Abstract

Penyakit kardiovaskuler masih merupakan masalah di Indonesia salah satunya adalah Infark Miokardial Akut (IMA). Patofisiologis AMI karena rective oxigen spesies  (ROS) melibatkan stimulasi sitokin pro inflamasi salah satunya adalah  interleukin-1 yang berakibat peningkatan ekspresi COX-2. Penelitian ini bertujuan mengetahui efek ekstrak etanol ganggang hijau dalam menurunkan ekspresi COX-2 pada tikus IMA yang diinduksi isoproterenol (ISO). Ekstraksi ganggang hijau dengan maserasi menggunakan etanol 96%. Tikus berat 200-250 g dibagi menjadi 6 kelompok masing-masing 6 ekor. Kelompok I kontrol, tikus tidak diberi ISO hanya diberi CMC Na, kelompok II kelompok ISO, tikus diberi CMC Na dan ISO, kelompok III, IV dan V tikus diberi ekstrak etanol ganggang hijau dosis 250, 500 dan 750 mg/KgBB peroral dan kelompok VI diberi melatonin 10 mg/KgBB. Perlakuan ekstrak selama 28 hari. Pada hari ke 29 dan 30 tikus diberi isoproterenol dosis 85 mg/KgBB selama 2 kali pemberian secara subkutan kecuali kelompok I. Pada hari ke 31 tikus diambil organ jantung kemudian dibuat preparat histologis dan dilakukan analisis ekspresi COX-2 dengan metode imunohistokimia dengan antibodi anti COX-2. Hasil penelitian menunjukkan bahwa pemberian ekstrak etanol ganggang hijau dosis 250, 500 dan 750 mg/KgBB dapat menurunkan ekspresi COX-2 pada tikus yang diinduksi isoproterenol.

Abstract

Angiogenesis is a neovascularization process which its function is to supply nutrient and oxygen for cell survival. Pasak bumi is one of the Indonesian original plants having anti cancer activity. Extract of methanol, buthanol, kloroform, and water akar pasak bumi was proved to have cytotoxic effect on cancer cell culture. This research aim to know activity of antiangiogenic extract of metanol akar pasak bumi at chorioallantoic membrane of chicken embryo. Antiangiogenic activity test conducted by divide 8-9 days old egg embrio into 8 groups. Group I as paper disc control, group II as bFGF control, group III as bFGF control + DMSO 0.8% solvent, group IV, V, VI, VII, and VIII as  groups of treatment. Each groups of treatment were gave by bFGF 1ng/µL extract of metanol akar pasak bumi respectively with concentration 20, 40, 60, 80 and 100 µg/mL concentration. After 3 days incubation, then the chorioallantoic membrane macroscopic and microcospically analized. The antiangiogenic activity extract of metanol akar pasak bumi at chorioallantoic membrane bFGF induce at used concentration, that is start from 40 µg/mL concentration. The increases of the concentration also increase the prohibition activity to angiogenesis.

Abstract

The leaves of Dewa (Gynura procumbens (Lour) Merr), is utilized in traditional medicine as anticancer, had been reported to contain flavonoid derivatives and supposed to have cytotoxic properties. Hence, these particular substances might be useful to be developed as anticancer in the future. Research on ethanol extract of Gynura procumbens (Lour) Merr leaves as anticarcinogen has been carried out, using toxicity assay model to vero, myeloma and lymphocyte cells. This research applied varied concentrations and adriamycin as a positive control. Observation to the cell growth inhibition was carried out for 4 days. The result showed that the dominant substance (the third spot of paper chromatogram) inhibit the growth of vero and myelome cells but lymphocyte cell. The LC50 of this substance to myelome, vero and lymphocyte cells were < 82.8 g/ml, 103.86 g /ml and > 2650 g/ml, respectively, whereas the LC50 of adriamycin to the cell lines were < 0.015 g /ml. Therefore, substances obtained from Gynura procumbens (Lour) Merr leaves might be developed against cancer.Key word: Gynura procumbens (Lour) Merr, mieloma

Abstract

Pasak bumi is one of the Indonesian original plants having anti cancer activity. One compound of akar pasak bumi, i.e eurycomanone was proven to have cytotoxic effect on cancer cell culture by apoptosis induction on HeLa cells. Therefore, this study was aimed to isolate and identify euycomanone from akar pasak bumi and to look at the anti angiogenic activity of the compounds on corio alantois membrane (CAM) of chicken embryo induced by bFGF.The akar pasak bumi was ground and was macerated with methanol. The macerate was fractioned by mean of VLC. Six fractions was obtained. The second fraction exhibited two spots on a TLC system, one spot was cochromatographed with eurycomanone as the reference standart and another spot exhibited blue fluorecence at 254 nm. This second fraction, then, was chromatographed on a preparative scale TLC to separate isolate A. Anti angiogenic test was performed for Isolate A at concentration series of 5, 10, 15, 20, 40 μg/mL respectively. Egg embrio (8-9 days) were divided into 8 groups. The first group was implanted paper disc only, the second group were given bFGF on Tris HCl buffer and the third group were given bFGF (on Tris HCl buffer) + 0,8 % DMSO. The fourth to eighth group were induced by 10 ng bFGF and the test preparation at the respective concentration. After incubation at 39 °C for 3 days, each egg was opened, the content of each was aspired and discarded, the CAM sticked to its shell was examined macroscopically and microscopically for the formation of new blood vessels.Based on spectral data of UV, IR, 1H NMR, 13C NMR and MS of isolate A and comparison with the spectral data of the standart, the isolate A was eurycomanone. Eurycomanone inhibited angiogenesis at concentration of 10 μg/mL.Key words: antiangiogenic, angiogenesis, E.longifolia, eurycomanone,CAM.

Abstract

PPermasalahan yang ada di masyarakat Dusun Taruban Wetan, Tuksono, Sentolo, Kulonprogo, Yogyakarta salah satunya adalah kurangnya pengetahuan masyarakat tentang cara bercocok tanam tanpa menggunakan media tanah dan pestisida di area yang seadanya. Maka program kerja KKN Reguler UAD  difokuskan pada teknik bercocok tanam tanpa menggunakan media tanah dan pestisida (Hidroponik) pada area sempit. Program tersebut bertujuan untuk memberdayakan masyarakat dusun Taruban Wetan mengenai teknik bercocok tanam Hidroponik. Metode dari pelaksanaan program KKN  ini meliputi : pengetahuan masyarakat, teknik sederhana dan praktek secara langsung. Hasil pencapaian program pelatihan dari kegiatan KKN ini adalah : 1) Peningkatan pengetahuan masyarakat mengenai teknik bercocok tanam Hidroponik, 2) Masyarakat dapat terampil menggunakan barang bekas menjadi media tanam Hidroponik, 3) Masyarakat dapat memanfaatkan area sempit sebagai tempat bercocok tanam Hidroponik.

Abstract

Posyandu merupakan wadah yang dapat digunakan untuk upaya pencegahan dan penanggulangan masalah kesehatan baik gizi, kesehatan ibu dan anak, Pola hidup bersih sehat, imunisasi dan sebagainya.  Lewat model penimbangan dan pemantauan gizi tiap bulan juga dapat dijadikan sebagai alat surveilans gizi yang tujuan akhirnya adalah upaya pencegahan dan penanggulangan masalah gizi balita. Jika pelaksanaan Posyandu baik maka target pragram akan terpenuhi, akan tetapi jika pelaksanaan posyandu kurang lancar maka target tidak akan bisa terpenuhi. Dalam praktek di masyarakat pelaksanaan Posyandu masih mengalami beberapa kendala antara lain kemampuan kader yang masih kurang dalam kegiatan penyuluhan kesehatan kepada masyarakat. Tujuan pengabdian ini yaitu untuk meningkatkan kemampuan kader posyandu dalam pengukuran dan penimbangan balita serta status gizi ibu dan balita. Perlu juga peningkatan kemampuan penyuluhan kader, mengingat keterbatasan tenaga penyuluh di puskesmas Depok 3. Hal ini berimplikasi pada penanganan permasalahan kesehatan ibu dan balita lebih cepat. Metode yang digunakan dalam pelatihan ini yaitu dengan pemberian kegiatan pelatihan-pelatihan untuk peningkatan skill kader Posyandu disertai dengan mengadakan lomba penyuluhan kepada masyarakat dan  pemberian hadiah berupa ung pembinaan untuk melengkapi melengkapi sarana pra sarana yang menunjang kegiatan Posyandu. Melalui kegiatan ini diharapkan peran Posyandu sebagai ujung tombak pemantauan status gizi di masyarakat dapat lebih optimal. Hasil dari kegiatan ini meningkatkan kemampuan kader dalam memberikan penyuluhan kepada masyarakat dengan metode ang lebih menarik sehingga meningkatkan kepercayaan diri kader untuk bisa berbagi kepada masyarakat tentang kesehatan. Pelatihan ini meluas karena juga diberikan kepada kader posyandu lansia dan juga posbindu.

Abstract

Free radicals give a negative effect. According to earlier research, Spyrogyra and Ulva lactuca L. have an anti-oxidant activity. This research aim to knowing the extract potential of green algae (Ulva lactuca) with variety concentration solvent as anti-oxidant by DPPH method, and finding the price of IC50 ethanol extract which compared with Galat acid. Extraction technic was maseration with ethanol solvent 60%,75%, and 96% which is implemented a qualitative test of extract active compound of U. lactuca Antioxidant activity of U. lactuca extract measured with spektrofotometri by using DPPH (1,1-difenil-2-pikrilhidrazil) method to get percentation of free radical resistor. The analytical result using SPSS, statistically got 95% credibility (p=0,05). This research showed that activity of antioxidant ethanol extract 96% of U. lactuca was higher than ethanol extract 60% and 75%. Activity of antioxidant ethanol extract 60%,75%, and 96% which confirmed by value of IC50 continously are 441,70 mg/ml ; 242,10 mg/ml dan 2,04 mg/ml. Whereas the value of IC50 galat acid standard is 3,92 x 10-3mg/ml. Based on tests of conducted antioxidant activity, the gallic acid has a strong antioxidant power, U. lactuca have a very weak antioxidant power

Abstract

Antioksidan merupakan senyawa yang dapat menghambat reaksi oksidasi dengan mengikat radikal bebas dan molekul yang sangat reaktif. Spirogyra sp. merupakan contoh spesies yang hidup di perairan tawar, sedangkan Ulva lactuca di perairan laut. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan sebagai penangkap radikal bebas ekstrak etanol ganggang hijau Spirogyra sp. dan Ulva lactuca dengan metode DPPH (1,1-difenil-2-pikrilhidrazil). Ekstrak etanol serbuk ganggang hijau Spirogyra sp. dan Ulva lactuca dibuat dengan dengan metode maserasi. Uji aktivitas antioksidan dilakukan dengan metode DPPH secara spektrofotometri visibel. Asam galat digunakan sebagai kontrol positif. Aktivitas penangkapan radikal bebas dinyatakan dalam ES50. Hasil penelitian menunjukkan bahwa ekstrak etanol Spirogyra sp. dan Ulva lactuca memiliki aktivitas sebagai penangkap radikal bebas. Nilai ES50 Spirogyra sp. sebesar (92,83±0,58) μg/mL dan Ulva lactuca sebesar (1376,65±7,80) μg/mL, sedangkan asam galat memiliki harga ES50 sebesar (0,73 ± 0,04) μg/mL

Abstract

Background : Nowadays, cardiovascular disease is increasing in number each year. It is triggered by high cholesterol diet consumed by urban people. Green algae (Ulva lactuca L.) contains phytomelatonin levels that can ease cholesterol. Objective : To know about the effect of giving green algae ethanolic extract in male strain Wistar mice's cholesterol level. Methods : This research involved 36 male strain Wistar mice divided to six groups. Negative control injected with 2 ml/200 gBW lard, positive control injected with 0,18 mg/200 gBW simvastatin and lard, group III, V, V each given lard and injected with ethanolic extract of green algae at a dose of 50 mg/kgBW, 100 mg/kgBW and 200 mg/kgBW respectively, and normal control given AD 2 dietary treatment. Each group was given the same treatment peroral in 56 days and the blood was taken every 14 days through sinus orbitalis. The blood cholesterol level was analyzed by using enzymatic CHOD-PAP method. Data obtained were statistically examined using Two way ANOVA , Kolmogorov Smirnov, Levene's method and LSD with confidence level at 95%. Results : The blood cholesterol level were not significanly different between the negative and normal control. The result of cholesterol level for 14, 28, and 42 days for all group did not increase compared to negative control, but cholesterol level in group with 200 mg/kgBW dose extract indicated significant difference with negative control for 56 day. Conclusion : Giving ethanolic extract of green algae for 56 days decreased blood cholesterol level in male rats.

Abstract

Halal medicine is an interesting topic to always discuss because it is a priority choice for Muslim consumers, one of which is halal capsules. Currently, molecular biology techniques such as real-time polymerase chain reactions are rapidly developing, including for the analysis of non-halal components based on DNA sequences. This study aimed to validate the quantitative PCR method for identifying DNA in gelatin-based products and to apply the confirmation method designed for capsule samples on the market circulating in Yogyakarta to prove the halalness of these samples. Validation of the porcine DNA detection analysis method on standard extraction of porcine gelatin using primer pairs obtained in previous studies. Validated methods are used for testing market capsule shells. The qPCR method using D-loop primers is specifically capable of amplifying porcine gelatin DNA up to a concentration of 0.5 pg/µL, with a CV value in the amplification response of porcine gelatin DNA isolates (1000 pg/µL) of 0.85% which meets the test criteria using the PCR. Three samples of commercial soft capsules tested gave a positive amplification response, meaning that the samples tested contained porcine DNA, and one negative sample, which probably had non-porcine gelatin. The application of this method is also very useful for ensuring the authenticity of the capsule shell, especially from cross-contamination and counterfeiting.
