Abstract

Phytopythium vexans isolated from potatoes rhizospher in Ngablak, Magelang, Central Java was reported having a potency to cause potato tuber rot in in vitro test. Some fungicides had been intensively applied in this area to control potato late blight even using sublethal dose. This study investigated resistance response of the pathogen against several fungicides which were Mancozeb, Dimethomorph, mixture of Mancozeb and Mefenoxam, and mixture of Oxathiapiprolin and Famoxadone. The experiment was conducted to observe the sensitivity response of pathogen against those fungicide and the experiment repeated with one of the isolates which growth in sublethal concentration to observe the resistance response. Both experiment data was analyzed by Probit Analysis. On sentivity test, data showed that P. vexans were insensitive against Dimethomorph, but it was sensitive against Mancozeb, mixture of Mancozeb and Mefenoxam, as well as mixture of Oxathiapiprolin and Famoxadone. On Mancozeb treatment, mycelia were inhibited 100% at the sugested concentration. However, at the sublethal concentration the mycelial collony grew abnormally. Pathogen isolate from the sublethal concentration were used as material for the resistance response study. The effect of the sublethal application of Mancozeb to P. vexans caused resistance to Mancozeb itself and cross resistance against Dimetomorph and mixture of Oxathiapiprolin and Famoxadone. It was proven by higher value of predicted EC50 and EC95 on the second experiment compared to the predicted EC50 and EC95 from the first one. The resistant P. vexans isolate was able to produce sporangium and chlamydospore.

Abstract

Shallot (Allium cepa var. aggregatum.) is a horticultural plant that is widely consumed in the world. However, the productivity of shallots in Indonesia is still relatively low, if compared to the actual optimum production potential of shallot. Shallot cultivation in Indonesia often experiences many problems. One of the problems is twisted diseases caused by Fusarium sp. This research aimed to study the effect of the application of organic material enriched with Bacillus in suppressing the development of twisted disease of shallot. This study was arranged in Randomized Complete Block Design (RCBD) with 5 treatments namely (A) compost + Bacillus velezensis isolate B-27, (B) compost + Bacillus cereus isolate RC76, (C) B. velezensis isolate B-27+B. cereus isolate RC76+compost, (D) compost + Trichoderma asperellum and (E) control (compost 1 ton/ha) with 5 replications on glasshouse treatment and 3 replications on field treatment. The results showed that the combination of B. velezensis in compost effectively reduced the incidence of twisted disease, the number of Fusarium spp. colonies, and the number of infected bulbs by Fusarium sp. Besides, the combination of compost with microbial agents showed better results than compost single treatment.

Abstract

Ultraviolet-B (UV-B) light is known to induce plant resistance in suppressing disease infection. Its ability to induce plant resistance has been widely used as an inducer of plant resistance. This study aims to determine the ability of UV-B light in suppressing shallot twisted disease, potency of plant resistance induction through accumulation of peroxidase enzymes, salicylic acid, chlorophyl content and to observe shallot growth after treatment. This study applied UV-B light with a duration of 65 mW/m2 for 4 hours each day for 3, 5 and 7 days, for inoculated and non-inoculated plants. Fungicide (Benomyl) was used as a comparison for inoculated plants. The results showed that UV-B irradiation of 4 hours/day for 7 days was able to reduce the incidence of shallot twisted disease, but the incubation period among treatments was not significantly different. UV-B irradiation of 4 hours/day, for 7 days was able to increase salicylic acid content, and maintain the same chlorophyll content as the negative control, however the peroxidase enzyme content was not significantly different. Treatment of UV-B 4 hours/day for 7 days irradiation did not inhibit the growth and production of shallot plants.

Abstract

The twisted disease is one of the essential diseases in shallots caused by Fusarium spp. This study aimed to study pathogenicity and identify Fusarium species isolated from shallot plants with twisted symptoms in Nganjuk and Bantul areas. The Fusarium isolates were identified and then tested for pathogenicity levels and the effect of the hormones GA3 and IAA on shallot symptoms. Molecular identification using NF2 and NF4 successfully identified one isolate of Fusarium oxysporum, three isolates of F. acutatum, and three isolates of F. solani. Each of these species produces different symptoms. Pathogenicity test showed that all isolates had disease incidence reaching 100%, except isolates of F. solani1 causing wilt and F. solani3 causing twisted have the lower disease incidence were 77.8% and 77.7%, respectively. The investigation caused twisted shallot related to different symptoms was tested using the Thin Layer Chromatography (TLC) method. The result indicates that all isolates did not find IAA hormone. In contrast, the hormone GA3 was found in F. solani2 and F. solani3 isolates, caused bulb rot and twisted disease, respectively. Detection of IAA, GA3, and other hormones in shallot plants showed different symptoms should be studied further.

Abstract

Shallots are a type of plant used as a flavoring agent and are often affected by twisted disease caused by Fusarium spp. This study aims to determine the ability of organic material enriched by Trichoderma asperellum in controlling twisted disease, increasing production, and suppressing the population of Fusarium spp. in the soil. The research showed that compost and chicken manure enriched with T. asperellum in a ratio (200:1) were able to reduce the disease incidence compared to controls, in addition to increasing production and suppressing the population of Fusarium spp. in the soil. The colonies of Fusarium spp. in the soil were correlated with disease incidence in each treatment. The highest incidence of twisted disease was followed by the number of Fusarium spp. colonies. The microbial composition did not affect disease suppression when observed using a dependent method using PCR-RISA. The microbial composition with the dependent method using PCR RISA did not affect the suppression of the twisted disease in the shallot plants.

Abstract

This research is about the involvement of trade unions in changing wage policies through social advocacy from the social welfare perspective. The urgency of conducting this research is to reveal how KASBI's involvement as a worker's union encourages changes in wage policies to ensure better labor welfare through advocacy against the Job Creation Law and advocacy against Minister of Manpower Regulation Number 2 of 2022. This research uses a descriptive qualitative method. Data were collected from February to June 2023 through online and offline interviews involving three informants selected by the purposive sampling technique. The research results reveal that KASBI engages politically to fight for the interests of the labor group, including obtaining guarantees for basic needs. The advocacy process involves planning a strike, consolidating alliances, holding a National Coordination Meeting (Rakornas), conducting a judicial review of the Constitutional Court, holding mass demonstrations, submitting hearings, asking the government for access to draft laws, and writing studies. Supporting factors of involvement and advocacy are credibility and financial independence. Meanwhile, the obstructing factor is the incapability of the governments to openness to the aspirations and interests of the labor group. This research has benefits and contributions to the study of social welfare, especially social welfare in the industrial sector, social policy, and social legislation. The results of this research can be a reference for social workers in carrying out advocacy in the policy realm, as well as a reflection for stakeholders in formulating policies that better accomodate worker’s aspirations.

Abstract

Chitala lopis, also known as giant featherback, is a fully protected fish species on Indonesian official list due to the susceptibility to exploitation. Several studies showed that the conservation of aquatic resources, such as giant featherback, could be achieved through the implementation of fish farming practices and optimization of feed use. Therefore, this study aimed to determine effect of different feeding rate on growth pattern, performance, and condition factor of the first generation of giant featherback. The first generation of fish was collected from the natural reproduction of broodstock in the earthen ponds. A total of 5 samples were then randomly divided into three treatment groups and reared in the nine net cages installed in the earthen pond. The stocking density in each group was three fish per cage, and the treatments were carried out in triplicates. Feed given comprised fish bycatch as control (T0) (5% of the total weight) and freshwater shrimp, Caridina sp. with two different feeding rates, namely 5% (T1) and 7% (T2) of the total weight. The mean initial weight of all the samples in this study was 41 3.85 g. The results showed that the highest survival and absolute growth rates (29.96.38 g, 78 11%) were obtained in T1, but were not significantly different (p0.05). Furthermore, the highest average relative growth rate was also found in T1 (76.2818.61%) and significantly different compared to other treatments (p0.05). Growth pattern of fish at the beginning of the experiment was negative allometric (0.54), showing a low correlation (0.03), but became positive allometric (4.17) at the end, with moderate correlation (0.66). The results also showed that the highest condition factor was obtained in T2 (0.780.03), and there was a significant effect (p0.05). Based on the results, Caridina sp. had a positive and significant impact on the weight growth of the first generation of giant featherback compared to other studies. Keywords:CultivationFeeding rateGiant featherbackCaridina sp.Rasbora sp.
