Abstract

Aucubin is an iridoid glycoside known to have various pharmacological activities. Current research data shows that the pharmacological activities are very diverse and complex, so bibliometric analysis is used to facilitate understanding and provide references for further research directions. Publications related to the pharmacological activities of aucubin were obtained through the Web of Science Core Collection (WoSCC) database, which were then analyzed using Vosviewer 1.6.20 software. A total of 152 publications were obtained from the period 2013-2023, consisting of 142 articles and ten review articles. The articles came from 45 countries, 204 institutions, 862 authors, and 42 journals. Based on keyword analysis, the most frequently appearing pharmacological activities were "antioxidant" and "anti-inflammatory" that can be the primary development directions in the future.

Abstract

Class 1 and 2 histone deacetylase inhibitors (HDACI) have been reported as novel therapeutic approaches to treat neurodegenerative disorders, depression, anxiety, and cognitive deficits. HDACI ameliorated deficits in cognition and stress-related behaviors in a wide range of neurologic and psychiatric disorders. Preclinically, behavioral bioassay can be used to predict the influence of new compounds for treatment of these illnesses. Curcumin and its new analogues PGV-0 and PGV-1 have been reported to inhibit HDAC2. However, reports regarding the effect of curcumin and its analogues on memory and cognitive function, anxiety, and social interaction behavior are as yet to be examined. Mice were divided into control and treated groups. Brain disorder was induced by oral administration of 10% ethanol in sodium-CMC for 7 days. Curcumin, PGV-0, PGV-1, and sodium butyrate (as positive control) were then given orally once a day for 21 days. The behavior tests of social interaction, open field, radial 8-arm-maze, and passive avoidance were performed on day 29. To increase dissolution and bioavailability of the compounds, they were formulated in a self-nano emulsifying drug delivery system (SNEDDS). Brains were isolated and analyzed using PCR to investigate the expression of genes related to neurobehavioral disorders hdac2, trkB, and bdnf. In different doses, curcumin, PGV-0, and PGV-1 increased social interaction capability, declined anxiety level, and improved long-term memory and cognitive function. The mechanism proposed is: HDACI curcumin and its analogues (PGV-0 and PGV-1) that keep the histone protein in acetylation state increase bdnf expression. The increased trkB expression is increasing the activation of the bdnf gene because trkB is the primary receptor of bdnf that supports the survival of existing neurons and encourages the growth and differentiation of new neurons and synapses. Thus, those mechanisms could improve long-term memory and cognitive function, increase social interaction, and reduce anxiety in ethanol-induced mice with brain disorders.

Abstract

The prevalence rate of mortality in colorectal cancer is comparatively elevated. A standard or effective treatment has not been developed to resolve this issue. Utilizing Muntingia calabura, which is known to contain rutin with anti-cancer properties, is one of the strategies that can be implemented. The present study focuses on the potential therapeutic effects of M. calabura as an anti-cancer agent in colorectal cancer. Specifically, this study investigates the anti-cancer properties of rutin against colorectal cancer. The results of this study showed that the ethanolic extraction of M. calabura contained approximately 0.727 mg/mL of rutin. Additionally, the cytotoxicity assessment revealed that the rutin fraction was moderately cytotoxic, with a value of 70.808 μg/mL. The results of molecular docking experiments demonstrated a high degree of accuracy in predicting the interaction between rutin and β-catenin. Furthermore, the quantitative reverse transcription polymerase chain reaction (qRT-PCR) assay revealed a significant reduction in the expression of the β-catenin gene within the cells exposed to the extract. In conclusion, M. calabura extract exhibits anti-cancer properties by influencing the WNT signaling pathway in CT26 cells.

Abstract

BACKGROUND: The 4T1 cells, a triple-negative breast cancer (TNBC) cell line, exhibit high malignancy and metastatic potential. As a primary treatment for TNBC, doxorubicin has limitations, including drug resistance mechanisms and severe side effects such as cardiotoxicity. Pentagamavunone-1 (PGV-1) exhibits antiproliferative and antimetastatic effects, induces prometaphase arrest, triggers cell senescence, and enhances reactive oxygen species (ROS) modulation, which may help overcome doxorubicin resistance. The selective cytotoxicity of PGV-1 against cancer cells suggests that it has a role in reducing systemic toxicity. Therefore, in this study, the anticancer effects of doxorubicin combined with PGV-1 was investigated.METHODS: Monolayer/2D and spheroid/3D models of 4T1 cells were used to assess the effects of PGV-1, doxorubicin, and their combination. MTT assay was used to evaluate the cytotoxicity, colony formation assay was used to measure persistent antiproliferative effects, and spheroid volume analysis was performed to assessed tumor growth inhibition. Senescence-associated beta-galactosidase (SA-β-gal) assay determined cellular senescence.RESULTS: The combination of PGV-1 and doxorubicin significantly enhanced cytotoxicity, with IC50 values of 0.57 µM and 4.88 µM, respectively (p=0.000). A strong synergistic effect was observed, leading to persistent suppression of cancer cell proliferation and an 80% reduction in colony formation (p=0.007). In the 3D spheroid model, combination treatment significantly reduced spheroid volume (p=0.002) more effectively than monotherapy, indicating superior growth inhibition and cytotoxicity. It also increased SA-β-gal, the senescence marker (p=0.010).CONCLUSION: The combination of PGV-1 and doxorubicin demonstrated potent anticancer effects in 4T1 monolayers and spheroid models by enhancing cytotoxicity and inducing cellular senescence. Abstract

The P. major plant has long been utilized in traditional herbal medicine and is widely accessible in Indonesia and globally. Extensive research has demonstrated its efficacy in wound healing through both clinical and preclinical studies. Gel formulations are recognized for establishing a conducive moist environment essential for wound healing, thus rendering them suitable for topical wound treatment. This study assesses the stability of HPMC gel base, carbomer 940, and their combination as carriers for the ethanol extract of P. major leaves. Stability testing involved a freeze-thaw cycle test at temperatures of 4°C and 40°C for three cycles. The findings indicate no significant alteration in the organoleptic properties and pH of the gel formulations before and after stability testing. However, t-test analysis reveals a noteworthy decrease in viscosity for carbomer and its combination, albeit not for HPMC alone. In conclusion, HPMC exhibits superior stability as a gel base compared to Carbomer 940 and their combination.

Abstract

Produk jamur tiram merupakan komoditas pertanian yang memiliki potensi ekonomi tinggi karena kandungan gizi yang baik dan ketersediannya melimpah, pengolahan jamur tiram segar sering menghadapi tantangan seperti umur simpan yang pendek dan fluktuasi harga di pasaran, sehingga perlu adanya suatu pengolahan untuk memberikan nilai tambah. Tujuan penelitian ini yaitu untuk menganalisis nilai tambah dan keuntungan dalam pengolahan jamur tiram (Pleurotus ostreatus) menjadi kaldu jamur di PT. Mitra Jamur Indonesia serta mengidentifikasi strategi pemasaran yang relevan. Metode penelitian yang digunakan meliputi metode kuantitatif dan kualitatif. Analisis nilai tambah dilakukan dengan menggunakan metode Hayami, analisis SWOT digunakan untuk merumuskan strategi pemasaran berdasarkan faktor internal dan eksternal. Hasil penelitian menunjukkan (1) Pengolahan jamur tiram menjadi kaldu jamur menghasilkan nilai tambah yang positif sebesar Rp. 31.685/kg, dengan rasio nilai tambah sebesar 52%, (2) Keuntungan pada produksi kaldu jamur sebesar Rp. 22.256.506/kg, (3) PT. Mitra Jamur Indonesia berada pada posisi kuadran I Strategi Agresif, strategi yang diusulkan mencakup optimalisasi media sosial, inovasi kemasan, serta kolaborasi dengan distributor untuk memperluas jangkauan pasar.

Abstract

Kegiatan pengabdian kepada masyarakat ini bertujuan untuk mengoptimalkan pemanfaatan lahan sempit di kawasan perkotaan, khususnya di Kelurahan Karangrejo, Sumbersari, Jember, melalui penerapan teknologi vertiminaponik. Vertiminaponik adalah sistem tumpang sari yang menggabungkan budidaya tanaman hortikultura dengan perikanan dalam satu unit sistem vertikal. Sistem ini memanfaatkan ruang vertikal untuk menanam sayuran, sementara kolam ikan menyediakan nutrisi alami bagi tanaman. Kegiatan ini melibatkan masyarakat setempat dalam proses pelatihan dan implementasi sistem vertiminaponik, termasuk pemeliharaan tanaman dan ikan serta pemasaran hasil pertanian dan perikanan. Program ini juga bertujuan untuk meningkatkan ketahanan pangan keluarga, mengurangi ketergantungan pada produk luar daerah, serta memberikan peluang ekonomi baru melalui penjualan hasil pertanian dan ikan lele. Hasil kegiatan menunjukkan bahwa penerapan sistem vertiminaponik berhasil meningkatkan produksi pertanian meski dengan keterbatasan lahan. Masyarakat telah menunjukkan kemampuan untuk merawat sistem secara mandiri, dan beberapa di antaranya telah mendapatkan pendapatan tambahan. Keberhasilan ini menunjukkan potensi teknologi vertiminaponik sebagai solusi untuk meningkatkan kemandirian pangan di daerah perkotaan dengan lahan terbatas. Selain itu, program ini mendukung tujuan pemerintah dalam mengembangkan pertanian organik dan berkelanjutan melalui pemanfaatan lahan yang lebih efisien.

Abstract

Triple-negative breast cancer (TNBC) is one of subtypes of cancer with the most associated death. With more than 20% of breast cancer patients having this subtype, an effective and safe chemotherapy is needed to ensure complete pathomorphological response. Currently, therapies for TNBC patients are divided into systemic therapy regimens for operable or non-operable diseases, with non-operable diseases mostly treated with targeted therapies such as pembrolizumab in tandem with other chemotherapy agents such as paclitaxel, gemcitabine or carboplatin. TNBC patients whom tumor is operable underwent additional step such as preoperative systemic therapy, which offers several advantages than chemotherapy alone. Neoadjuvant chemotherapy (NACT), which consist of mainly drugs from therapeutic classes such as anthracycline and taxanes, is used with limited success (23%) and decreased recurrence rates. Optimal method for treating TNBC is still lacking, therefore the need for further research regarding chemotherapy for TNBC with higher success rate and better prognosis for the patients is paramount.

Abstract

Amylum is widely used as an excipient in pharmaceutical formulations, one of which is inert and can be mixed with any drug without causing a chemical reaction. The aim of this research is to know the preparation method of cassava amylum (Manihot esculenta, Crantz) native and pregelatinized which is used as binder to dissolution rate of paracetamol tablet with granulation method. The native and pregelatinized amylum produced from cassava is carried out by a preliminary test to determine the nature of physical characteristics and to know the ability as a binder of the tablet. The paracetamol tablets produced from the two methods were tested dissolution using medium buffer phosphate at 37 ± 0,5 ° C (pH 5.8, 50 rpm). The results showed that cassava starch preparation method had an effect on dissolution rate of tablet. Pregelatinized amylum as binder on paracetamol tablet showed Q value (93,17 ± 0,01)% while native amylum showed value of Q (81,76 ± 0,20)%. The use of pregelatinized cassava starch as a binder on paracetamol tablet has better characteristic properties compared with native starch.

Abstract

Pembangunan perumahan di perkotaan yang sangat pesat, cenderung tidak mempertimbangkan faktor konservasi lingkungan dengan memberikan sumbangsih ruang terbuka hijau yang terbatas. Pekarangan merupakan lahan yang potensial untuk dikembangkan menjadi lahan pertanian produktif terutama untuk pemenuhan kebutuhan pangan yang bergizi bagi si pemilik. Keterbatasan lahan bukanlah hal yang menjadi hambatan untuk mengaktualisasi potensi nilai ekonomi. Lahan di lingkungan perumahan yang cenderung terbatas dapat dioptimalkan untuk ditanami tanaman yang memiliki nilai ekonomi tinggi seperti tanaman pangan, tanaman hias, tanaman obat dan tanaman penyuplai oksigen dalam jumlah besar. Salah satu model optimalisasi lahan terbatas untuk mendukung pertanian sederhana di lingkungan perumahan yaitu model smart vertical farming. Metode pelaksanaan kegiatan antara lain dengan memberikan pelatihan teknik vertical farming, praktik budidaya vertical farming, pembuatan model smart vertical farming dan cara melakukan perawatan. Paparan pelatihan dan praktik disampaikan oleh narasumber dari bidang agribisnis, sedangkan pemodelan smart vertical farming disampaikan oleh narasumber dari bidang teknik informatika. Opini yang berkembang di masyarakat bahwa menerapkan konsep vertical farming adalah sesuatu yang mahal dan rumit, nyatanya tidak semuanya benar. Dengan bermodalkan bibit tanaman, paralon atau botol bekas, maka masyarakat dapat membuat model pertanian vertikal. Model tersebut akan menjadi lebih efisien apabila menerapkan konsep pertanian cerdas berbasis sensor dalam pemantauan dan pengelolaan tanaman. Hasil dari kegiatan baik pelatihan, praktik, maupun pembuatan model smart vertical farming sangat diterima dengan baik oleh masyarakat.
