Abstract

Abstrak Latar belakang: Rennellia elliptica Korth. secara tradisional digunakan sebagai zat afrodisiak. Dilihat dari potensinya, bukti ilmiah ekstrak tumbuhan ini sebagai penghambat phosphodiesterase-5 masih belum dilakukan Tujuan: Penelitian ini bertujuan untuk mengetahui senyawa fitokimia yang terdapat pada ekstrak etanol 70% tanaman rennellia elliptica Korth. menggunakan LC-MS/MS dan aktivitas afrodisiakan secara in silico. Metode: Ekstrak etanol rennellia elliptica Korth. dianalisis menggunakan LC-MS/MS selanjutnya senyawa tersebut digunakan sebagai senyawa ligan dalam penambatan molekuler untuk mengetahui afinitas pengikatan konstituen fitokimia tersebut terhadap protein target phosphodiesterase 5 (PDB ID: 2H42). Hasil: Ditemukan 17 komponen senyawa antara lain golongan terpenoid, fenilpropanoid, antrakuinon dan flavonoid. Berdasarkan studi in silico dari 17 senyawa yang dianalisis dengan LC-MS/MS, Dulxanthone B (∆G=-9,52 kkal/mol) menunjukkan afinitas tertinggi untuk berikatan dengan target protein. Simpulan: Senyawa fitokimia dari rennellia elliptica Korth. dapat berfungsi sebagai terapi tambahan dalam penemuan obat untuk pengobatan yang menghambat phosphodiesterase 5. Abstract Background: Rennellia elliptica Korth. is traditionally used as an aphrodisiac substance. Judging from its potential, scientific evidence of this plant extract as a phosphodiesterase-5 inhibitor has yet to be conducted. Objective: . This research aims to determine the phytochemical compounds contained in the 70% ethanol extract of the Rennellia elliptica Korth plant. using LC-MS/MS. Method: The ethanol extract of rennellia elliptica Korth. was analyzed using LC-MS/MS and then the compounds were used as ligand compounds in molecular docking to determine the binding affinity of the phytochemical constituents to the target protein phosphodiesterase 5 (PDB ID: 2H42). Results: 17 compound components were found, including terpenoids, phenylpropanoids, anthraquinones and flavonoids. Based on in silico studies of 17 compounds analyzed by LC-MS/MS, Dulxanthone B (∆G=-9.52 kcal/mol) showed the highest affinity for binding to protein targets.. Conclusions: Phytochemical compounds from Rennellia elliptica Korth. may serve as an adjunct therapy in drug discovery for treatment that inhibits phosphodiesterase 5.

Abstract

Codo Village, located in Malang Regency, once held a serenization program. Local PKK women pioneered this program, whose actions were realized through a movement to plant lemongrass throughout the vacant village land to avoid the spread of dengue fever in 2016. A lack of knowledge about the potential of lemongrass plants caused a decline in enthusiasm for the serenization program, marked by many broken lemongrass plants. So, the DILAN (Simple Distillation) training provides a solution to revitalize the serenization program, starting with the formation of cadres, delivery of materials related to lemongrass plants and simple extraction of essential oils using the DILAN method, introduction to e-commerce, supervision and evaluation. The result of this program is that PKK women can produce marketable products from essential lemongrass oil, such as soap, aromatherapy candles, and carbolic acid. The products produced are codo asri and can be obtained from online sales sites. This program can increase the knowledge, productivity and abilities of the population to improve the community's economic status so that the revitalization program can be successfully implemented.

Abstract

Erectile dysfunction is one of the most common sexual problems in men. Drugs available on the market can be used to treat this problem, but the use of these drugs often causes unwanted side effects, the use of natural ingredients as an alternative treatment is increasingly popular, one of which is a herb made from chili jamu (Piper retrofractum Vahl). Metabolite profiling of the chili jamu plant turns out to be different depending on the location of growth, it is thought to result in differences in natural conditions and circumstances which result in differences in aphrodisiac activity. Cabe Jawa extracted with 96% ethanol was analyzed using LC / MS, detected as many as ten secondary metabolites namely, Salsolinol, Quercetin, Caffeoyl putrescin, Diferuloyl putrescine, Feruloyl tyramine, Hydroxychloroquine, Tetrahydropapaveroline, Piperine, N-Oleyl-Leucine, Capsaicin. From the results of in silico tests, it is known that piperine has activity as a phosphodiesterase type 5 (PDE5) inhibitor with binding energy and inhibition constant values of -8.62 kcal/mol and 0.485 uM. Keywords:          Piper retrofractum Vahl, Secondary metabolite, Aphrodisiac   Abstrak Disfungsi ereksi merupakan salah satu masalah seksual yang paling umum terjadi pada pria. Obat yang tersedia di pasaran dapat digunakan untuk mengatasi masalah ini, tetapi penggunaan obat tersebut seringkali menimbulkan efek samping yang tidak diinginkan., penggunaan bahan alami sebagai alternatif pengobatan semakin populer, salah satunya yaitu ramuan berbahan cabe jamu (Piper retrofractum Vahl). Metabolit profiling dari tanaman cabe jamu ternyata berbeda-beda tergantung dari lokasi tumbuhnya, hal tersebut diduga mengakibatkan perbedaan kondisi dan keadaan alam yang berakibat pada perbedaan aktivitas afrodisiak. Cabe jamu yang diekstrak dengan etanol 96% di analisa dengan menggunakan LC/MS, terdeteksi sebanyak sepuluh metabolite sekunder yaitu, Salsolinol, Quercetin, Caffeoyl putrescin, Diferuloyl putrescine, Feruloyl tyramine, Hydroxychloroquine, Tetrahydropapaveroline, Piperine, N-Oleyl-Leucine, Capsaicin. Dari hasil uji in silico diketahui bahwa piperine memiliki aktifitas sebagai penghambat phosphodiesterase type 5 (PDE5) dengan nilai energi ikat dan konstanta inhibisi sebesar -8,62 kkal/mol dan 0,485 uM. Abstract

AbstrakSwamedikasi merupakan salah satu cara masyarakat untuk mengatasi keluhan penyakit yang tergolong ringan. Tren ini semakin meningkat selama pandemi COVID-19 berlangsung seiring dengan adanya berbagai aturan pembatasan yang membuat hampir seluruh lapisan masyarakat membatasi kegiatan di luar rumah. Sangat penting untuk masyarakat memiliki pengetahuan yang memadai, baik tentang obat modern dan tradisional, untuk menjamin efektivitas obat sehingga kesembuhan atau perbaikan kesehatan yang diharapkan dapat tercapai. Berdasar hal tersebut diselenggarakan  pengabdian kepada masyarakat dalam bentuk penyuluhan guna meningkatkan pemahaman masyarakat mengenai swamedikasi obat modern dan tradisional. Penyuluhan berbasis online dengan metode ceramah dan diskusi melalui webinar melibatkan 305 peserta dari berbagai daerah di Indonesia. Penilaian pemahaman masyarakat melalui pre-test sebelum dan post-test setelah materi webinar diberikan. Hasil analisis menunjukkan bahwa tingkat pengetahuan masyarakat umum meningkat signifikan (p < 0,05) yang ditandai dengan nilai rata-rata pre-test sebesar 54,47 dan nilai rata-rata post-test sebesar 74,89. Sama halnya dengan tingkat pengetahuan tenaga kefarmasian juga meningkat signifikan (p < 0,05) dengan nilai rata-rata pre-test sebesar 75,15 dan nilai rata-rata post-test sebesar 86,59. Berdasar hasil tersebut dapat disimpulkan bahwa pengabdian kepada masyarakat ini berpengaruh terhadap tingkat pengetahuan masyarakat umum dan tenaga kefarmasian mengenai obat modern dan tradisional sehingga dapat menjamin efektivitas pengobatan dan didapatkan mutu kesehatan yang maksimal. Kata kunci: COVID-19; kesehatan masyarakat; obat modern; obat tradisional. AbstractSelf-medication is one way for the community to deal with complaints of ailments that are classified as mild. This trend has increased during the COVID-19 pandemic along with various restrictions that have made almost all levels of society restrict activities outside the home. It is very important for the community to have adequate knowledge, both about modern and traditional medicine, to ensure the effectiveness of the drug so that the expected recovery or health improvement can be achieved. Based on this main problem, the author provides community service in the form of counseling to increase public understanding regarding modern and traditional drug self-medication. Online-based counseling using lecture and discussion methods via webinars involved 305 participants from various regions in Indonesia. Assessment of public understanding through pre-test before and post-test after the webinar material was given. The results of the analysis show that the level of knowledge of the general public has increased significantly (p < 0.05) as indicated by the average pre-test score of 54.47 and the average post-test score of 74.89. Likewise, the level of knowledge of pharmaceutical staff also increased significantly (p < 0.05) with an average pre-test score of 75.15 and an average post-test score of 86.59. Based on these results, it can be concluded that this community service influences the level of knowledge of the general public and pharmaceutical staff regarding modern and traditional medicine so that it can guarantee the effectiveness of treatment and obtain maximum health quality. Keywords: COVID-19; public health; modern medicine; traditional medicine.

Abstract

Background: Red ginger (Zingiber officinale var. rubrum) and aromatic ginger (Kaempferia galanga L.) are traditional Indonesian medicinal plants with proven pharmacological activity, but their poor aqueous solubility requires formulation strategies to enhance solubilization and physical stability. Objective: This study aimed to optimize and evaluate a Self-Nanoemulsifying Drug Delivery System (SNEDDS) combining red ginger and aromatic ginger. Methods: The optimization was conducted by varying the oil-to-Smix ratio and extract concentration, while the internal Smix composition (Tween 80: PEG 400 = 1:2.8 w/w) remained constant to isolate the effect of oil–surfactant proportion. Virgin coconut oil (VCO), Tween 80, and PEG 400 were used as the oil phase, surfactant, and co-surfactant, respectively. Three formulas were prepared based on oil-to-Smix (w/w): Formula A (1:4.68, 500 mg extract), Formula B (1:10.6, 500 mg extract), and Formula C (1:10.6, 460 mg extract). Physical characterization included visual observation, centrifugation, particle size, polydispersity index (PDI), and transmittance before and after thermodynamic stability tests. Results: Formula A was unstable and exhibited precipitation. Before stability testing, Formula C showed improved dispersion, with a 41.5% reduction in droplet size and a 79.9% decrease in PDI relative to Formula B. After stability testing, Formula C demonstrated markedly better performance, including an 85.5% smaller droplet size, 81.4% lower PDI, and a 2.4% increase in transmittance compared with Formula B. These quantitative improvements indicated that Formula C exhibited more optimal solubilization capacity and physical stability. Conclusion: This study concludes that extract concentration and the oil-to-Smix ratio significantly influence SNEDDS characteristics, and Formula C represents the most favorable formulation for further development.

Abstract

Erectile dysfunction can be treated by pharmacological therapy with phosphodiesterase-5 inhibitors (PDE5-I). One plant that has a function as a phosphodiesterase-5 inhibitor is Kigelia africana (Lam.) Benth. Research on Kigelia africana (Lam.) Benth. An extract as a Phosphodiesterase type 5 (PDE 5) inhibitor is still lacking. This research aims to determine the phytochemical compounds in the 70% ethanol extract of the pericarpium, fructus and semen of Kigelia africana (Lam.) Benth. with LC-MS/MS and to determine the ligand-protein interaction through in-silico studies. In previous studies, no one compared each part of the Kigelia africana (Lam.) Benth. as an aphrodisiac. The results of the identification of the 70% ethanol extract of Kigelia africana (Lam.) Benth. found several alkaloids, flavonoids, iridoids, phenolics, polyphenols, coumarins, steroids and fatty acids. Based on the results of molecular docking on 70% ethanol extract of pericarpium, fructus and semen of Kigelia africana (Lam.) Benth., 18 compounds, 20 compounds and 18 compounds were obtained sequentially, which were analyzed using LC-MS/MS. The sitosterol compound with (∆G = -8.90 kcal/mol) was identified in the three parts of the 70% ethanol extract sample of kunto bimo fruit, which showed the highest affinity for binding to the target protein compared to sildenafil with (∆G = -8.68 kcal/mol). Sitosterol compound has the same amino acid residues as the native ligand, namely ILE A:824, TYR A:664, ALA A:779, ILE A:768, ALA A:767, LEU A:765, ASN A:661, HIS A:613, PHE A:786, VAL A:782, LEU A:804, MET A:816, PHE A:820 in hydrophobic bonds. Phytochemical compounds from Kigelia africana (Lam.) Benth. It has the potential as an alternative additional therapy and a promising source for the discovery of new drugs as aphrodisiacs targeting PDE5.

Abstract

Pemanfaatan tanaman obat untuk melawan COVID-19 menjadi tren di masyarakat beberapa tahun terakhir. informasi mengenai penggunaan tanaman obat untuk melawan COVID-19 menyebar dengan cepat sehingga dikhawatirkan dapat meningkatkan kesalahan informasi dan dapat menimbulkan permasalahan kesehatan baru akibat penggunaan obat tradisional yang kurang tepat. Kegiatan pengabdian masyarakat ini bertujuan untuk meningkatkan pemahaman masyarakat terkait pemanfaatan obat tradisional serta cara pembuatannya yang baik untuk melawan COVID-19. Kegiatan ini dilakukan dalam bentuk ceramah secara daring yang melibatkan 47 orang guru dan pengelola Lembaga Pendidikan Lukman Al Hakim Hidayatullah, Surabaya. Penilaian terhadap tingkat pengetahuan para peserta dilakukan melalui pre-test dan post-test menggunakan instrumen berupa sepuluh pertanyaan pilihan ganda dalam format respons tunggal tentang obat tradisional untuk melawan COVID-19 dan cara pembuatan obat tradisional yang baik. Diperoleh peningkatan nilai rata-rata post-test (75,83) dibanding pre-test (51,39) yang menunjukkan bahwa adanya peningkatan pengetahuan para peserta webinar tentang obat tradisional untuk melawan COVID-19 beserta cara pembuatannya yang baik.

Abstract

Desa Ngadisari, Kabupaten Probolinggo memiliki beragam tanaman liar yang berpotensi dimanfaatkan untuk pengobatan. Beberapa diantaranya adalah adas, jarak, kecubung, keningar, purwoceng, krangean, kayu ampet, pulosari, dan pronojiwo. Beragamnya tanaman yang ada di sekitar masyarakat setempat merupakan kekayaan tersendiri dari daerah ini, namun tidak banyak masyarakat Desa Ngadisari yang memanfaatkannya secara baik. Hal ini terjadi karena aspek kekurangpraktisan yaitu bahan-bahannya harus diramu terlebih dahulu, selain itu banyak kios yang menjual obat modern di lokasi terpencilpun. Berdasarkan latar belakang di atas, maka dilakukan pelatihan pengembangan produk jamu dan simplisia dari tanaman lokal Bromo bagi masyarakat Suku Tengger, khususnya di Desa Ngadisari, Probolinggo. Produk tersebut berupa jamu godogan, teh herba dan simplisia yang dapat digunakan oleh masyarakat ataupun dipasarkan dengan nilai ekonomi tinggi dan bermanfaat bagi kesehatan. Hal ini dapat menciptakan peluang usaha baru bagi masyarakat Desa Ngadisari sehingga meningkatkan perekonomian dan taraf kesehatan masyarakat desa tersebut, sehingga hal ini bisa membuat lingkungan baik.
