Abstract

The Sesquiterpene lactones, enhydrin and uvedalin, are the chemotype and subtype compounds of the yacon leaf. They are known to have anti-diabetic activity, but their mechanisms are not clear. One of the mechanisms of anti-diabetic agents is to inhibit the activity of the dipeptidyl peptidase 4 enzyme (DPP-4). The research aimed to determine the inhibition of the extract, enhydrin, and uvedalin isolated from the yacon leaf against the DPP-4 enzyme activity. Yakon leaves were extracted using 70% ethanol by the maceration method. Enhydrin and uvedalin were obtained from previous research. The inhibitory activity of the DPP-4 enzyme was then determined by the fluorescence assay method using a multi-well plate reader. Sitagliptin was used as standard inhibitor of DPP-4 enzyme. The ethanol extract of yacon leaves inhibits the DPP-4 enzyme with an IC50 of 856.9 ppm. The percentage inhibition of the DPP-4 enzyme by the enhydrin and uvedalin at a concentration of 250 ppm was 2.37 and 35.16%, respectively. The inhibitory potency of the pure isolated compounds was not substantially greater than that of the crude extract itself. This led us to the conclusion that the overall contribution of enhydrin and uvedalin to the extract's DPP-4 inhibitory activity is actually modest, suggesting the presence of other active compounds within the extract. However, it may provide a new approach to the treatment of type 2 diabetes.

Abstract

Atopic Dermatitis (AD) is a chronic inflammatory skin condition marked by impaired skin barrier function, immune system irregularities, and microbial imbalance. This research examines the efficacy of Lactiplantibacillus plantarum, a probiotic recognized for its established safety and advantageous characteristics, in the management of atopic dermatitis (AD). The pathophysiology of AD encompasses genetic, environmental, and immunological components, with dysbiosis of skin and gut microbiota being essential. L. plantarum has shown beneficial effects in controlling gut and skin microbiota, improving skin barrier integrity, and modulating immunological responses via processes including short-chain fatty acid (SCFA) synthesis, cytokine regulation, and competitive exclusion of pathogens. Clinical investigations demonstrate that L. plantarum supplementation can diminish the severity of atopic dermatitis, as assessed by the SCORAD index, and enhance skin barrier integrity. The effectiveness of probiotic therapy may be affected by genetic, nutritional, and environmental variables. This review emphasizes the promise of L. plantarum as a therapeutic agent for AD, while also discussing the limitations and future possibilities for probiotic-based therapies in managing this intricate condition.

Abstract

Penelitian ini bertujuan mengetahui peran zink pada kesembuhan luka dalam kondisi berat badan berlebih. Dalam penelitian digunakan 20 ekor tikus Sprague Dawley jantan berumur tiga bulan (150-200 g). Tikus dibagi secara acak menjadi empat kelompok (A, B, C, dan D), masing-masing kelompok terdiri atas lima ekor. Kelompok A dan B diberi pakan normal, sedangkan kelompok C dan D diberi pakan lemak tinggi. Setelah dua bulan perlakuan pakan, seluruh tikus dioperasi untuk membuat luka irisan pada kulit, kemudian luka ditutup dengan jahitan. Luka jahitan pada kelompok A dan C diberi aplikasi topikal unguenta vaseline album (tanpa zink), sedangkan luka jahitan pada kelompok B dan D diberi aplikasi topikal unguenta zink 10% dengan bahan dasar vaselin album. Lima hari setelah operasi, seluruh tikus diambil darahnya untuk dianalisis terhadap jumlah leukosit total kemudian diikuti pengambilan jaringan kulit untuk pemeriksaan histopatologis dan penghitungan jumlah leukosit di jaringan. Hasil penelitian menunjukkan adanya interaksi yang signifikan antara pakan dan aplikasi topikal terhadap jumlah leukosittotal dalam darah (P0,05). Jumlah leukosit di jaringan mengalami penurunan setelah aplikasi topikal zink, meskipun hasilnya tidak signifikan (P0,05). Analisis histopatologis menunjukkan epidermis sudah tertutup pada seluruh tikus perlakuan, namun demikian sebagian tikus masih menunjukkan dermis yang terbuka, yaitu kelompok A sebanyak 60%, kelompok B sebanyak 60%, kelompok C sebanyak 60%, dan kelompok D sebanyak 40%. Dapat disimpulkan bahwa proses kesembuhan luka setelah pemberian topikal zink pada tikus yang diberi pakan lemak tinggi menunjukkan hasil yang baik, yaitu ditandai dengan penutupan lapisan epidermis dan dermis, serta menurunnya jumlah leukosit di jaringan.

Abstract

Muhammadiyah is a da'wah organization that devotes itself to teaching life based on tawhid and renewal, amar ma'ruf nahi mungkar. Muhammadiyah's education reform aims to unify religion and values and advance the education system. The Council of Education is working hard to improve Amal Usaha Muhammadiyah's (AUM) management in education, especially in the financial management of elementary, junior high, and high school. AUM financial management has improved, but not optimally because AUMDik in Banyuwangi Regency still has not operationalized its financial reporting through the Madinapay website. The results of the current situation analysis of LPPK East Java are (1) LPPK has made efforts to realize effective, efficient and accountable financial governance by creating a budget plan program for Muhammadiyah East Java schools and making financial reporting applications; (2) LPPK has not completed the application with manual guidance; (3) LPPK has carried out system socialization to Muhammadiyah Schools, but the results are not optimal because Muhammadiyah schools in East Java do not use the application until now. The results of service activities at AUMDik in Banyuwangi Regency, especially school treasurers, 1) partners have increased their ability to operate the Madinapay website, 2) some partners already have an integrated financial system but have not been integrated with stakeholders.

Abstract

Costus speciosus or Pacing has been investigated to have cytotoxic effect on several human cancer cells. Current was conducted to determine the cytotoxic activity of ethanol extract of the Pacing rhizome (EP) on SKOV-3 ovarian cancer cells, to investigate EP’s effect on BAX and p53 expression, and predict the inhibition activity on intrinsic pathway. EP were extracted using maceration method with 70% ethanol. The cytotoxic effect was performed using MTT Assay with various concentrations (375 μg/mL, 250 μg/mL, 62.5 μg/mL, 31.25 μg/mL, and 7.8125 μg/mL). The ability of EP to induce apoptosis in SKOV-3 cells was measured using flow cytometry. The BAX and p53 gene expressions on SKOV-3 cells were detected with RT-qPCR after treatment of EP with concentrations of ¼ IC50, ½ IC50, and IC50. The ability of diosgenin to interact with BAX and p53 can be seen from the interaction with the MCL-1 protein and was predicted with molecular docking. The results showed that IC50 of cytotoxic activity was 69.143 μg/mL. EP could induce apoptosis on SKOV-3 cells and can induce BAX and p53 protein expressions. The docking results show that diosgenin has the potential to act as an antagonist for the MCL-1 protein, it can increase apoptosis.Keywords: apoptotic, Costus speciosus extract, SKOV-3, p53, BAX.

Abstract

Diabetes mellitus (DM) memiliki hubungan yang erat dengan perubahan pola makan dan gaya hidup yang tidak sehat. Jumlah penderita diabetes di Indonesia terus meningkat. Minat masyarakat dalam mengonsumsi tanaman obat semakin berkembang karena pasien lebih memilih produk alami yang dianggap lebih aman. Penelitian tentang tanaman obat sebagai agen antidiabetes juga menunjukkan tren yang meningkat. Studi ini bertujuan untuk memetakan penelitian yang menggunakan tanaman obat terkait diabetes di Indonesia. Data subjek penelitian diperoleh dari Scopus, dan perangkat lunak statistik RStudio dengan paket bibliometric digunakan untuk menganalisis dan memvisualisasikan data. Hasil analisis menunjukkan bahwa pertumbuhan publikasi di bidang ini mencapai 36,25% per tahun. Jumlah penulis bersama per dokumen adalah 4,93 dengan tingkat kolaborasi internasional sebesar 15,12%. Sebanyak 1.548 artikel dengan 4.877 penulis diterbitkan di 429 jurnal dan prosiding terpilih. Elya B (n = 35), Universitas Airlangga (n = 435), dan IOP Conference Series: Earth and Environmental Science (n = 92) merupakan penulis, afiliasi, dan penerbit paling produktif. Dengan jumlah sitasi per artikel sebesar 7,78 menunjukkan bahwa penelitian dari Indonesia berkontribusi pada penelitian lain. Penulis artikel di Indonesia sebagian besar (89,73%) berkolaborasi dengan peneliti sesama di Indonesia, sementara sisanya (10,27%) berkolaborasi dengan peneliti di luar negeri seperti Jepang, Malaysia, Thailand, China, dan Korea. Dapat disimpulkan bahwa kuantitas dan kualitas penelitian tentang tanaman obat untuk DM di Indonesia semakin meningkat dan berkontribusi pada penelitian lebih lanjut untuk pengelolaan diabetes. Kata Kunci: Bibliometric, Diabetes Mellitus, Tumbuhan Obat, Scopus, Rstudio Diabetes mellitus (DM) strongly associated with changes in unhealthy diet and lifestyles. The number of diabetic patients in Indonesia continues to increase. Public interest in consuming medicinal plants was developing because patients prefer on natural products and are considered safer. Research on medicinal plants as antidiabetic agents also shows an increasing trend. This study aims to map research using medicinal plants related to diabetes in Indonesia. Data on research subjects were retrieved from Scopus and the bibliometric package in RStudio statistical software was used to analyze and visualize the data. The results of the analysis showed that the growth of publications in this field was 36.25% per year. The number of co-authors per document was 4.93 with an international co-authorship rate of 15.12%. A total of 1548 articles with 4877 authors were published in 429 selected journals and proceedings. Elya B (n = 35), Universitas Airlangga (n = 435), the IOP Conference Series: Earth and Environmental Science (n = 92) was the most productive author, affiliation, and publisher. With the number of citations per article of 7.78, it shows that this research from Indonesia contributes to another research. The authors of the articles in Indonesia mostly (89.73%) collaborated with fellow researchers in Indonesia, while the rest (10.27%) collaborated with researchers abroad such as Japan, Malaysia, Thailand, China, and Korea. It can be concluded that the quantity and quality of research on medicinal plants for DM in Indonesia is increasing and contributing to further research for diabetes management.

Abstract

Mangga Arumanis merupakan tanaman yang banyak diminati di indonesia karena rasanya yang manis dan harga terjangkau. Penelitian ini bertujuan: (1) menganalisis tantangan risiko usahatani mangga di Kelurahan Mimbaan Kecamatan Panji Kabupaten Situbondo (2) menentukan keputusan yang tepat dalam menghadapi risiko di Kelurahan Mimbaan (3) menganalisis cara memitigasi risiko. Metode yang digunakan adalah metode kualitatif dengan studi kasus eksploratif. Sampel ditentukan berdasarkan jenis kelamin, umur, Pendidikan terakhir, pekerjaan sampingan, dan lama usahatani. Dengan sejumlah 13 petani mangga.  Metode analisis data menggunakan metode ERM dan SWOT. Hasil penelitian menunjukkan: (1) terdapat tantangan risiko seperti ketidakpastian cuaca, belum ada peran kelompok tani, dan peminat petani muda (2) berdasarkan matriks ERM terdapat 15 jenis risiko diantaranya, tiga jenis risiko dengan tingkat keparahan kecil, lima jenis risiko dengan tingkat keparahan menengah & besar, dan dua jenis risiko dengan tingkat keparahan sangat besar (3) berdasar analisis analisis diketahui bahwa usahatani mangga di Kelurahan Mimbaan berada pada kuadran I yang mengindikasikan bahwa stragetgi SO (Strenght, Opportunity) adalah strategi alternatif yang baik. Strategi tersebut mencakup pemanfaatan peluang pasar ekspor, pemasaran produk offline online, serta kerjasama dengan industri olahan mangga. Penelitian ini juga membeikan wawasan mengenai mitigasi risiko melalui diversifikasi usaha secara  vertikal dan manajemen yang lebih baik untuk meningkatkan daya saing petani mangga.

Abstract

Usahatani tembakau menjadi mata pencaharian banyak petani Curahsuri, namun usaha ini mempunyai banyak risiko. Penelitian ini bertujuan: (1) mengidentifikasi risiko paling dominan yang berpengaruh pada usahatani tembakau di Desa Curahsuri Kecamatan Jatibanteng (2) menyusun upaya penanggulangan risiko yang muncul dalam usahatani tembakau di Desa Curahsuri (3) mengetahui strategi yang diterapkan pada usahatani tembakau di Desa Curahsuri. Metode yang digunakan adalah metode deskriptif dan kualitatif. Sampel ditentukan sebanyak 30 petani tembakau. Metode analisis data menggunakan metode ERM dan SWOT. Hasil penelitian menunjukkan: (1) terdapat 3 macam jenis risiko pada posisi level risiko sangat tinggi, yaitu:. dengan kode A2 (faktor iklim/cuaca), C3 (keterbatasan pengetahuan teknologi), dan E3 (persaingan). (2) berdasarkan matriks risiko dengan kode A2 memerlukan penanggulangan dengan tindakan yang dapat diterapkan petani dengan menunda jadwal awal tanam, dan melakukkan konversi lahan. Kode C3 memerlukan penanggulangan dengan memberikan pelatihan penggunaaan teknologi untuk meningkatkan efisiensi tenaga kerja. Pada kode E2 memerlukan penanggulangan dengan peran pemerintah memberikan kebijakan yang kondusif terkait harga tembakau menjadi stabil dan petani tidak dirugikan. (3) berdasarkan analisis SWOT  diketahi bahwa usahatani tembakau di Desa Curahsuri berada pada kuadran I yang mengindikasikan bahwa strategi SO (Strenght, Opportunity) adalah strategi alternatif yang baik. Strategi tersebut Mejalin kemitraan dengan usahatani tembakau dapat memberikan kepastian pasar dan harga, memanfaatkan kualitas dan kuantitas produksi seperti memperbaiki budidaya dan penggunaan teknologi, dan mengikuti pelatihan dengan komuditas unggulan kelompok tani seperti pelatihan budidaya.

Abstract

Leptospirosis presents with nonspecific clinical features and requires time‐consuming laboratory tests for gold standard diagnosis. This study aims to design and characterize the recombinant LipL32 from synthetic gene and assess its performance as an antigen for detecting leptospirosis. The antigen was developed by cloning the LipL32 gene conserved portion of Leptospira interrogans serovar Icterohaemorrhagiae strain Langkawi. The immunoinformatic was used to characterize the developed rLipL32. Western blot results using anti‐histidine revealed a band of rLipL32 protein at ~40 kDa. Subsequently, it was used to examine the IgM antibody on human sera by using ELISA. The IgM‐LipL32 ELISA was evaluated using 67‐positive and 25‐negative sera and compared with a commercial ELISA. With a cut‐off value of 0.8, it showed 85.7% sensitivity, 83.3% specificity, a 48% positive prediction value (PPV), and 97% negative prediction value (NPV), indicating modest performance compared to existing commercial kits. The rLipL32 is a potential antigen for detecting IgM using ELISA; however, for use in low incidence areas, a confirmation test is crucial.
