Abstract

Penelitian ini bertujuan mengetahui perbedaan intensitas berahi sapi aceh antara yang disinkronisasi berahi dengan prostaglandin F2 alfa (PGF2α) dan berahi alami. Dalam penelitian ini digunakan 20 ekor sapi aceh betina yang dibagi atas dua kelompok. Kriteria sapi yang digunakan adalah umur 5-8 tahun, mempunyai bobot badan 150-250 kg, dan mempunyai minimal dua siklus reguler. Sapi yang digunakan mempunyai skor kondisi tubuh dengan kriteria baik, yaitu 3 atau 4 pada skala skor 5. Pada Kelompok I (KI) sapi disinkronisasi berahi mengunakan PGF2α sebanyak 5 mg/ml secara intramuskular. Pada kelompok II (KII) sapi dibiarkan memperlihatkan gejala berahi alami. Penilaian intensitas berahi dilakukan dengan memberi skor 1, 2, dan 3, berdasarkan kriteria yang dibuat oleh Kune dan Solihati (2007). Hasil penelitian menunjukkan tidak ada perbedaan intensitas berahi sapi aceh baik yang disinkronisasi berahi dengan PGF2α dan sapi yang mengalami berahi alami dengan skor intensitas berahi masing-masing adalah 2,40±0,84 dan 2,70±0,48.

Abstract

The objective of this research is to know Aceh cattle follicle dynamic under environmental heat stress condition. This research was conducted on December 2012 until August 2013. Animals used in this research were 20 Aceh cattle aged 5-8 years old, weighing of 150-250 kg, and had at least 2 regular cycles. All cattle used were clinically in good body condition score. The cattle were divided into two groups of 10, used in two different period of times (December-January and July-August) to examine the effect of environmental heat to their follicle dynamic. Each group is separated into two different keeping management; one group was kept in pens while the others in the pasture, 5 cattle in each management group. Research timing was based on information acquired from Indrapuri Agency for Meteorology, Climatology, and Geophysics (BMKG) which predicted that extreme weather would last from July to August while December to January would be relatively normal in Aceh region. All cattle’s estrous cycle were synchronized by 5 mg/mL of PGF2α intramuscularly (Lutalyse™, Pharmacia Upjohn Company, Pfizer Inc.). Ultrasonography examination was performed to monitor ovary follicle’s growth and dynamic during one cycle. Days during ovulation marked by standing heat was regarded as Day 0 of estrous cycle. Follicular dynamic examination during estrous cycle on December-January and also on July-August, both penned or pastured cattle showed the follicle waves that was 3 follicle growth waves. The size of follicle growth on first wave (1st DF) on cattle kept in pen on July-August and December-January were relatively similar. The size of follicle growth on first wave for cattle kept in pasture on July-August and December-January were relatively similar. To conclude, Aceh cattle’s follicle dynamic is not change during environment heat stress condition and is not affected by different cattle-keeping management.

Abstract

Penelitian ini bertujuan mengetahui pengaruh pemberian ekstrak vesikula seminalis terhadap penurunan konsentrasi progesteron serta diameter korpus luteum pada tikus putih. Dalam penelitian ini digunakan 30 ekor tikus putih (Rattus norvegicus) betina dewasa, galur Wistar, berumur 3-4 bulan dengan berat badan antara 200-250 g dan dibagi atas dua kelompok (K1 dan K2) masing-masing diberi 25 μg cloprostenol dan 0,2 cc ekstrak vesikula seminalis secara intravaginal pada hari ke-7 kebuntingan. Tiga ekor tikus masing-masing kelompok dibunuh pada jam ke-0, 3, 6, 12, dan 26. Pemeriksaan progesteron dilakukan menggunakan metode enzymelinkedimmunosorbantassay (ELISA). Konsentrasi progesteron pada kelompok perlakuan PGF2α dan ekstrak vesikula seminalis pada lima periode waktu pengukuran yakni jam ke-0, 3, 6, 12, dan 26 memperlihatkan perbedaan yang signifikan (P0,05). Ekstrak vesikula seminalis mempunyai kemampuan yang sama dengan PGF2α komersial dalam menurunkan diameter korpus luteum yang ditandai secara mikroskopis dengan berkurangnya vaskularisasi darah menuju ovarium (P0,05). Disimpulkan bahwa ekstrak vesikula seminalis mempunyai kemampuan yang lebih baik dibandingkan dengan PGF2α komersial dalam menurunkan konsentrasi progesteron tikus putih dan mempunyai kemampuan yang sama dengan PGF2α komersial dalam menurunkan diameter CL.

Abstract

The aim of this research was to find out in vitro antiplasmodium activity of Malacca leaves (Phyllantus emblica) ethanolic extract against Plasmodium falciparum growth. In this study, Plasmodium culture contained 5% parasitemia in ring stage was cultured using candle jar method and antiplasmodial activity test was carried out using microculture. The treatments were divided into 7 groups with four repetitions. K1 as negative control group was given Roswell Park Memorial Institute (RPMI), while K2 as positive control group was given artesdiaquine. Groups K3, K4, K5, K6, and K7 group was added with 100 µg/mL, 75 µg/mL, 50 µg/mL, 25 µg/mL, and 5 µg/mL of Malacca leaves ethanolic extract, respectively. Antiplasmodial activity was determined by inhibition concentration of 50% parasite growth (IC50). The data were analyzed using ANOVA and followed by Duncan test. The average of parasitemia level in group K1, K2, K3, K4, K5, K6, and K7 were 55.25±15.62, 8.50±2.52, 8.50±3.00, 9.25±0.95, 9.00±2.70, 9.79±2.06, and 10.75±2.22, respectively. The average of inhibition percentage in group K1, K2; K3; K4; K5; K6; and K7 were 0.00±0.00%, 84.62±4.55%; 84.62±5.43%; 83.26±1.73%; 83.71±4,90%; 82.35±3,73%; and 80.54±6.83%, respectively (P0.01). The results showed that the administration of malacca leaves ethanolic extract significantly affect (P0.01) the inhibition of Plasmodium growth as compared to group K1 (negative control). Probit analysis reveals the IC50 value was 3.889 µg/mL. In conclusion, all doses of malacca leaves ethanolic extract used in this study was able to inhibit Plasmodium falciparum growth with IC50 value was 3.889 µg/mL.

Abstract

ABSTRACT The aim of this research was to asses the acute toxicity of ethanolic extract of malaka leaves using lethal dose 50 (LD50) on mice (Mus musculus). Twenty male mice weighing between 20-30 g were randomly divided into 4 groups (group K1-K4) of 5 mice each. All mice in group K1, K2, K3, and K4 were administered ethanolic extract of malaka leaves with the dose of 2, 4, 8, and 16 g/kg bw, respectively. Single dose of ethanolic extract of malaka leaves were given by oral gavage prior to clinical observation . The observation period was 14 days post administration, for sign of toxicity symptom, weight loss, and mortality. The result showed that no mortality was observed in the experimental animals during this study. Slight reduction of body weight was observed in group K2, K3, and K4, and no toxicity sign was found during fourteen days of observation. The LD50 of ethanolic extract of malaka leaves was higher than 16 g/kg body weight, thus, the substance was practically non toxic substance.

Abstract

This study’s purpose is to determine the effect of supplementation of willow bark extract (1000 mg/L) in drinking water and 15% cassava leaf powder in feed on broiler weight gain. This study was designed using completely randomized design (CRD) in which 20 broiler chickens were divided into four treatment groups with five broiler per each treatment. Control group (P0) was only given commercial feed, P1 (commercial feed + cassava leaf powder), P2 (commercial feed + willow bark extract), and P3 (commercial feed + cassava leaf powder and willow bark extract). Data were analyzed using analysis of variance and followed by Duncan test. The results showed that P2 significantly different (P0.01) compare to P0 and P3, and different (P0.05) from P1. Group P1 was different (P0.05) from group P3, but P3 showed no significant different (P0.05) to P0 on weight gain. whereas feed intake was similar among treatments, however food conversion was differ among treatments. In conclusion, combination of willow bark extract and cassava leaf powder could not increase broiler chickens weight gain.

Abstract

This study aims to determine the efficacy of jatropha curcas L. cream in the maturation phase of healing of mice leather injuries. This study used nine rats with 3 treatments, ie treatment group I smeared cream base (P1), treatment group II smeared 0.1% sulfadiazine cream (P2) and treatment group III smeared jatropha curcas 10% (P3). The result of ANAVA test of treatment I, treatment II and treatment III had significant effect (P 0,05) on fibroblast cells but each treatment group on collagen density level showed significant effect (P 0.01). Duncan test results on fibroblasts, P3 was significantly different (P 0.05) with P2 and P3, whereas the results on P3 collagen density were significantly different (P 0.01) with P2 and P1. The conclusion that 10% jatropha gum cream for 10 days can decrease the number of fibroblast cells and increase the amount of collagen thus accelerate the process of maturation phase of healing of mice leather injuries.

Abstract

The experiment was conducted to compare estrous performance between kacang and PE goats after estrous inducted with PGF2 alpha (PGF2α). Fifteen female goats were used in this study and divided into two groups. Characteristics of goat used are clinically healthy, never giving birth, 1,5-3,0 years of age, and have undergone at least two times the regular cycle. Group I (KI) consisted of 10 kacang goats and group II (KII) consisted of 5 PE goats. All goats in both groups were estrous synchronized with 1 ml PGF2a intramuscularly. Estrous observation was done visually three times a day at 8 a.m., 12 p.m., and 4 p.m. Assessment of estrous intensity based on scoring method. The data of intensity and onset were analyzed by Mann Whitney test and estrous duration analyzed by t test. The result showed that intensity, onset, and duration of estrous in KI vs KII were 2.47±0.21 vs 2.25±0.00, 38.20±2.39 vs 84.40±8.53 hours, and 40.40±2.27 vs 46.40±4.56 hours respectively. It concluded that there was different estrous performance between kacang and PE goats following estrous synchronized with PGF2α.

Abstract

This study was aimed to determine effect of bandotan (Ageratum conyzoides) leaf extract on the carcass weight and abdominal fat of broiler. A total of 15 chicken with the age of twenty five days old were used in this study and allotted into 3 groups with 5 chickens per groups. Broiler in group P0 (as a control) were not given bandotan leaf extract,whereas broiler in group 1 (P1) and group 2 (P2) was given 3 ml and 5 ml of bandotan leaf extract, respectively. The extract was orally given once daily for 5 consecutive days. Results of analysis of variance (Anova) showed that bandotan leaf extract did not significantly (P0.05) affect carcass weight and abdominal fat of broiler. The average percentage of carcass weight in groups P0, P1, dan P2 were 73.30±3.54, 71.80±3.06, and 70.33±9.18%, respectively and the average percentage of abdominal fat in group P0, P1, dan P2 were 1.99±0.68, 2.30±0.46, amd 1.96±0.18%, respectively. Based on these results it can be concluded that bandotan leaf extract could not increase carcass weight and reduce abdominal fat of broilers.

Abstract

 This research aims to know the testicular histopathology of mice that have been exposed to cigarette smoke and given red watermelon extract dosage 22 mg and 44 mg. Experimental research using completely random design (RAL) with 4 group treatments. The negative control group (KN). The positive control group (KP) which have been exposed to cigarette smoke. Treatment group 1 (K1) which have been exposed to cigarette smoke and given the red watermelon extract 22 mg. Treatment group 2 (K2) which have been exposed to cigarette smoke and given the red watermelon extract 44 mg. Exposed to cigarette smoke conducted in the morning for an hour to burn one cigarette and red watermelon extract given in the afternoon. This was done for 30 days. On day 31, mice were sacrificed and their testis were taken to weighed and made histology slide. The data were analyzed using ANOVA in one direction and continued with Duncan. The results of this research showed that the extract of red watermelon for 30 days looks a highly significant effect (P 0.05) to the diameter of the seminiferous tubules, the number of spermatogonia cells, primary spermatocytes cells, spermatid cells. While on the weight of the testes showed no real effect (P 0.05). Red watermelon extract dosage 22 mg dan 44 mg of the mice exposed cigarette smoke can prevent cell damage in mice testis.
