Abstract

ABSTRACT Introduction: Diarrhea is one health problem in the world with mortality and morbidity rates are high, both in developed and developing countries. This research aims to detect Shigella sp in the stool of children with diarrhea using culture method and PCR. Methods: This research is a descriptive with cross-sectional design. A total of 50 samples were obtained from four health centers in Makassar city, i.e Barabaraya, Pampang, Antang Perumnas and Tamangapa from May - July 2016 with rectal swab method. For culture test, the samples were inoculated on medium SSA. While with PCR test, primer icsA (526 bp) was used. Results: The results indicate that, from 50 samples tested by using culture method, there is no sample (0%) posi- tive detected Shigella sp. While the PCR method there 6 samples (24%) positive detected Shigella sp. Conclusion: The sensitivity of PCR in the detection of Shigella sp from the level of dilution 100 - 10-3. Al together, it can be inferred that PCR method is more accurate method detect Shigellasp than other culture methode. Keywords: Diarrhea, Shigellasp, Culture, PCR

Abstract

ABSTRACT Introduction : Diarrheal diseases are  a serious issue on health in developing countries and the liquid cause’s morbidity and mortality in children that cause the diarrhea pathogenic bacte- ria including Escherichia coli.  This research aims to detect E. coli in stool diarrhea patient of the  child with culture method and  Polymerase Chain Reaction. Methods : This research is descriptive research design with cross-sectional approach. The sampling was collected from some public health centers namely Pampang, Barabarayya, Antang  Perumnas, Tamangapa public health centers. Specimen testing process was conducted at Microbiology laboratory of the Teaching Hospital University of Hasanuddin to identify E. coli bacterium with the culture and  to detect eae gene and bfp with PCR technique. Results : The results indicate that from50 samples, there  are  15 samples (30%) of samples positively detection E. coli using culture method and  PCR  test using eae and  bfp primer found 20 (40%) and  1 sample (2%) positive enteropathogen E. coli. Conclusion : PCR  methods indicated the  result of E. coli bacteria results faster and more accurate that other  culture methods. Keywords : Diarrhea, Escherichia coli, Polymerase Chain Reaction

Abstract

Avian Influenza (AI) is an infectious disease caused by the influenza type A virus. The highly pathogenic AI (HPAI) H5N1 outbreak in Indonesia has occurred since 2003 until now. Education, biosecurity, vaccination, elimination, diagnostic, and surveillance are strategy to prevent and control AI virus (AIV) infection. Providing antiviral drug can be used as an alternative to control AIV in poultry, but it will be limited if resistance occurs. This study aims to determine the resistance to neuraminidase inhibitors (NAIs) (oseltamivir) and M2 ion channel inhibitors (amantadine) of HPAI H5N1 virus isolated from poultry in Sulawesi during 2017-2018. This research was conducted by whole-genome sequencing (WGS) with the next generation sequencing (NGS) (Illumina) technique on 5 poultry virus isolates. Molecular analysis was performed by multiple alignments and amino acid prediction using the MEGA X program. Antiviral resistance of oseltamivir and amantadine was assessed based on analysis of NA and M2 proteins compared to reference isolates from Sulawesi in NCBI. Based on the NA protein analysis, no mutations were found at positions 119, 275, 293, and 295, indicating that all the samples and reference isolates from Sulawesi are still sensitive to oseltamivir. Whereas at positions 26, 27, 30, 31, and 34 of M2 protein, there was a V27I mutation in Sulawesi reference isolate in 2016 and the combination of V27A and S31N mutations in 2 research isolates in 2018, which indicate possible resistance to amantadine. In conclusion, there is amantadine resistance of HPAI-H5N1 virus isolated from poultry in Sulawesi, 2018.

Abstract

Objective: The aims of this research is to notice maternal and perinatal outcomeon pregnant women withcovid-19 infection at Dr. Wahidin Sudirohusodo Hospital During the Period of April-July 2020.Method: This research is using descriptive method which is by taking data from the pregnant women withcovid-19 infection at Dr. Wahidin Sudirohusodo Hospital during the period of April 1st – July 12th 2020.The data covers the age, gestational age, amount of parity, symptoms, pregnancy complication, supportiveexamination results such asneutrofil score, limfosit, CT-scan Thorax, method of delivery, baby’s APGARscore, covid baby status, maternal death, perinatal death.Results: There are 26 pregnant patients characteristic subjects with covid-19 infection, mostly 20-35 yearsold, aterm gestational (>37 weeks), multiparity, low educational background, unemployed, mostly with nosymptoms, increasing limfositneutrofil ratio rate >3,13. Good maternal outcome, there are no maternal deathin this research. Perinatal outcome from 24 babies, 5 babies are confirmed covid-19 positive and all are ingood condition.Conclusion: This research concludes good maternal and perinatal outcome

Abstract

Infectious disease still remains a serious problem in Indonesia and worldwide, it leads to use of uncontrollable anti-microbial treatment with cause to drug resistance. With unsupported condition for using a sensitivity test prior to antimicrobial treatment, a common guide for choosing an antimicrobial agent for infection of specific specimen is needed. The aims of this research is to determine the pattern of bacteria, more specific specimen is urine. This research was an observational Experimental consecutive sampling study, sample for this research were taken from inpatient room of Wahidin Sudirohusodo Hospital of Makassar during 2019-2020. The largest number of pathogenic bacteria that potentially becoming infection agent in inpatient room of Wahidin Sudirohusodo Makassar was E.coli, P.Aureginosa and Acinetobacter. Abstract

Candida spp merupakan infeksi jamur tersering pada manusia yang dapat menyebabkan infeksi primer dan sekunder pada penderita tuberkolosis paru. Penelitian ini bertujuan untuk mengukur keakuratan (sensitifitas dan spesifitas ) Multiplex PCR untuk mendeteksi Candida spp. Desain penelitian adalah observational studi dengan jumlah sampel 55 sputum BTA positip yang dipilih berdasarkan propotional ramdoms sampling. Pengambilan sputum, data faktor biomedis meliputi: umur, jenis kelamin, riwayat tentang penyakit tuberkulosis, riwayat penyakit lainnya, riwayat pengobatan dikumpulkan oleh petugas laboratorium yang terlatih. Analisis data digunakan untuk mengukur sensitifitas dan spesifitas Multiplex PCR untuk mendeteksi Candida spp. Hasil penelitian menunjukkan bahwa kelompok umur 29-33 tahun dan 40-45 tahun memiliki resiko terinfeksi oleh jamur Candida spp, sebanyak 7 orang (20%). 6 orang (17.1 %) laki-laki dari kelompok umur 29-33 tahun dan 1 orang (2.8 %) dari kelompok umur 40-45 tahun sedangkan untuk perempuan terdapat 3 orang (8.5 %) dari kelompok umur 46-51 tahun memilki resiko untuk terinfeksi jamur Candida spp.Untuk Multiplex PCR memiliki sensitivitas dan spesifitas yaitu 100%. Multiplex PCR mendeteksi Candida albicans, Candida krusei, Candida parapsilosis, Candida tropicalis, dan Candida dubliniensis. Dari penelitian ini dapat disimpulkan bahwa untuk mendeteksi Candida spp dari sputum pasien tuberkulosis dapat menggunakan Multiplex PCR yang merupakan metode identifikasi yang sensitif, akurat, dan cepat. Oleh karena itu kami menyarankan untuk dilakukan penelitian tentang identifikasi Candida spp dari berbagi sumber specimen.

Abstract

Background: The selection process in medical schools should be able to identify individuals that mostprobably will successfully complete their education. In Faculty of Medicine Hasanuddin University (FMHU)the selection processes are Seleksi Nasional Masuk Perguruan Tinggi Negeri), SBMPTN (Seleksi BersamaMasuk Perguruan Tinggi Negeri), JNS (Jalur Non Subsidi) and International students. Looking at the variousselection methods in FMHU, this study aimed to see how each selection method related with students’academic achievement.Methods: This study compared academic achievement in two semesters at three academic admission students(2014, 2015 and 2016), for four admission processes. The data consist of 308 students from batch of 2014, 308students from batch of 2015, and 320 students from batch of 2016. The analysis was conducted in descriptiveanalysis, One Way ANOVA, and followed by Post-hoc Bonferroni Test.Results: There was significant difference of students GPA for each type of admission. The highest academicachievement was obtained by students from International and SNMPTN admission while the lowest academicachievement was obtained by students from SBMPTN and JNS.Conclusion: Students from International and SNMPTN admission had the highest academic achievementcompared to SBMPTN and JNS. This study showed that there should be an evaluation of the assessment usedin SBMPTN and to continue the use of SNMPTN as one of the main admission route at FMHU.

Abstract

Background: According to 2018 World Health Organization (WHO) data globally, an estimated 17.3% of the population has inadequate zinc intake, with estimates ranging from 5.7% in Oceania to 7.6% in Europe, 9.6% in America and the Caribbean, highest in Africa (23.9%) and Asia (19.6%). Zinc is important for the function of a number of enzymes and growth hormones during pregnancy. In pregnant women, the relative zinc concentration decreases up to 35% due to the influence of hormonal changes and the transport of nutrients from mother to baby. Objectives: The purpose of this study was to identify the effect of giving zinc tablets to pregnant women with zinc deficiency in the third trimester on body weight and length of babies born at the Makassar City Health Center. Methods: This type of research is True Experimental with a pretest-posttest design with a control group. The sample in this study was 62 samples of third trimester pregnant women, and the sampling technique used was purposive sampling. Measurement of zinc levels in third trimester pregnant women using the Elisa reader kit at the Research Laboratory of the Hasanuddin University Teaching Hospital. The research instruments were in the form of a research explanation sheet, respondent's consent sheet, respondent's checklist sheet, and the mother's zinc tablet consumption control sheet for 14 days. Results: Judging from the average value of newborns in pregnant women who did not have zinc deficiency, the average value of birth weight in pregnant women with zinc deficiency was 15.70 g/dL and 18.95 g/dL. zinc deficiency with a value (p < 0.05), while pregnant women with zinc deficiency have an average birth length of 10.00 g/dL and mothers who do not have a deficiency of 19.87 g/dL with a value (p < 0.05). So, it can be concluded that giving zinc tablets to pregnant women in the third trimester has an effect on Birth Weight (BBL) and Birth Length (PBL). Conclusion: Giving zinc tablets has an effect on increasing zinc levels in third trimester zinc deficiency pregnant women and increasing birth weight and length of the baby.

Abstract

ABSTRACT Streptococcus pneumoniae, a gram-positive bacterium of normal flora but can be a pathogenic bacterium that causes pneumonia if the immune system declines. Community pneumonia ranks fourth and top ten diseases treated per year. Populations that are susceptible to pneumonia are children aged less than 2 years, old age. The Purpose aim to detect ply genes in sputum with suspected pneumonia in elderly as a virulence factor of S. pneumoniae by culture and PCR. This research method is a descriptive study to detect the virulence factor of the S. pneumoniae ply gene in the sputum of patients with suspected pneumonia in the elderly with the culture and PCR methods. Conventionally (culture method) obtained 1 sample from 56 positive samples of S. pneumoniae gram positive bacteria on the molecular method (PCR) there were 7 positive samples of S. pneumoniae bacteria from 56 samples. In detecting ply genes in sputum with suspected pneumonia in elderly as a virulence factor from S. pneumoniae bacteria were culturally detected 1 positive sample from 56 samples and by PCR method detected 7 positive samples from 56 samples. In detecting ply genes using culture and PCR it can be said that the PCR method is more accurate, fast and precise in detecting the S. pneumonia gene in sputum samples with suspected pneumonia in elderly.

Abstract

Objective: This study aims to determine the support / involvement of the husband in the anxiety level of third trimester multigravida pregnant women with mild malaria. Method: The research design used was quasi experimental with pre-test post-test design with a study of 85 respondents with consecutive sampling techniques in three trimester multigravida pregnant women with mild malaria in Timika Health Center, Timika Jaya Health Center, Pasar Sentral Health Center and Wania Health Center in Mimika Regency Papua Province in November - December 2019 using the Hamilton Anxiety Rate Scale (HARS) questionnaire to measure anxiety levels, a husband's support / involvement questionnaire to measure a husband's support / involvement in pregnancy and preparation for cohesion, a questionnaire level of knowledge of malaria sign and danger in pregnancy. Providing education on the signs and dangers of malaria in pregnancy. The analysis of this study uses the Wilcoxon test. Result: statistical test results show that there are differences in anxiety levels in pregnant women with malaria before and after education, with the average score of anxiety levels tending to decrease, moderate and mild anxiety.
