Abstract

Dewasa ini, di Indonesia eksplorasi untuk mencari bahan pangan alternatif pengganti beras masih terus dilakukan. Kandungan makronutrien dan kajian glisemik indeks dari umbi ganyong (Canna edulis Kerr.), walur (Amorphophallus variabilis), porang (Amorphophallus Oncophyllus Prain), suweg (Amorphophallus campanulatus Bl ) dan uwi (Dioscorea alata L.) telah dilakukan pada penelitian ini dan bertujuan untuk memberikan dasar ilmiah pemilihan pangan alternatif, terutama bagi penderita diabetes mellitus dan obesitas. Pengukuran kandungan makronutrien meliputi kandungan karbohidrat mereduksi dan tidak mereduksi, protein dan serat dilakukan mengacu pada metode AOAC (1990), sedangkan glisemik indeks ditetapkan secara in vivo menggunakan hewan percobaan tikus jantan galur Wistar. Glisemik indeks sampel umbi-umbian seluruhnya lebih rendah dari glisemik indeks beras (72,8). Harga glisemik indeks sangat dipengaruhi oleh kandungan serat, sedangkan pengaruh kandungan karbohidrat terhadap harga glisemik indeks tidak dapat diamati. Walur dan porang memiliki kandungan serat yang besar, yaitu masing-masing 15,09% dan 11,27%, sedangkan harga glisemik indeks masing-masing sekitar 20,6 dan 16,9. Kata kunci: glisemik indeks, makronutrien, karbohidrat, serat, protein

Abstract

Bengkoang has long been used by ancestors as a raw material for cosmetics. In Indonesia, it is usually eaten raw, sometimes with salt, lemon juice and powdered chili. However, scientific evidences that support the use of bengkoang have not been widely published. Phytosterols from the bengkoang root have been isolated and identified based on their NMR spectrum data and mass spectrum. The result of the structure elucidation informed that ß-sitosterol and stigmasterol are major components of phytosterol in bengkoang root. Phytosterol content was about 0.02% of dry weight bengkoang or 2.76 % in petroleum ether extract of bengkoang, with a ratio of ß-sitosterol and stigmasterol (65:35). Therefore, bengkoang can be further exploited as a raw material of phytosterol supplement.

Abstract

Bengkoang telah banyak digunakan dalam industri kosmetika sebagai whitening agent. Berdasarkan penelitian Lukitaningsih (2009), bengkoang mengandung 6 senyawa aktif yang mampu berperan sebagai whitening agent dengan menghambat aktivitas enzim tirosinase dari jamur Aspergillus oryzae (TyrAo). Namun interaksi senyawa aktif bengkoang dalam menghambat enzim tirosinase belum dapat diketahui. Interaksi senyawa-senyawa aktif bengkoang dengan enzim TyrAo dapat diketahui dengan studi komputasional (in silico). Pemodelan interaksi senyawa aktif bengkoang dengan enzim TyrAo dilakukan dengan metode homology modeling dan molecular docking. Homology modeling dilakukan untuk memodelkan struktur tiga dimensi (3D) enzim tirosinase Aspergillus oryzae (TyrAo) melalui template berupa protein homolog yang sudah diketahui struktur 3D-nya yaitu enzim TyrAb (PDBID: 2Y9X). Model TyrAo digunakan sebagai target makromolekul dalam metode molecular docking. Metode molecular docking merupakan metode untuk menggambarkan posisi ligan (senyawa-senyawa aktif bengkoang) pada sisi aktif reseptor (model TyrAo). Berdasarkan docking yang dilakukan diketahui bahwa residu-residu yang banyak berpengaruh pada interaksi ligan pada sisi aktif adalah residu Thr275 yang berinteraksi secara ikatan hidrogen dengan ligan dan residu His294 yang berinteraksi secara hidrofobik pada cincin aromatik ligan. Penelitian in silico dan in vitro yang telah dilakukan memiliki korelasi (R2) sebesar -0,8366. Korelasi ini menandakan bahwa aktivitas senyawa-senyawa aktif pada bengkoang dalam menghambat enzim TyrAo memiliki hasil yang serupa pada penelitian yang  dilakukan secara in silico dan in vitro.

Abstract

The form of liposome formulations is one of strategies to overcome the limitations of anthocyanin extract stability of roselle calyx (Hibiscus sabdariffa L.). In this study, antioxidant comparative analysis of antioxidant activity and total anthocyanin content from rosella extract and its liposomes form was conducted. Preparation of extract made by maceration using the best solvent consisted of methanol : formic acid (97:3) (v/v). From this procedure, the obtained extract was about 26.40% and antioxidant activity was 19.99%. Liposome formulations of methanol extract from roselle calyx were prepared by reverse phase method using rosella extract as aqueous phase and egg lecithin as the lipid phase. The stability of exctract and liposome form of rosella calyx was compared. The stability study showed that there was a reduction of antioxidant activity (DPPH test) 11.25 % for the roselle extract, while the liposome form was 0.93% after 30 days storage in low temperature (0-50C) and saturated nitrogen. In addition, total anthocyanin content (pH differential method test) decreased approximately 39.12 % for  the roselle extract and 0.72% for the liposome form. More over, the liposome had lower level of irritation than the form of extract. Therefore, it can be concluded that anthocyanin encapsulated by liposome was more stable, safe, and effective than the anthocyanin of the rosella calyx extract.

Abstract

The aim of the study was the determination of glycaemic index (GI) of local tubers such as Uwi (Dioscorea alata), Gadung (Dioscorea hispida), dan Talas (Colocasia esculenta). Tubers were prepared as raw flour which were administered orally to the rats. Blood were taken at 0; 0,5;  1 and 2 hour after tubers administration. The dose was 2,5 kg/kg body weight (BW) whereas glucose was administered as a standart treatment. Area Under Curve (AUC) was calculated after plotting the graph of time versus glucose levels. Glucose levels were determined using GOD-PAP kit. GI values were calculated as a ratio of AUC (0-2hr) of tubers to glucose. Porang was moderate while the other tubers were low GI.

Abstract

Bengkoang or Pachyrhizus erosus has been used as a traditional cosmetic material since many years ago. Until now, many cosmetic preparations using bengkoang as a main material have been produced by cosmetic industries, mainly for skin care, whitening and sunscreen preparation. However, content of vitamin A, vitamin C and alpha hydroxyl acid has not been discovered yet. These compounds are essential in skin care process because of their activities in cell regeneration, antioxidant as well as peeling of dead skin cell layer. The aim of this research is to provide information about the content of three compounds in bengkoang. Vitamin A has been analysed by spectrophotometer, vitamin C analysed using titration method and alpha hydroxyl acid analysed using gas chromatography. The result showed that concentrations of vitamin A and vitamin C are 179.21 ± 8.19 ppm and 0.31 ± 0.06 %, respectively. Meanwhile the content of alpha hydroxyl acid was 0.80 ± 0.01% (measured as glycolic acid).

Abstract

This study aims to determine the bioactivity of L.domesticum as cosmetic ingredient. The extraction was prepared by maceration with ethanol 96 % and ethyl acetate to obtain ethanolic extract of fruit flesh (BLE), ethanolic extract of fruit peel (KLE), ethyl acetate extract of fruit flesh (KLE) and fruit peel (KLEA). Then, determination of compounds was conducted by thin layer chromatography (TLC) and analysed of total phenolic and flavonoid content with colorimetric method. The antioxidant capacity was measured using radical scavenging assay of 1,2-diphenyl-2-picrylhydrazyl (DPPH), β-carotene bleaching assay (BCB) and ferric reducing antioxidant power assay (FRAP). The inhibitory effect of skin degradation enzymes (anti-aging) was carried out using elastase and collagenase assay. The tyrosinase inhibitory effect was determined using mushroom tyrosinase. Based on TLC result, extract of L.domesticum containing phenolic, flavonoids, terpenoids and steroids compounds. The largest of total flavonoid content was obtained in KLE (4,76±0,26 %b/b EQ) and phenolic total content in BLEA (6,4±0,15 %b/b EG). Sequence of antioxidant activity measured using DPPH method was BLEA> KLE> BLE> KLEA; while using BCB method and FRAP method were respectively KLEA>BLEA> KLE> BLE; and BLEA> KLE> BLE> KLEA. KLE and KLEA extracts showed the most potent to remaining activity of the enzyme elastase and collagenase. In addition, BLE extract has the higest tyrosinase inhibition than others. Based on these results it can be concluded that the extracts of the fruit flesh and fruit peel of L. domesticum can be used as a cosmetic active ingredient for anti-aging and anti-tyrosinase.

Abstract

The present of chlorophyll is not expected in the isolation process of plant active constituent. Electrocoagulation is a potential dechlorophyllation method. This research aims to know the effectivity of electrocoagulation in the dechlorophyllation process of Cosmos caudatus Kunth., Morinda citrifolia, and Mangifera indica L. leaves methanolic extract as well as the effect to the total phenolic content. Electrocoagulation are performed using copper, silver, aluminum and iron plates as the electrode. Dechlorophyllation by extraction using n-hexane is performed as reference. The % absorbance of chlorophyll and % of total phenolic content of dechlorophyllated samples are measured spectrophotometrically. Electrocoagulation process reduces % absorbance of chlorophyll in Cosmos caudatus Kunth., Morinda citrifolia, and Mangifera indica L. leaves methanolic extract. The more the duration of electrocoagulaton process the lower the % absorbance of chlorophyll obtained. % absorbance of chlorophyll of the electrocoagulated extract according to paired t-test (P=0.95) are significantly different with the previous. One way ANOVA continued with LSD (P=0.95) shows that the % absorbance of chlorophyll from the electrocoagulated extract are significantly different with those extracted with n-hexane. Electrocoagulation process reduces total phenolic content along with duration of electrocoagulation.

Abstract

Not many researches on a fruit of limpasu (Baccaurea lanceolata Muell. Arg ) were found, while the use by tribal hamlets in Kalimantan already became a daily routine before they went into the field with the way made powder and smeared on the face. Therefore, the research on active substances needs to be done. For this study, we prepared two isolates which coded as K2 and CE, K2 from qualitative test negative isolates phenolic groups was identified by spray FeCl3, whereas qualitative test positive isolates were coded as CE and detected by a nitrogen-containing reagents dragendorf spray. The K2 isolate from SPF test resulted an SPF value of 3.2 at a concentration of 0.2 mg/ml, while CE isolate was given a value of SPF 5.0 at a concentration of 0.2 mg/ml. According to sensitivity skin test on rabbit skin, both K2 and CE isolated were given the value below 1, indicated the isolates were found to not irritate. This study proposes the potential from Baccaurea lanceolata Muell.Arg fruit isolates to be developed as a sunscreen for UV-protection.Keywords : Baccaurea lanceolata Muell.Arg, SPF test, sensitivity skin test

Abstract

Osteoporosis is a common problem in menopause woman. The main cause is lack of estrogen hormone. Commonly, prevention therapy is by consuming high calcium milk, but it is not effective. Bali orange’s peel (Citrus maxima Merr.) is a waste material but known contains phytoestrogen according to previous study. Considering of this result, fortification of high calcium milk and Bali orange’s peel is expected to be an effective solution for osteoporosis in menopause woman. This research began with extraction of Bali orange’s peel (BPE) using ethanol 70% by maceration method. Ovariectomized Sprague Dawley female rats as the model of post menopausal woman were treated by BPE for 28 days. The doses of BPE was given to rats is 500 and 1000 mg/KgBW combined with high calcium milk. Bone density was determined using digital microradiography, the profile showed the increase of bone density in group that treated with combination of BPE 1000 mg/Kg BW and high calcium milk compare to control and given only milk groups. Docking molecular showed that BPE’s active compound which are hesperidin and naringin have interaction with estrogen receptor α and β. Docking score of naringin with ER α and β are -19,97; -18,99 respectively. Meanwhile the docking score of hesperidin with ER α and β are -19,98; +49,92 respectively. Overall, the result of this research showed that fortification of BPE with high calcium milk has good prospect to develop as effective therapy of osteoporosis.Keywords : Citrus maxima Merr., phytoestrogen, osteoporosis, high calcium milk, estrogen receptor
