Abstract

Lung cancer has the highest mortality rate of all cancers. In its advanced stages, this disease is associated with poor survival rates and quality of life. The most common subtype of lung cancer is adenocarcinoma which is frequently associated with mutations in the epidermal growth factor receptor (EGFR), a protein crucialfor cell proliferation. The presence of EGFR mutations plays a key role in targeted therapy using tyrosine kinase inhibitors (TKIs). Afatinib is a second-generation TKI that offers several advantages over the first-generation. Afatinib is effective against complex and uncommon EGFR mutations, making it a commonly usedoption for patients with lung adenocarcinoma. However, afatinib is also associated with a higher incidence of adverse drug reactions (ADRs) compared to other TKIs, which may impact therapeutic outcomes. This study aimed to assess the quality of life (QoL) of advanced lung adenocarcinoma patients with EGFR mutations andthe prevalence of ADRs associated with afatinib treatment at Dr. Sardjito General Hospital in Yogyakarta. The study employed an observational cross-sectional design. Prospective data collection was conducted for two months, from January to February 2025 at the Oncology Department following ethical approval. A totalof 15 patients were enrolled in this study. The most commonly identified ADRs were acne (45%), diarrhea (20%), paronychia (20%), and oral mucositis (15%). The mean QoL scores was 81.38 ± 10.01 for the functional scale, 15.93 ± 8.48 for the symptom scale, and 72.78 ± 19.53 for global health status.

Abstract

The Sesquiterpene lactones, enhydrin and uvedalin, are the chemotype and subtype compounds of the yacon leaf. They are known to have anti-diabetic activity, but their mechanisms are not clear. One of the mechanisms of anti-diabetic agents is to inhibit the activity of the dipeptidyl peptidase 4 enzyme (DPP-4). The research aimed to determine the inhibition of the extract, enhydrin, and uvedalin isolated from the yacon leaf against the DPP-4 enzyme activity. Yakon leaves were extracted using 70% ethanol by the maceration method. Enhydrin and uvedalin were obtained from previous research. The inhibitory activity of the DPP-4 enzyme was then determined by the fluorescence assay method using a multi-well plate reader. Sitagliptin was used as standard inhibitor of DPP-4 enzyme. The ethanol extract of yacon leaves inhibits the DPP-4 enzyme with an IC50 of 856.9 ppm. The percentage inhibition of the DPP-4 enzyme by the enhydrin and uvedalin at a concentration of 250 ppm was 2.37 and 35.16%, respectively. The inhibitory potency of the pure isolated compounds was not substantially greater than that of the crude extract itself. This led us to the conclusion that the overall contribution of enhydrin and uvedalin to the extract's DPP-4 inhibitory activity is actually modest, suggesting the presence of other active compounds within the extract. However, it may provide a new approach to the treatment of type 2 diabetes.

Abstract

Atopic Dermatitis (AD) is a chronic inflammatory skin condition marked by impaired skin barrier function, immune system irregularities, and microbial imbalance. This research examines the efficacy of Lactiplantibacillus plantarum, a probiotic recognized for its established safety and advantageous characteristics, in the management of atopic dermatitis (AD). The pathophysiology of AD encompasses genetic, environmental, and immunological components, with dysbiosis of skin and gut microbiota being essential. L. plantarum has shown beneficial effects in controlling gut and skin microbiota, improving skin barrier integrity, and modulating immunological responses via processes including short-chain fatty acid (SCFA) synthesis, cytokine regulation, and competitive exclusion of pathogens. Clinical investigations demonstrate that L. plantarum supplementation can diminish the severity of atopic dermatitis, as assessed by the SCORAD index, and enhance skin barrier integrity. The effectiveness of probiotic therapy may be affected by genetic, nutritional, and environmental variables. This review emphasizes the promise of L. plantarum as a therapeutic agent for AD, while also discussing the limitations and future possibilities for probiotic-based therapies in managing this intricate condition.

Abstract

Penelitian ini bertujuan mengetahui peran zink pada kesembuhan luka dalam kondisi berat badan berlebih. Dalam penelitian digunakan 20 ekor tikus Sprague Dawley jantan berumur tiga bulan (150-200 g). Tikus dibagi secara acak menjadi empat kelompok (A, B, C, dan D), masing-masing kelompok terdiri atas lima ekor. Kelompok A dan B diberi pakan normal, sedangkan kelompok C dan D diberi pakan lemak tinggi. Setelah dua bulan perlakuan pakan, seluruh tikus dioperasi untuk membuat luka irisan pada kulit, kemudian luka ditutup dengan jahitan. Luka jahitan pada kelompok A dan C diberi aplikasi topikal unguenta vaseline album (tanpa zink), sedangkan luka jahitan pada kelompok B dan D diberi aplikasi topikal unguenta zink 10% dengan bahan dasar vaselin album. Lima hari setelah operasi, seluruh tikus diambil darahnya untuk dianalisis terhadap jumlah leukosit total kemudian diikuti pengambilan jaringan kulit untuk pemeriksaan histopatologis dan penghitungan jumlah leukosit di jaringan. Hasil penelitian menunjukkan adanya interaksi yang signifikan antara pakan dan aplikasi topikal terhadap jumlah leukosittotal dalam darah (P0,05). Jumlah leukosit di jaringan mengalami penurunan setelah aplikasi topikal zink, meskipun hasilnya tidak signifikan (P0,05). Analisis histopatologis menunjukkan epidermis sudah tertutup pada seluruh tikus perlakuan, namun demikian sebagian tikus masih menunjukkan dermis yang terbuka, yaitu kelompok A sebanyak 60%, kelompok B sebanyak 60%, kelompok C sebanyak 60%, dan kelompok D sebanyak 40%. Dapat disimpulkan bahwa proses kesembuhan luka setelah pemberian topikal zink pada tikus yang diberi pakan lemak tinggi menunjukkan hasil yang baik, yaitu ditandai dengan penutupan lapisan epidermis dan dermis, serta menurunnya jumlah leukosit di jaringan.

Abstract

Muhammadiyah is a da'wah organization that devotes itself to teaching life based on tawhid and renewal, amar ma'ruf nahi mungkar. Muhammadiyah's education reform aims to unify religion and values and advance the education system. The Council of Education is working hard to improve Amal Usaha Muhammadiyah's (AUM) management in education, especially in the financial management of elementary, junior high, and high school. AUM financial management has improved, but not optimally because AUMDik in Banyuwangi Regency still has not operationalized its financial reporting through the Madinapay website. The results of the current situation analysis of LPPK East Java are (1) LPPK has made efforts to realize effective, efficient and accountable financial governance by creating a budget plan program for Muhammadiyah East Java schools and making financial reporting applications; (2) LPPK has not completed the application with manual guidance; (3) LPPK has carried out system socialization to Muhammadiyah Schools, but the results are not optimal because Muhammadiyah schools in East Java do not use the application until now. The results of service activities at AUMDik in Banyuwangi Regency, especially school treasurers, 1) partners have increased their ability to operate the Madinapay website, 2) some partners already have an integrated financial system but have not been integrated with stakeholders.

Abstract

Costus speciosus or Pacing has been investigated to have cytotoxic effect on several human cancer cells. Current was conducted to determine the cytotoxic activity of ethanol extract of the Pacing rhizome (EP) on SKOV-3 ovarian cancer cells, to investigate EP’s effect on BAX and p53 expression, and predict the inhibition activity on intrinsic pathway. EP were extracted using maceration method with 70% ethanol. The cytotoxic effect was performed using MTT Assay with various concentrations (375 μg/mL, 250 μg/mL, 62.5 μg/mL, 31.25 μg/mL, and 7.8125 μg/mL). The ability of EP to induce apoptosis in SKOV-3 cells was measured using flow cytometry. The BAX and p53 gene expressions on SKOV-3 cells were detected with RT-qPCR after treatment of EP with concentrations of ¼ IC50, ½ IC50, and IC50. The ability of diosgenin to interact with BAX and p53 can be seen from the interaction with the MCL-1 protein and was predicted with molecular docking. The results showed that IC50 of cytotoxic activity was 69.143 μg/mL. EP could induce apoptosis on SKOV-3 cells and can induce BAX and p53 protein expressions. The docking results show that diosgenin has the potential to act as an antagonist for the MCL-1 protein, it can increase apoptosis.Keywords: apoptotic, Costus speciosus extract, SKOV-3, p53, BAX.
