Abstract

Latar belakangBiopsi aspirasi jarum halus/fine needle aspiration biopsy (FNAB) pada lesi tiroid mempunyai keterbatasan dalam mendiagnosis lesi indeterminate karena belum dapat ditentukan sifat biologiknya. Hasil FNAB menentukan penatalaksanaan pasien sehingga perlu dilakukan pulasan imunositokimia untuk meningkatkan akurasi. Cytokeratin 19 (CK19) merupakan penanda yang sensitif untuk karsinoma papiler tiroid (KPT), namun masih jarang dilakukan pada spesimen FNAB. Penelitian ini bertujuan untuk mengetahui peran imunositokimia CK19 dalam meningkatkan akurasi diagnostik lesi indeterminate pada FNAB nodul tiroid.MetodePenelitian ini merupakan desain potong lintang terhadap 42 kasus FNAB tiroid yang terdiri atas 11 kasus (26%) lesi jinak, 12 kasus (29%) atypia of undetermined significance (AUS), 10 kasus (24%) suspicious, dan 9 kasus (21%) KPT yang telah dilakukan operasi. Sampel didapatkan dari arsip Departemen Patologi Anatomik FKUI/RSCM tahun 2014-2015. Dilakukan pulasan imunositokimia CK19. Ekspresi dinilai menggunakan skor berdasarkan kurva receiver operating characteristic (ROC) dan sebagai baku emas adalah diagnosis histopatologik dari spesimen operasi.HasilPada 42 kasus yang dipulas dengan CK19 terdapat 22 kasus indeterminate yang terdiri atas 13 kasus yang terekspresi positif kuat dan 9 kasus yang menunjukkan ekspresi positif lemah/negatif. Dari 13 kasus yang terekspresi positif kuat, 11 kasus menunjukkan histopatologik ganas (berasal dari 7 kasus suspicious dan 4 kasus AUS) dan 2 kasus histopatologik jinak (berasal dari 1 kasus AUS dan 1 kasus suspicious). Sedangkan 9 kasus yang berekspresi positif lemah/negatif terdiri atas 8 kasus histopatologik jinak (berasal dari 6 kasus AUS dan 2 kasus suspicious) dan 1 kasus histopatologik ganas (berasal dari 1 kasus AUS). Dengan menggunakan tabel 2x2 diketahui bahwa pulasan imunositokimia CK19 pada lesi FNAB kategori indeterminate mempunyai sensitivitas 92%, spesifisitas 80%, nilai prediksi positif 85%, nilai prediksi negatif 89% dan akurasi diagnostik 86%.KesimpulanPulasan CK19 dapat digunakan sebagai penanda untuk membedakan karsinoma papiler tiroid dengan nodul jinak tiroid pada spesimen FNAB lesi indeterminate.

Abstract

Studi epidemiologi yang menghubungkan diet minyak ikan diet dengan risiko kanker kolorektal telah banyak dilaporkan walaupun hasinya tidak konsisten. Kebanyakan laporan menunjukkan adanya hubungan yang positif. Penelitian ini dilakukan untuk memahami efek penghambatan minyak ikan yang diperoleh dari sisa produk pengalengan ikan pada ekspresi penanda inflamasi dari preneoplasia kolorektal mencit yang diinduksi oleh azoxymetane (AOM) dan dekstran natrium sulfat (DSS). Dalam penelitian ini, mencit Balb/c diinduksi dengan AOM 10 mg/kg berat badan diikuti dengan pemberian 1% DSS selama seminggu. Minyak ikan diberikan secara oral dengan dosis 1,5 mg, 3 mg, dan 6 mg pada setiap mencit percobaan per hari. Ekspresi PGE2 diamati pada sel epitel kripta pada mukosa kolon. Pada bulan kedua, ekspresi PGE2 menurun dengan pemberian dosis sedang (3 mg/hari) dan tinggi (6 mg/hari) minyak ikan. Sedangkan pada bulan ketiga dan keempat, penurunan ekspresi PGE2 diamati karena pemberian minyak ikan dosis rendah, sedang dan tinggi (p=0,010 dan 0,005 m). Kesimpulannya, pemberian dosis sedang minyak ikan pada mencit yang diinduksi AOM/DSS telah menurunkan ekspresi PGE2 pada bulan kedua adalah dosis yang paling efektif.

Abstract

BackgroundUrothelial carcinoma is the most common bladder malignancy in men. Risk factors include smoking, chemicals exposure, radiation,schistosoma hematobium infection. P53 mutation is the most common mutation in urothelial bladder carcinoma that cause p53 proteinaccumulation which are detected through immunohistochemical examination. The aim of this study is to analyze the differencesbetween p53 expression in low and high grade urothelial and the carcinomas association of p53 expression with tumor stage.MethodsThis was a cross sectional study with 47 cases (22 cases of low grade and 25 cases of high grade) urothelial carcinoma in AnatomicalPathology Department Faculty Medicine of Universitas Indonesia/Cipto Mangunkusumo Hospital (FKUI/RSCM) in 2009-2017. Allcases stained with p53 antibody with immunohistochemical methods with positive cut off ≥20%.ResultsP53 positive expression are found in 33 samples, 20 cases of high grade and 13 cases in low grade; 22 cases of Nonmuscle invasivebladder cancer and 11 cases of Muscle invasive bladder cancer.ConclusionThe expression of p53 in low grade and high grade urotelial carcinoma of the bladder and its association with tumor stage showed nosignificant difference.

Abstract

ABSTRACTEvaluation of changes in antibody orders is a common exclusion in vaccination strategies because of the method ofanalysis. The fact that protection by antibodies produced by the Severe Acute Respiratory Syndrome Coronavirus-2(Sars-Cov-2) infection both naturally and through vaccines will decrease in less than one year, is a challenge for acountry with a large population like Indonesia to run its vaccination program. The main challenge is whether thevaccination strategy adopted will overcome the vaccine barrier and the race between the rate of vaccination and therate of viral mutation and antibody reduction. In addition to antibodies, the adaptive immune system is also run by Tcells that are included in the cell-mediated immune system (CMI) group. In patients with asymptomatic or mildsymptoms of Coronavirus 2019 (Covid-19), T and CMI responses are known to appear in some patients who are notknown to have been exposed to Sars-Cov-2 before. This evidence suggests that the adaptive catastrophe for SarsCov-2 has been acquired by ordering memory T cells and may last longer than previously thought. For countries withlarge populations, this will certainly help overcome the limitations of vaccines and the time needed to implement theirvaccination strategies.

Abstract

Breast cancer remains a significant cause of cancer-related mortality among women globally, highlighting the importance for preventive strategies targeting early molecular events. BRCA1 plays a critical role in maintaining genomic stability through DNA repair mechanisms. However, the potential of soybean phytochemicals to modulate BRCA1 activity at the molecular level, particularly through computational approaches, has not been extensively explored. This study aimed to evaluate the chemopreventive potential of soybean phytochemicals targeting the BRCA1 protein using an in silico approach. A total of 32 compounds were prepared and docked into the BRCA1 binding site using Autodock Tools 1.5.7, followed by interaction analysis and visualization, prediction of pharmacokinetic and toxicity profiles using SwissADME, pkCSM, and ProTox. The results showed that the top compounds exhibited binding energy ranging from -6.04 to -8.07 kcal/mol, which were lower than the reference compound. Interaction analysis revealed stable binding with key amino acid residues, including Met1775, Leu1839, and Lys1702 through hydrogen and hydrophobic interactions. Among the evaluated compounds, daidzin showed the most balanced profile in terms of binding affinity, interaction relevance, and favorable ADMET properties. This study provides a systematic in silico evaluation of soybean phytochemicals targeting BRCA1 and highlights their potential as candidates for breast cancer chemoprevention.

Mucin-1 expression in endometrial tissue of Macaca nemestrina during mid-luteal phase after controlled-ovarian hyperstimulation. Biodiversitas 22: 1927-1933. Endometrial receptivity is one of the factors for successful implantation in pregnancy. Controlled ovarian hyperstimulation (COH) is a required step of in vitro fertilization (IVF), one of the standard procedures to overcome infertility. Exogenous gonadotropin hormones from COH provoke the secretion of estrogen and progesterone from the ovaries in higher amounts. The supraphysiological environment could impact the endometrial receptivity of the implantation process. Mucin-1 (MUC1) can be used as a marker to indicate alterations in the endometrial tissue. Therefore, this study aimed to investigate the alteration of mucin-1 expression in endometrial tissue of Macaca nemestrina after COH protocol. This study used endometrium tissue of M. nemestrina embedded with paraffin as tissue blocks. The subjects were 15 female macaques in reproductive age (8-10 years old) with a history of producing offspring. These macaques were classified into four groups based on the COH protocols, which consist of administering gonadotropin-releasing hormone (GnRH) agonist and recombinant FSH (r-FSH) with dosages of 30 IU, 50 IU, 70 IU (intervention groups), and no r-FSH (control group). The stimulations were administered for 14 days during the mid-luteal phase. Moreover, tissue staining was done by using immunohistochemistry. Mucin-1 expression was analyzed manually for glandular and luminal compartments and automatically for the stromal compartment by Red Green Blue (RGB) measure plugin as a color image from ImageJ software. The expression of mucin-1 was counted semi-quantitatively as H-score. Mucin-1 expression differences in endometrial tissue were assessed by one-way ANOVA with a significant difference found in the glandular part and insignificant results found in luminal and stromal parts. Pearson correlation test was also performed to determine the relationship of steroidal hormones and mucin-1 expression in glandular and luminal compartments. Both correlations were statistically insignificant. However, a positive correlation was found with the glandular compartment and a negative correlation with luminal compartment. As the subject species, M. nemestrina is limited for research utilization in our country, small number of subjects becomes a limitation of this study. Further studies need to be conducted to deepen the understanding of the regulation of mucin-1 expression, particularly by the altered concentrations of steroidal hormones as a consequence of COH.
