Abstract

Objective: To compare the administration of sambiloto tablets (AS201-01) and dihydroartemisinin-piperaquine phosphate tablets in pregnant mice infected by P. berghei on the expression of MIF in the placenta.Materials and Methods: Experimental laboratory research, 24 pregnant mice were divided into 4 groups with randomization, ie. non-infected, placebo, sambiloto (AS201-01) and DHP groups. On day 9 P. berghei was infected, on day 11 the treatment was given, day 15 the surgery was performed, placental samples were taken, immunohistochemical staining was given, and MIF expression was assessed.Results: The expression of MIF in the group not infected with P. berghei had the lowest mean, while the highest mean was found in the placebo group. Uninfected groups were not significantly different from sambiloto (AS201-01) tablet group. Sambiloto tablet group (AS201-01) had lower MIF expression than DHP group, but it was not significantly different. Sambiloto tablet group (AS201-01) showed lower MIF expression than placebo. MIF expression in DHP group was lower than that in placebo group. From the lowest, the MIF expressions were as follows: group not infected with P. berghei, group receiving sambiloto (AS201-01) tablet, DHP group and placebo group.Conclusion: MIF expression in the placenta of pregnant mice infected with P. berghei and receiving sambiloto (AS20-01) tablets was not different from those receiving DHP tablets.

Abstract

ABM (Agaricus blazei Murill) is a basidiomycetes fungus. ABM is used by people for the treatment of diabetes, antihypertention, anticholesterol, anticancer, and immunostimulant. ABM contains terpene, steroids, agaritine, vitamin C, vitamin E, and betaglucane. In this research, ABM extract was tested as an anti-breast cancer in vitro using MCF-7 breast cancer cells. The extract was obtained from the multistage extraction process of several solvents in turn, the solvent used, among others, n-hexane, dichloromethane (DCM), chloroform, ethyl acetate, butanol, and water. The results of the research were the obtained IC50 value from n-hexane extract 247,17 μg /ml; extract DCM 227μg/ml ; chloroform extract 215,64 μg /ml ; extract of ethyl acetate 234,9 μg/ml ; butanol extract 500,78 μg/ml; while the water extract was inactive. Based on these results can be considered for further research to fractionate in order to know which class compounds have the potency as anticancer within the extracts.Key words : Agaricus blazei, multistage extraction, MCF-7 cells.

Abstract

Dusun Kiringan-Jetis, Bantul, merupakan desa wisata jamu gendong dengan penduduk mayoritas berprofesi sebagai pengrajin jamu. Jamu yang mereka jual secara turun-temurun terbatas dalam bentuk jadi dan setengah jadi. Perlu adanya peningkatan diversifikasi produk jamu untuk meningkatkan nilai jualnya. Kegiatan pengabdian pada masyarakat ini bertujuan untuk meningkatkan ketrampilan usaha dalam meningkatkan kualitas dan mengembangkan produk jamu yang akan dijual. Untuk dapat mencapai tujuan yang diinginkan dari kegiatan ini maka dilakukan upaya: (1) Edukasi dan pelatihan ketrampilan membuat es krim herbal dan (2) edukasi dan pelatihan ketrampilan membuat inovasi kemasan produk jamu yang aman dan menarik.Tim pelaksana kegiatan ini terdiri dari dosen dan mahasiswa S1 Fakultas Farmasi Universitas Airlangga dan Universitas Ahmad Dahlan. Dari kegiatan tersebut terlihat antusias para peserta pelatihan, peningkatan pengetahuan dan ketrampilan dalam pembuatan es krim herbal dan inovasi kemasan produk. Hal ini terlihat dari peningkatan skor post-testsetelah dilakukan pelatihan.Besar harapan para peserta kegiatan untuk dapat mengaplikasikan pengetahuan yang didapat sehingga dapat meningkatan perekonomian Dusun Kiringan-Jetis, Yogyakarta

Abstract

Cempaka kuning (Michelia champaca) which is belongs to Magnoliaceae family, has been traditionally used for treatment fever. This research was conducted to evaluate antimalarial activity of Cempaka kuning stembark extract against Plasmodium falciparum 3D7 in vitro. Cempaka kuning stembark was extracted with n-hexane, chloroform and methanol yielded n-hexane extract, chloroform extract and methanol extract, followed by antimalarial activity assay of these extracts. Phytochemical screening showed that n-hexane extract contain volatile oil and terpenoids, chloroform extract contain Volatile oil, terpenoids and fiavonoids, then methanol extract contain volatile oil, terpenoids, flavonoids, tannins and glycosides. Result of this study also showed that n-hexane extract, chloroform extract and methanol extract were active against P. alciparum 3D7 in vitro, with an IC50 value of 0.36, 0.24 and 1.00 µg/mL. The findings indicate that Cempaka kuning stembark extracts possess strong antimalarial activity and are prospective to be developed as antimalarial.

Abstract

Artocarpus champeden Spreng. (Moraceae) known as “cempedak”, was traditionally used for antimalarial remedies in Indonesia. Study to determine antimalarial activity of ethanol extract of cempedak stembark was carried out. Artocarpus champeden Ethanol Extract (ACEE) was administered twice per day (multiple dose) and once per day (single dose) orally. The study was carried out by modification method of the “4 Days Suppressive Test” originally described by Peter. The result showed that ACEE administered twice per day inhibited parasites growth (ED 0.19 mg/kg BW) higher than once per day (ED 6.0 mg/kg BW) and 10 mg/kg body weight of dose administrated twice per day was more effective than 100 mg/kg body weight once per day.

Abstract

Background: Hepatitis c virus infection is a global health problem which chronically infected 71 million people in the world. This infection has a risk of becoming liver cirrhosis and hepatocellular carcinoma. Since the current HCV therapy has been developed by direct-acting antivirals (DAA), however, most patients get limited access due to the high cost. Therefore, further development anti-HCV agent still greatly needed. Ruta angustifolia is a natural resource which was reported to possess anti-HCV activity. Ribavirin is an antiviral agent used to treat several virus infections, either DNA or RNA. Ribavirin was known to inhibit HCV infection by regulated immune system in host cells and interfering the replication of HCV by inhibit HCV RdRp. Objective: The current study evaluated the combination treatment of R. angustifolia extracts and ribavirin by in vitro culture cells of Huh 7it. Method: The study was conducted under an invitro cell culture of Huh 7it and infected with JFH1a. Result:  The result demonstrated an enhancement effect of the extract by increasing the anti-HCV activity 3.5-fold higher compared to ribavirin alone. The 50% inhibitory concentration of ribavirin by single treatment was 10.43 ± 0.18 µg/mL, while in combination with Ruta angustifolia extract was 2.80 ± 0.03 µg/mL. Further analysis of the combination by CompuSyn software mediated a synergistic effect among the combination with a combination index value of 0.691. Conclusion: These results suggested that combination of Ribavirin and Ruta angustifolia should be considered in developing anti-hepatitis C virus agents.

Abstract

Desa Ngadisari, Kabupaten Probolinggo memiliki beragam tanaman liar yang berpotensi dimanfaatkan untuk pengobatan. Beberapa diantaranya adalah adas, jarak, kecubung, keningar, purwoceng, krangean, kayu ampet, pulosari, dan pronojiwo. Beragamnya tanaman yang ada di sekitar masyarakat setempat merupakan kekayaan tersendiri dari daerah ini, namun tidak banyak masyarakat Desa Ngadisari yang memanfaatkannya secara baik. Hal ini terjadi karena aspek kekurangpraktisan yaitu bahan-bahannya harus diramu terlebih dahulu, selain itu banyak kios yang menjual obat modern di lokasi terpencilpun. Berdasarkan latar belakang di atas, maka dilakukan pelatihan pengembangan produk jamu dan simplisia dari tanaman lokal Bromo bagi masyarakat Suku Tengger, khususnya di Desa Ngadisari, Probolinggo. Produk tersebut berupa jamu godogan, teh herba dan simplisia yang dapat digunakan oleh masyarakat ataupun dipasarkan dengan nilai ekonomi tinggi dan bermanfaat bagi kesehatan. Hal ini dapat menciptakan peluang usaha baru bagi masyarakat Desa Ngadisari sehingga meningkatkan perekonomian dan taraf kesehatan masyarakat desa tersebut, sehingga hal ini bisa membuat lingkungan baik.

Abstract

Background: Artocarpus altilis leaf extract (AAL.E) was separated by VLC, and six fractions were obtained. Fraction 5 (AAL.E.5) showed antimalarial activity with an IC50 value of 3.71 µg/mL. Objective: This study aimed to determine the antimalarial activity of AAL.E.5 subfractions against P. falciparum, the mechanism of action against Plasmodium Falciparum Malate quinone oxidoreductase (PfMQO), and the active substances. Methods: The AAL.E.5 was separated by open-column chromatography and eluted with chloroform-methanol gradient elution in order of increasing polarity. The antimalarial activity of all subfractions was assessed using a lactate dehydrogenase (LDH) assay against P. falciparum and the mechanism of action of the PfMQO enzyme. The profiles of the most active subfractions were analyzed using High-Performance Liquid Chromatography (HPLC). Results: The separation of fraction 5 (AAL.E.5) yielded 11 subfractions (AAL.E.5.1–AAL.E.5.11). Screening antimalarial activity at 10 μg/mL in this subfraction showed that only five subfractions (AAL.E.5.6-AAL. E.5.10) inhibited P. falciparum and two subfractions (AAL.E.5.6 and AAL.E.5.10) inhibited the PfMQO enzyme. Only subfraction 6 (AAL.E.5.6) inhibited both, with IC50 values of 6.609 µg/mL and 20.34 µg/mL. The thin layer chromatography profile of AAL.E.5.6 revealed reddish-orange spots, indicating the presence of flavonoid compounds, and was also presumed from the UV-visible to HPLC chromatogram for band I in the 300 – 400 nm range and band II in the 240–285 nm range. Conclusion: Subfraction 6 has antimalarial activity against P. falciparum and is thought to have a mechanism of action in PfMQO. Based on the TLC, HPLC, and UV-Vis spectra, subfraction 6 was assumed to be a flavonoid.

Abstract

The study was aimed to determine the anti-inflammatory activity of water extract of the Luvunga sarmentosa stem in an animal model. Twenty-five Wistar rats were divided into five groups (n=5). Group 1 was administered 0.9% normal saline (negative control), group 2 was administered 150 mg/kg diclofenac sodium (positive control), and groups 3 to 5 were administered 50, 300, and 550 mg/kg BW of L. sarmentosa extract, respectively. Carrageenan was injected subcutaneously into each rat's subplantar region of the left hind paw. The paw volume was measured using a plethysmometer. The results showed that the water extract of L. sarmentosa stem (doses of 50, 300, and 550 mg/kg BW) significantly reduced the paw edema volume from the 4th to 5th hour compared to the negative control. The percent inhibition of edema at the 5th hour is 47.45; 46.95; 50.39%. The first phase of the edema (1st and 2nd hour) was not affected by the extract. Meanwhile, diclofenac sodium decreased paw edema volume from the 1st to 5th hour with a percent inhibition of 95.90% at the 5th hour. The histopathology result is relevant to the percentage inhibition of edema. Treatment with L. sarmentosa extract showed slight improvement, destruction of epidermal tissue, hyperkeratotic skin, and subepidermal edema. Meanwhile, positive control showed no inflammatory signs with normal keratin, subepidermal, and subcutaneous layers. The water extract of L. sarmentosa stem has anti-inflammatory activity. This extract effectively reduces the paw edema volume in the late phase with decreased neutrophil infiltration.

Abstract

Luvunga sarmentosa, commonly known as saluang belum, is widely used in Kalimantan to relieve pains, rheumatism, boost the immune system, and fever. The research on the free radical scavenging and analgesic effect of the L. sarmentosa stem extract has not been reported. This study aimed to evaluate the free radical scavenging and analgesic activity of the ethanol extract of L. sarmentosa. The L. sarmentosa stem was extracted using 70% ethanol and tested for free radical scavenging using the 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) and analgesic activity, acetic acid-induced writhing test, and hot plate test in an animal model. The results showed that the 70% ethanol extract of the L. sarmentosa had an anti-free radical scavenging and analgesic activity. The extract has weak free radical scavenging with an IC50 value of 293.45 µg/mL. Analgesic activity using the writhing test indicated that the extract significantly reduced the writhes count after oral administration in a dose-dependent manner compared to the negative control. Extract at a dose of 550 mg/kg BW can reduce the writhing test by 67.60% compared to others. In contrast, the diclofenac sodium reduced the number of writhes by 74.74%. While in a hot plate, the extract at a dose of 550 mg/kg BW produced a maximum possible analgesia (MPA) of 17.64%, lower than the MPA of diclofenac sodium (51.01%). Analgesic activity of the extract has higher inhibition on the writhing test than on the hot plate. The extract could be responsible for the peripheral mechanism by inhibiting the prostaglandin biosynthesis.
