Abstract

Mesenchymal Stem Cells Wharton's Jelly (MSC-WJ) has become a new hope for Alzheimer's disease due to its capability to suppress Amyloid-beta (Aβ) plaques formation. It is found that people with Alzheimer's disease have a lower expression of Sirtuin-1 because of the oxidative stress that was produced by Aβ plaques. Objective: To observe the Sirtuin-1 gene in Alzheimer's disease rat models that AlCl3 was inducing after being given MSC-WJ. Methods: This was an experimental study with a post-test-only control group design, including three groups with a total sample of 18 samples of Alzheimer RNA rat models. Group-1without AlCl3 and MSC-WJ; Group-2 with AlCl3; Group-3 AlCl3 and MSC-WJ. Mean value of the Sirtuin-1 gene was obtained by the Image-J application using a semiquantitative method which was then tested using the One Way ANOVA test. The study was significant if the p-value < 0,05. Results: This study found that the mean ± standard error value of the Sirtuin-1 gene in three groups were 0.18±0.04; 0.16±0.03; and 0.36±0.06 respectively. The statistical test showed a significant difference between the three groups (p = 0.014). Conclusion: There was an increased Sirtuin 1 gene expression in Alzheimer’s rat Model after being given Mesenchymal Stem Cells Wharton’s Jelly.Keywords:  Alzheimer, MSC-WJ, Sirtuin-1

Abstract

Latar Belakang: Jumlah penderita diabetes melitus gestasional terus meningkat di seluruh dunia. Kondisi hiperglikemia yang terjadi pada masa kehamilan dapat memicu terjadinya stress oksidatif yang dapat menstimulasi peningkatan ekspresi Tumor Necrosis Factor-α (TNF-α). Ekstrak daun kemangi (Ocimum basilicum) mengandung zat antidiabetes dan antioksidan. Penelitian ini bertujuan untuk melihat efektifitas ekstrak daun kemangi (Ocimum basilicum) terhadap ekspresi TNF-α pada tikus model diabetes melitus gestasional. Objektif: Untuk mengetahui pengaruh pemberian ektrak daun kemangi (Ocimum basilicum) terhadap ekspresi TNF-α pada tikus model diabetes melitus gestasional. Metode: Penelitian ini merupakan penelitian eksperimental dengan menggunakan metode post test only control group design. Sampel penelitian menggunakan bahan biologis tersimpan dari RNA tikus yang telah diinduksi streptozotocin 40 mg/KgBB sebagai animal model DMG pada manusia yang disimpan di Lab Biomedik FK Unand. Hasil: Ekspresi TNF-α pada kontrol negatif, kontrol positif, perlakuan satu dan perlakuan dua berturut-turut adalah 1,01, 1,06, 0,95, dan 0,61. Kesimpulan: Menunjukkan adanya pengaruh pemberian ekstrak daun kemangi (Ocimum basilicum) terhadap ekspresi TNF-α pada tikus model diabetes melitus gestasional.

Abstract

The increased production of beta-amyloid marks Alzheimer's disease. PPAR-γ shows involvement in Alzheimer's disease. PPAR-γ Gene is suspected to reduce beta-amyloid plaques and reduce inflammation and oxidative stress in Alzheimer's disease. Mesenchymal Stem Cells Wharton's Jelly (MSC-WJ) is expected to increase the PPAR-γ Gene expression. Objective:  To observed the PPAR-γ Gene expression in Alzheimer's Mice. Methods: The mice are induced using AlCl3 and were given MSC-WJ. This study was an experimental study with a post-test-only control group design on 18 samples of RNA from Alzheimer's mice that were divided into 3 groups. Negative control group (K-), positive control group (K+), and experimented group (P). The mean of PPAR-γ gene expression was obtained by comparing the PPAR-γ gene with the GAPDH gene. This study used semiquantitative methods using ImageJ. Data analysis was used with the Kruskal-Wallis test. It is significant if the value is <0,05. Results: The mean ratio of PPAR-γ gene expression that was obtained in K-, K+, and P were 0.12, 0.06, and 0.08, respectively. There were significant differences between each group, with a p-value of 0,023 (p<0,05). Conclusion: Giving MSC-WJ could increase the PPAR-γ gene expression. Further study using real-time PCR is highly needed to increase the MSC-WJ treatment in patients with Alzheimer's disease.Keywords:  Alzheimer disease, MSC-WJ, PPAR- γ

Abstract

Objective: This study aimed to evaluate the therapeutic potential of Bone Marrow-derived Mesenchymal Stem Cells (BM-MSCs) in mitigating amyloid plaque formation in Alzheimer's disease induced by Aluminum Chloride (AlCl3) in rats. Methods: Rats were divided into three groups: negative control, positive control (AlCl3-induced), and treatment (AlCl3 + BM-MSCs). Cognitive function was assessed five days post-induction, and amyloid plaques were quantified using Congo Red staining in the cortex and hippocampus. Results: A significant reduction in amyloid plaque was observed in rats treated with BM-MSCs. The proportion of stained areas decreased from 1.88 to 1.73 in the cortex and from 1.61 to 1.47 in the hippocampus, compared to AlCl3-only controls. Conclusion: BM-MSCs demonstrated a moderate but statistically significant reduction in amyloid plaque in a rat model of Alzheimer's disease, indicating their therapeutic potential.

Abstract

Latar belakang: Rinosinusitis kronis (RSK) merupakan inflamasi pada mukosa hidung dan sinus paranasal. Rinosinusitis kronis berdasarkan perbedaan ekspresi sitokin dan pola remodeling inflamasi dibedakan menjadi dua fenotipe, RSK dengan polip dan RSK tanpa polip. Transforming growth factor–?1 (TGF–?1) merupakan salah satu sitokin yang berperan pada remodeling jaringan.       Tujuan: Mengetahui ekspresi gen TGF–?1 pada RSK dengan polip dan RSK tanpa polip. Metode: Penelitian analitik komparatif menggunakan desain potong lintang (cross sectional comparative study) pada 12 responden RSK dengan polip dan 12 responden RSK tanpa polip. Sampel diambil saat operasi Bedah Sinus Endoskopi Fungsional (BSEF) pada jaringan polip dan mukosa sinus etmoid atau maksila. Semua sampel dilakukan pemeriksaan ekspresi gen dengan metode Real Time Polymerase Chain Reaction (RT-PCR). Data dianalisis dengan SPSS, dikatakan bermakna jika p<0,05. Hasil: Ekspresi TGF-?1 pada RSK tanpa polip lebih tinggi (18,63±24,58) dibandingkan RSK dengan polip (2,82±4,02). Secara statistik perbedaan antara RSK polip dan RSK tanpa polip ini bermakna (p<0,05). Kesimpulan: Terdapat peningkatan ekspresi TGF-?1 pada RSK tanpa polip yang bermakna secara statistik dibandingkan dengan RSK dengan polip.

Abstract

The incidence of diabetes continues to increase worldwide. The state of hyperglycemia during pregnancy can trigger oxidative stress which can result in hypoxic conditions in the tissues characterized by an increase in HIF-1α. Basil which contains antioxidants is expected to reduce the expression of HIF-1α. This study aims to determine the effect of the administration of basil on the expression of HIF-1α in gestational diabetes mellitus model rats. To determine the effect of the administration of basil extract (Ocimum basilicum) on the expression of HIF-1α in gestational diabetes mellitus model rats. This research is experimental research conducted using the post test only control group design method. The research sample used biological material stored from rat RNA that had been induced by streptozotocin 40 mg/kgBB as an animal model of DMG in humans stored in the Biomedical Lab, FK Unand. The results of this study indicate the effect of the administration of basil extract (Ocimum basilicum) on the expression of HIF-1α in gestational diabetes mellitus model rats. Researchers used conventional PCR tools and still manually used ImageJ to look for HIF-1α expression values from electrophoresis results, so the possibility of bias still exists.

Abstract

Elementary school students are consumers who are among those who consume the most food and beverages. Food and drinks sold around the school contain many hazardous substances such as dyes that have been banned from circulating (Rhodamine B in iced drinks, jelly drinks, and so on) and other additives such as formalin which is used as a chewy substance in meatballs sold in carts and restaurants that can cause acute poisonings such as allergies, coughs and chronic poisonings such as cancer of the liver, kidneys and other organs. The purpose of this activity is to increase knowledge about substances in food and an introduction to blood group examination. This activity was carried out at SD Negeri 13 Tungkal Selatan Village in North Pariaman District. The method of activity is in the form of counseling and demonstration. According to the survey before the blood group examination, it turned out that most of the elementary school students did not know their blood type. Before the blood group examination, the students were also given knowledge about determining a person's blood type. After this counseling was carried out, respondents became more aware of the impact of food/snacks on health, and respondents knew their respective blood types. Of the 64 students, there are 22 A, 17 B, 12 AB, 13 O. To avoid problems related to food and blood type. There is a need for continuous health education for elementary school students.
