Abstract

Kumis kucing (Orthosiphon stamineus) is a medicinal plant that can treat various diseases. Kumis kucing has been known to contain bioactive compounds, namely polyphenol, alkaloid, and terpenoid which have antioxidant, antibacterial, antiviral, anti-inflammatory, anti-allergic and anti-cancer activities. This study aims to determine the phytochemical content, antioxidant activity, and antibacterial activity of the cat's whiskers leaf extract. Kumis Kucing leaf extract was obtained by sonication-maceration method. Antioxidant activity test of cat's kumis leaf extract was tested using the CUPRAC and ABTS methods. The results showed that the kumis kucing’s leaf extract contains bioactive compounds of polyphenol, alkaloid, and terpenoid. The highest average antioxidant activity was produced by the purple phenotype cat whiskers leaf extract using the ABTS method, which was 168.68 µg TE/g dry weight. The highest average antibacterial activity in Escherichia coli  strain was obtained from leaf extract of purple phenotype cat whiskers (U2) with an inhibition zone diameter of 3.2 mm.

Abstract

Celosia cristata, an edible ornamental plant, is a potential floricultural commodity that needs further improvement to increase its agro-morphological characters and polyphenol content. Induced mutagenesis using ethyl methane sulphonate (EMS) is an effective tool to increase genetic diversity that has been applied in many plant species. This study aimed to assess the morphological diversity, polyphenol content, and antioxidant activities of C. cristata mutagenized by EMS in the M2 generation. A total of 230 M2 plants generated from the M1 generation were evaluated in this study and the polyphenols content and antioxidant activities analysis were conducted on fifteen selected M2 plants. Polyphenols content was analyzed using the Folin-Ciocalteu method and colorimetric method with slight modification, and the antioxidant activities investigated using 2,2’-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay and ferric reducing antioxidant power (FRAP) assay with minor changes. There are six subpopulations with the highest diversity of quantitative characters in the M2 population in quantitative characters, i.e. C2-17-1.0%, C2-1-0.7%, C2-20-2.0%, C2-25-0.7%, C2-1-0.9%, and C2-1-0.7%. Changes in the shape and color of leaves, stems, and flowers of C. cristata were also observed in the M2 population. C2.1, C2.6, and C2.12. 2.1, C2.6, and C2.12 are potential plants derived from EMS mutagenesis with the highest polyphenol content and antioxidant capacity in the M2 population. In conclusion, induced mutation using EMS can enhance the agro-morphological diversity, polyphenols content along with the antioxidant activities of C. cristata, and demonstrate the successful mutation breeding program.

Abstract

Abstract:  Curcuma aeruginosa Roxb. is one of the most widespread medicinal plants in Southeast Asia, one of which is Indonesia. The purpose of this study was to determine the phytochemical content, antibacterial activity against Escherichia coli, and the level of cytoxicity in the rhizome of C. aeruginosa from five different genotypes. The maceration technique was utilized to extract samples. Using qualitative techniques, the presence of secondary metabolites in the sample extract was assessed. While the extract's cytotoxicity was verified using the brine shrimp lethality test, its antibacterial activity against E. coli was determined using the disc diffusion method. The qualitative phytochemical analysis revealed that the five rhizome genotypes included triterpenoids but lacked phenols, quinones, and alkaloids. Increasing the concentration of extract samples increased the lethality against Artemia salina shrimp larvae, but resulted in an LC50 value high enough to render it cytotoxically ineffective. Compared to the positive control, the antibacterial activity test resulted in a very tiny diameter of the clear zone, indicating that the C. aeruginosa had limited antibacterial activity against E. coli.Abstrak: Temu hitam termasuk ke dalam salah satu tanaman obat yang tersebar luas di Asia Tenggara, salah satunya Indonesia. Tujuan dari penelitian ini adalah mengetahui kandungan fitokimia, aktivitas antibakteri terhadap bakteri Escherichia coli, dan tingkat toksisitas pada rimpang temu hitam (Curcuma aeruginosa Roxb.) dari lima genotipe yang berbeda. Ekstraksi sampel dilakukan dengan metode maserasi. Ekstrak sampel ditentukan adanya senyawa metabolit sekunder dengan metode kualitatif. Sementara sitotoksisitas ekstrak ditentukan dengan metode brine brine shrimp lethality test, sementara untuk antibakteri terhadap E. coli dilakukan dengan metode difusi cakram. Hasil uji fitokimia kualitatif menunjukkan kelima genotipe rimpang temu hitam mengandung triterpenoid dan tidak mengandung fenol, kuinon serta alkaloid. Peningkatan konsentrasi sampel temu hitam meningkatkan daya bunuh terhadap larva udang Artemia salina, namun menghasilkan nilai LC50 yang cukup tinggi sehingga memiliki sitotoksisitas yang rendah. Uji aktivitas antibakteri menghasilkan diameter zona bening yang sangat kecil dibandingkan kontrol positifnya, dengan demikian rimpang temu hitam memiliki aktivitas antibakteri pada E. coli yang rendah.

Abstract

Krokot (Portulaca grandiflora) merupakan tanaman hias yang belum banyak diketahui dan dimanfaatkan secara optimal oleh masyarakat. Tanaman ini juga bermanfaat sebagai obat dan kosmetik (bahan baku). Krokot (P. grandiflora) mengandung beragam metabolit sekunder, antara lain alkaloid, flavonoid, tanin, dan lainnya. P. grandiflora diketahui memiliki potensi sebagai antioksidan. Aktivitas antioksidan dari salah satu bagian untuk satu jenis tanaman dapat berbeda karena kandungan fitokimia dari setiap bagian tanaman juga berbeda. Review ini menjelaskan kandungan fitokimia dan kadar antioksidan dari tanaman P. grandiflora dari artikel ilmiah. Artikel yang dituju bersumber dari jurnal internasional, jurnal nasional terakreditasi sinta. Terdapat banyak kandungan fitokimia dari P. grandiflora dan berpotensi bekerja sebagai antioksidan. Berdasarkan artikel dan senyawa fitokimia yang didapat tanaman P. grandiflora berpotensi sebagai antioksidan.

Abstract

Temulawak merupakan tumbuhan dengan nilai ekspor tertinggi. Temulawak juga dapat tumbuh serta beradaptasi di berbagai habitat. Temulawak bermanfaat sebagai antiinflamasi dan ekstrak kurkuminoid temulawak berpotensi sebagai antikanker. Penelitian ini bertujuan untuk mengetahui efek toksisitas dan kandungan fitokimia dari tiga (3) varietas temulawak, yaitu Cursina 1, Cursina 2, dan Cursina 3. Metode yang digunakan untuk uji toksisitas secara kuantitatif yaitu dengan brine shrimp lethality test (BSLT). Senyawa yang toksik berdasarkan metode BSLT seringkali berkorelasi positif sebagai antikanker. Uji toksisitas dalam penelitian ini ditentukan oleh nilai LC50 atau mortalitas larva udang yang telah terpapar sampel selama 24 jam. Selain itu, dilakukan uji skrining fitokimia secara kualitatif. Skrining Fitokimia adalah cara mengidentifikasi bioaktif yang belum tampak dalam suatu uji yang dapat memisahkan bahan alam yang mengandung fitokimia. Skrining ini terdiri dari uji alkaloid, fenol, flavonoid, saponin, tannin, triterpenoid dan steroid. Ketiga Ekstrak (Cursina 1, Cursina 2, dan Cursina 3) menunjukkan aktivitas ketoksikan dalam uji toksisitas jika ekstrak dapat menyebabkan kematian 50% hewan uji pada konsentrasi <1000 ppm. Hasil analisis fitokimia temulawak positif mengandung metabolit sekunder seperti alkaloid, fenol, flavonoid, saponin, tanin, dan triterpenoid.  Hal ini ditunjukkan dengan perubahan warna pada larutan sampel temulawak setelah perlakuan. Berdasarkan uji ANOVA, hasil yang signifikan memiliki nilai p<0,05. Namun data yang diperoleh tidak signifikan karena menghasilkan nilai p>0,05. Suatu ekstrak dikatakan sangat toksik jika  LC50 ≤ 30 mg/L, dikatakan toksik jika nilai LC50 ≤ 1.000 mg/L, dan dikatakan tidak toksik jika nilai LC50 >1.000 mg/L. Dapat disimpulkan bahwa sampel Cursina 3 memiliki tingkat toksisitas yang lebih besar dibandingkan Cursina 1 dan Cursina 3.

Abstract

Previous studies have shown that bitter leaf (Vernonia amygdalina Del.) has the ability as an antioxidant and an α-glucosidase inhibitor. Still, the difference in antioxidant activity and α-glucosidase inhibition based on the leaves and stems parts have not yet been determined. The effect of flavonoid-enriched extract on antioxidant activity and inhibition of α-glucosidase has not yet been determined. This research aimed to assess the impact of flavonoid-enriched extract from the leaves and stems part of the bitter leaf. The leaves and stems part of the bitter leaf were extracted using Soxhlet apparatus with 80% methanol and then underwent successive fractionation with petroleum ether, chloroform, and ethyl acetate. The crude extract and the fraction were concentrated and followed by the determination of total flavonoid, total phenolic, antioxidant activity, α-glucosidase inhibition activity, and calculated the IC50 of α-glucosidase inhibition. This research showed that chloroform-ethyl acetate leaf fraction was the best fraction with the higher total flavonoid (24.091±0.972 mg QE/g DW), total phenolic (84.299±4.589 mg GAE/g DW), diphenylpicrylhydrazyl (DPPH) antioxidant activity (33.881 μM TE/g DW), ferric reducing antioxidant power (FRAP) antioxidant activity (312.022±1.745 μM TE/g DW) and α-glucosidase inhibition activity with an IC50 value 1.23 mg/mL.

Abstract

Mutation induction by colchicine is one method to improve plant genetic diversity (polyploidy). This study aims to 1) obtain growth regulators composition suitable in inducing callus, 2) obtain the LC50 value in callus of P. amboinicus, 3) to determine the effect of colchicine on callus growth, 4) to get shoots (putative mutant) varian, regenerating from indirect organogenesis (callus) in MV1. The study is conducted at the Laboratory of Agricultural Seed Management, Plantation Research, and Development Center from June 2018 to June 2019. The study uses a complete randomized design with four treatments and three replications. Explants use ware the nodes. Callus induction uses MS0 medium with 0.5 ppm 2.4-D, 2 ppm 2.4-D, 0.5 ppm 2.4-D + 1 ppm BAP, and 0.5 ppm 2.4-D + 0.5 ppm TDZ treatment. Mutation induction was performed using colchicine concentration at 0%, 0.02%, 0.04%, and 0.06% treatments. The regeneration media is an MS0 medium. The results show the best combination of plant growth regulators to induce the callus is 0.5 ppm 2.4-D. The LC50 colchicine concentration is at 0.0275%. However, this phenotype is reversible. An additional subculture period is required to move to normal morphology

Abstract

Javanese turmeric (Curcuma xanthorrhiza Roxb.) is important for study due to its increasing economic value in many aspects including marker-associated compounds for curcuminoids and xanthorrhizol. This systematic review aimed to summarize and find information about C. xanthorrhiza starting from its pharmacological activity and mechanism as an anticancer, anti-inflammatory, and antioxidant extraction process, and cultivation techniques. The data were obtained from relevant journals in national and international scientific databases PubMed, Scopus, Google Scholar, Web of Science, SINTA, ScienceDirect, and Wiley Online from 2000 to 2023. Results showed that from the perspective of anti-inflammatory pharmacological activities, xanthorrhizol inhibits pain response and inflammatory response. Its antioxidant activity showed a contribution to inhibiting oxidation which also successfully inhibits cancer cell proliferation. The extraction method of xanthorrhizol and curcuminoid compounds with Ultrasonic-Assisted Extraction (UAE) 20-2000 kHz performs better than other methods. C. xanthorrhiza cultivation techniques under shading produce better rhizomes. Based on research on cultivation techniques, extraction methods, and pharmacological activities, C. xanthorrhiza has an important role in the future.

Abstract

Cardamom (Amomum cardamomum) is a herbal plant with various phytochemical compounds such as phenols, starch, tannins, terpenoids, flavonoids, and antioxidants. Cardamom of the same type but originating from different regions can produce variations in the content of metabolites. This study aimed to select cardamom accession containing high starch, chlorophyll, and antioxidant capacity. The research was conducted at Biopharmaca Experimental Garden, Cikabayan IPB University from March to August 2022 using four cardamom accessions, i.e., Bogor Hijau, Bogor Merah, Ciamis and Sukabumi. Three leaves were taken for each accession, with three replications. The leaves taken were numbers 5, 6, and 7 counted from the tops of the cardamom plant. The methods used in this study were the phenol-sulfuric acid method, the DMSO method, the DPPH method, and the CUPRAC method. The results showed that the highest starch content was produced by the cardamom of Bogor Merah accession, with an average of 3.63 g (100 g)-1 FW. The highest total chlorophyll content was found in the cardamom of Bogor Hijau accession, with an average of 1.9 mg g-1 FW. At the same time, the highest antioxidant capacity using the DPPH method was produced by the cardamom of the Sukabumi accession with an average of 1.28 μmol TE g-1 FW. In comparison, the highest antioxidant capacity of the CUPRAC method was produced by the cardamom of the Bogor Hijau accession with an average of 5.71 µmol TE g-1 FW. Thus, it is recommended to use Bogor Merah dan Bogor Hijau accession for further evaluation. Keywords: accession, antioxidant activity, cardamom leaves, chlorophyll, starch

Abstract

Xylocarpus granatum is mangrove plant that traditionally used as face powder in Central Sulawesi, Indonesia which related to antioxidant, antiglycation and tyrosinase inhibition activities. This study aimed to evaluate the potency of X. granatum as a tyrosinase inhibitor, antiglycation, and antioxidant. The leaves, stem, stem bark, fruit flesh, fruit peel, and kernel of X. granatum were extracted using ethanol then their tyrosinase inhibition, antiglycation, and antioxidant were evaluated. Tyrosinase inhibition activity was evaluated using in vitro assay with L-tyrosine and L-DOPA as the substrate of monophenolase and diphenolase. Antiglycation activity was studied by measuring the excitation and emission fluorescence from glucose and fructose reaction with Bovine Serum Albumin. Antioxidant activity was measured using the DPPH radical scavenging assay. The result showed that the ethanolic extract of fruit flesh has higher potency as tyrosinase inhibitor (IC50 of 393.8 mg/L and IC50 of 448 mg/L, respectively for monophenolase and diphenolase). Antiglycation assay showed that the ethanolic extract of stem bark provides the strongest antiglycation activity with an IC50 of 118.1 mg/L. Meanwhile, fruit peel provides the strongest antioxidant activity with an IC50 of 5.5 mg/L. Fractionation of ethanolic extracts of each part of X. granatum tree yield fractions with lower bioactivity compared to the crude extract. Moreover, stem extract and fractions from two different locations (Tarakan and Kendari) tend to have different bioactivities strengths.  The stem part of X granatum could be developed as new raw material of cosmetic product in Indonesia, while ethanol as the solvent for extraction, and the different bioactivity of stem extract from different location can be the consideration for the industry to standardize the extract prior to production of final product.
