Abstract

Puji dan syukur kami panjatkan kepada Tuhan Yang Maha Esa atas rahmat dan hidayahNya, Jurnal Jamu Indonesia Volume 4 No 1 tahun 2019 dapat diterbitkan. Jurnal Jamu Indonesia merupakan media untuk diseminasi hasil penelitian yang berkaitan dengan jamu di Indonesia. Jamu telah dikenal lama penduduk Indonesia sebagai salah satu pengobatan. Berbagai tanaman di Indonesia sangat berpotensi sebagai jamu. Penelitian tanaman obat dari bagian daun, batang, dan rimpang tanaman telah banyak dilakukan dan terus berkembang. Naskah dalam edisi ini membahas khasiat antidiabetes dari ekstrak biji nangka dan jamu JAMSI, penggunaan amilum dari sukun sebagai bahan pembuatan tablet kunyah ekstrak daun jambu biji, dan khasiat antibakteri dari tanaman paku Stenochlaena palustris dan Pteridium caudatum. Selain aspek khasiat, pada edisi ini juga membahas penggunaan sistem informasi untuk melakukan manajemen pengetahuan berbasis ontologi terkait dengan tumbuhan obat dan senyawa yang dikandungnya. Redaksi mengucapkan terima kasih kepada seluruh penulis atas partisipasinya dalam berbagi hasil penelitiannya melalui publikasi di Jurnal Jamu Indonesia. Redaksi juga mengucapkan terima kasih kepada seluruh reviewer atas partisipasinya dalam meningkatkan kualitas tulisan naskah jurnal di edisi ini. Semua hasil karya tersebut akan diarsipkan sebagai dokumentasi berharga demi terjaganya informasi yang dapat menjadi sumber pengembangan ilmu pengetahuan di masa depan. Abstract

Penelitian ini bertujuan untuk membuktikan khasiat JAMSI, Jamu antidiabetes PT. Mahkotadewa Indonesia, secara preklinis pada tikus diabetes yang diinduksi dengan streptozotocin. Tikus jantan Sprague Dawley sebanyak 36 ekor dibagi secara acak menjadi 6 kelompok (masing-masing kelompok 6 tikus): kelompok normal, kelompok kontrol diabetes (diinduksi + air saline), kelompok kontrol positif (diinduksi + Glibenclamide 2.5 mg/kg BB), dan tiga kelompok perlakuan (diinduksi + JAMSI dengan dosis 0.27, 0.81, dan 1.62 ml/kg BB). Perlakuan diberikan selama 28 hari setelah diinduksi dengan streptozotocin. Pemberian JAMSI 1.62 ml/kg BB terbukti mempengaruhi penurunan kadar glukosa darah dan hemoglobin terglikosilasi (HbA1C) dibandingkan kelompok diabetes. Hasil ini mengindikasikan bahwa JAMSI berkhasiat sebagai antidiabetes.

D.C.) terhadap ketinggian tempat, khasiat antihiperglikemia daun kelor, khasiat antimikroba tanaman paku, dan khasiat jamu antidiabetes secara in vivo pada tikus. Redaksi mengucapkan terima kasih kepada seluruh penulis atas partisipasinya dalam berbagi hasil penelitiannya melalui publikasi di Jurnal Jamu Indonesia. Abstract

This study evaluated the free radical scavenging activity in ethanolic extracts from 20 accessions of Curcuma aeruginosa. The radical scavenging activity of the extract accessions was investigated with 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical. IC50 values for DPPH radical scavenging activity ranged from 89.81 to 505.65 µg mL-1. Based on IC50 values, twenty accessions of C. aeruginosa can be divided into three groups: strong (two accessions); moderate (seventeen accessions); and low (one accession) of DPPH scavenger. Sukoharjo (SH) and Muara Bungo (MB) showed promising accessions for antioxidant potential, thus these accessions important to selection for future breeding program in pharmaceutical products.

Abstract

Orthosiphon aristatus (called kumis kucing in Indonesia) is a valuable herb for diabetes mellitus treatment. In this study, LC-MS/MS and PCA analyses were used to investigate the metabolite profile, classify O. aristatus extracts, and assess the inhibitory activity of a-glucosidase and the probable bioactive compounds through in silico study. Results showed that the methanol and ethanol extracts of O. aristatus were active in α-glucosidase inhibitory activity. Both extracts contained 86 compounds as known from the LC-MS/MS analysis. PCA analysis identified 10 metabolites that correlated with α-glucosidase inhibitory activity. Results of in silico analysis obtained rosmarinic acid compound potentially act as anti-diabetic activity, which can be developed for further research.

Abstract

Glucose oxidase (GOD) is an oxidoreductase enzyme that catalyzes the oxidation of glucose to gluconolactone and hydrogen peroxide. Then, gluconolactone will be hydrolyzed to gluconic acid. The wide application of gluconic acid in various industries has increased production demand. However, glucose concentrations higher than 40% (w/w) inhibited the conversion of glucose to gluconic acid due to a decrease in the oxygen solubility concentration at pH 6, 30℃, and 1 bar pressure. Therefore, decreasing the value of Km is predicted to reduce saturation and enhance gluconic acid production. This study aimed to analyze the interaction between the IPBCC.08.610 GOD mutant with β-D-Glucose in improving gluconic acid production by decreasing the Km value. Mutations were performed in silico using Chimera and then docked using AutoDock Vina. The mutations resulted in distinct ligand poses in the binding pocket, different -OH conformations of the ligands, and changes in the T554M/D578P mutant’s hydrophobicity index (554 mutated from threonine to methionine, and 578 mutated from aspartate to proline), and decreased ΔG and Km values in the H559D mutant (559 mutated from histidine to aspartate), D578P and T554M/D578P. This decrease might strengthen the ligand-receptor interaction, increasing gluconic acid production. The H559D was the best mutant to increase production based on the ΔG, Km value, and stability due to the addition of hydrogen bonds.

Abstract

Abstract:  Plants produce a variety of secondary metabolites, one of which is essential oils that contain a variety of volatile compounds and are useful for humans. Cymbopogon nardus contains volatile compounds that can inhibit the proliferation of cancer cells. This research aims to explore the antiproliferation activity of C.nardus’ essential oils with different concentrations against breast cancer Michigan Cancer Foundation-7 (MCF-7) cells. Antiproliferation test was carried out with total cells method using trypan blue and cells were calculated using a microscope. Gas Chromatography-Mass Spectrometer (GC-MS) was performed to identify the volatile compounds. The results showed that the Inhibitory Concentration 50 (IC50) value was 359.6 ppm with an inhibition percent of 44.49% at 170 ppm. Meanwhile, inhibition percent against Vero normal cell was 29.04%, compared to Doxorubicin 35.23%. The dominant volatile compound in C. nardus’ essential oil were Geraniol and Citronellol.Abstrak Tanaman menghasilkan berbagai macam metabolit sekunder, salah satunya dalam bentuk minyak atsiri yang mengandung berbagai macam senyawa volatil yang berguna bagi manusia. Cymbopogon nardus mengandung senyawa volatil yang dapat menghambat proliferasi sel kanker. Penelitian ini bertujuan untuk mengetahui aktifitas antiproliferasi minyak atsiri C. nardus dengan beberapa konsentrasi terhadap sel kanker payudara Michigan State Foundation-7 (MCF-7). Uji antiproliferasi dilakukan dengan metode total sel menggunakan trypan blue dan sel dihitung menggunakan mikroskop. Gas Chromatography-Mass Spectrometer (GC-MS) dilakukan untuk mengidentifikasi senyawa volatil. Hasil menunjukkan bahwa konsentrasi penghambatan 50 (IC50) sebesar 359.6 ppm dengan persen penghambatan sebesar 44.49% pada konsentrasi 170 ppm. Sementara itu, persen ihibisi terhadap sel normal Vero sebesar 29.04%, dibandingkan dengan Doksorubisin yang sebesar 35.23%. Senyawa volatil dominan yang terdapat pada C. nardus adalah Geraniol dan Citronellol.

Abstract

Kumis kucing (Orthosiphon stamineus) is a medicinal plant that can treat various diseases. Kumis kucing has been known to contain bioactive compounds, namely polyphenol, alkaloid, and terpenoid which have antioxidant, antibacterial, antiviral, anti-inflammatory, anti-allergic and anti-cancer activities. This study aims to determine the phytochemical content, antioxidant activity, and antibacterial activity of the cat's whiskers leaf extract. Kumis Kucing leaf extract was obtained by sonication-maceration method. Antioxidant activity test of cat's kumis leaf extract was tested using the CUPRAC and ABTS methods. The results showed that the kumis kucing’s leaf extract contains bioactive compounds of polyphenol, alkaloid, and terpenoid. The highest average antioxidant activity was produced by the purple phenotype cat whiskers leaf extract using the ABTS method, which was 168.68 µg TE/g dry weight. The highest average antibacterial activity in Escherichia coli  strain was obtained from leaf extract of purple phenotype cat whiskers (U2) with an inhibition zone diameter of 3.2 mm.

Abstract

Celosia cristata, an edible ornamental plant, is a potential floricultural commodity that needs further improvement to increase its agro-morphological characters and polyphenol content. Induced mutagenesis using ethyl methane sulphonate (EMS) is an effective tool to increase genetic diversity that has been applied in many plant species. This study aimed to assess the morphological diversity, polyphenol content, and antioxidant activities of C. cristata mutagenized by EMS in the M2 generation. A total of 230 M2 plants generated from the M1 generation were evaluated in this study and the polyphenols content and antioxidant activities analysis were conducted on fifteen selected M2 plants. Polyphenols content was analyzed using the Folin-Ciocalteu method and colorimetric method with slight modification, and the antioxidant activities investigated using 2,2’-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay and ferric reducing antioxidant power (FRAP) assay with minor changes. There are six subpopulations with the highest diversity of quantitative characters in the M2 population in quantitative characters, i.e. C2-17-1.0%, C2-1-0.7%, C2-20-2.0%, C2-25-0.7%, C2-1-0.9%, and C2-1-0.7%. Changes in the shape and color of leaves, stems, and flowers of C. cristata were also observed in the M2 population. C2.1, C2.6, and C2.12. 2.1, C2.6, and C2.12 are potential plants derived from EMS mutagenesis with the highest polyphenol content and antioxidant capacity in the M2 population. In conclusion, induced mutation using EMS can enhance the agro-morphological diversity, polyphenols content along with the antioxidant activities of C. cristata, and demonstrate the successful mutation breeding program.

Abstract

Abstract:  Curcuma aeruginosa Roxb. is one of the most widespread medicinal plants in Southeast Asia, one of which is Indonesia. The purpose of this study was to determine the phytochemical content, antibacterial activity against Escherichia coli, and the level of cytoxicity in the rhizome of C. aeruginosa from five different genotypes. The maceration technique was utilized to extract samples. Using qualitative techniques, the presence of secondary metabolites in the sample extract was assessed. While the extract's cytotoxicity was verified using the brine shrimp lethality test, its antibacterial activity against E. coli was determined using the disc diffusion method. The qualitative phytochemical analysis revealed that the five rhizome genotypes included triterpenoids but lacked phenols, quinones, and alkaloids. Increasing the concentration of extract samples increased the lethality against Artemia salina shrimp larvae, but resulted in an LC50 value high enough to render it cytotoxically ineffective. Compared to the positive control, the antibacterial activity test resulted in a very tiny diameter of the clear zone, indicating that the C. aeruginosa had limited antibacterial activity against E. coli.Abstrak: Temu hitam termasuk ke dalam salah satu tanaman obat yang tersebar luas di Asia Tenggara, salah satunya Indonesia. Tujuan dari penelitian ini adalah mengetahui kandungan fitokimia, aktivitas antibakteri terhadap bakteri Escherichia coli, dan tingkat toksisitas pada rimpang temu hitam (Curcuma aeruginosa Roxb.) dari lima genotipe yang berbeda. Ekstraksi sampel dilakukan dengan metode maserasi. Ekstrak sampel ditentukan adanya senyawa metabolit sekunder dengan metode kualitatif. Sementara sitotoksisitas ekstrak ditentukan dengan metode brine brine shrimp lethality test, sementara untuk antibakteri terhadap E. coli dilakukan dengan metode difusi cakram. Hasil uji fitokimia kualitatif menunjukkan kelima genotipe rimpang temu hitam mengandung triterpenoid dan tidak mengandung fenol, kuinon serta alkaloid. Peningkatan konsentrasi sampel temu hitam meningkatkan daya bunuh terhadap larva udang Artemia salina, namun menghasilkan nilai LC50 yang cukup tinggi sehingga memiliki sitotoksisitas yang rendah. Uji aktivitas antibakteri menghasilkan diameter zona bening yang sangat kecil dibandingkan kontrol positifnya, dengan demikian rimpang temu hitam memiliki aktivitas antibakteri pada E. coli yang rendah.
