Abstract

Objective: This study aims to map the distribution of the Aspartate 614 to Glycine (Asp614Gly) mutation in the spike protein of the Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) circulating in West Sumatra, to understand the extent of its spread within the local population. Methods: RNA samples were taken from 98 confirmed COVID-19 cases across various area in West Sumatra. Genetic amplification and next-generation sequencing (NGS) were performed to identify the presence of the Asp614Gly mutation. The frequency of mutations was analyzed using bioinformatics software. Results: Out of the 98 samples analyzed, approximately 97% exhibited the Asp614Gly mutation. The geographic distribution of this mutation showed significant variation among the districts of West Sumatra, with the highest prevalence in urban areas. Conclusion: These findings indicate that the Asp614Gly mutation is highly prevalent in West Sumatra. Associated with increased virus transmissibility, this mutation dominates the circulating virus strains in the region, highlighting the need for ongoing genetic surveillance to guide public health interventions and vaccination strategies.

Abstract

Konsumsi air yang cukup sesuai rekomendasi berperan penting bagi fungsi kognitif dan kesehatan anak. Akan tetapi, masih dijumpai anak sekolah dasar yang belum memahami manfaat mencukupi kebutuhan air tubuh. Program pengabdian ini bertujuan menilai sekaligus meningkatkan pengetahuan tentang hidrasi pada siswa kelas 4 dan 5 di sekolah dasar full day berbasis alam. Mitra program adalah SDIT Alam Ar-Royyan di Kota Padang. Metode yang digunakan berupa edukasi interaktif selama tiga sesi selama tiga minggu. Setiap sesi diawali pengarahan dan pre-test, dilanjutkan penyampaian materi melalui media audio visual, permainan, serta tanya jawab. Post-test dan penguatan materi dilaksanakan dua hari setelahnya. Instrumen penilaian menggunakan kuesioner cetak dan dianalisis secara deskriptif. Sebanyak dua puluh tiga siswa mengikuti pertemuan pertama, dengan tujuh belas siswa menyelesaikan seluruh rangkaian program. Hasil menunjukkan peningkatan pengetahuan yang bermakna yaitu sebesar 96,1% siswa mengalami kenaikan skor pengetahuan dan 76,5% berpengetahuan “baik”. Ukuran kelompok yang kecil mendukung interaksi, fokus, dan umpan balik yang lebih personal. Hambatan utama adalah penjadwalan dan ketidakhadiran sebagian siswa. Penetapan jadwal melalui nota kesepahaman dengan bagian kurikulum, pelibatan orang tua dan guru melalui diskusi kelompok terarah, serta pemantauan kebiasaan minum harian di kelas diperlukan untuk keberlanjutan program.

Abstract

Breast cancer remains one of the leading causes of death in women worldwide, with high incidence and mortality rates. The abnormal expression of the small non-coding RNA, microRNA-10b (miR-10b), has been associated with breast cancer progression. miR-10b is involved in various biological processes, including proliferation, metastasis, and drug resistance. However, the underlying mechanism by which miR-10b contributes to breast cancer progression remains unclear. This study aimed to investigate the effect of miR-10b on the expression of HOXD10 and PI3K genes, as well as its impact on cell viability and proliferation in breast cancer cells. In this study, T47D breast cancer cells were transfected with mimic miR-10b using the FANA-oligo transfection method at concentrations of 1, 2.5, and 5 µM. Cell viability was assessed using the MTT assay, while HOXD10 and PI3K gene expression levels were measured using RT-qPCR and ELISA. Data were analysed using paired t-tests with SPSS, and results were considered statistically significant at p < 0.05. The results showed that mimic miR-10b induced a dose-dependent increase in cell proliferation. Notably, only HOXD10 gene expression was significantly affected by the transfection, while PI3K gene expression remained unchanged. Cells transfected with 5 µM miR-10b mimic exhibited the highest proliferation rate compared to non-transfected controls (p < 0.05). In conclusion, miR-10b promotes breast cancer cell proliferation by downregulating HOXD10 expression, independent of PI3K, suggesting it is a potential target for further investigation.

Abstract

Our community service activity aimed to increase knowledge and awareness about the dangers of per- and polyfluoroalkyl substances (PFAS), known as “forever chemicals,” which are commonly found in household products, food packaging, and cosmetics. Long-term exposure to PFAS has been associated with numerous serious health effects, including an increased risk of several cancers, disruption of hormone homeostasis, liver toxicity, impaired immune function, and adverse reproductive and developmental outcomes.  The participants included housewives and adolescent girls (n = 20) who attended Padang Pasir Community Health Center, East Padang, Padang on 6 September 2025. An interactive educational session was conducted using a pre-test–post-test design to evaluate changes in knowledge and awareness. Analysis of participants who completed both assessments showed an improvement in knowledge following the intervention, with the greatest increases observed in understanding of healthy cooking utensils (50%) and the presence of persistent chemicals in cosmetics (50%). However, evaluation of awareness levels revealed only a minimal increase in median scores (from 57 to 60), which was not statistically significant (p = 0.221). We conclude that a focused educational session is effective in improving participants’ cognitive knowledge, but a more comprehensive and sustained approach is needed to promote meaningful behavioral change and deeper awareness, particularly since practicality and convenience remain major considerations when choosing household products, food packaging, and cosmetics.
