Abstract

Lemongrass traditionally can be used as a mixture of water bath for rheumatic patients, antiseptic medicine, headache reducer, unbend the leech bite and overcome insect bite, but, the bioactivity potential of citronella lemongrass still remains unknown. The content of citronella lemongrass is known to have secondary metabolites such as flavanoid, phenolic, and terpenoid. The aim of this study is to determine the LC50 value (Lethality Concentration 50) in the root, stem, and leaves part of citronella lemongrass. Citronella lemongrass is extracted with maceration method using 96% ethanol solvent. Rendement value of leaf extract is greater than the value of stem and root extract, which is equal to 8.69% for stem extract, 3.73% for root extract and 9.70% for leaves extract. A toxicity test was performed by using the BSLT method, shrimp larvae were inserted into the test solution with each different solution concentration. LC50 values were obtained based on the calculation of percent mortality of shrimp larvae by using probit analysis. LC50 ethanol extract 96% of citronella lemongrass leaf is 67.841 μg/ml, citronella lemongrass stem is 31.604 μg/ml and citronella lemongrass root is 10.489 μg/ml. LC50 value shows that ethanol extract 96% of root, stem, and leaf of citronella lemongrass have no bioactivity potential due to the value of LC50 is greater than 1000 μg/ml. Abstract

The formation of prostaglandins and free radicals in the body occurs in response to inflammation. The use of non-steroidal synthetic drugs to overcome the effects of free radicals often causes harmful side effects to the body. Natural ingredients that have high antioxidant potential, such as curcumin, ginger are expected to overcome this problem. This study aims to measure the antioxidant potential of ginger nanocurcuminoid preparations coated with palmitic acid, by observing the lipid peroxide profile in the liver of inflammatory Spague-Dawley rats. SpagueDawley mice were induced with 1% carrageenan to trigger inflammation, then the lipid peroxide levels were measured after 24 hours. Measurement of lipid peroxide levels was carried out using a spectrophotometer with a wavelength of 532 nm. The size of the nanocurcuminoid preparation coated with palmitic acid was 561.53 nm with an IP value of 0.309. Lipid peroxide levels in the curcuminoid extract group at a dose of 100 mg / kg BW and the nanocurcuminoid group at a dose of 250 mg / kg BW were 0.27x10-4 nmol / g and 1.22 x10-4 nmol / g respectively. These showed that the antioxidant potential of temulawak nanocurcuminoids at a dose of 250 mg / kg BW was 114 times higher than that of curcuminoid extract at a dose of 100 mg / kg BW.

Abstract

FTIR spectroscopy combined with principal component analysis and discriminant analysis were used for identification and authentication of jahe merah from its related species namely jahe emprit and jahe gajah. FTIR spectra of all samples were recorded in the wavenumber range 4000-400 cm-1 and then subjected for preprocessing signal such as normalization and baseline correction. Combination of FTIR spectra with some chemometrics method such as principal component analysis and discriminant analysis were used to distinguish the three varieties of ginger. By using discriminant analysis, the three varieties of ginger were classified according to its variety. The developed method could be used for identification and authentication of jahe merah.ABSTRAKSpektroskopi FTIR dan kombinasinya dengan analisis komponen utama dan analisis diskriminan telah digunakan untuk identifikasi dan autentikasi jahe merah dari dua spesies yang berkerabat dengannya yaitu jahe emprit dan jahe gajah. Seluruh contoh dibuat spektrum FTIR pada kisaran bilangan gelombang 4000-400 cm-1 dan diberi perlakuan pendahuluan seperti normalisasi dan koreksi garis dasar. Kombinasi spektrum FTIR dengan beberapa metode kemometrik seperti analisis komponen utama dan analisis diskriminan digunakan untuk membedakan ketiga varietas jahe. Analisis diskriminan dapat mengelompokkan ketiga varietas jahe sesuai dengan jenis varietasnya. Metode yang dikembangkan ini dapat digunakan untuk tujuan identifikasi dan autentikasi jahe merah.

Abstract

The leaves of Plectranthus amboinicus (Lour.) Spreng. (Lamiaceae) is a perennial plant that contained flavonoids compounds with have antidiabetic properties. The aim of the work was to select the best solvent for extraction of the flavonoid compounds from torbangun leaves. A statistical simplex-centroid mixture design for the water, ethanol, and hexane solvents has been used to the extraction of extract yield, flavonoids content, and inhibitory α-glucosidase activity of torbangun leaves.  The ethanol-water extracts showed the highest extract yield with value of 15.92%. The ethanol-hexane extracts presented the highest total flavonoids content, 96.30 mg QE/g, followed by hexane extract (82.91 mg QE/g). The ethanol extract recorded higher inhibitory of α-glucosidase activity, with value of 25.53%, followed by hexane extract (25.21%), and ethanol-water extract (23.02%). The weak correlation between the α-glucosidase inhibition and total flavonoids content showed that flavonoids contribute non-significantly to the α-glucosidase activity.

Abstract

Turmeric is a rhizome plant used as traditional Indonesian medicine. Generally, the extraction process influences the efficacy of turmeric in curing a disease. This study compares the thin layer chromatography profile (TLC) of turmeric extract based on variations in the extraction method. The extraction process was carried out using 70% ethanol solvent in fresh rhizomes and turmeric simplicia. The study used maceration, sonication, and microwave assistance techniques. The thin layer chromatography profiles were analyzed using curcuminoid standards in visible and UV light visualization (254 and 366 nm). The results showed higher yields in the fresh turmeric extracts through maceration extraction, sonication, and microwave-assisted techniques, respectively. The thin layer chromatography pattern using eluent chloroform dichloromethane (32.5: 67.5 v / v) showed the extract was separated into 3 - 4 compounds, with different curcuminoid thicknesses in each treatment. The thin layer chromatography profile with the highest quality of curcuminoid content was found successively in sonication simplicia,  maceration, and microwave simplicia extracts, as well as sonication fresh turmeric extract, maceration, and microwaves. The high yield of turmeric extract does not indicate an increase in the quality of the curcuminoid levels produced.Keywords: Turmeric, Thin Layer Chromatography, curcuminoids, extraction modificationKunyit merupakan tanaman rhizoma yang telah dimanfaatkan dalam pengobatan tradisional Indonesia. Khasiat kunyit dalam menyembuhkan suatu penyakit dipengaruhi oleh proses ekstraksinya. Penelitian ini bertujuan membandingkan profil kromatografi lapis tipis (KLT) ekstrak kunyit berdasarkan variasi metode ekstraksi. Proses ekstraksi dilakukan menggunakan pelarut etanol 70% pada rimpang segar dan simplisia kunyit. Teknik ekstraksi yang dilakukan adalah secara maserasi, sonikasi dan bantuan gelombang mikro. Profil kromatografi lapis tipis dianalisis dengan menggunakan standar kurkuminoid pada visualisasi sinar tampak serta UV  (254 dan 366 nm). Hasil penelitian menunjukkan rendemen yang lebih tinggi terdapat pada jenis ekstrak kunyit segar daripada simplisianya, secara berturut-turut melalui teknik ekstraksi maserasi, sonikasi, dan bantuan gelombang mikro. Pola kromatografi lapis tipis ekstrak dengan menggunakan eluen kloroform:diklorometan (32,5:67,5 v/v) menunjukkan bahwa ekstrak terpisah menjadi 3 - 4 senyawa, dengan  ketebalan kurkuminoid yang berbeda pada masing-masing perlakuan ekstraksi. Profil kromatografi lapis tipis dengan kualitas kadar kurkuminoid tertinggi terdapat pada ekstrak simplisia secara sonikasi, disusul dengan ekstrak simplisia secara maserasi dan gelombang mikro, serta ekstrak kunyit segar secara sonikasi, maserasi, dan gelombang mikro. Rendemen yang tinggi dari ekstrak kunyit tidak mengindikasikan kualitas kadar kurkuminoid yang dihasilkan semakin baik.Kata kunci: Kunyit, Kromatografi Lapis Tipis, kurkuminoid, modifikasi ekstraksi

Abstract

Temulawak merupakan salah satu tanaman obat Indonesia yang termasuk dalam keluarga Zingiberaceae. Kurkuminoid merupakan salah satu senyawa bioaktif dalam temulawak yang dilaporkan memiliki aktivitas biologi. Pada penelitian ini dilakukan evaluasi kandungan kurkuminoid dan penghambatan enzim α-glukosidase dari ekstrak empat aksesi temulawak asal Jawa Barat (Sukabumi), Jawa Tengah (Wonogiri dan Karanganyar), Jawa Timur (Ngawi), dan satu ekstrak asal Bogor-Jawa Barat. Simplisia temulawak tersebutdiekstraksi secara bertahap dengan etanol dan n-heksan. Kandungan kurkuminoid diukur dengan menggunakan metode HPLC. Aktivitas penghambatan enzim α-glukosidase dilakukan secara in vitro dengan microplate reader pada 410 nm. Aksesi temulawak asal Wonogiri menunjukkan kandungan kurkuminoid tertinggi, dengan hasil pengukuran antara 35,57- 85,19 mg/g. Nilai IC50 untuk aktivitas penghambatan enzim α-glukosidase antara 333,27̵908,35 μg/mL, dengan IC50 terbaik adalah temulawak aksesi Wonogiri.Berdasarkan penelitian ini, aksesi temulawak asal Wonogiri sangat baik dalam hal kandungan kurkuminoid dan aktivitas penghambatan enzim α-glukosidase dibandingkan dengan aksesi asal Sukabumi, Karanganyar dan Ngawi, maupun dengan varietas asal Bogor.

Abstract

The present study aimed to evaluate different agro-morphological traits among twenty genotypes of Curcuma aeruginosa Roxb. and three varieties of Curcuma zanthorrhiza Roxb. Agro-morphological data traits were investigated based on qualitative and quantitative parameters from PPVFRA descriptors with modification. All the recorded data was analyzed through SPSS 16.0 and R 3.4.2 for ANOVA and similarity analysis, respectively. Significant differences (P <0.05) were observed in the traits studied of the habit of the rhizome, the shape of the rhizome, length of primary rhizome, number of mother rhizome, plant height, pseudostem diameter, number of leaves, leaf length, leaf width, and number of shoots. Hierarchical cluster analysis (HCA) classified the genotypes into three groups. The principal component analysis (PCA) were showed consistent with results of the HCA. These genotypes could be used as novel genotype in agricultural biochemistry for breeding programs.

Abstract

Torbangun (Plectranthus amboinicus) is a medicinal plant belonging to the Lamiaceae family. In Batak society, the torbangun leaves are consumed to increase breast milk production. The phytochemical variability can be improved by gamma-ray irradiation for plant breeding programs. In this work, a non-targeted metabolomics method was followed to evaluate the compounds profile of irradiated mutant and wild-type on leaves of P. amboinicus. GC-MS analyses evaluated the mutant putative (derived from gamma-ray irradiation) and wild-type plantlets. The resulting of GC-MS data were analyzed with chemometric analysis using hierarchical cluster analysis (HCA). The chemical diversity was showed in mutant and wild-type P. amboinicus. The major metabolite in plantlet wild-type were hydroxymethylfurfural (35.15%), brevifolin (7.69%), 3-Dihydro-3,5-Dihydroxy-6-Methyl-4H-Pyran-4-one (DDMP) (13.13%), stigmasterol (6.51%) and ferruginol (8.63%). However, the compound of DDMP (19.57%), neophytadiene (8.47%), linolenic acid (9.10%), ferruginol (7.61%), stigmasterol (14.14%) and gamma-sitosterol (7.08%) were identified in mutant plantlet. HCA showed diversity between wild-type and mutant plantlets, which highest content of DDMP and stigmasterol and lowest of hydroxymethylfurfural of mutant plantlet.

Abstract

Genetic enhancement in vegetatively propagated crops can be done through mutation induction. Colchicine-induced mutation is one of the methods that can be employed to increase plant genetic diversity. This study aimed to determine the effect of colchicine on the performance and genetic parameters of MV3 generation of Plectranthus amboinicus (L.). This study was conducted at the Laboratory of Agricultural Seed Management, Plantation Research and Development Center, from June 2018 to June 2019. Nodes and shoots were used as explants. Mutation induction was performed using colchicine at concentrations of 0%, 0.02%, 0.04%, and 0.06%. Explant regeneration and subculture were done on MS0 medium. The number of plantlets yielded were 59 (0% concentration of colchicine), 60 (0.02%), 81 (0.04%), and 80 plantlets (0.06%), respectively. Results indicated that colchicine-induced mutation in an in vitro culture was able to generate high genetic diversity in both quantitative and qualitative characters of the plantlets. At the concentration of 0.04%, colchicine produced the highest frequency of putative mutants (28.4%). Genetic parameters in MV3 generation of P. amboinicus plantlets showed that five quantitative characters, i.e. plantlet height, number of leaves, number of shoots, leaf length, and number of roots had high heritability values at a concentration around the LC50 value (0.0275%).

Abstract

Penelitian ini bertujuan untuk membandingkan aktivitas antioksidan dan kandungan fenolik dari rimpang temulawak dan temu ireng. Aktivitas antioksidan ditentukan dengan menggunakan metode DPPH (1, 1-diphenyl-2-picrylhydrazyl). Kandungan fenolik total ditentukan dengan menggunakan metode Folin-Ciocalteu. Ekstrak etanol 70% dari rimpang temulawak memiliki aktivitas antiosidan (IC50, 167.03 µg/ ml) yang lebih baik dibandingkan dengan ekstrak etanol 70% rimpang temu ireng aeruginosa (IC50, 406.52 µg/ ml). Kandungan fenolik total dari ekstrak etanol 70% rimpang temulawak (139.16 mg TAE/ g) lebih tinggi dibandingkan dengan ekstrak etanol 70% rimpang temu ireng (51.49 mg TAE/ g). Terdapat korelasi yang kuat antara aktivitas antioksidan dan kandungan fenolik total dari ekstrak temulawak dan temu ireng.
