Abstract

Penelitian ini bertujuan untuk mengetahui untuk mendaptkan gambaran tentang prevalensi kejadian Escherichia coli penghasil ESBL yang resisten terhadap cephalosporin generasi III pada pasien RSUP Dr. Wahidin Sudirohusodo Makassar dan prevalensi genotif CTX-M Escherichia coli penghasil ESBL yang resisten terhadap cephalosporin generasi III pada pasien RSUP Dr. Wahidin Sudirohusodo Makassar. Penelitian dilaksanakan diruang perawatab rawat inap, ruang ICU, ruang HCU, ruang PICU, ruang IGD anak, Infection Center, laboratorium patologi klinik RSUP Dr. Wahidin Sudirohusodo dan laboratorium mikrobiologi klinik RSP Universitas Hasanuddin Makassar periode Juni 2018 – Agustus 2018. Penelitian ini menggunakan desain eksperimental laboratorium dengan teknik pengambilan sampel secara consecutive sampling. Bakteri Escherichia coli diisolasi dari 50 pasien. Pengujian dilakukan meliputi uji sensitivitas antibiotika dengan mengunakan metode difusi agar Kirby-bauer dan vitek 2 compact, uji deteksi ESBL dengan metode Double Disc Synergy Test (DDST), Phenotypic Confirmatory Test dan Uji genotip CTX-M Polymerase Chain Reaction (PCR). Hasil penelitian menunjukkan uji sensitivitas antibiotika pada 50 sampel klinik (Urin, Sputum, Darah, Feses, Pus dan Jaringan) yang diuji terhadap antibiotika golongan sefalosporin generasi III yang telah mengalami resistensi sebesar 40 isolat (80%) baik pada Ceftriaxone, Cefotaxime dan Ceftazidine. Pada uji fenotip dengan metode DDST dan Vitek 2 compact ditemukan 40 sampel (80%) positif ESBL pada Antibiotika Cefotaxime+As.Klavulanat (30 ųg/10 ųg ) maupun Ceftazidine+As.Klavulanat (30 ųg/10 ųg ). Pada uji genotip CTX-M di temukan 41 sampel (82%) memiliki gen CTX-M penghasil ESBL

Abstract

Introduction: Health literacy is one of health determinants  public health development goals. The aim of this research is to determine the effect of precede-proceed education model on health literacy of patients suffer from hypertension in the working area of Pattingalloang Health Centre Makassar. Method: The research employed quasy-experimental method using nonequivalent control design. The research sample was 40 people. The sample was determined using purposive sampling. Data were obtained using questionnaire and interview then it is analyzed by Mann-Whitney and Spearman statistical test. Results: The results showed that: (1) There was differences in the functional health literacy between  hypertension patient group provided with an education and group without education intervention; (2) There was no significant difference in communicative and critical health literacy between group with education intervention and group without education intervention. However, proportion of health literacy of patient with education intervention was better; (3) there was a relation between respondent characteristics, health service access, health information access, and social support that the patient obtained and health literacy. Conclusion: The availability of education to hypertension patients by modifying the precede-proceed model were giving influence to health literacy. This study suggest that involvement of health workers expecially for nurses, in an effort to improve health literacy of patients with counseling, home visite for patients and their families

Abstract

The important genetic factor about susceptibility of oxidative stress in Pulmonary Tuberculosis (PTB) has not been determined. The aim of this study was to analyse the association between polymorphism glutamatecysteine ligase catalytic subunit (GCLC) genes with susceptibility of oxidative stress in PTB in Makassar population of Indonesian. The Methods use is a case-control study was performed on 25 paired subjects with or without PTB, These subjects were selected from result of BTA stain from sputum subjects. The following polymorphisms were genotyped by PCR-RFLP of −129C/T in the GCLC gene. Genotype frequencies and allelic frequencies were analysed. The Results was found There were significant differences in the distribution of genotype frequencies for polymorphism −129C/T in GCLC gene between PTB and non-PTB subjects. The distribution of the allelic frequencies of these genes also showed significant difference between the two groups. Conclusions is The genetic polymorphisms in GCLC −129C/T are associated with susceptibility of Oxidative stress on PTP subject.

Abstract

Mycobacterium tuberculosis is an acid-resistant bacterium that causes tuberculosis and can be detected using a variety of methods. This study aimed to determine the comparison of the detection results of Mycobacterium tuberculosis using the molecular rapid test (MRT) and immunochromatographic method. This research was conducted at Batara Siang Hospital, Pangkajene, and Kepulauan Regencies, in July-September 2019 using 100 samples. This study is cross-sectional research, applying the chi-square test for analysis. The results of statistical tests revealed a significant difference between the results of tests using molecular rapid tests and immunochromatography (<0.001). The sensitivity, specificity, positive probability value, negative probability value, and accuracy of the immunochromatography method against the molecular rapid test (MRT) were 91.3%, 100%, 100%, 93.1%, and 96%, respectively.

Abstract

ABSTRACK Background: Health workers are one of the groups most at risk of being infected with Covid-19 due to their intense interactions with patients. Therefore, it is necessary to make efforts to increase the endurance of the pandemic by administering vaccines.Purpose: This study aims to determine the effectiveness of giving phase III booster vaccinations for health workers against increasing Covid-19 antibody titers.Methode: This study belongs to the type of descriptive quantitative research, the sampling technique is carried out using the slovin formula.Results: The results showed that constantly (100%), giving booster vaccinations to health workers can increase the effectiveness of the Covid-19 vaccine that has previously been given and is able to strengthen the antibody that has been awakened.  Conclusion: The administration of the phase III booster vaccine can increase the titer of the Covid-19 antibody so that the risk of transmission of the corona virus can be suppressed.Suggestion: Other puskesmas can carry out phase-III booster vaccinations for health workers as a whole

Abstract

Pendahuluan: Kualitas udara dalam lingkungan fasilitas kesehatan termasuk ruangan bersalin menggambarkan kondisi kesehatan lingkungan rumah sakit dan dianggap memiliki patogenisitas yang dapat menyebabkan infeksi yang berkaitan dengan rumah sakit. Angka kejadian infeksi banyak ditemukan dalam ruangan yang memiliki mikroorganisme di udara yang lebih tinggi seperti ruang bersalin. Metode: Penelitian ini menggunakan studi dekstriptif observasional dengan rancangan studi cross-sectional dengan melakukan pengamatan, pengukuran kualitas mikrobiologi udara dan identifikasi terhadap bakteri aerob di udara. Identifikasi koloni bakteri aerob yang tumbuh pada media blood agar menggunakan alat Matrix-Assisted Laser Desorption Ionization-Time Of Fight Mass Spectrometry (Maldi-Tof MS). Data yang diperoleh kemudian dilakukan analisis dengan menggunakan aplikasi Statistical for Social Science (SPSS). Analisis hubungan antara suhu dan kelembaban dalam ruangan terhadap nilai konsentrasi mikroorganisme di udara menggunakan uji korelasi Rank Spearman’s. Hasil: konsentasi mikroorganisme di udara dalam ruangan bersalin melebihi nilai standar baku berdasarkan Permenkes RI No.1204/Menkes/SK/X/ tahun 2004. Bakteri-bakteri aerob yang teridentifikasi di udara antara lain Staphylococcus arletae, Staphylococcus gallinarum, Bacillus cereus grup, dan Brevibacillus spp. Kesimpulan: Kelembaban udara sangat mempengaruhi pertumbuhan mikroorganisme di udara. Suhu ruangan pada nilai standar maksimal dan kelembaban dalam nilai standar minimal akan menjaga konsentrasi bakteri udara dalam ruangan memenuhi nilai standar mikroorganisme dalam ruangan.

Abstract

The high demand for N95 masks, especially during the COVID (Coronavirus disease)-19 pandemic, has caused shortages worldwide. This study aimed to examine the sterilization ability of the portable sterilizer prototype for N95 masks and its effect on the filtration ability and changes in air resistance on the N95 mask in order to thrift personal protective equipment (PPE) use during a shortage. The sample used was an N95 mask type 1860. The mask was contaminated with 0.6-0.8 MFU Staphylococcus aureus and Escherichia coli. The sterilization methods used were Ultraviolet Germicidal Irradiation (UVGI), Heat at 75°C, and a combination of both from 1 to 120 minutes. Next, the masks were cultured in a nutrient agar medium. For aerosol penetration and air resistance tests, masks were tested before and after the sterilization process, lasting from 5 to 60 minutes. This prototype sterilizer with Heat effectively killed E. coli and S. aureus starting from 3 minutes. The filtration ability of the N95 mask was maintained at >95% even after the sterilization process with 75°C heat, UVC, or a combination of both for up to 60 minutes. There was also no significant difference in air resistance between new masks and masks that had been sterilized using a portable sterilizer. This prototype sterilizer with Heat at 75°C can effectively sterilize against both gram-positive and negative bacteria in the N95 mask without reducing the aerosol filtration ability and changing the air resistance of the N95 mask.

Abstract

This study aims to analyze the determinants of the occurrence of anemia in pregnant women. The study used an observational analytic design with a case control with analysis using bivariate and multivariate methods. The results show that a significant relationship with the incidence of anemia in third trimester pregnant women, namely pregnancy distance, KEK status, compliance with Fe Consumption, Achievement of ANC visits and History of exposure to Covid19. The factors that most influenced the incidence of anemia were Compliance with Fe Tablet Consumption, mother's KEK status and history of exposure to Covid19. Age and history of premarital counseling were not significant to the incidence of anemia. Conclusion compliance with consumption of Fe tablets is the most influential factor on the incidence of anemia in third trimester pregnant women at new normal era. Abstract

Leprosy, caused by Mycobacterium leprae, is a chronic infectious disease that impacts the skin and peripheral nerves, causing long-term disability. The invasion of M. leprae into the body triggers immunologic responses and single single-nucleotide polymorphisms in cytokine-encoding genes may influence predisposition and susceptibility, possibly predicting the incidence of leprosy reactions. The aim of this study was to assess the gene polymorphism of interleukin-10 promoter IL-10 −819C/T in leprosy patients, leprosy patients with erythema nodosum leprosum (ENL) reaction, and household contacts. A total of 54 individuals were included, with 18 in each group. Skin smear and histopathologic examinations were used to confirm the diagnosis of leprosy and ENL. The polymerase chain reaction and restriction fragment length polymorphism (PCR-RFLP) technique was used to determine the polymorphism. The results confirmed the presence of polymorphism of which all TT, CT, and CC genotypes presented. The TT genotype was most prevalent in household contacts (94.4%) followed by ENL (50%), and leprosy patients (44.4%). The CT genotype was most frequently detected in leprosy patients (50%), followed by ENL cases (44.4%), and household contacts (5.56%). In contrast, CC was mostly presented in ENL cases (5.56%), only 1% in leprosy patients, and absent among household contacts. Although the most prevalent allele in all three groups was the T allele, the C allele presented in 27% and 30% of ENL and leprosy patients, respectively and only 5% in household contact individuals. This study suggests that the polymorphism variations of IL-10 −819C/T are higher in leprosy and ENL patients compared to household contacts. Since this data is preliminary, larger studies are needed.

Abstract

The main challenge in the management of Tuberculosis (TB) is diagnosing quickly and accurately, especially Latent Tuberculosis Infection (LTBI). LTBI detection was carried out using the Tuberculin Skin Test (TST) and Interferon Gamma Release Assay (IGRA). In TB endemic areas, these two examinations have limitations, so current research is directed at finding specific antigens for the diagnosis of LTBI. One of the potential proteins is Antigen 85A (Ag85A) Mycobacterium tuberculosis (Mtb) encoded by Fibronectin-binding protein A (FbpA). The Ag85 complex induces the proliferation of T-cells and interferon-gamma in most healthy individuals infected with Mycobacterium tuberculosis, Mycobacterium leprae, and BCG-vaccinated mice, making it a potential antigen. This study aims to clone and produce recombinant protein Ag85A from Mtb in Escherichia coli BL21. The methods used were ligation to the pET-32a expression vector, transformation to Escherichia coli BL21, and production of protein by IPTG induction. Characterization of recombinant clones by colony PCR and sequencing. The results obtained were that the fbpA gene isolated from Mtb clinical isolate had been amplified, and the PCR product was 900 bp. The production of Antigen 85A has been successfully carried out and produces 44 kDa.
