Abstract

This study examines the role of Brachionus sp protein on the interleukin-6 pro-inflammatory immune system in Cantang grouper infected with Viral Nervous Necrosis (VNN). The purpose of this study was to determine the protein content of Brachionus sp. which has the potential as an antivirus and to find out the benefits of Brachionus sp. on the expression of Interleukin-6 as an indicator of increased fish body defense system against VNN. Testing of Brachionus protein by injection at doses of 35 µl, 105 µl, and 170 µl /150 gram Cantang grouper. The results showed that the lowest decrease was at a dose of 105 µl/150-gram Cantang groupers.  In addition, Brachionus sp. contains 3 protein bands with molecular weights of 122.73 kDa, 75.49 kDa, and 13.77 kDa.

Abstract

Sedimentasi merupakan salah satu dinamika yang terjadi di kawasan pesisir untuk mengetahui kondisi pantai tersebut. Pesisir perairan Lekok merupakan salah satu kawasan perairan terbuka yang memiliki dinamika perubahan pesisir sangat fluktuatif. Studi sebaran jenis sedimen yang dilakukan secara terstruktur pada lokasi ini digunakan untuk memberikan informasi mengenai proses transportasi dan pengendapan sedimen berdasarkan distribusi ukuran butir sedimen. Penelitian ini bertujuan untuk mengetahui ukuran butir sedimen secara horizontal di pesisir perairan Lekok. Penelitian ini dilakukan pada sembilan titik sampling dengan pengambilan sampel berupa sedimen dasar secara acak dengan menggunakan metode grab sampler. Selanjutnya dilakukan analisis lebih lanjut dengan metode gravimetri. Berdasarkan hasil penelitian diketahui bahwa karakteristik sedimen dalam Pesisir perairan Lekok umumnya didominasi oleh tiga jenis sedimen yaitu: lempung berpasir, lempung dan lempung lanau. Nilai sorting berada dalam kondisi well-sorted hingga kondisi moderately well-sorted di mana kondisi sedimen didominasi dengan well sorted. Nilai skewness berada pada nilai skewed positively dimana nilai menunjukkan hasil positif. Nilai kurtosis berada pada leptokurtic dimana sedimen dari stasiun 1 ke stasiun 9 homogen.

Abstract

Pencemaran mikroplastik di ekosistem perairan telah menjadi perhatian global karena dampaknya terhadap biota laut, termasuk ikan yang merupakan sumber pangan utama bagi manusia. Penelitian ini bertujuan untuk mengkaji pengaruh cemaran mikroplastik terhadap profil darah ikan bandeng (Chanos chanos) di perairan utara Jawa Timur. Sampel ikan bandeng diambil dari beberapa lokasi lokasi yang memiliki tingkat pencemaran mikroplastik berbeda. Analisis mikroplastik dalam jaringan ikan dilakukan menggunakan metode mikroskopis dan spektroskopi, sementara analisis profil darah mencakup pemeriksaan parameter hematologis seperti jumlah eritrosit, leukosit, kadar hemoglobin, dan hematokrit. Hasil penelitian menunjukkan adanya akumulasi mikroplastik dalam jaringan ikan bandeng di seluruh lokasi pengambilan sampel, dengan konsentrasi tertinggi ditemukan di area dengan aktivitas industri dan pemukiman padat. Pengaruh mikroplastik terhadap profil darah ikan terlihat dari peningkatan jumlah leukosit dan penurunan kadar hemoglobin yang signifikan pada ikan yang terpapar mikroplastik dalam jumlah tinggi. Hal ini mengindikasikan adanya respon stres dan inflamasi akibat paparan mikroplastik, yang berpotensi memengaruhi kesehatan ikan dan kemampuan adaptasinya terhadap kondisi lingkungan. Penelitian ini menunjukkan bahwa cemaran mikroplastik dapat berdampak negatif pada sistem hematologi ikan bandeng, yang berimplikasi pada kesehatan ekosistem dan keamanan pangan bagi manusia. Temuan ini mendukung perlunya tindakan mitigasi pencemaran mikroplastik di perairan pesisir serta pengembangan kebijakan pengelolaan lingkungan yang lebih efektif.

Abstract

Penelitian ini bertujuan untuk mengevaluasi kualitas perairan di Pantai Utara Jawa Timur berdasarkan parameter suhu, salinitas, pH, Total Dissolved Solids (TDS), Total Suspended Solids (TSS), oksigen terlarut (DO), amonia, nitrat, dan ortofosfat. Penentuan lokasi sampling dilakukan menggunakan metode purposive random sampling di tiga lokasi, yaitu Kabupaten Sidoarjo, Gresik, dan Lamongan. Hasil pengukuran menunjukkan suhu perairan berkisar antara 29,5–31,7 °C, sesuai dengan standar optimal untuk biota laut (28–32 °C). Salinitas dan pH (7,3–7,6) berada dalam batas wajar, mendukung kehidupan organisme akuatik. Nilai TDS di bawah 1000 ppm, sementara rata-rata TSS adalah 10,4 mg/L, jauh di bawah ambang batas 20–80 mg/L. Konsentrasi DO berkisar 4,7–6,97 mg/L, hampir memenuhi standar optimal > 5 mg/L. Kandungan amonia (0,007–0,088 mg/L) dan nitrat (0,051–0,095 mg/L) berada dalam batas aman. Konsentrasi ortofosfat (0,025–0,123 mg/L) melampaui ambang batas 0,015 mg/L di beberapa lokasi, kemungkinan disebabkan oleh limbah domestik. Hasil penelitian ini menunjukkan kondisi perairan Pantai Utara Jawa Timur mendukung ekosistem akuatik, meskipun potensi ancaman dari aktivitas manusia memerlukan pengelolaan lebih lanjut untuk menjaga keberlanjutan ekosistem

Abstract

Modern strategies incorporate preventive measures such as extracellular products, as soluble proteins with antigenic properties, as well as generating a broad immune response. The aim of research is to determine and analyze the effect of the extracellular product of E. tarda on enhancing the immunity of Nile tilapia (O. niloticus). The research design with treatment durations of d0, 3rd, 6th, 9th day, and control (A, B, C, D, K). ECP vaccination was injected intraperitoneally at a dose of 0.1 ml/fish. The parameters analyzed are the expression of IL-1, IL-6, and IFN-, phagocytic activity, leukocyte differential, total white blood cells, and relative percent survival. The results of phagocytosis activity were highest in treatment C at 84.18% 1.81. The highest leukocyte differential in treatment C was lymphocytes at 56% 2.00, monocytes at 28% 2.00, and 16% 0.00, with a total leukocyte count of 29 x 10^3 cells 2.64, and RPS values ranging from 83.33% to 100% over 9 days of post-vaccination maintenance. Then, post-vaccination, there was an increase in the expression levels of the cytokine genes IL-1 and IL-6, but there was no increase in the expression levels of the IFN- gene compared to the control in Nile tilapia. Abstract

Kamal Beach in the Madura Strait is suspected to have experienced heavy metal contamination due to ship recycling industry activities. Heavy metals have a negative impact on the aquatic environment, aquatic biota and even humans. This study aims to analyze the impact of heavy metal content in Kamal Beach, especially heavy metals Fe, Zn, and Cu as heavy metals found in the region on the histopathological aspects of the gill and liver organs of aquatic biota, namely mullet fish (Crenimugil seheli). The levels of heavy metals Fe, Zn, and Cu were analyzed using the AAS (Atomic Absorption Spectometry) method in accordance with SNI 6989.4:2009. Hiatopathological analysis of the gill and liver organs was carried out using the scoring method where from the results of the analysis it was known that the damage that occurred in the gill organs was in the form of edema, hyperplasia, lamellar fusion, and necrosis while damage to the liver organs included degeneration, necrosis, and congestion. Damage to the gill and liver organs of mullet fish is caused by the high content of Zn and Cu in water and sediment.

Abstract

Madura strait known as the second busiest shipping lanes in Indonesia. Human activities on the environment can influence the marine ecosystem derived from the household, fishery processing and capture fisheries. It can pollute coastal environments, one of which is heavy metal exposure such as Cd, Hg, Ca, As and Pb. These metals are greatly impacting the life of sea biota. The objective of this research, therefore, is to examine the hepatopancreas damage rate of shrimp Neocallichirus karumba due to the exposure of heavy metal Pb in Madura Strait. Method of research is by analyzing Pb content in water, sediment and shrimp by taking sample from 3 different stations. Hepatopancreas damage is then analyzed with SEM-EDX. Result of research indicates that at Station A, Pb pollution in water is the biggest and counted for 0.25 ppm, while those in sediment and shrimp are 5.85 ppm and 1.24 ppm. At Station B, Pb pollution in water is 0.19 ppm, whereas that in sediment is 5.51 ppm, but that in shrimp is 1.04 ppm. At Station C, Pb pollution in water counts for 0.18 ppm, and in sediment, it stands for 5.5 ppm but 0.02 ppm for shrimp. Result of analysis against hepatopancreas damage is explained as follows. At Station A, vacuolization is 20 % and Pb content in organ is 0.520 ppm. At Station B, the parameters are 10% and 0.196ppm. At Station C, it includes 15% and 0.173ppm. Organ damage is straightforwardly related to Pb content in water and sediment. Abstract

Pesticides are recognized as serious pollutants in the aquatic environment with the potential to cause genotoxic on the aquatic organism, especially fish. The micronucleus (MN) assay has been used to evaluate genotoxicity of many compounds in polluted ecosystems such pesticides. The aim of this study to determine genotoxic effect of methomyl-based pesticide on tilapia (Oreochromis niloticus). Fish were exposed to six different concentrations base on range finding test (0 ppm, 3.2 ppm, 4.2 ppm, 6.5 ppm, 8.7 ppm and 10 ppm) of methomyl-based pesticide. The micronucleus were collected from peripheral blood erythrocyte of fish after 96 h exposure. Peripheral blood samples smears were stained with Giemsa, MN frequencies were counted and statistically analyzed using one-way ANOVA. The result of this study showed after 96 hours exposed to methomyl-based pesticide, at concentration 0 ppm causes 0% mortality, at concentration to 3.2 ppm causes 30% mortality, at concentration 4.2 ppm causes 60% mortality, at concentration 6.5 ppm causes 70% mortality, at concentration 8.7 ppm causes 80% mortality, at concentration 10 ppm causes 100% mortality of fish test. Lethal Concentration 50 (LC50 - 96 hours) of methomy-base pesticide towards tilapia (O. niloticus) is 4.015 ppm. Through micronuclei assay during 96 hour exposure of methomyl-based pesticide, the result shows that frequencies of micronuclei in erythrocyte of fish test at concentration at 0ppm is 12‰, 18‰ and 16‰; at concentration at 3.2ppm is 33‰, 26‰ and 29‰; at concentration at 4.2ppm is 41‰, 38‰ and 46‰; at concentration at 6.5ppm is 68‰, 81‰ and 82‰; at concentration 8.7ppm is 133‰, 130‰ and 137‰; at concentration 10ppm is 163‰, 166‰ and 156‰. It revealed that methomyl-based pesticide exposure induced after 96 h significantly (P<0.05) increased genotoxic potentials simultaneous with increased concentration.Keywords: Genotoxic, Methomyl, Micronucleus Assay, Pesticide, Tilapia.

Abstract

Koi herpesvirus (KHV) is a virus that usually infects carp and koi causing mass mortality, as well as economic and social loss. The results of varied test are often found in its development. Allegedly, there are mutation or genetic variation of KHV in Indonesia. Based on these problems, this research aims to understand the genetic variation and kindship of KHV isolate in the carp (Cyprinus carpio) in Lombok Island of West Nusa Tenggara Province based on thymidine kinase sequence. Water quality of fish pond was checked. KHV clinical symptom was checked visually in each fish sample, then sacrifices the organ (gill) for PCR analysis and DNA sequencing. The research results show that clinical symptoms that appear are pale fish gill and bleeding on the surface of the body. The temperature in the research site shows an optimum range for KHV development. The results electrophoresis of carp suspected with KHV shows five positives out of a total six samples. Genetically, KHV isolates from Lombok Island of West Nusa Tenggara Province is identical with the GenBank isolate code KHV-J/U/I and Indo_0C05WNT2.Keywords: Carp, Koi herpesvirus, Phylogenetic, Thymidine kinase.

Abstract

The viral nervous necrosis (VNN) is betanodavirus of the family Nodaviridae with acute infection and associated with high levels mortality up to 100% of numerous larval-stage marine and freshwater fish species. The danger signals of viral infection that are capable of activating APCs (Antigen Presenting Cells), furthermore produced molecules protein receptors such as interferon and heat shock proteins. Stimulating interferon type I (IFN I) induce several antiviral molecules, further binding the actin cytoskeleton to reach the site of infection. This study demonstrated a correlation between increased levels of Hsp70 (heat shock proteins) and actin filamentous (β-actin) within invasion wild-type isolate of ssRNA VNN from Asian Seabass (L. calcalifer) juvenile. Furthermore, expression of Hsp70 and actin as an indicator or biomarker of stressed states in fish. The reverse transcriptase polymerase chain reaction (RT-PCR) method used to finding expression Hsp70 and β-actin. Whereas nested RT-PCR used for VNN genes. The up-regulation of Hsp70 was observed on the brain tissue higher than eye tissue of sample positive infected VNN. Whereas different expression of β-actin, β-actin receptor expression tends to be stable in the organs of the brain and eye, both invaded VNN or normal, while on the eye, the ratio slightly increased based on the intensity of the band. The research shows that altered expression of heat shock protein 70 and beta-actin gene receptors in the target organs that response to invasion VNN at Asian seabass (L. calcalifer) juvenile. Alteration expression of heat shock protein 70 and beta-actin can be used as an indicator or biomarker of stress cells, especially from VNN invasion at fish.
