Abstract

Logam berat merupakan zat pencemar yang sangat berbahaya bagi sistem lingkungan hidup karena bersifat toksik, tidak dapat terurai secara alami dan cenderung terakumulasi didalam perairan dan tubuh organisme. Akumulasi logam berat Hg, Cu dan Pb dalam biota dapat berdampak buruk bagi kesehatan manusia yang mengonsumsinya dalam konsentrasi tertentu seperti gangguan sistem saraf, merusak ginjal bahkan kematian. Penelitian ini bertujuan untuk menentukan kandungan logam berat merkuri (Hg), tembaga (Cu) dan timbal (Pb) pada kerang hijau (Perna viridis) dan batas aman konsumsinya di wilayah Pelabuhan  Tanjung Mas dan perairan daerah Mangunharjo, Kota Semarang. Metode penelitian yang digunakan yaitu metode deskriptif eksploratif. Teknik Pengambilan sampel dan penentuan lokasi menggunakan metode purposive sampling. Pengambilan sampel dilakukan pada bulan Juli 2020. Analisis kandungan logam berat menggunakan alat AAS (Atomic Absorption Spectrophotometer) dan Mercury Analyzer. Hasil penelitian menunjukkan bahwa rata-rata kandungan logam berat pada wilayah Pelabuhan Tanjung Mas lebih tinggi yaitu (<0,002 mg/Kg) untuk (Hg), (1,864 mg/Kg) untuk (Cu) dan (0,697 mg/Kg) untuk (Pb), sementara daerah Mangunharjo memperoleh hasil yang lebih rendah yakni (1,646 mg/Kg) untuk (Cu) dan ( 0,253 mg/Kg) untuk (Pb) serta (<0,002 mg/Kg) untuk (Hg). Berdasarkan baku mutu menurut Keputusan Menteri Lingkungan Hidup No.22 Tahun 2021, tingkat pencemaran logam berat di Pelabuhan Tanjung Mas dan Perairan Mangunharjo sudah melampaui batas yang sudah ditetapkan dan batas maksimum daging kerang hijau yang dapat dikonsumsi untuk berat badan rata-rata 60 kg yaitu ± 2,15 kg dan 2,55 kg per minggu untuk masing masing lokasi.Heavy metals are pollutants that are very dangerous for the environmental system because they are toxic, cannot be decomposed naturally and tend to accumulate in waters and organisms' bodies. The accumulation of heavy metals Hg, Cu and Pb in biota can have a negative impact on human health who consume them in certain concentrations such as nervous system disorders, kidney damage and even death. This study aims to determine the content of heavy metals mercury (Hg), copper (Cu) and lead (Pb) in green mussels (Perna viridis) and  its safe limit for consumption in the Tanjung Mas Harbor area and the waters area of Mangunharjo, Semarang City. The research method used is descriptive exploratory method. Sampling technique and location determination using purposive sampling method. Sample was taken in July 2020. The analysis of heavy metal content was conducted using AAS (Atomic Absorption Spectrophotometer) and Mercury Analyzer. The results showed that the average heavy metal content in the Tanjung Mas Port area was higher (<0.002 mg/Kg) for (Hg), (1.864 mg/Kg) for (Cu) and (0.697 mg/Kg) for (Pb), while the Mangunharjo area obtained lower yields (1.646 mg/Kg) for (Cu) and (0.253 mg/Kg) for (Pb) and (<0.002 mg/Kg) for (Hg). Based on the quality standard according to the Decree of the Minister of the Environment No. 22 of 2021, the level of heavy metal pollution in both locations has exceeded the predetermined limit and the maximum limit of green mussel that can be consumed for an average body weight of 60 kg from both locations is ± 2,15 kg and 2,55 kg per week.

Abstract

Research has made efforts to contain the spread of the Covid-19 virus in the work area of ​​PT. Rosaria Bakery. This study aims to determine how occupational safety and health programs are implemented in an effort to prevent the spread of the covid-19 virus in the work environment of PT. Rosaria Bakery has formed a Covid-19 Team which has established Working Guidelines for the New Normal Conditions. This work guide contains several modules that must be adhered to by all employees and other related parties to PT. Rosaria Bakery, including the Work From Home module, the driving module, the receiving guests and packages module, the meeting module, the dining module (canteen), the prayer prayer module, the janitor module and the Work Guide. The purporse of this study is to monitor the implementation of occupational safety and health programs carried out by workers in an affort to protect all employees. And this method han an employee as a resource person who is working some where by isong validation and reliability test.

Abstract

Rhizophora apiculata is a type of mangrove that has the ability to adapt to extreme environmental conditions such as temperature, low oxygen levels, and high salinity. This adaptability affects the production of secondary metabolites. Information about the antibacterial activity of this mangrove against MDR (Multi-Drug Resistant) bacteria is still very limited. The content of secondary metabolites produced by mangrove R. apiculata is also expected to affect antioxidant activity against free radicals. The purposes of this study were to examine the presence of bioactive compounds by phytochemical tests and to evaluate the antibacterial activity against MDR bacteria such as Pseudomonas aeruginosa, Escherichia coli, Bacillus cereus, and Bacillus subtilis; and antioxidants property of R. Apiculata leaves that were collected from Teluk Awur, Jepara. The leaves were extracted using the multilevel maceration method with solvent sequence n-hexane, ethyl acetate and methanol. Metabolite finger printing using TLC method was carried out to detect the presence of alkaloids, flavonoids, saponins, phenolics, quinones, steroids, and triterpenoids. The antibacterial test was carried out using agar well diffusion method while the antioxidant test was carried out using the DPPH method. The results of the phytochemical test showed that there were groups of alkaloids and steroids in the n-hexane solvent; alkaloids, phenolics, and steroids in ethyl acetate solvent; as well as alkaloids, flavonoids, phenolics, and saponins in methanol solvents. The results of this study indicate that R. apiculata from Teluk Awur Coastal Waters, Jepara had no potential as an antibacterial against MDR (Multi-Drug Resistant) bacteria, however the methanol extract has the potential to be used as an antioxidant with an IC50 value of 85.999 ppm. The bioautography showed that compounds from the phenol group, flavonoids, triterpenoids and b-carotene pigments acted as antioxidant agents in the leaf extract of R. apiculata.   Rhizophora apiculata merupakan salah satu jenis mangrove yang memiliki kemampuan beradaptasi terhadap kondisi lingkungan ekstrem seperti suhu, kadar oksigen rendah dan salinitas tinggi. Kemampuan beradaptasi tersebut mempengaruhi produksi matabolit sekunder. Informasi mengenai kemampuan aktivitas antibakteri mangrove jenis ini melawan bakteri MDR (Multi Drug Resistent) masih sangat terbatas. Kandungan metabolit sekunder yang dihasilkan mangrove R. apiculata ini juga diharapkan dapat mempengaruhi aktivitas antioksidan melawan radikal bebas. Tujuan dari penelitian ini adalah untuk mengkaji kandungan senyawa bioaktif serta aktivitas biologis berupa antibakteri melawan bakteri MDR Pseudomonas aeruginosa, Escherichia coli, Bacillus cereus dan Bacillus subtilis; dan antioksidan dari ekstrak daun mangrove R. apiculata asal perairan Teluk Awur, Jepara. Sampel diekstraksi menggunakan 3 pelarut berbeda (n-heksana, etil asetat dan metanol) dengan metode maserasi bertingkat. Analisis metabolit sidik jari dilakukan menggunakan plat KLT untuk mengetahui kandungan senyawa alkaloid, flavonoid, saponin, fenolik, kuinon, steroid dan triterpenoid. Uji antibakteri dilakukan menggunakan metode sumuran sedangkan uji antioksidan dilakukan menggunakan metode DPPH. Hasil uji fitokimia menunjukkan terdapat golongan senyawa alkaloid dan steroid pada pelarut n-heksana; alkaloid, fenolik dan steroid pada pelarut etil asetat; serta alkaloid, flavonoid, fenolik dan saponin pada pelarut metanol. Hasil penelitian ini mengindikasikan bahwa R. apiculata asal perairan Teluk Awur, Jepara tidak potensial sebagai antibakteri melawan bakteri MDR (Multi Drug Resistent), akan tetapi ekstrak metanol potensial digunakan sebagai antioksidan dengan nilai IC50 85,999 ppm. Tahapan bioautografi menunjukkan bahwa senyawa dari golongan fenol, flavonoid, triterpenoid dan pigmen b-karoten berperan sebagai agen antioksidan pada ekstrak daun R. apiculata.

Abstract

Seagrass ecosystems are limited by several environmental factors, namely temperature, light, salinity, depth, primary substrate, nutrients and movement of seawater (waves, currents, tides). The type of substrate can cause differences in the composition of seagrass species and affect differences in the fertility and growth of each kind of seagrass. Seagrasses are suitable for living on muddy, sandy, loam, or substrates with coral fragments and rock crevices. This study aims to determine the ecological index of seagrass in Prawean Beach, Jepara. The research was conducted using an exploratory descriptive method. The study was carried out in November 2021. The materials used in this study were sediment and seagrass samples. The research was carried out in November 2021. Identification of seagrass species using the LIPI method and grain size analysis of sediments using the granulometric method. PCA analysis aims to determine the variables of ecological factors that affect the ecological index of seagrass. PCA analysis will show necessary information regarding the relationship between variables in one graph. The results obtained four seagrass types: Thalassia hemprichii, Enhalus acoroides, Cymodocea serrulata and Cymodocea rotundata. The species density value is classified as relatively dense and the species cover value is classified as less rich/unhealthy with the type of sediment in the form of fine sand. PCA analysis resulted in 3 main factors formed, namely: F1) Enhalus acoroides cover, gravel, silt, temperature, pH and current; F2) Sand and salinity; and F3) Density and cover of Cymodocea rotundata.
