Abstract

Endometriosis merupakan penyakit yang bersifat progresif, kronik, rekuren dan hingga saat ini etiologi pastinya belum diketahui. Gejala yang ditimbulkan endometriosis bervariasi mulai dari asimptomatik, nyeri pelvis hingga infertilitas. Beberapa studi terakhir di bidang biologi molekular menunjukkan adanya kemungkinan endometriosis sebagai penyakit epigenetik dan salah satu gen yang diketahui mengalami perubahan epigenetik adalah gen HOXA10. Gen HOXA10 berperan mengatur reseptivitas endometrium selama proses implantasi. Keberhasilan proses implantasi merupakan salah satu faktor yang berperan pada fertilitas wanita dan Gen HOXA10 memiliki peranan penting dalam proses implantasi tersebut. Metilasi gen HOXA10 mengalami peningkatan yang signifikan pada endometrium eutopik wanita yang mengalami endometriosis. Pada endometrium wanita yang mengalami endometriosis terdapat penurunan ekspresi HOXA10 yang signifikan dan hal tersebut mengindikasikan adanya gangguan pada reseptivitas uterus yang dapat berperan dalam menurunnya tingkat kesuburan.

Abstract

MikroRNA merupakan bagian dari keluarga besar non-coding RNA yang secara negatif meregulasi ekspresi gen pengkode protein paska transkripsi melalui pasangan basa antara 5’ seed region miRNA dengan 3’ untranslated region dari mRNA. Berbagai studi telah menunjukkan adanya peran penting miRNA pada perkembangan kanker. Keluarga miR-200 termasuk mikroRNA yang berperan dalam inisiasi hingga tahap metastasis kanker. miR-200 terdiri dari 5 anggota yaitu miR-200a, -200b, -200c, -141, dan -429. Keluarga miR-200 berperan pada tiap tahapan metastasis yaitu mulai dari pertumbuhan tumor, angiogenesis, invasi ke jaringan sekitar, transisi dari epitelial ke mesenkimal, migrasi sel tumor, intravasasi sel tumor, kemampuan survival sel tumor di dalam  sirkulasi, dan ekstravasasi sel tumor serta kolonisasinya.

Abstract

Malignant peripheral nerve sheath tumor (MPNST) is a soft tissue sarcoma, which is difficult to distinguish from other spindle cell sarcomas. MPNST is hostile, with a high recurrence, and tends to metastasize hematogenously, especially to the lungs. A phase of the metastasis is a degradation of the extracellular matrix, where Matrix Metalloproteinase (MMP) plays an essential role in this process. Gelatinase-type MMP, MMP-2 and MMP-9, can degrade basal membrane and fibrillar collagen to open the invasion pathway. MMP-2 can degrade more collagen and non-collagen extracellular matrix than MMP-9. Therefore, the study aimed to see the relationship between MMP-2 overexpression and histopathological malignancy grading and other clinical prognostic variables. The study was conducted by immunohistochemical staining of MMP-2 in 39 cases, consisting of 19 cases of low-grade MPNST and 20 cases of high-grade MPNST. Subsequently, an analysis of the relationship between MMP-2 overexpression and the malignancy grading and clinical variables was performed, such as age, sex, and tumor size and location. MMP-2 overexpression was seen in 19 (95%) cases of high grade and three (15.8%) cases of low-grade MPNST (p 0.000). The study also found a significant relationship between MMP-2 overexpression and histopathology grading, which may be helpful to define the prognosis.

Abstract

BACKGROUND: Plasma cell neoplasm (PCN) is a neoplastic plasma cell proliferation which includes solitary bone plasmacytoma (SBP), extramedullary plasmacytoma (EMP) and multiple myeloma (MM). Bartl classifies the degrees of PCN as low, intermediate and high. The aim of this study is to find the correlation between tumor suppressor gene p53 (TP53) expression and Ki-67 proliferation with Bartl malignancy degree of PCN. Therefore earlier PCN diagnostic method to prevent the development of PCN into MM can be found.METHODS: Thirty-two PCN cases were classified into three groups based on Bartl’s degrees of malignancy. TP53 and Ki-67 immunohistochemical staining were performed on samples and the percentage of positivity was evaluated.RESULTS: The Bartl’s low degree of malignancy was found in 10 MM cases (31.2%), intermediate degree in  5 SBP cases (15.6%) and high in 2 SBP and EMP cases (6.2%). TP53 expression was obtainable at 4% of low, 16% of intermediate and 10% of high degree. There was a significant difference between TP53 expression in low and intermediate degree (p=0.004). Mean proliferation index of Ki-67 was 57% in low, 44.6% in intermediate, and 32.6% in high degree. There was no significant difference of Ki-67 proliferation indexes among the group (p=0.339).CONCLUSION: Increasing expression TP53 was in accord with Bartl’s degrees of malignancy, especially in low and intermediate degree, but there was no significant difference between Ki-67 proliferation index and Bartl’s degrees of malignancy.KEYWORDS: plasmacytoma, myeloma, TP53, Ki-67, Bartl classification

Abstract

There are many methods for development of mammae cancer animal model, one of which is chemical induction using carcinogenic agent, DMBA. This research aimed to explore the influence of dose and time regimens of DMBA on development of mammae carcinogenesis on Sprague dawley female rats. The first study was 50 rats treated with 20 mg/kg bw of DMBA orally for eleven times at twice a week. Morphological evaluations were conducted with mammae palpation for 15 weeks and then all of rats were sacrificed for collecting mammae organs for histological analysis using hematoxylin-eosin staining. The results showed that the first and the latest nodules appeared at the fourth-week and the fourteenth-week after ending DMBA induction, respectively, in which the most often nodule appearances were at the seventh-week. The number of nodule incidence and multiplicity were by 74% and 2 noduls/rat, respectively. Histological analysis of mammae glands determined that they fell under in Ductal Carcinoma Invasive (DCIV) category. The second study was 25 rats gavaged orally with DMBA at dose 20 mg/kg bw for five times every three days. After palpating for 15 weeks, the results showed that no nodule was observed but the histological analysis demonstrated developing of mammae gland carcinogenesis reaching about 60% Ductal Carcinoma Insitu (DCIS) and 40% Ductal Carcinoma Invasive (DCIV) stages. Based on the results of this study can be concluded that the dose and frequency of DMBA will affect the successful development of mammary gland carcinogenesis. In DMBA induction with low frequency, no data showed the incidence and multiplicity of tumor, but histopathologic level carcinogenesis can be distinguished. In DMBA induction with high frequency, incidence and multiplicity of tumor data can be obtained but can not be distinguished histopathologically.Keywords: DMBA, doses, carcinogenesis, mammae, histopatology

Abstract

ABSTRACTBreast cancer is the second mostly found disease after cervix cancer to Indonesian woman. Research on the effect of Ulva fasciata Delile and Turbinaria decurrensBory ethyl acetate extract feeding on the Adenocarcinoma mammaecell proliferation on C3H mice has beenconducted. Thirty experimental mice were devided into 6 treatment groups, i.e.: K-I, negative control; K-II, solvent control; K-III, given U. fasciata ethyl acetate extract dose of 41.32 mg/20 g BW (body weight); K-IV, given U. fasciataethyl acetate extract dose of 82.64 mg/20 g BW; K-V, given T. decurrensethyl acetate extract dose of 24.29 mg/20 g BW; and K-VI, given T. decurrens ethyl acetate extract dose of 48.59 mg/20 g BW. All treatments were given during 21 days. The mice body weight and tumor volume were measured and collected subsequently at every two days. Tumor doubling time was calculated at the end of research. Results showed that there wasa highly tumor growth inhibition on the K-IV shown by the high doubling time value. Tumor growth inhibition decreased according to the sequence as follows: K-IV (U. fasciata dose of 82.64 mg/20g BW), K-V (T. decurrensdose of 24.29 mg/20 g BW), K-VI (T. decurrensdose of 48.59 mg/20 g BW) which equal to K-II (solvent control), K-III (U. fasciatadose of 41.32 mg/20 g BW), K-I(negative control).

Abstract

Treatment for ovarian carcinoma is still far from optimal, animal models are still needed to study human epithelial ovarian cancer. Animal models of ovarian cancer are very important for understanding the pathogenesis of the disease and for testing new treatment strategies. Ovarian carcinogenesis models in mice have been modified and repaired to produce preneoplastic lesions and neoplastic ovaries that are pathogens resembling human ovarian cancer. Although spontaneous ovarian tumors in mice have been reported, some of the shortcomings of existing studies preclude their use as animal models of ovarian cancer. Because of this, many efforts have been made to develop animal models that are relevant for ovarian cancer. Experimental animal models are developed accurately to represent cellular and molecular changes associated with the initiation and development of human ovarian cancer. Accurate experimental models have significant potential in facilitating the development of better methods for early detection and treatment of ovarian cancer. Several animal models of ovarian cancer have been reported, including manipulation of various reproductive factors or exposure to carcinogens. The latest advance in ovarian cancer modeling is using genetically engineered mice.

Abstract

Background: In colorectal carcinoma (CRC), myofibroblast are the main component cells in tumour stroma which have an important role in the metastases process. The precentage between carcinoma and desmoplastic stroma known as carcinoma percentage (CP), can be used as an independent predictor metastases. D2-40/Podoplanin (PDPN) known as a spesific marker for lymphatic endothelial cell (LEC), which used to assess lymphatic microvessel density (LMVD) and lymphatic vessel invasion (LVI). This study aims to determine correlation and association between CP, LMVD and LVI with the metastases process to lymph node (LN).Method: CP assessment conducted on 44 samples of adenocarcinoma not otherwise specified (NOS) colorectal were divided into 22 cases with CP-High and 22 cases with CP-Low and examination D2-40/Podoplanin to assess LMVD and LVI. The statistical test is performed to find the correlation between CP and LMVD, as well as the relationship between CP, LVI and metastasis KGB.Results: There were a strong correlation between CP and LMVD intratumoral and peritumoral area with the negative correlation. There were a significant association (p=0,00) between LMVD (intratumoral and peritumoral area) with the LVI. There was a significant association between LVI and lymph node metastases (p = 0,03). Intratumoral area showed significant association with lymph node metastases (p = 0,04), whereas peritumoral area showed no significant association (p = 0,17).Conclusion: CP examination in histopathology specimen can be used to predict high/low rate of tumour cells metastases to the lymph node, based on a strong correlation between CP and LMVD.

Abstract

Latihan fisik anaerobik merupakan jenis latihan fisik berdurasi singkat dengan intensitas tinggi. Latihan fisik anaerobik tidak menggunakan oksigen sebagai sumber energi. Keadaan anaerobik menghasilkan asam laktat sehingga terjadi peningkatan kadar laktat darah.  Latihan fisik anerobik selain dilakukan secara kontinu, juga dapat dilakukan secara intermiten. Tetapi, hingga saat ini, sedikit penelitian mengenai latihan fisik anerobik yang dilakukan secara intermiten dan kadar laktat darah. Penelitian inibertujuan mengukur kadar laktat darah tikus Wistar setelah latihan fisik anaerobik intermiten selama 4 dan 12 minggu. Enam belas tikus Wistar dibagi menjadi dua kelompok,yaitu kelompok kontrol dan perlakuan anaerobik. Tiap kelompok perlakuan anaerobik dibagimenjadi dua subgrup,yaitu subgrup 4-minggu dan 12-minggu.Pada kelompok anaerobik dilakukan latihan fisik mengunakan treadmill T-6000 dengan kecepatan 35 m/menit selama 15 menit,dan setiap 5 menit diistirahatkan selama 90 detik. Pengukuran kadar laktat dilakukan dengan kit L-lactate (PAP) Randox (LC 2389). Kadar laktat darah kelompok perlakuan latihan fisik anaerobik intermiten sekama 4 minggu mengalamipeningkatan yang bermakna (p=O.OOO;p<0.05) dibandingkan kelompok kontrolnya (4.04±0.09mmoi/L dibanding 1.53±0.16mmoi/L). Kadar laktat darah post exercisekelompok perlakuan anaerobik intermiten selama 12 minggu mengalami peningkatan bermakna (p=O.OOO; p<0.05) dibandingkan kelompok kontrol12 minggu (3.91±0.03mmoi/L dibanding 2.53±0.03mmoi/L, A=1,38mmoi/L). Penelitian ini menunjukkan latihan fisik anaerobik intermiten selama 4 maupun 12 minggu menyebabkan perubahan kadar laktat darah tikus Wistar.
