Abstract

We collected the culturable heterotrophic bacteria from oligotrophic high mountain lake habitats and tested their capability to induce ice formation. Direct plating was carried out using low-nutrient medium at a temperature of between 3 and 4°C. As many as 84 isolates were recovered from glacial ice and natural biofilm growing on granite rocks surface. Six out of 84 isolates were capable of expressing the ice-nucleation phenotype. After autoclaving the cell suspension at 121°C for 15 min, isolate J78 was still able to retain the ability for ice formation. Heat-stable ice nuclei produced by ice-nucleating active bacteria have potential applications in biotechnology. Characterization of INA bacteria was performed employing live-dead Gram staining and molecular methods. Universal primers for Bacteria (S-D-Bact-0008-b-S-20 and S-D-Bact-1524-a-A-18) were used for PCR to amplify almost the full length of the 16S rRNA genes of selected INA isolates. Restriction fragment length polymorphism analysis resulted in 2 unique patterns, as represented by J43 and J83, respectively. Based on DNA sequencing of 16S rRNA gene, isolate J43 (GeneBank accession no. AJ864852) was closely related to Pseudomonas mephitica (99.2% sequence similarity) and Janthinobacterium lividum (99% similarity), whereas isolate J83 (GeneBank accession no. AJ864859) showed 100% sequence identity to Pseudomonas fluorescens.

Abstract

The aim of this study was to evaluate the effect of micro-encapsulated synbiotic application at different frequencies for luminous disease control in white shrimp (Litopenaeus vannamei). The luminous disease is caused by Vibrio harveyi. In this experiment, a synbiotic which was a combination of the probiotic Bacillus sp. NP5 RfR and the oligosaccharides from sweet potato (Ipomea batatas L.) jago variety was apllied. The synbiotic was encapsulated by spray drying method. The in vivo experiment was conducted by supplementing the shrimp’s diet with the micro-encapsulated synbiotic for 40 days. Treatments included the administration micro-encapsulated synbiotic in different frequencies i.e. once a week (A), twice a week (B), daily (C), and without micro-encapsulated synbiotic (control treatment). The control treatment consisted of positive (K+) and negative (K-) controls. After 30 days period, all of the shrimp were challenged by intramuscular injection of pathogenic V. harveyi RfR at a concentration of 106 CFU ml-1 except the negative control. The treatment C resulted in significantly higher survival rate (SR), specific growth rate (SGR), and immune responses than those of the controls, whereas the feed conversion ratio (FCR) was lower than the controls. In addition, the population of Bacillus sp. NP5 RfR and total bacterial count (TBC) in the intestines increased, whereas the population of V. harveyi RfR and the total vibrio count (TVC) were lower compared to the controls.

Abstract

In this study, fish feed supplemented by probiotic, prebiotic and synbiotic were applied to enhance the immune response and survival rate of the fish against Streptococcus agalactiae infection. Treatments used in this study, were as follows: feed containing 1% (v/v) of probiotic (C), feed containing 2% (v/v) of prebiotic (D) and feed containing mixed of 1% (v/v) of probiotic + 2% (v/v) of prebiotic (E, defined as synbiotic). Control fish (both positive, A; and negative, B; treatments) were fed by fish feed without containing neither probiotic nor prebiotic. In this study, test fish used monosex (all male) red tilapia with the average of body weight of 13,43±2,97 gram. The fish were fed by supplemented feed within first 30 days pre injection. After that, the treated fish were challenged by 109 CFU/ml of S. agalactiae. Nile tilapia fed by synbiotic (treatment E) had higher immune response (haemoglobin, neutrofil, and phagocyte indices) than control, but were not significantly different than those of treatment C, and D. After challenged test by S. agalactiae, treatment C, D and E resulted significantly higher resistance than that of control. The survival rate of fish fed by supplemented feed containing probiotic (C), prebiotic (D) and synbiotic (E) were 76%, 76% and 80%, respectively; higher than than positive control (50%). The results showed that addition of probiotic, prebiotic and synbiotic in fish feed could increase immune response and survival rate against S. agalactiae.Keywords : tilapia, probiotic, prebiotic, synbiotic, Streptococcus agalactiae

Abstract

Intensification of shrimp farming has led to problems of water quality and development of pathogenic bacteria. The excess feed and fecal deposited in the bottom of the pond undergo ammonification and result in excess of ammonia formation in pond water and sediment. The purposes of this research was to investigate the impact nitrifying bacteria application on the controlling of pathogenic Vibrio sp. bacteria and toxic nitrogen gasses. Twelve (12) transparent glass bottles (effective volume of 3 L) were used in this research. Tested probiotic was purchased in free market and producer claims to contain Nitrosomonas sp., Nitrosococcus sp., Nitrobacter sp., Bacillus sp., Aerobacter sp., and Pseudomonas sp. The tested media was sea water containing Vibrio sp., TAN, NO2 and NO3 of 54.07 ± 2.93 mg/L; 6.33 ± 0.17 mg/L; 2.43 ± 0.04 mg/L; and 0.46 ± 0.01 mg/L respectively. The treatment of probiotic was 0 mg/L as a control; 0.1 mg/L; 0.2 mg/L; and 0.4 mg/L with 3 replications. In regard to concentration of Vibrio sp., NH3 and NO2 gasses, treatment doses of 0.1 mg/L (A) resulted a save level within 4 days after treatment, but to more secure in the practical work for shrimp farm, the doses of 0.2 mg/L (B) is suggested.

Abstract

Super-intensive fish culture activities can lead the deterioration of water quality. Biofloc technology application can reduce the ammonia wastes and converts into bacterial biomass that can be used as a food source for fish. This study aimed to analyze the influence of heterotrophic bacterial cell concentrations in water and feed supplementation to improve the culture performances of catfish (C. gariepinus) on biofloc-based culture system. The experiments were conducted within 42 days consisted of five treatments, namely: (K-): system without biofloc, (K+) system with biofloc, (A) biofloc + L1k cells (102 CFU mL-1), (B) biofloc + L1k cells (104 CFU mL-1), and (C) biofloc + L1k cells (106 CFU mL*). The results showed that the growth performance of catfish cultured in biofloc system with the addition heterotrophic bacterial cell concentrations at 104 CFU mL-1 showed the best results compared to other treatments, with the value of survival rate was 92.67% ± 6.92, feed conversion ratio was 0.90 ± 0.07, and daily growth rate of 6.10% ± 0.09. Bacterial cells abundance were ranging from 104 CFU mL-1 up to 108 CFU mL-1, either with or without the addition of hetero-trophic bacterial cells. Abstrak Penerapan teknologi bioflok mampu mengurangi limbah amonia menjadi biomassa bakteri yang dapat dijadikan sebagai sumber pakan bagi ikan. Penelitian ini bertujuan untuk menganalisis pengaruh konsentrasi sel bakteri heterotrofik da-lam air dan pakan suplemen untuk meningkatkan pertumbuhan ikan lele dumbo (Clarias gariepinus) pada sistem berba-sis bioflok. Percobaan dilakukan dengan lima perlakuan, yaitu: (K-) Tanpa bioflok, (K +) Bioflok, (A) Bioflok + L1k (102 CFU mL-1), (B) Bioflok + L1k (104 CFU mL-1), dan (C) Bioflok + L1k (106 CFU mL-1). Empat hari sebelum dilakukan pemeliharaan (H-4) diinokulasikan bakteri heterotrofik sebanyak 10 ml m-3 air dengan konsentrasi sesuai perlakuan dan molase cair 10 g ke media pemeliharaan. Pemeliharaan ikan dilakukan selama 42 hari, dengan frekuensi pem-berian pakan 2 kali sehari dan tingkat pemberian pakan 5% dari biomassa ikan. Pemeliharaan ikan dilakukan selama 42 hari dengan frekuensi pemberian pakan 2 kali sehari dan tingkat pemberian pakan 5% dari biomassa ikan. Penambahan sel bakteri L1k ke dalam media budidaya dilakukan seminggu sekali sebanyak 10 ml m-3 dengan konsentrasi sel 102, 104, dan 106 CFU ml-1. Penambahan molase dilakukan setiap hari ke media bioflok dengan rasio C:N akhir sebesar 15:1. Kinerja pertumbuhan ikan yang diamati meliputi parameter kelangsungan hidup, pertumbuhan, rasio konversi pakan, populasi sel bakteri total dan sel bakteri L1k. Hasil penelitian menunjukkan bahwa kinerja produksi ikan lele dumbo pada perlakuan bioflok lebih baik dibanding tanpa bioflok. Penambahan sel bakteri heterotrofik L1k 104 CFUml-1 menunjukkan hasil terbaik dengan nilai tingkat kelangsungan hidup 92,67% ± 6,92, rasio konversi pakan 0,90 ± 0,07, dan laju pertumbuhan harian 6,10% ± 0,09. Kelimpahan sel bakteri total berkisar dari 104 CFU mL-1 hingga 108 CFU mL-1, baik dengan maupun tanpa penambahan sel bakteri heterotrof.

Abstract

The aimed of this research was to evaluate the effects of dietary probiotic microcapsules B.cereus P22 and S. lentus (L1k) at different dose and frequency on growth performance, immune response and resistance of African catfish infected with A. Hydrophila. Probiotics used in this study were B. cereus P22 and S. lentus L1k encapsulated by spray draying method. The research was carried out for 56 days with eight treatments and four replications. The treatments were K- (negative control), K+ (positive control), A (feed supplemented with 0,5% of microencapsulated probiotic, fedevery day), B (feed supplemented with 0,5% of microencapsulated probiotic, fed once every three days), C (feed supplemented with 1% of microencapsulated probiotic, fed every day), D (feed supplemented with 1% of microencap-sulated probiotic with an administration once every three days), E (feed with 2% of microencapsulated probiotic with an administration every day) and F (feed with 2% of microencapsulated probiotic with an administration once every three days). On day 42, all of the fish except K- were challenged by intramuscular injection of A. hydrophila (108 CFU ml-1). In 40 days after infection, there were no significant difference on survival rate (SR) between treatments (p> 0.05). Treatment E displayed the higher growth rate (4,54±0,02%), total probiotic B. cereus (P22) and S . lentus (L1k) (4.06± 0.09 log CFU g-1; 4.02±0.08 log CFU g-1) than other treatments; whereas treatment D showed the best feed conversion rasio(1.191±0.013), and treatment F offered the highest total bacterial count (7.11±0.53 log CFU g-1). An administration of 2% microencapsulated probiotic in every day frequency demonstrated the better growth performance, immune res-ponse and resistance of African catfish to A. hydrophila compare with other treatments. Abstrak Penelitian ini bertujuan untuk mengkaji dosis dan frekuensi pemberian mikrokapsul probiotik melalui pakan terhadap kinerja pertumbuhan, respons imun, dan resistensi ikan lele yang diinfeksi Aeromonas hydrophila. Penelitian ini meng-gunakan Bacillus cereus P22 dan Staphylococcus lentus L1k yang telah dienkapsulasi melalui metode spray draying. Penelitian ini dilaksankan selama 56 hari dengan delapan perlakuan dan empat ulangan, terdiri atas perlakuan K- (kon-trol negatif), K+ (kontrol positif), A (pakan+mikrokapsul probiotik dosis 0,5% frekuensi setiap hari), B (pakan+mikro-kapsul probiotik dosis 0,5% frekuensi tiga hari sekali), C (pakan+mikrokapsul probiotik dosis 1% frekuensi setiap hari), D (pakan+mikrokapsul probiotik dosis 1% frekuensi tiga hari sekali), E (pakan+mikrokapsul probiotik dosis 2% fre-kuensi setiap hari) dan F (pakan+mikrokapsul probiotik dosis 2% frekuensi tiga hari sekali). Ikan diuji tantang dengan A. hydrophila pada hari ke 42 dengan kepadatan 108 CFU ml-1 secara intramuscular (kecuali K- diinjeksi dengan phos-phate buffer saline). Setelah 40 hari pascainjeksi, laju sintasan tidak menunjukkan perbedaan yang signifikan antarperlakuan (p>0,05).Perlakuan E menunjukkan laju pertumbuhan (4,54±0,02%) dan total probiotik B. cereus (P22) dan S. lentus (L1k) (4,06±0,09 log CFU g-1; 4,02±0,08 log CFU g-1) tertinggi; sementara perlakuan D memberikan hasil nisbah konversi pakan terbaik (1,191±0,013), perlakuan F menunjukkan total bacterial count tertinggi (7,11±0,53 log CFU g-1). Hasil penelitian menunjukkan bahwa dosis 2% yang diberikan setiap hari memberikan hasil yang lebih baik dalam meningkatkan laju pertumbuhan, respons imun, dan resistensi ikan lele terhadap A. hydrophila.

Abstract

Vaccination is one of the most effective control measure in preventing Streptococcus agalactiae infection. In this study, vaccine coatings were prepared with certain biomaterials such as chitosan, skim milk, and maltodextrin at concentration 1 % or 10 %, and further freeze-dried. Vaccine selected (in vitro) by tests: viability cell, solubility, protein concentration and protein molecular weight. The result of in vitro test showed that chitosan coating at doses 1 % and 10 % were the best in solubility and protein concentration test. Vaccine coated with chitosan was administrated again by injection (in vivo) in nile tilapia (Oreochromis niloticus) before challenged by S. agalactiae. This test consists of six treatments and three replications, i.e. the fish were injected with liquid vaccine (A), Phospat Buffered Saline (PBS) (B), vaccine with chitosan coating 1 % (C), vaccine with chitosan coating 10 % (D), chitosan 1 % (E), and chitosan 10 % (F). Parameters were observed including survival rate, relative percent survival (RPS), antibody titre, and total leucocyte. The results showed that treatment C showed a significant difference (P < 0,05) in survival rate 92.22 ± 3.85 %, RPS 85.21 ± 7.20 %, and antibody titre highest result, but not significant (P > 0,05) to increase total leukocyte in experimental fish against S. agalactiae infection. In conclusion, vaccine freeze dry with chitosan coated 1 % is effective to improve immunity system of nile tilapia. Abstrak Vaksinasi merupakan salah satu tindakan paling efektif dalam pencegahan infeksi Streptococcus agalactiae. Pada penelitian ini, vaksin disalut dengan bahan biomaterial (kitosan, susu skim, dan maltodekstrin) dengan dosis 1% atau 10%, dan dikeringbekukan. Seleksi vaksin dilakukan secara in vitro dengan melihat uji viabilitas sel, kelarutan, kon-sentrasi protein, dan berat molekul protein. Hasil seleksi menunjukkan bahwa vaksin dengan penyalut kitosan 1% dan 10% berada pada urutan kedua terbaik pada uji kelarutan dan konsentrasi protein. Vaksin yang disalut kitosan diuji-kan kembali secara in vivo melalui injeksi pada ikan nila (Oreochromis niloticus) sebelum diuji tantang dengan bakte-ri S. agalactiae. Uji (in vivo) terdiri atas enam perlakuan dan tiga ulangan yakni perlakuan ikan diinjeksi dengan vaksin cair (A), Phosphat Buffered Saline (PBS) (B), vaksin yang disalut kitosan 1% (C), vaksin yang disalut kitosan 10% (D), kitosan 1% (E), dan kitosan 10% (F). Parameter yang diamati meliputi nilai sintasan, relative percent survival (RPS), titer antibodi, dan total leukosit. Hasil penelitian perlakuan C menunjukkan hasil berbeda nyata (P < 0,05) dengan nilai sintasan 92,22 ± 3,85%, RPS 85,21 ± 7,20%, dan titer antibodi C juga menunjukkan hasil paling tinggi, tetapi tidak berbeda nyata (P > 0,05) untuk meningkatkan jumlah leukosit dalam percobaan melawan infeksi bakteri S. agalactiae. Dengan demikian, vaksin kering beku yang disalut kitosan 1% efektif untuk meningkatkan kinerja imunitas ikan nila.

Abstract

Asian sea bass is one of economically important commodities in aquaculture. However, its culture often challenged by vibriosis infection which resulted in lower production. The study aimed to evaluate the effectiveness of the ambon banana plant powder application in feed as an effort to control the disease caused by Vibrio alginolyticus infection. Asian sea bass with total length of 7.48 ± 0.45 cm. Five different treatments were applied in this experiment, i.e. positive control, negative control, prevention, curative, and antibiotic application. The challenge test is carried out on the 15th day. Challenge test was performed with intramuscularly injection of V. alginolyticus at the cell density of 106 CFU/fish. The results showed the amount of feed consumption after the challenge test was highest in the curative and antibiotic control. However, these results were not significantly different (P>0.05) among preventive and curative treatments but were significantly different from controls (P<0.05). Fish treated with supplemented feed showed the highest total erythrocytes, total leukocytes, hemoglobin, respiratory burst, and lysozyme activity compared to controls. IL-1b gene expression increased after the challenge test with the highest level of expression in the curative treatment. It can be concluded that the administration of ambon banana plant powder (3 g/100 g diet) could control of V. alginolyticus infection in Asian sea bass and resulted 70.00% survival rate on prevention and 83.33% on curative treatments.

Abstract

Pathogenic Vibrio spp. have been implicated as being one of the major causes of the disease problems in penaeid shrimp culture. For many pathogens, the outcome of the interaction between host and bacterium is strongly affected by the bacterial population size. Efforts for rapid molecular detection is to isolate specific genes which was owned by the pathogenic luminescent Vibrio and used it as the marker molecular in the rapid diagnosis of the disease. Haemolysin and gyrase gene known as one of the specific genes possessed pathogenic bacteria including Vibrio. The objectives of this study were to determine the specificity of designed specific primer from local isolate. Specificity test was performed for haemolysin primer (IAVhF1 and IAVhR1), gyrase primer (IAGyrF1 and IAGyr2) and commercial detection kit. The specificity test comparing the PCR result of pathogenic Vibrio DNA template as positive control, and DNA template from non pathogenic luminescent Vibrio, as well as pathogenic non Vibrios. The result showed that at annealing temperature of 60°C, haemolysin primer was more specific in detecting pathogenic Vibrio for penaeid shrimp than that of gyrase primer and commercial detection kit.

Abstract

Koi Herpesvirus (KHV) is a malignant virus infecting the goldfish and koi in all stadia and cause mortality up to 95%. The purpose of this study was to determine the potency and efficacy of inactivated-vaccine in addition with adjuvant against KHV in koi fish. The viral propagation was done using a KF-1 cell line in 25 cm3 flask. The cultured virus was harvested on 12 days post inoculation, and then the harvested virus was inactivated with 0.1% formalin as inactivated-vaccine. Three hundred of test fish (10.38 ± 1.25 g) maintained in 126 L of plastic containers with aeration, and fed with pellets twice a day. After 14 days of adaptation, the fish were divided into five treatments (A= vaccine; B= vaccine + Complete Freund’s Adjuvant; C= vaccine + Incomplete Freund’s Adjuvant; K+= positive control, and K-= negative control) and each treatment has four replicates. Vaccine was given by injecting intramuscularly of 0.1 mL per fish. All fish were challenged by injecting intramuscularly of 0.1 mL of KHV virus with concentration of 104.58 TCID50/mL after 21 days post vaccination. The results showed that the B treatment had higher (P<0.05) values of hematocrit level, lysozyme activity, and titer of antibody compared with positive control. In addition, the survival of fish in B treatment also had the highest percentages and significantly different compared to other treatments (P<0.05). The conclusion of this research was the application of inactivated KHV vaccine in 0.1% formalin with the addition of Complete Freund’s Adjuvant through the injection dose 0.1 mL fish-1 in 104.58 TCID50/mL capable to enhance the immune responses and raised the optimal protection of KHV antibody in koi fish.
