Abstract

ABSTRAK. Pertumbuhan dan kualitas Aglaonema dipengaruhi oleh beberapa faktor di antaranya media tanam dan aktivitas hormonal. Penggunaan media tanaman alternatif selain pakis dan hormon seperti sitokinin penting untuk diketahui. Tujuan penelitian ialah untuk mengetahui pengaruh kombinasi tiga komposisi media tumbuh sebagai media alternatif serta pemberian sitokinin terhadap pertumbuhan dan kualitas  Aglaonema Fit Langsit. Penelitian dilakukan di Rumah Kaca Fakultas Pertanian Universitas Padjadjaran mulai Bulan Mei sampai dengan Agustus 2008. Komposisi media tanam yang digunakan yaitu arang sekam, cocopeat, dan zeolit dengan perbandingan (2:2:1), (3:2:1), (4:2:1), dan sebagai kontrol digunakan pakis, humus, pasir malang, dan cocopeat (2:1:1:1) yang dikombinasikan dengan pemberian sitokinin dengan konsentrasi 0, 50, dan 100 µl/l yang disemprotkan pada daun setiap 2 minggu sekali. Hasil percobaan menunjukkan bahwa kombinasi arang sekam, cocopeat, dan zeolit (3:2:1) disertai sitokinin 50 µl/l memberikan pengaruh yang lebih baik dibandingkan perlakuan lainnya dalam memperpanjang dan memperlebar ukuran daun, sehingga dapat meningkatkan kualitas tanaman Aglaonema menjadi lebih rimbun dan kompak karena semakin meningkatnya ukuran daun.ABSTRACT. The growth and quality of Aglaonema is affected by several factors such as growing media and hormones. The using of alternative growing media and hormones such as cytokinin are urgently needed to be identified. The aim of experiment was to find out the effect of the combination of three growing media composition as an alternative growing medium with cytokinin on the growth and quality of Aglaonema Fit Langsit. This experiment was conducted in Greenhouse at the Faculty of Agriculture, Padjadjaran University from May to August 2008. The compositions of growing media consisted of the mix of carbonated rice hulls, cocopeat, and zeolite at three combinations (2:2:1, 3:2:1, and 4:2:1) and a control consisted of the mix of fern, humus, malang sands, and cocopeat (2:1:1:1). They were combined with cytokinins of 0, 50, and 100 µl/l. The results showed that the composition of carbonated rice hulls, cocopeat, and zeolite (3:2:1) combined with 50 µl/l cytokinin gave a better result than other treatments in increasing leaves length and leaves width, so it increases the quality of Aglaonema become more dense and compact due to by increasing the leaf size.

Abstract

Post-harvest loss is an obstacle for tomato farmers that cause tomato farming being unprofitable. Utilization of mutant genes prolong tomato fruit shelf life through crossing is one of the best solutions to improve the fruit quality. The objective of this study was to determine the success of cross-pollination between commercial tomato varieties and shelf life of mutant tomatoes. Two mutant tomato lines Sletr1-1 and Sletr1-2, and one wild type strain were crossed with four commercial varieties (Intan, Mutiara, Ratna, and Mirah ). The results showed that the success rate of crossing ranged from 77.50% to 100%, with an average of 95.76%. The average maximum growth potential of seeds produced was 89.17% with the lowest value of 75% and the highest 98.5%. The most top success of crosses was built by a combination of crossing Ratna x Sletr1-2, and the combination of Mirah x Sletr1-1 showed the lowest percentage of the crossing. The Intan showed the highest success as female parent of crossing, while Mirah was the lowest production of the crossing. The plant height of all hybrid was smaller compared to the commercial varieties.

Abstract

Penurunan kualitas selama penyimpanan merupakan permasalahan yang seringkali ditemui pada pascapanen buah jambu kristal. Oleh karena itu, diperlukan penanganan yang tepat. Penanganan pascapanen yang umum digunakan adalah dengan penggunaan kemasan. Tujuan penelitian ini adalah untuk mengkaji hubungan berbagai kemasan dan masa simpan terhadap nilai kekerasan, total padatan terlarut (TPT) dan kadar air buah jambu kristal. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) pola faktorial dengan 4 ulangan. Faktor pertama adalah pengemasan (tanpa kemasan (kontrol), plastik wrapping, koran) dan faktor kedua adalah masa simpan (0, 4 dan 8 hari). Data yang diperoleh kemudian diolah dengan analisis sidik ragam (ANOVA), selanjutnya diuji Duncan dengan taraf 5%. Hasil yang didapatkan menunjukkan tidak terjadi interaksi antara jenis kemasan dan masa simpan terhadap nilai kekerasan, TPT dan kadar air buah jambu kristal. Namun, jenis kemasan koran menampilkan nilai kekerasan buah jambu kristal yang lebih rendah daripada perlakuan tanpa kemasan (kontrol) dan plastik wrapping, sedangkan masa simpan 0 hari memperlihatkan nilai kekerasan buah jambu kristal yang lebih tinggi dibandingkan dengan masa simpan 4 dan 8 hari. Nilai kekerasan buah mengalami penurunan setelah 4 hari penyimpanan. Penggunaan kemasan berupa plastik wrapping dan koran tidak lebih baik dibandingkan dengan tanpa pengemasan (kontrol) terhadap nilai kekerasan pada buah jambu kristal.ABSTRACT Losses during storage is often encountered in postharvest of crystal guava fruit. Therefore, proper handling is needed. Packaging is the most commonly performed postharvest handling. The purpose of this study was to examine the relationship of various packagings and storage periods to the values of firmness, total soluble solids (TSS) and water content of crystal guava fruit. This study used factorial arrangement of completely randomized design (CRD) with four replications. The first factor was packagings (without packaging (control), wrap plastic and paper) and the second factor was storage periods (0, 4, and 8 days) in room temperature (±250C). The data obtained then processed using analysis of variance (ANOVA), then duncan test was carried out with a level of 5%. The results showed no interaction between the types of packaging and storage periods to the values of firmness, TSS and water content of crystal guava fruit. However, the paper packaging presented the firmness value which was lower than without packaging (control) and wrap plastic, whereas the 0 day storage showed the firmness value fruit which was higher than the 4 and 8 days storage. Value of fruit firmness decreased after 4 days of storage. The use of packaging in the form of wrap plastic and paper were not better than without packaging (control) to the firmness of the crystal guava fruit.

Abstract

Semakin tingginya permintaan bahan baku kentang untuk french fries khususnya, perlu diimbangi oleh peningkatan suplai kentang dengan kualitas yang memenuhi persyaratan skala industri. Aplikasi dosis pupuk NPK yang tepat serta umur panen yang sesuai merupakan strategi dalam meningkatkan kualitas kentang. Penelitian bertujuan untuk menentukan  dosis pupuk NPK dan umur panen yang dapat menghasilkan ubi kentang dengan kualitas olah terbaik. Percobaan dilakukan dengan menggunakan Rancangan Acak Kelompok pola faktorial dengan dua faktor dan tiga ulangan. Faktor pertama, yaitu dosis pupuk NPK (50, 100, 150% dosis rekomendasi) dan umur panen (90, 105, 120 HST). Hasil penelitian memperlihatkan bahwa perlakuan dosis pupuk NPK dan umur panen tidak memperlihatkan interaksi. Kadar air dan gula reduksi tidak dipengaruhi oleh perlakuan dosis pupuk dan umur panen. Perlakuan dosis pupuk 150 kg N ha-1, 225 kg P ha-1, dan 225 kg K ha-1 menunjukkan kadar pati ubi tertinggi dengan tingkat kerenyahan french fries paling disukai. Perlakuan umur panen 120 hari setelah tanam (HST) memberikan kadar pati dan tingkat kesukaan warna dan kerenyahan french fries paling tinggi. AbstractThe potato demand is increasing, as a raw material for french fries in particular, yet followed by potato improvement to fulfil the industry requirement. The application of NPK fertilizer in appropriate dosage and harvesting time is the essential strategy to increase potato quality. This study aimed to determine the proper dosage of NPK and harvesting time to obtain the best characteristics of potato tuber for processing. The experimental design used a factorial randomized block design consisting of two factors; NPK fertilizer dosages (50, 100, 150% recommended dosage) and harvesting time (90, 105, 120 DAP). NPK fertilizer dosage had no interaction with harvesting time. Tuber water content and reducing sugars were not affected by NPK fertilizer dosage and harvesting time. The treatment of 150 kg N ha-1, 225 kg P ha-1 and 225 kg K ha-1 had the best tuber starch content, and after processing to be french fries, it showed the most favored crispness. Meanwhile, harvesting time at 120 DAP resulted in the highest tuber starch content, french fries colour and crispness score. Giving a certain dose of NPK fertilizer will affect the quality of potato tubers. The right harvesting time affects on the potato tuber quality.

Abstract

Rendahnya produksi kentang di Indonesia disebabkan oleh ketersediaan benih kentang bermutu hasil dari metode konvensional yang kurang memadai. Metode kultur jaringan mampu menghasilkan bibit bermutu dan bebas virus, dalam jumlah banyak serta waktu yang singkat. Penelitian ini bertujuan untuk mendapatkan konsentrasi 6-benzylaminopurine (BAP) dan Naphthalene Acetic Acid (NAA) terbaik pada berbagai eksplan untuk pertumbuhan tunas meriklon kentang (Solanum tuberosum L.) Kultivar Atlantik secara in vitro. Percobaan dilaksanakan pada November 2016 sampai Februari 2017 di Laboratorium Kultur Jaringan Teknologi Benih Fakultas Pertanian Universitas Padjadjaran. Rancangan Acak Lengkap (RAL) digunakan sebagai rancangan percobaan yang terdiri dari dua unit percobaan terpisah yaitu unit eksplan meristem interkalar dan meristem apikal, masing-masing delapan perlakuan dan empat ulangan. Media Murashige and Skoog (MS) digunakan sebagai media dasar dengan penambahan BAP (0,0 mg L-1; 1 mg L-1; 1,5 mg L-1; 2 mg L-1) dan NAA (0,0 mg L-1; 0,5 mg L-1). Hasil menunjukkan konsentrasi BAP 1 mg L-1 merupakan perlakuan yang paling baik dalam menghasilkan jumlah tunas, cabang, daun dan buku pada eksplan meristem interkalar. Pada eksplan meristem apikal, penambahan BAP dan NAA belum mampu meningkatkan pertumbuhan eksplan. Meristem interkalar yang diberi auksin dan sitokinin berpotensi menjadi bahan alternatif eksplan meriklon kentang selain meristem apikal. The production of potato in Indonesia is low due to the limited number of high quality potato seeds produced from the conventional methods. The tissue culture methods can be used to produce high quality and virus-free seeds in more reliable number in a short time. Randomized Completely Design (RCD) was used as the experimental design consisted of two separate trial units namely intercalary meristem and apical meristem explants units with eight treatments and four replications of each. Murashige and Skoog (MS) was used as the base media added with BAP i.e. 0.0 mg L-1; 1 mg L-1; 1.5 mg L-1; 2 mg L-1 and NAA 0.0 mg L-1 and 0.5 mg L-1. Results showed that concentration of BAP 1 mg L-1 generated the best results on the number of shoots, branches, leaves and nodes on meristem intercalary explant. Meanwhile on apical meristem explant, the addition of BAP and NAA had not yet improved the explant growth. Intercalary meristem added with auxin and cytokinin is potential as alternative of mericlone explants material of potato besides the apical meristem.

Abstract

This study aimed for knowing and obtaining the best concentration of kinetin and NAA interaction effects in influencing the shoot induction, knowing how the plant growth regulators in induction mediastill affect the shoot additionin the MS0media and also knowing the largest number of roots in rooting media for shallot by in vitro. The experiment was conducted at Laboratory of Tissue Culture Seed Technology, Faculty of Agriculture, Padjadjaran University, during January 2011 until May 2011. This experiment divided in 3 stages, namely shoot induction stage, shoot subculture to MS0 media stage and shoot subculture to rooting media stage. Experimental method used in the shoot induction stage was factorial Completely Randomized Design with three replications. The first factor was the kinetin with four levels,0, 1, 2, and 3 mg L-1. The second factor was the NAA with three levels, as 0, 0.01, and 0.1 mg L-1. Basic media used for each treatment was MS. The experiment result showed there was an interaction between kinetin and NAA on shoot induction stagewith the plantlet height, leaf number, and shoot addition. The best result for leaf number was gained from interaction with 2 mg L-1 kinetin without NAA,while the treatment of 2 mg L-1 kinetin with 0.01 mg L-1 NAA gave a better interaction for theshoot addition variable.

Abstract

This study was aimed at determining the concentration of several types of cytokinins and auxin for the induction of turmeric shoots in vitro. The research was conducted at the Tissue Culture Seed Technology Laboratory, Faculty of Agriculture, Padjadjaran University, Jatinangor. The study was conducted from October 2017 to February 2018. The source of planting material is in the form of shoots from the turmeric rhizome. The source of explants or planting material came from the field collected at the Tissue Culture Seed Technology Laboratory, Faculty of Agriculture, Padjadjaran University. Explants were taken from rhizome buds with a size of 0.6-2.0 cm. The experiment used a Completely Randomized Design which was analyzed using the Student’s T-test method. The number of experimental and control groups in this study were seven groups. Variation in treatment with different BAP, thidiazuron, zeatin, and NAA concentrations in each group. The results show that Thidiazuron 1 mgL-1 + NAA 1 mgL-1 gives better results on the percentage of live explants and number of shoots on turmeric plants (Curcuma domestica Val.) Clones 41 at the age of 14 weeks after planting.RESPONS EKSPLAN KUNYIT PADA SITOKININ DAN AUKSIN DALAM MEDIA MURASHIGE DAN SKOOGPenelitian ini bertujuan untuk mendapatkan salah satu konsentrasi dari beberapa jenis sitokinin dan auksin untuk induksi tunas kunyit secara in vitro. Penelitian dilakukan di Laboratorium Kultur Jaringan Teknologi Benih, Fakultas Pertanian, Universitas Padjadjaran, Jatinangor. Waktu pelaksanaan penelitian ini dimulai pada awal bulan Oktober 2017 sampai bulan Februari 2018. Sumber bahan tanam berupa tunas dari rimpang tanaman kunyit. Sumber eksplan atau bahan tanam berasal dari lapangan yang dikoleksi di Laboratorium Kultur Jaringan Teknologi Benih, Fakultas Pertanian, Universitas Padjadjaran. Eksplan diambil dari mata tunas rimpang dengan ukuran 0,6-2,0 cm. Percobaan menggunakan Rancangan Acak Lengkap yang dianalisis menggunakan metode Student’s T-test. Jumlah kelompok eksperimen dan kontrol dalam penelitian ini adalah tujuh kelompok. Variasi perlakuan dengan penambahan konsentrasi BAP, thidiazuron, zeatin, dan NAA yang berbeda pada setiap kelompok. Hasil penelitian ini menunjukkan bahwa Thidiazuron 1 mgL-1 + NAA 1 mgL-1 memberikan hasil yang lebih baik pada persentase eksplan hidup dan jumlah tunas pada tanaman kunyit (Curcuma domestica Val.) klon 41 pada umur 14 MST (Minggu Setelah Tanam).

Abstract

This study was aimed at obtaining the type and concentration of cytokines as well as the optimal concentration of auxin for the multiplication of turmeric shoots in vitro. The trial was conducted in September 2017 until February 2018 at the Tissue Culture Seed Technology Laboratory of the Faculty of Agriculture, Padjadjaran University. The materials used in this study were Murashige and skoog modified multiplication medium, jelly, sterile distilled water, HgCl2, 70% alcohol, clorox, tween 80 and fungicide and bactericidal. Growth Regulating Substances (GRS) used are BAP, TDZ, Zeatin and NAA. The explant source used was derived from the shoot of turmeric clones from Bogor. The experimental design in this study was Completely Randomized Design (CRD) with seven treatments and four replications. The planting medium used was Murashige Skoog Modification (MS) with the addition of Benzyl Amino Purine (BAP) 9 mg L-1, Thidiazuron (TDZ) 1 mg L-1, Zeatin 0.1 mg L-1 and NAA (0.01 mg L-1, 1 mg L-1). The results show that the combination of BAP 9 mg L-1 and NAA 0.01 mg L-1 produced the highest shoot induction at 12 weeks after planting. Giving zeatin 0.1 mg L-1 and NAA 1 mg L-1 produces the highest shoot length at 12 weeks after planting.PENGUJIAN MULTIPLIKASI EKSPLAN KUNYIT DENGAN PENAMBAHAN AUKSIN DAN SITOKININ PADA MODIFIKASI MEDIAPenelitian ini bertujuan untuk mendapatkan jenis dan konsentrasi sitokinin serta konsentrasi auksin yang optimal untuk multiplikasi tunas kunyit secara in vitro. Percobaan dilaksanakan pada bulan September 2017 sampai Februari 2018 di laboratorium Kultur Jaringan Teknologi Benih Fakultas Pertanian Universitas Padjadjaran. Bahan yang digunakan dalam penelitian ini adalah murashige dan skoog modified multiplication medium, agar-agar, aquades steril, HgCl2, alkohol 70%, clorox, tween 80, fungisida, dan bakterisida. Zat Pengatur Tumbuh (ZPT) yang digunakan yaitu BAP, TDZ, Zeatin, dan NAA. Sumber eksplan yang digunakan yaitu berasal dari tunas kunyit klon asal Bogor. Rancangan percobaan pada penelitian ini yaitu Rancangan Acak Lengkap (RAL) dengan tujuh perlakuan dan empat ulangan. Media tanam yang digunakan yaitu Murashige Skoog Modifikasi (MS) dengan penambahan Benzyl Amino Purine (BAP) 9 mg L-1, Thidiazuron (TDZ) 1 mg L-1, Zeatin 0,1 mg L-1, dan NAA (0,01 mg L-1, 1 mg L-1). Hasil percobaan menunjukkan bahwa kombinasi BAP 9 mg L-1 dan NAA 0,01 mg L-1 menghasilkan induksi tunas tertinggi pada 12 MST. Pemberian zeatin 0,1 mg L-1 dan NAA 1 mg L-1 menghasilkan panjang tunas tertinggi pada 12 MST.

Abstract

Perkembangan teknologi budidaya tanaman saat ini sudah berkembang dengan cepat. Salah satu teknologi yang sedang dikembangkan adalah teknologi budidaya tanaman tanpa tanah atau dengan menggunakan system hidroponik. Banyak jenis hidroponik yang salah satunya adalah metode Nutrient Film Technique (NFT). Tujuan dilakukanya kegiatan ini adalah untuk mengenalkan teknologi terbarukan budidaya tanaman tomat ke masyarakat, sehingga pengetahuan masyarakat akan meningkat. Kegiatan ini dilakukan di Desa Cileunyi Kulon, Kecamatan Cileunyi, Kabupaten Bandung, dengan metode survey dan sosialisasi pengenalan teknologi budidaya tomat melalui sistem NFT kepada masyarakat.Program ini berhasil menginformasikan kepada masyarakat desa mengenai teknik budidaya tanaman tomat menggunakan teknologi NFT dengan hasil yang memuaskan. Respon peserta penyuluhan yang antusian dan merasa tertarik dengan teknologi tersebut mendorong peserta untuk bersedia untuk melakukan budidaya tanaman tomat dengan teknologi NFT serta bekerja sama untuk dapat menghasilkan produk tomat dengan teknologi tersebut secara berkelanjutan.

The quality component measurement of horticultural products becomes more important to achieve better quality products also to obtain uniform product. Recently, measurement by non-destructive methods turns out to be more needed, because consumers become more selective and demand producers to develop a quick, effective and accurate quality assessment system. Near Infrared Spectroscopy (NIRS) can measure plant contents quickly, relatively economical in a bigger scale, and importantly non-destructive. The measurement of quality components of green bitter gourd, such as firmness, water content, total soluble solid, and color, are important but have not been widely conducted. This research was conducted from June to August 2013 at Post Harvest Laboratory, Faculty of Agriculture, Padjadjaran University, Sumedang, Indonesia with PLS regression modeling method. The results show R-value and standard error of 0.91 and 0.47 for water content, 0.93 and 1.03 for firmness, and 0.95 and 0.49 for a* value of fruit color.
