Abstract

Vibriosis was a crucial problem and a primer infection causing mortality of humpback grouper.Virulence factor of vibrio, such as vili, is an adhesion molecule protein based on haemaglutination test toblood serum of humpback grouper. This research was aimed to characterize an adhesion molecule of viliprotein of V. alginolyticus on epithelial cells of Humpback grouper as haemaglutinin protein. Themethods are laboratory exploration by isolate epithelial cells of Humpback grouper, characterize a receptorand test vili protein to adhesion molecule that is epithelial cell of Humpback grouper. The result of proteinhaemaglutinin test to vili V. alginolyticus show that an adhesion molecule with a molecule weight of38.98 kDa is an adhesin vili V. alginolyticus and a haemaglutinin. The adhesion pattern of V.alginolyticus on epithelial cells of Humpback grouper exposed to protein haemaglutinin of vili show bothdiffuse and localize pattern. Research conclusion was the haemaglutinin protein of vili V. alginolyticuswas an adhesin having a role to both virulence process of bacteria and attachment process to receptor ofepithelial cells of Humpback grouper.Keywords: adhesion molecule, epithelial cell, vili, V. alginolyticus, C. altivelis

Abstract

Cantang grouper (Epinephelus sp.) is a fish with a high economic value and has been widely cultivated. VNN disase, caused by Betanodavirus infection, can lead to mass mortality of cantang grouper. Precautions considered effective in preventing Viral nervous necrosis (VNN) disease include vaccination. This study aimed to determine the effect of administering recombinant Chlorella vulgaris protein nanovaccine based on chitosan hybrid nanoparticles in increasing the immune response of cantang grouper infected with VNN. The study consisted of five treatments: K+ (cantang grouper infected with VNN), K- (healthy cantang grouper), T1 (healthy cantang grouper given 33 µl of nanovaccine and then challenged with VNN), T2 (healthy cantang grouper given 66 µl of nanovaccine and then tested with a VNN challenge), and T3 (healthy cantang grouper given 112 µl of nanovaccine and then tested with a VNN challenge). The parameters observed were haematology, nanovaccine Relative percent survival (RPS), and Survival rate (SR) levels. The study results showed that administering the nanovaccine increased the immune response of the fish, as indicated by an increase in haematology response, RPS, and SR in cantang grouper compared to the treatment without nanovaccine administration. Abstract

Viral Nervous Necrosis (VNN) and Vibriosis are contagious diseases that can affect grouper fish from the larval to adult stages and cause significant economic losses for farmers. The owner brought the sunuk grouper to BKIPM Mataram to determine the fish's health. Since the grouper arrived at the floating net cage, the owner reports it has sustained bodily injuries. The grouper is separated from the healthy fish and placed in the area where the sick fish are. During maintenance, the fish exhibits a decrease in appetite, wounds on the mouth, operculum, and body from the head to the tail, low mobility, and swimming upside-down. The physical examination revealed that the fish was swimming limply in an inverted position and had lesions on various body parts. According to virological and bacteriological laboratory tests, the sunuk grouper infected with Vibriosis and VNN can be affected by the condition of fish that have been sick since they first arrived. Also, stress, contact with infected fish, and a decreased appetite result in sunuk grouper contracting the disease. Due to a disease, a fish's immune system does not function optimally. This condition makes sunuk susceptible to several infectious diseases.

Intensive ponds often experience a decrease in water quality and cause ponds to bloom due to the fertility of the pond bottom due to the buildup of organic material originating from leftover feed, feces and the remains of dead shrimp. The aim of this research is to determine the relationship between water quality and phytoplankton abundance in intensive pond types with different densities in Bayeman Village, Probolinggo Regency. The research was carried out from January to March 2023. Samples were taken using the purposive sampling method in 6 different pond plots. Phytoplankton analysis was carried out at the Laboratory of the Sand Source Freshwater Fisheries Technical Implementation Unit and the Hydrobiology Laboratory of the Fish Resources Section of the Faculty of Fisheries and Marine Sciences. Measurement of in situ water quality parameters, namely temperature, pH, brightness, DO, TDS, light intensity, salinity and ex situ nitrate, phosphate and ammonia using spectrophotometric methods. The correlation test results of physico-chemical parameters related to phytoplankton abundance have a strong and heterogeneous relationship only with the orthophosphate parameter

Abstract

The Brantas River has many functions for both humans and the aquatic organisms in it. The type of fish that is often found in the Brantas River in Surabaya is the red bader fish (Barbonymus altus). Hematology is a sensitive parameter and can be used as a marker in assessing water conditions. The aim of this research is to analyze the hematological profile of red bader fish (Barbonymus altus) to assess the condition of the Brantas River, Surabaya. The research was carried out at 3 locations on the Brantas River in Surabaya with 3 sampling times with an interval of 1 month starting in January – March 2022. The parameters observed were water quality (temperature, TSS, TDS, pH, DO, BOD, ammonia and heavy metals Hg) and hematological parameters (erythrocytes, hemoglobin, hematocrit, leukocytes and micronuclei). Analysis of the relationship between water quality parameters and hematology using the Correspondence Correlation Analysis (CCA) method. The results show that there are water quality parameters that do not meet quality standards and there are abnormal hematological parameters. CCA analysis shows that there is a correlation between water quality and hematological parameters with moderate concentrations (erythrocytes, hemoglobin and hematocrit) and strong concentrations (leukocytes and micronuclei).

Abstract

The goldfish (Carassius auratus) has become one of the most sought-after ornamental fish today. The continuously rising market demand for goldfish as pets and decorative objects has driven the growth of the goldfish (Carassius auratus) farming industry. However, fish farmers still face difficulties in controlling the health of the fish and the quality of their water. The aquatic environment plays a crucial role in the growth of goldfish (Carassius auratus). Therefore, environmental management and fish health are essential in goldfish (Carassius auratus) farming. After the PKM Team analyzed the existing issues, the activities carried out to address these issues include the implementation of an Environmentally Friendly Recirculating Aquaculture System (RAS) in the goldfish (Carassius auratus) farming ponds. In this PKM program, water filtration will consist of mechanical filters, biological filters, chemical filters, and UV light. The results of the community service activities conducted by the PKM Team with their partners include increased knowledge, improved skills, increased production capacity, and increased profitability. Abstract

The Porong River is one of the rivers that faces high anthropogenic impacts, such as surfactant waste from detergents. The hematology of Javanese carp (Barbonymus gonionotus) serves as a sentinel organism that can be used as a biomarker for the waters of the Porong River. The aim of this study is to analyze the water quality status of the Porong River polluted with surfactants using the STORET method, as well as its impact on the hematology of Javanese carp using regression and correlation analysis. The research employed a descriptive survey method. The results of the study indicated that the Porong River was categorized as moderately polluted. The average highest surfactant concentration reached 7.2 mg/L, leading to an increase in TSS to 107.95 mg/L and a decrease in DO to 1.80 mg/L. Surfactants exhibited a negative correlation with erythrocytes (91.2%), hematocrit (47.7%), and hemoglobin (60.6%), indicating that the higher the surfactant concentration, the lower the number of erythrocytes, hematocrit levels, and hemoglobin levels in Javanese carp. Conversely, there was a positive correlation between surfactants and leukocytes (56.7%) and blood glucose (66.7%), suggesting that the higher the surfactant concentration, the higher the number of leukocytes and blood glucose levels in Javanese carp.

Abstract

Abstract Humpback grouper (Cromileptes altivelis) is a commodity to be cultivated because it has a high price, but the grouper highly susceptible to Vibriosis (Vibrio harveyi). The research objective was to see the function of immune cells of interleukin-4 (IL-4) are formed due to the induction of immunogenic proteins of Vibrio harveyi. Research procedures include Vibrio harveyi immunogenic protein isolation, SDS-PAGE electrophoresis, elektroelusi and dialysis, clinical exam of Vibrio harveyi immunogenic proteins, and immunohistochemical to look at immune cells (cells IL-4) with using a secondary antibody labeling of anti IL-4 biotin conjugate. The results showed that Vibrio harveyi protein found have immunogenic properties is 51.16 kDa. This protein is an adhesin protein. Protein 51.16 kDa was purified by electroelution and the protein will be used as a vaccine administered by intraperitoneal injection in grouper (Cromileptes altivelis) at a dose of 31.6 ug protein / 150 g of fish weight. In organ tissues of the brain, eyes and kidneys of induced fish by immunohistochemical examination, It was detected Immune cells (cells IL-4) using a secondary antibody labeling of anti IL-4 biotin conjugate. Tues formed IL-4 can be seen on the network grouper indicated by a change in tissue color with an inverted microscope examination of the golden brown color is formed on the IL-4 immune cells. It show the cross-reaction between 51.16 kDa immunogenic protein induced in fish with anti IL-4 biotin conjugate. The conclusions is the protein of Vibrio harveyi with molecular weight of 51.16 kDa is an immunogenic protein which able to induce formation of IL-4 in grouper.

Abstract

AbstractNanochloropsis oculata is one of kind green algae that overload in Indonesia ocean. The purpose of this research  was to know the potency antioxidant of Nannochloropsis oculata extract as in the Humback Grouper. The research was conducted by  experimently and used direct random sampling as design with 4 experiments and 3 repeats  i.e control fish, fish was  added Nannochloropsis oculata extract, Vibrio alginolyticus - infected fish, and Vibrio alginolyticus - infected fish that was added Nannochloropsis oculata extract. The result showed, fish that added by Nannchloropsis oculata extract made a different influence which was very clear to the decreasing of  free radical in the body. Free radical (malonaldehid) in the Vibrio alginolyticus -  infected fish was 4, 04 µg/100 mg which fish was added by Nannochloropsis oculata extract, the free radical level was decrease into 3, 29 µg/100 mg massa.
