Abstract

Shallot (Allium wakegi Arakic.v. Lembah Palu) is a horticulture commodity prospective as a very good business opportunity, since becomes the major ingredient of fried shallot, the most unique souvenir of Palu. The productivity however, is generally low due to lack in availability of good seed and seed production technology. The purpose of this study is to develop seed production technology through multiplication of somatic embryos. This assessment is the second part of research stages regarding the development of shallotsomatic embryo technology in Central Sulawesi. The first stage of the research is setrilization and callus induction of Lembah Palu shallot. This second stage is maturation dan germination of somatic embryos.The experiment was arranged as factorial completely randomized design, comprising of two factors. The first factor was 2,4-D concentration; consisting of three levels i.e 0.00, 0.25 and 0.50 mg/L. The second factor is kinetin concentration with the same levels as in 2,4-D. Each treatment combination was replicated three times, thus there were 27 experimental units. The result showed that addition of kinetin in MS basal media strongly affected the percentage of mature embryos and the percentage of mature embyos producing shoots, with the highest rate was obtained from the media fortified with 0,50 mg/L kinetin (26,913% and 43,318% respectively). Addition of 2,4-D to the MS basal media tended to inhibit the rate of regenerating shoot from the embryos. There is no interaction between 2,4-D and kinetin concentrations in affecting maturation and germination of the somatic embryos.

Abstract

Production and management technologies are needed to produce great seeds. This study aims to obtain the location and length of time to store upland rice seeds. The study was conducted in Tamarenja and Kalukubula villages. Germination testing was carried out at the Seed Science and Technology Laboratory of the Faculty of Agriculture from May to December 2019. The seeds used were newly harvested seeds from the field. The treatment used a randomized two-factor block design. The first factor was storage locations: indoor Tamarenja, outdoor Tamarenja, indoor Kalukubula and outdoor Tamarenja. The second factor consisted of two storage times, namely: one month and two months. The treatments repeated six times, so there were 48 experimental units where each experimental unit contains 50 seeds, and the whole seeds used were 2,400. Data analysis used Fisher's test and continued with 1% Tukey’s HSD test. The results showed that the best storage location was in Kalukubula, both outdoors and indoors. The best storage time was two months. Those produced the highest germination capacity and growth potential and faster germination time, percentage of moldy seeds, and moldy sprouts.

Abstract

Before crossing plants, it's critical to understand their morphological characteristics. Differences in plant morphological features distinguish the advantages of each plant. The objective of this study was to determine the morphological characteristics of superior purple and local red corn cultivated at the height of 22 meters above sea level, as well as their adaptability. Kalukubula Village, Sigi Biromaru District, Sigi Regency, Central Sulawesi was the site of this study. The research took place between October 2020 and February 2021. A Randomized Block Design was used in this investigation, with two corn kinds as treatments: The experiment was repeated 16 times with purple superior corn and red corn, yielding 32 experimental units. The quantitative data was examined using ovarian analysis (ANOVA) followed by a 5% Tukey HSD test to assess the difference between the two corns. Plant height, number of leaves, leaf length, stem diameter, age of male flower appearance, harvest age, fruit diameter with husk, length of cob, number of seeds per fruit, the weight of kernel per fruit, and dry kernels production of tons/ha were all found to have a higher growth rate than purple corn kernels. The qualitative characters of superior purple corn kernels include: leaf veins and leaf bases are purple, stems are purplish-green, female flowers are white with anger, the husk is purplish-green, and kernels are purple-black. Local red corn include white leaf veins, light green leaf bases, purple stems, light green female flowers, purple husk, and red kernels.

Abstract

The relationship between several characters of yields is very important for developing effective selection criteria. The total correlation between the result character and the result component may be misleading as a result of its relationship with other characters. Separating the total correlation into direct and indirect effects provides a more significant explanation of the cause of the relationship between dependent variables such as yield and independent variables such as yield components. This study aims to determine several traits that can be used to make selections indirectly and directly to increase the yield of upland rice. This research has been carried out on agricultural land in the village of Tamarenja (Kalama), Sindue District, Donggala Regency. Starting from August to December 2019, using a Randomized Block Design (RBD) with seven genotypic cultivars and three groups of cultivars so that there were twenty one experimental plots. The results showed that the number of leaves, leaf length, number of tillers, number of productive tillers, flowering age, thickness of seeds and weight of 1000 grains had a positive and significant correlation to yield. Flowering age, 1000 grain weight, and number of tillers are some of the characteristics that have a high direct effect and significant correlation to upland rice yield.

Abstract

Increasing the number of inhabitants in Indonesia led to the limited agricultural land so that the paddy fields become narrow and limited, the limited paddy fields caused agricultural production in Indonesia to decline while demand increased. Tidal land cultivation is still rarely performed due to the high salt content (NaCl). The purpose of this study is to obtain upland rice cultivars that are tolerant of salinity stress. The study was conducted in November to December 2019 at the Seed and Science Technology Laboratory of the Faculty of Agriculture, University of Tadulako. The study was arranged using a completely randomized design (CRD) of two factors where the first factor consisted of two levels, namely Logi, and Tako, while the second factor consisted of five levels, namely 0.5%, 0.6%, 0.7%, 0.8%, and 0.9% were repeated three times. The results showed that tako cultivars have a good tolerance to salinity stress compared to logi cultivars, tako cultivars have the fastest germination time and germination capacity above 80%. At a concentration of 0.5% can not inhibit the growth of upland rice, but at a concentration of 0.9% can inhibit the growth of upland rice.

Abstract

This study aimed to determine the effect of different seed sizes and planting media on the growth of clove seedlings. This research was conducted in the experimental station of the Faculty of Agriculture, Tadulako University from January to July 2019. This research was in the form of field trials and was designed with a split plot design with the main plot being the type of planting media consisting of 1) sawdust, 2) compost and 3) cocopeat. The subplots consist of large, medium, and small seeds. There were nine treatment combinations and each combination was repeated three times so that in total there were 27 experimental units. The results showed that there was an interaction effects between seed size and type of planting media. The best seed size treatment is found in large seed sizes. Sawdust media produces plant height, number of leaves aged 2,4,6 MST, stem diameter and wet weight of plants aged 8 MST higher than other planting media.

Abstract

The expansion of rice planting areas in the future needs to be carried out outside Java, as fertile land in Java is shrinking due to land conversion. Most of the land outside Java consists of marginal land, such as areas with high salinity. High salinity levels in the soil cause a decrease in plant germination, growth, and production. The research aimed to examine the tolerance of each cultivar at each level of NaCl concentration, to obtain cultivars that were tolerant to salinity stress, and to determine the NaCl concentration that suppresses the germination of local upland rice. The research was structured using a completely randomized design with a two-factor factorial pattern. The first was the six upland rice cultivars, namely, dongan, jahara, Pulu Konta, pomegranate, kalendeng, and pulu tau leru, while the second factor was the NaCl concentration consisting of 0%, 0.2%, 0.4%, 0.6%, 0.8% and 1%. Thus, there were 36 experimental units, which were repeated three times so that there were 108 experimental units. The results of the research showed that there was no interaction with all germination variables; cultivar and concentration had a significant effect on maximum growth potential, germination capacity, germination time, plumule length, radicle length and dry weight of sprouts, while cultivar affected the wet weight of sprouts. Kalendeng has a high salinity tolerance. The use of NaCl concentrations of up to 0.6% has been able to reduce the germination capacity of local upland rice. The conclusion of the research showe that there was no interaction between all germination variables.

Abstract

The objective of this research was to find the optimal combination of soil loosening timing and liquid organic fertilizer dosage to enhance the yield of Lembah Palu shallot variety. The study was conducted in Kayumalue Ngapa village, Central Sulawesi, from November 2019 to January 2020. A two-factorial randomized block design was employed. The first factor was the soil loosening time: loosening at 1, 3, 5 weeks after planting and loosening at 2, 4, 6 weeks after planting. The second factor was the application of liquid organic fertilizer: 150 ml/ha, 300 ml/ha, and 450 ml/ha. The findings revealed that the interaction between soil loosening time and liquid organic fertilizer dosage significantly affected the plant height parameter. Particularly, loosening the soil at 1, 3, 5 weeks after planting along with a 150 ml/ha dose of liquid organic fertilizer produced higher plant height. Moreover, the soil loosening time of 1, 3, 5 weeks after planting resulted in improved plant height, tuber diameter, and fresh tuber weight. Similarly, the liquid organic fertilizer of 150 ml/ha demonstrated positive effects on plant height.

Abstract

This study examined the effects of bamboo shoot extract and storage duration on the viability of shallot seeds of Palu Valley variety. The experimental design was a factorial randomized block with two factors: bamboo shoot extract concentrations (10, 20, and 30 ml/liter of water) and seed storage durations (30, 40, 50, and 60 days). This resulted in twelve treatments, each replicated three times. Data were analyzed using analysis of variance, followed by an Honest Significant Difference test at the 5% level to compare treatment means. Results showed that bamboo shoot extract at 20 ml and 30 ml significantly increased the number of leaves in the first week and plant height in the first and second weeks. A 60-day storage duration positively affected germination, leaf number, plant height, and root volume.

Abstract

Seaweed is a fishery commodity that has high economic value because it contains carrageenan which can be used in the pharmaceutical, food, cosmetics, and industrial fields. Central Sulawesi Province is the second largest seaweed producer in Indonesia after South Sulawesi Province. The genetic diversity of seaweed in Central Sulawesi has not been studied much. Therefore, molecular-based characterization activities to identify the genetic variation of seaweed in the province are urgently needed. The purpose of the study was to optimize PCR techniques based on targeted start codon markers (SCoT markers) to identify the genetic diversity of seaweed accessions from Central Sulawesi Province. The results showed that the parameters for seaweed PCR amplification have been optimized. Of the fifteen SCoT primers, 10 of them can work to amplify seaweed DNA collected from several regions in Central Sulawesi. This is shown by the production of PCR result amplicons. PCR results also indicate polymorphism from the seaweed samples tested, although some seaweed samples have not been successfully amplified. The parameters of the PCR technique still need to be improved, especially the concentration of DNA prints, so that valid PCR results will be obtained for use in identifying seaweed genetic diversity.
