to suppress Amyloid-beta (Aβ) plaques formation. It is found that people with Alzheimer's disease have a lower expression of Sirtuin-1 because of the oxidative stress that was produced by Aβ plaques. Objective: To observe the Sirtuin-1 gene in Alzheimer's disease rat models that AlCl3 was inducing after being given MSC-WJ. Methods: This was an experimental study with a post-test-only control group design, including three groups with a total sample of 18 samples of Alzheimer RNA rat models. Group-1without AlCl3 and MSC-WJ; Group-2 with AlCl3; Group-3 AlCl3 and MSC-WJ. Mean value of the Sirtuin-1 gene was obtained by the Image-J application using a semiquantitative method which was then tested using the One Way ANOVA test. The study was significant if the p-value < 0,05. Results: This study found that the mean ± standard error value of the Sirtuin-1 gene in three groups were 0.18±0.04; 0.16±0.03; and 0.36±0.06 respectively. The statistical test showed a significant difference between the three groups (p = 0.014). Conclusion: There was an increased Sirtuin 1 gene expression in Alzheimer’s rat Model after being given Mesenchymal Stem Cells Wharton’s Jelly.Keywords:  Alzheimer, MSC-WJ, Sirtuin-1

Abstract

Cervix cancer, also known as cervical cancer, is a type of cancer that is caused mainly by oncogenic Human Papillomavirus (HPV), which attacks the cervix. One of the plants that can be used in medicine is clove. Objective: To analyzed the effect of a fraction of ethyl acetate eugenol of clove flower for the proliferation and migration of HeLa cells. Methods: This was an in vitro experiment. Ethyl acetate fraction was tested in vitro with the MTT Assay method. A scratch Assay is done to determine the migratory power of cancer cells to Trypan Blue to see (dead cells/living cells). Data collection techniques were carried out using experimental observation techniques. The data obtained in the MTT test is the percentage of inhibited HeLa cells. Scratch Assay results were analyzed with ImageJ software and the Trypan Blue method using the TC10 tool. Results: The MTT Assay test obtained an IC50 value of 24.51 µg/ml, the average migration of treatment cells 303 and the average control group 715 cells. The number of proliferating treatments 6.14 x 105 and control 12.1 x 105. Conclusion: Based on its potential, clove flower can be a candidate for cervical cancer cell therapy (HeLa Cell Line) in the future.Keywords:  anti-cervical cancer, cervical cancer, cell proliferation, MTT assay, scratch assay

Abstract

Diabetes mellitus (DM) is a metabolic disease characterized by blood glucose levels above normal or hyperglycemia caused by defects in insulin secretion, insulin action, or both. This study aims to determine the effect of wharton’s jelly mesenchymal stem cells on blood glucose levels in hyperglycemic Wistar rats (Rattus norvegicus). This research is a laboratory experimental study using a post-test control group design. This research was conducted on 18 male white rats (Rattus norvegicus) which were divided into 3 groups, namely the normal/negative group (K-), the positive control group (K+) which was induced by alloxan, and the three treatment groups (P) which were induced by alloxan, alloxan and given a single dose of 3x106 cells of Wharton’s jelly mesenchymal stem cells for one month. Blood glucose levels were measured using a glucometer every week, from the first week to the 5th week. Data analysis used the Kruskal-Wallis and Mann-Whitney tests. Mean of blood glucose level in the last week with a glucometer in the (P) group was 85.33 mg/dL, lower than that in the (K+) group, namely 551.5 mg/dL. However, the (K-) group was the lowest compared to the (P) and (K+) groups, namely 72.83 mg/dL The data showed that there was a significant difference in blood glucose levels between groups (p<0.05). The conclusion of the study was that the administration of Wharton’s jelly mesenchymal stem cells had an effect on reducing blood glucose levels in hyperglycemic rats

Abstract

Dalam skala global, kanker menempati peringkat kedua sebagai pembunuh. Salah satu bentuk pengobatan kanker adalah radioterapi, yang menggunakan radiasi pengion dosis tinggi untuk menghancurkan sel kanker dan mengurangi ukuran tumor. Peneliti berharap dapat mempelajari lebih lanjut tentang demografi pasien radioterapi yang dirawat di RSUP Dr. M. Djamil Padang pada tahun 2019 dan 2020 melalui penelitian ini. Pendekatan sampel lengkap digunakan dalam penelitian ini, yaitu deskriptif retrospektif. Pasien yang menerima terapi radiasi untuk kanker memiliki berbagai diagnosis, termasuk kanker serviks (19%), payudara (18%), nasofaring (15%), rektum (8%), dan paru-paru (8%). Penelitian ini menemukan bahwa mayoritas pasien adalah ibu rumah tangga (36,71%), berpendidikan SMA (58,86%), dan berusia antara 45 dan 49 tahun (18,57%). Kanker stadium IV termasuk karsinoma sel skuamosa serviks, karsinoma payudara invasif, karsinoma sel skuamosa nasal, adenokarsinoma rektum, dan adenokarsinoma paru merupakan yang paling umum dan memiliki temuan patologi anatomi yang tinggi (46,20%). Kesimpulannya mayoritas distribusi frekuensi pasien radioterapi pada usia 45-49 tahun, perempuan, pasien berasal dari Kota Padang, lulusan SMA, pekerjaan ibu rumah tangga, diagnosis kanker pada kanker serviks, dan stadium IV yang tertinggi dalam penggunaan radioterapi serta squamous cell carcinoma.

Abstract

Latar Belakang: Tuberkulosis merupakan penyakit menular yang disebabkan oleh Mycobacterium tuberculosis yang menduduki peringkat kedua penyebab kematian terbanyak. Faktor yang menyebabkan tingginya angka kejadian adalah keterlambatan dalam penegakan diagnosis. Pemeriksaan baku emas untuk tuberkulosis adalah kultur Lowenstein-Jensen yang membutuhkan waktu cukup lama untuk pertumbuhan koloni bakteri. Objektif: Penelitian ini bertujuan untuk mengetahui sensitivitas dan spesifisitas dari RT PCR menggunakan kit RT PCR yang bermerk Kit Crown Lab TB Dx dibandingkan dengan Kultur Lowenstein-Jensen. Metode: Penelitian ini menggunakan metode pendekatan cross-sectional dengan teknik purposive sampling sebanyak 87 sampel dari data sekunder yang diolah dan dianalisis dengan rumus uji diagnostik. Hasil: Hasil RT PCR didapatkan nilai positif 44 orang (50,6%) dan nilai negatif 43 orang (49,4%). Kultur Lowenstein- Jensen didapatkan nilai positif 43 orang (49,4%) dan nilai negatif 44 orang (50,6%). Hasil perbandingan kedua metode didapatkan nilai sensitivitas sebesar 97%, spesifisitas 95%, nilai duga positif 95%, dan nilai duga negatif 97% Kesimpulan: Kesimpulan dari penelitian ini Real Time PCR memiliki sensitivitas, spesifisitas, dan nilai duga positif dan negatif yang tinggi dan memiliki kelebihan waktu pemeriksaan yang lebih cepat dibandingkan pemeriksaan dengan Kultur Lowenstein-Jensen.

Abstract

Atherosclerosis is a leading cardiovascular disease characterized by the buildup of plaques within arterial walls. The aim of this study was to investigate the potential of peptides derived from Lytechinus variegatus spines as novel therapeutic agents for atherosclerosis using an in silico approach. Key proteins involved in atherosclerosis were selected as target proteins: vascular endothelial growth factor receptor (VEGFR), protein kinase B (AKT1), epidermal growth factor receptor (EGFR), mitogen-activated protein kinase 8 (MAPK8), and endothelin-1 (ET-1). Comprehensive analysis involving ligand and protein preparation, toxicity, and allergenicity assessments, absorption, distribution, metabolism and excretion (ADME) predictions, and molecular docking were conducted to evaluate the safety, pharmacokinetic properties, binding affinity (kcal/mol), root mean square deviation (RMSD) (Å), as well as a 2D and 3D visualization. Toxicity predictions revealed that peptide 9 was non-toxic and non-allergenic, with a lethal dose 50 (LD50) of 3,000 mg/kg, indicating its safety. Peptide 9 demonstrated the most promising results, effectively inhibiting VEGFR2 (-10,90 kcal/mol), AKT1 (-10,56 kcal/mol), EGFR (-9,82 kcal/mol), MAPK8 (-9,64 kcal/mol), and ET-1 (-11,41 kcal/mol) with strong binding affinities and specificity. These interactions suggested that peptide 9 from Lytechinus variegatus spines may serve as a competitive multitarget inhibitor, offering potential multitarget therapeutic activity against atherosclerosis. Peptide 9 also had high water solubility and did not affect the concentration or excretion of other drugs or compounds, minimizing the risk of drug-drug interactions.

Abstract

Tujuan: mengetahui pengaruh pemberian ekstrak daun sungkai dalam menghambat proliferasi dan migrasi sel HeLa. Metode: Penelitian ini merupakan penelitian eksperimental yang menggunakan 24 disk kultur sel HeLa yang konfluens. Sampel dibagi menjadi 4 kelompok (K,P1,P2 dan P3). Kelompok perlakuan diberikan ekstrak daun sungkai dengan konsentrasi IC25, IC50, dan IC75 yang diinkubasi selama 72 jam. Analisis data menggunakan uji One Way ANOVA dan Post Hoc Bonferroni. Hasil: Rerata jumlah sel proliferatif yang didapatkan pada kelompok K, P1, P2, dan P3 berturut-turut adalah 12,1 x 105, 8,94 x 105, 6,14 x 105 dan 5,04 x 105. Rerata migrasi jumlah sel yang berkumpul pada zona scratch pada kelompok K, P1, P2, dan P3 berturut-turut adalah 49,.0, 237,8, 210,5, dan 100,7. Didapatkan perbedaan yang bermakna tiap kelompok pada uji proliferasi dan migrasi dengan nilai p = 0,000 (p < 0,05). Kesimpulan: Terdapat pengaruh pemberian ekstrak daun sungkai terhadap proliferasi dan migrasi sel HeLa. Perbedaan bermakna ditemukan pada kelompok kontrol dan kelompok perlakuan dengan konsentrasi ekstrak IC25, IC50, dan IC75.

Abstract

Background: Childhood malnutrition stems from inadequate intake and disease; synbiotics (probiotics+prebiotics) can modulate gut microbiota and immune function, aligning with SDG-3 targets through cost-effective prophylaxis and immunomodulation. Objective: To evaluate onion (Allium cepa Linn.)-derived inulin combined with probiotic-rich Sumatran dadih as supportive therapy for pediatric malnutrition. Methods: Narrative literature review of human and relevant preclinical studies on synbiotics, inulin, dadih, gut microbiota, and immune modulation, with selection based on relevance and methodological quality. Results: Evidence suggests synbiotics improve microbiota diversity, short-chain fatty acid production, nutrient absorption, and reduce inflammation and infection rates. Onion-inulin functions as a fermentable prebiotic; dadih provides robust local lactic-acid bacteria strains. Yet data are heterogeneous, sample sizes small, and dosing/formulation poorly standardized; safety reporting is variable. Conclusion: An onion-inulin + dadih synbiotic is a plausible, context-appropriate adjuvant for managing childhood malnutrition. Rigorous, controlled trials should optimize dose, strain selection, and formulation, and confirm efficacy and safety for scalable implementation.

Abstract

Tujuan : mengetahui hubungan indeks massa tubuh dengan kadar glukosa darah puasa dan tekanan darah pada tenaga kependidikan Fakultas Kedokteran Universitas Andalas. Metode : Jenis penelitian ini merupakan penelitian analitik dengan desain penelitian cross-sectional dengan subjek penelitian sebanyak 37 orang yang dilaksanakan di Fakultas Kedokteran Universitas Andalas pada tanggal 08 September 2021. Metode pengambilan sampel ini dengan total sampling, analisis data hubungan indeks massa tubuh dengan kadar glukosa darah puasa dan tekanan darah menggunakan uji korelasi Spearman. Hasil: Nilai median indeks massa tubuh sebesar 24,22 Kg/, nilai rerata 24,94 Kg/, nilai minimum sebesar 16,23 Kg/ dan nilai maksimum 40,15 Kg/. Nilai median kadar glukosa darah puasa 127 mg/dL, nilai rerata 143,65 mg/dL, nilai minimum sebesar 93 mg/dL dan nilai maksimum 399 mg/dL. Nilai median dari tekanan darah 126 mmHg,nilai rerata 128,7 mmHg,  nilai minimum 94 mmHg, dan nilai maksimum 213 mmHg. Tidak terdapat korelasi bermakna antara indeks massa tubuh dengan kadar glukosa darah puasa (p=0,203) ataupun pada indeks massa tubuh dengan tekanan darah (p= 0,097) pada subjek penelitian. Kesimpulan: tidak terdapat korelasi bermakna antara indeks massa tubuh dengan kadar glukosa darah puasa dan antara indeks massa tubuh dengan tekanan darah pada tenaga kependidikan pada Fakultas Kedokteran Universitas Andalas.
