Abstract

The usage of doxorubicin (DOX) as an anticancer drug in cancer patient may cause several side effects. One of that is cardiotoxicity by inducing the expression of nitric oxide synthase which may release nitric oxide (NO) resulting reactive oxygen species (ROS) in the cardiac. DOX needs to be combined with antioxidant since it could supressed ROS in the cardiac and reduce cardiomyopathy. Kelor (Moringa oleifera) is known as the source of antioxidant. This study aim to observe the treatment effects of ethanolic extract of kelor (EEK) on histopathology profile and concentration of NO in rats cardiac. The result from the hematoxylin-eosin staining showed that EEK improved the histopathology profile of rats’ cardiac. Compared with the DOX-only treatment, the structure of cardiac muscle cells treated by ethanolic extract of kelor is more well-arranged and the cells’ nucleus still visible. Concentration of NO was measured by cardiac puncture method. The result showed that the concentration of NO was decrease in line with increasing dose levels of EEK in combination with DOX. But at rats only given with EEK, the concentration of NO is quite high. In conclusion, EEK could be a cochemotherapy agent by reducing the cardiotoxicity effect of DOX.Keywords : doxorubicin, Moringa oleifera, nitric oxyde, histopathology

Abstract

Cyclophosphamide (CPA) is an effective chemotherapeutic agent, but has side effect, causing DNA damage (genotoxic). Papaya leaf (Carica papaya L.) is known has flavonoid compound, quercetin. Quercetin is known has DNA protecting effect (antigenotoxic effect) by metabolism modulation. Thus, the aim of this research is to investigate the antigenotoxic effect of ethanolic extract of papaya (Carica papaya L.) leaf (EEPL) on CPA induced mice. The antigenotoxic effect was evaluated by mammalian in vivo micronucleus test. EEPL was orally administered as single treatment at dose 1000 mg/kgBW and in combination with CPA 50 mg/kgBW at dose 250 mg/kgBW; 500 mg/kgBW; and 1000 mg/kgBW. Molecular docking using PLANTS on CYP 3A4 was performed to explore the antigenotoxic effect mechanism. The three different combination dose of EEPL with CPA significantly (P<0.05) decreased the amount of micronucleated polychromatic erytrhocyte (MNPCE)/1000 polychromatic erythrocyte (PCE) and also increased % PCE/(PCE+normochromatic erythrocyte (NCE)), compared with single dose of CPA. Nevertheless, the antigenotoxic effect wasn’t significant compared with each combination dose. The docking score result showed quercetin (-82,41) has more potent interaction to CYP 3A4 than cyclophosphamide (-70,16) and both of them has similar active site at amino acid residue Ile 369 and Thr 309. The results obtained indicated that EEPL at dose 250 mg/KgBB is the optimal dose as antigenotoxic agent by interaction between quercetin with CYP 3A4 based on molecular docking.Keywords: antigenotoxic, Carica papaya L., MNPCE, in vivo

Abstract

Background: Hylocereus polyrhizus has activities as antimicrobial agent, anti-hypercholesterolemia, anti-diabetic (diabetes mellitus), cardiovascular risk reduction, health supplement, and melanoma cell suppression. The extracts from the peels of H. polyrhizus were able to increase phagocytic ability, cell numbers and leukocytes and to influence relative spleen weights in the formation of body immune system in male rats. The fruit peels contained phenolics, flavonoids, carotenoids, and anthocyanins. This study investigated the active compounds of H. polyrhizus peels, which are able to increase immune system of human body.Materials and method: In vitro assay was applied to examine the active compounds, identified as lupeol, obtained from isolated extract of red dragon fruit for their lymphocyte proliferation and nitric oxide (NO)-producing activities. Lymphocyte proliferation assay was performed with 3-4.5-dimethylthiazol-2-yl)-2.5-diphenyltetrazolium bromide (MTT) method. The cell control was lymphocyte cell suspension in RPMI medium added with phytohaemaglutinine (PHA). The NO measurement was conducted with nitric solvent and Greiss reagent.Results: The ANOVA analysis of the average optical density (OD) of lymphocyte proliferation showed that the addition of isolated lupeol at the concentrations of 6.25, 12.5, 25, 50 and 100 µg/mL were able to improve lymphocyte proliferation and activate the NO production in the rats with treatment of positive control. Conclusion: Isolated lupeol at concentrations of 6.25, 12.5, 25, 50 and 100 µg/mL revealed significant difference with medium control and cell control. It was able to increase effects on lymphocyte proliferation and NO production. Therefore, the lupeol which was isolated might have high potential to be an immunostimulant.Keywords: Hylocereus polyrhizus, lupeol, lymphocyte proliferation, nitric oxide production

Abstract

Sterculia quadrifida, R.Br. is one of the plants that grows on the mainland of Timor, East Nusa Tenggra. Sterculia quadrifida, R.Br. commonly known as Peanut tree in English and faloak in Indonesian has various pharmacological activities and has been widely used in traditional medicine. This review article aimed to provide a comprehensive overview of the potential of the faloak plant as an herbal medicine by looking at various aspects such as ethnobotany, phytochemicals, pharmacology and toxicity. Literature searches are carried out in scientific databases that are accepted worldwide such as ScienceDirect, Scopus, PubMed, Springer Link, Wiley Online Library and advanced searches on Google scholars, books, abstracts, theses, scientific reports, and several non-impact and non-indexed journals with the search keywords "Sterculia quadrifida" or "Peanut tree" or "Red fruit kurajong" or "Faloak". From the various literature that has been collected, faloak has been shown to have pharmacological activities such as anticancer, antioxidant, antifungal, immunomodulatory, antiviral, antibacterial, antidiabetic, and antipyretic. The existence of this activity cannot be separated from the chemical compounds contained therein such as flavonoids, alkaloids, terpenoids, phenols, tannins and saponins. Faloak also has low toxicity with LD50> 5000 mg/kg body weight rats.

Abstract

Meniran (Phyllanthus niruri L), adalah salah satu tanaman obat yang telah banyak digunakan dalam sediaan obat tradisional dengan indikasi meningkatkan daya tahan tubuh. Diduga terdapat hubungan yang kuat antara aktivitas antioksidan dan respon imun, terutama respon imun humoral yang diperantarai oleh aktivitas makrofag. Pembuatan ekstrak kering sebagai bahan baku sediaan obat tradisional berpotensi menimbulkan adanya perubahan aktivitas ekstrak dikarenakan adanya pemanasan pada prosesnya. Untuk itu, pada penelitian ini diuji aktivitas penangkapan radikal DPPH. Hasil penelitian menunjukkan bahwa Ekstrak kering meniran dengan kadar flavonoid total sebesar 0,79 ± 0,06 % EK, memiliki aktivitas penangkapan radikal DPPH dengan IC50 sebesar 74,28 µg/mL. Proses pengeringan dan penambahan bahan pengisi pada ekstrak dapat berpengaruh pada aktivitas penangkapan radikal DPPH.

Abstract

Penelitian ini bertujuan mengetahui peran zink pada kesembuhan luka dalam kondisi berat badan berlebih. Dalam penelitian digunakan 20 ekor tikus Sprague Dawley jantan berumur tiga bulan (150-200 g). Tikus dibagi secara acak menjadi empat kelompok (A, B, C, dan D), masing-masing kelompok terdiri atas lima ekor. Kelompok A dan B diberi pakan normal, sedangkan kelompok C dan D diberi pakan lemak tinggi. Setelah dua bulan perlakuan pakan, seluruh tikus dioperasi untuk membuat luka irisan pada kulit, kemudian luka ditutup dengan jahitan. Luka jahitan pada kelompok A dan C diberi aplikasi topikal unguenta vaseline album (tanpa zink), sedangkan luka jahitan pada kelompok B dan D diberi aplikasi topikal unguenta zink 10% dengan bahan dasar vaselin album. Lima hari setelah operasi, seluruh tikus diambil darahnya untuk dianalisis terhadap jumlah leukosit total kemudian diikuti pengambilan jaringan kulit untuk pemeriksaan histopatologis dan penghitungan jumlah leukosit di jaringan. Hasil penelitian menunjukkan adanya interaksi yang signifikan antara pakan dan aplikasi topikal terhadap jumlah leukosittotal dalam darah (P0,05). Jumlah leukosit di jaringan mengalami penurunan setelah aplikasi topikal zink, meskipun hasilnya tidak signifikan (P0,05). Analisis histopatologis menunjukkan epidermis sudah tertutup pada seluruh tikus perlakuan, namun demikian sebagian tikus masih menunjukkan dermis yang terbuka, yaitu kelompok A sebanyak 60%, kelompok B sebanyak 60%, kelompok C sebanyak 60%, dan kelompok D sebanyak 40%. Dapat disimpulkan bahwa proses kesembuhan luka setelah pemberian topikal zink pada tikus yang diberi pakan lemak tinggi menunjukkan hasil yang baik, yaitu ditandai dengan penutupan lapisan epidermis dan dermis, serta menurunnya jumlah leukosit di jaringan.

Abstract

Background: Neutropenia can increase morbidity in cancer patients because it can cause an increased risk of infection, delay or discontinue chemotherapy, and decrease the dose of cytotoxic agents which affect the patient's response and clinical outcome. The administration of G-CSF can increase ANC levels in neutropenic patients. The effectiveness of G-CSF was assessed by increasing ANC levels compared to before the administration of G-CSF.Objective: This study aims to determine the effect of G-CSF administration on ANC levels in cancer patients with neutropeniaMethods: This research was conducted at Dr. Kariadi General Hospital Semarang in October-December 2022 period with retrospective cohort method. The sample was cancer patients who experienced neutropenia and were given G-CSF.Results: Administration of G-CSF significantly increased ANC values (p-value 0.00 <0.05), increased by 921.07 ± 647.20 in the administration of filgrastim and 933.13 ± 558.89 in the administration of lenograstim and shortened the duration of ANC recovery (p-value 0.00 <0.05), recovery was 1.5 days shorter with filgrastim and 1.9 days with lenograstim.Conclusion: G-CSF Filgrastim and Lenograstim administration can increase ANC levels and shorten the duration of neutropenia treatment

Abstract

Anemia merupakan kejadian efek samping obat akibat pemberian kemoterapi pada pasien kanker Limfoma Non-Hodgkin (LNH). CHOP dan RCHOP merupakan kemoterapi lini pertama yang diberikan pada pasien kanker LNH. Anemia dapat digunakan sebagai salah satu prediktor kematian pada pasien kanker LNH. Perbandingan efek kemoterapi lini pertama berupa anemia belum pernah dilakukan, sehingga tujuan penelitian ini membandingkan kemoterapi CHOP dan RCHOP terhadap kejadian anemia pada pasien. Desain penelitian adalah Kohort retrospektif pada pasien kanker LNH yang dirawat di RSUP Dr. Kariadi Semarang selama periode Januari 2017-April 2022. Subjek penelitian adalah pasien kanker LNH yang memenuhi kriteria inklusi dan dibagi menjadi dua kelompok yaitu CHOP dan RCHOP. Anemia didefinisikan sebagai penurunan nilai Hemoglobin <14 g/dL pada pria dan <12g/dL pada wanita. Analisis statistik yang digunakan dalam penelitian adalah analisis non parametrik berupa Chi-Square. Pasien CHOP mempunyai risiko 0,943 kali mengalami kejadian anemia dibandingkan dengan pasien RCHOP (p-value=0,132, RR 0,943, IK 95%=0,879-1,012). Kesimpulannya, tidak ada perbedaan kemoterapi CHOP dan RCHOP terhadap kejadian anemia pada pasien kanker LNH.

Abstract

Curcumin, a diarylheptanoid, is the active compound isolated from the rhizomes of Curcuma species. Curcumin can modulate mechanisms in inflammatory-related immunomodulatory. The two methoxy groups in aromatic rings in the structure of curcumin play a role in the anti-inflammatory. Macrophages are crucial cellular components of innate immunity. For this reason, this review aims to assess the role of curcumin and its derivatives in the innate immune response of macrophage cells. Curcumin as anti-inflammatory releases inflammation mediators such as cytokines, nitric oxide, reactive oxygen species, and others. Curcumin regulates innate immune via NLRP3 inflammasomes, nuclear factor-kappa B, mitogen-activated protein kinase, and Janus kinase1/2‐signal transducer and activator of transcription protein1 signaling pathway. However, curcumin has limited clinical application, especially solubility and bioavailability. Therefore, a structure and formula modification of curcumin was developed. This review also discusses no evidence of curcumin's mutagenicity, genotoxicity, and toxicity. Thus, the brief information provided in this review investigates the role of curcumin in macrophage cells.

Abstract

The main objective of this article is to explore the available classified information on Zingiber aromaticum Val. Rhizome and its active compound, Zerumbone. Throughout history, this plant has been utilized to treat various ailments such as abdominal disorders, cough, anemia, malaria, jaundice, arthritis, and infections. Recently, Zerumbone, a sesquiterpene compound found in the rhizomes, has gained attention for its biological activity. This article reviews the current research on the pharmacological activities of Z. aromaticum Val. Rhizome, with a specific focus on its active component, Zerumbone.This review found that the antioxidant and antiinflammatory effects of Zerumbone play a role in the pharmacological effects exhibited by Z. aromaticum, such as antidiabetic, antiparasitic, anti-hyperlipidemia, antianxiety, and antiaging effects. Additionally, the correlation between the biological activity of Zerumbone and the pharmacological activity of Z. aromaticum extract is discussed. This review provides a valuable reference for developing and further researching Zingiber aromaticum Val. and Zerumbone in medical treatments.
