Abstract

This research aims to study the anatomical structure of macroalgae. Sample of macroalga was collected by using Line Intercept Transect (LIT) method with sampling quadrat. Macroalgae samples were dried and separated from the roots, stems, leaves and receptacles (if present). Samples were cut transversely and longitudinally at each section and observed under binoculared microscope of Olympus CX with monitor. Based on the histology, anatomical structure of macroalgae species can be divided into two tissues from outside to inside, namely cortex and medullary cells. The cortex is composed of one layer or more. The cortex is the area between the epidermis and the central column. The medulla cells only have one layer which is the largest layer. Based on the observation result of cells type from observed spesies Eucheuma denticilatum, Gracilaria arcuata, Hydropuntia edulis, G. salicornia, Hypnea valentiae, Turbinaria deccurens, and T. ornata, it shows that cells of medulla become small toward cortex.Indonesian title:  Karakteristik anatomi jenis makroalga dari Pulau Bombuyanoi, Kabupaten Bolaang Mongondow Timur, Sulawesi Utara

Abstract

Algae Kappaphycus alvarezii is one of the leading commodities from fisheries and marine sectors in Indonesia. This algae species has many benefits in the fields of marine biology, chemistry, pharmacy, and pharmacology, as well as an excellent nutritional content for health. It is cultivated in the coastal waters of Minahasan Peninsula, especially in Buku Village, Belang District, Southeast Minahasa Regency. This study was carried out to analyze bioactive compound in the algae. Samples of 2-weeks old were taken directly from the cultivation area. Screening test was done qualitatively while an advanced test was done using LC-HRMS method. The results showed 4 bioactive compounds detected in the sample, they are alkaloids, tannins/phenolics, saponins, and flavonoids. From the advanced test, it was found 10 major compounds in the sample, i.e. (2-ethylhexyl) phthalate, phthalic acid, D-(+)-proline, maraniol, stigmatellin Y, Pentyl isopentyl phthalate, methyl thiofentanyl, phenylethanolamine, acetyl hexamethyl tetralin, and fucoxanthin.Indonesian title: Senyawa bioaktif pada alga Kappaphycus alvarezii dari perairan Belang, Kabupaten Minahasa Tenggara

Abstract

Bitung City is one of the centers of fishery production in Indonesia. Analysis of capture fisheries resources, especially at the Bitung Oceanic Fishing Port (BOFP) as a fish landing center in Bitung city, needs to be done scientifically in order to find out the need for sustainable capture fisheries. The purpose of this research is to analyze the potential of the types of superior commodities that have the opportunity to be developed. Data was collected using survey methods and in-depth interviews with fishermen and local stakeholders. Analysis of the potential of fish resources was carried out using the surplus production method. From the results of the study, it is found that there are 8 (eight) types of leading commodities in BOFP which can be developed with different potentials. They are: 1) Marlin with a sustainable potential of 159 tons, utilization rate of 44%; 2) Tongkol with sustainable potential of 10.485 tons, utilization rate is 58%; 3) Kembung with a sustainable potential of 183 tons utilization rate of 61%; 4) Big-eye tuna with sustainable potential of 111 tons utilization rate of 69%; 5) Selar with sustainable potential of 450 tons utilization rate of 85%; 6) Yellow Fin Tuna with a sustainable potential of 15.251 tons utilization rate of 86%; 7) Layang with sustainable potential of 7.152 tons utilization rate of 95%, and 8) Cakalang.Indonesian title: Analisis komoditas unggulan dari sumber daya perikanan tangkap di Pelabuhan Perikanan Samudera Bitung, Sulawesi Utara

Abstract

Isolation of marine fungi symbiont of green algae Bornetella sp as a producer of antibacterial compounds has been carried out. This study aims to obtain symbiont fungi from green algae Bornetella sp which produces antibacterial compounds. The symbiont fungus was isolated using the direct planting method. Screening for the antibacterial activity of pure symbiont fungi isolates against Escheriscia coli and Staphylococcus aureus, and was carried out by the agar diffusion method. The isolation results obtained 4 isolates, but the one with the most potential to inhibit the growth of the tested bacteria was MFALM2. Molecular characterization showed that the MFALM2 isolate was identified as Aspergillus nomius with a 100% closeness level. Penelitian ini bertujuan untuk memperoleh jamur simbion dari alga hijau Bornetella sp yang menghasilkan senyawa antibakteri. Isolasi jamur simbion dilakukan dengan metode direct planting. Skrining aktivitas antibakteri isolat murni jamur simbion terhadap bakteri uji Escheriscia coli dan Staphylococcus aureus dilakukan dengan metode difusi agar.  Hasil isolasi diperoleh 4 isolat, namun yang paling berpotensi menghambat pertumbuhan bakteri uji adalah MFALM2. Karakterisasi molekuler menunjukkan bahwa isolat MFALM2 teridentifikasi sebagai Aspergillus nomius dengan tingkat keeratan sebesar 100%.Kata kunci: isolasi; jamur lau; Aspergillus nomius; alga hijau; Bornetella sp.

Abstract

Supervision of fishing vessels is one of the efforts to prevent violations in the fishery. The level of compliance of fishing vessels in accordance with the laws and regulations is influenced by the performance of fishery supervisors who act as the spearhead of surveillance activities because of their authority in issuing operational letters. The purpose of this study is to optimize the supervision of marine resources and fisheries in North Minahasa so that it is expected that the level of violations that occur can decrease and state losses can be prevented. The methodology used is (1) Descriptive analysis and calculation of the average weighting of the value of each factor that affects the performance of supervisors; (2) Spearman Rank Analysis to determine the level of relationship of each factor that affects the supervisor's performance level; (3) Analytical Hierarchy Process (PHA) method to improve the performance of fishery ship supervisors in North Minahasa which can also be applied in supervisory units or other regional working areas. From the results of this study can be stated the level of performance of fisheries supervisors in North Minahasa is good. Improving the performance of fishery boat supervisors in North Minahasa can be done by improving the seriousness of performance that can be done by awarding; improved proficiency that affects the quality of inspection results and improvement of facilities and infrastructure that is able to improve the speed of inspection results.Keywords:  Ship obedience; Supervisory performance; StrategyAbstrakPengawasan kapal perikanan merupakan salah satu upaya mencegah terjadinya pelanggaran bidang perikanan. Tingkat ketaatan kapal perikanan yang sesuai dengan peraturan perundang-undangan dipengaruhi oleh kinerja pengawas perikanan yang berperan sebagai ujung tombak kegiatan pengawasan karena wewenangnya dalam mengeluarkan surat laik operasi. Tujuan penelitian ini adalah untuk mengoptimalkan pengawasan sumberdaya kelautan dan perikanan di Minahasa Utara sehingga di harapkan tingkat pelanggaran yang terjadi dapat menurun dan kerugian negara dapat tercegah.  Metodologi  yang digunakan  adalah  : (1) Analisis deskriptif   dan   perhitungan   rata-rata   bobot   nilai   masing-masing  faktor yang berpengaruh terhadap kinerja pengawas; (2) Analisis Rank Spearman  untuk mengetahui  tingkat hubungan dari masing-masing  faktor yang berpengaruh  terhadap  tingkat  kinerja  pengawas; (3) metode Proses Hierarki Analitik (PHA) untuk meningkatkan kinerja pengawas kapal perikanan di Minahasa Utara yang juga bisa diterapkan di Satuan Pengawas atau Wilayah Kerja daerah lain. Dari hasil penelitian ini dapat dinyatakan tingkat kinerja dari pengawas perikanan di Minahasa Utara adalah baik. Peningkatan kinerja pengawas kapal perikanan di Minahasa Utara dapat dilakukan dengan cara meningkatkan kesungguhan kinerja yang bisa dilakukan dengan pemberian penghargaan; peningkatan kecakapan yang berpengaruh terhadap kualitas hasil pemeriksaan dan peningkatan sarana dan prasarana yang mampu meningkatkan kecepatan hasil pemeriksaan.KATA KUNCI: Ketaatan kapal; Kinerja pengawas; Faktor Internal dan Eksternal

Abstract

 Community structure and interest of coral fish associated with artificial reefs in The Bay of Tahuna Sangihe Islands Regency is carried out in relation to increased coral fish production in Tahuna Bay. In addition to sandy waters and seagrass so that environmental engineering is pursued with artificial reefs. Artificial reefs are deployed in 5 locations with a depth of about 8 and underwater observations are carried out in data retrieval using the Underwater Visual Census (UVC) method. The data is processed to get a profile of the community structure of coral fish that can be associated with artificial reefs. The composition of coral fish species consists of target fish 9 families; 14 species, major fish 6 families; 7 species and fish indicator 1 family; 1 species. Indicator fish are generally eaters of the ends of young branch corals, branch corals have not grown in artificial reef structures. The diversity index (H') of the moderate category (+2.5) with a high, stable community, evenness (E) level or classified as a relatively high, uniformity (S) high. A low dominance index (C) indicates no species dominates. Mann-Whitney analysis about the level of fish interest to artificial reefs structure both in morning and evening showed no significant difference, while the highest acquisition of Important Values Indices of coral fish in the morning and evening, is Dascyllus trimaculatus by + 73,15. Generally, artificial reef indicates that the coral fish community was maximum developed yet, it was suspected that the coral fish community is still in the stage of adjustment or adaptation to the new environmental structure.Keywords: Community Structure; Artificial Reef; Associate Fishes; gulf TahunaAbstrakStruktur komunitas dan ketertarikan ikan karang yang  berasosiasi dengan terumbu buatan di Teluk Tahuna Kabupaten Kepulauan Sangihe dilakukan dalam kaitan dengan peningkatan produksi ikan karang di Teluk Tahuna. Selain karena perairan berpasir dan lamun sehingga diupayakan suatu rekayasa lingkungan dengan penempatan terumbu buatan. Terumbu buatan diletakkan (deploy) di 5 lokasi dengan kedalaman sekitar 8 dan dilakukan pengamatan bawah air dan pengambilan data menggunakan metode Underwater Visual Census (UVC). Data diolah  untuk mendapatkan profil struktur komunitas ikan-ikan karang yang dapat berasosiasi dengan terumbu buatan. Komposisi jenis ikan karang terdiri dari ikan target 9 family; 14 spesies, ikan mayor 6 family; 7 spesies dan ikan indikator 1 family; 1 spesies. Ikan indikator yang umumnya pemakan ujung karang cabang muda, karang cabang belum bertumbuh di struktur terumbu buatan. Indeks keanekaragaman (H’) kategori sedang (+ 2,5) dengan kemerataan (E) tergolong tinggi, komunitas stabil, keseragaman (S) tinggi. Indeks dominansi (C) rendah mengindikasikan tidak ada spesies mendominasi. Hasil Analisis Mann-Whitney terhadap tingkat ketertarikan ikan terhadap terumbu buatan baik pagi dan sore hari menunjukkan tidak ada perbedaan yang nyata artinya terumbu buatan mampu menarik ikan karang untuk berlindung dan mencari makan, sedangkan perolehan tertinggi Index Nilai Penting ikan karang saat pagi dan sore hari yaitu Dascyllus trimaculatus sebesar 72.04% dan 74.27% Secara umum menunjukkan terumbu buatan belum berkembang maksimal, di duga komunitas Ikan karang masih dalam tahap penyesuaian atau adaptasi dengan struktur terumbu buatan di lingkungan yang baru.Kata kunci: struktur komunitas; terumbu buatan; ikan asosiasi; teluk tahuna

Abstract

Sharks are a group of cartilaginous fish that are vulnerable to overfishing.  Genetics approaches play an important role in shark conservation. Shark fishing has become the main activity of fishermen in several areas, including in North Sulawesi. This research is focused on the molecular aspects and conservation status of shark species obtained from the shark fin trade in Manado, North Sulawesi. COI gene was amplified using Fish BCL5 (for) and HCO219 (rev) primers. Nucleotide sequences of each sample were aligned with the closest sequences in the GenBank database using the BLAST (Basic Local Alignment and Search Tool) method. The conservation status of the shark species is carried through the IUCN (International Union for the conservation of nature red list) Red list website. Molecular identification results showed that the shark’s fins from Manado had high similarity with Carcharhinus falciformis (HM1 and HM2) and Carcharhinus melanopterus (HM3). According to IUCN red list data, the C. falciformis and C. melanopterus were categorized as vulnerable to extinction (VU).Keywords:  Molekuler; COI; Shark; Manado and IUCN Red list . AbstrakHiu merupakan kelompok ikan bertulang rawan yang sangat rentan terhadap dampak penangkapan secara berlebihan.  Informasi terkait genetik hiu  yang semakin terancam populasinya sangat berperan penting dalam upaya konservasi hiu. Penangkapan hiu telah menjadi aktivitas utama nelayan di beberapa daerah, termasuk di Sulawesi Utara. Penelitian ini difokuskan pada aspek  molekuler dan penentuan status konservasi spesies hiu menggunakan sampel sirip yang di dapatkan  dari perdagangan sirip  hiu, di kota Manado.  Amplifikasi gen COI dilakukan dengan menggunakan primer Fish BCL5 (for) dan HCO219 (rev). Sekuens nukleotida masing-masing sampel disejajarkan dengan nukleotida terdekat yang ada dalam database genbank menggunakan metode BLAST (Basic Local Aligment and Search Tool)  Penentuan status konservasi dilakukan melalui penelusuran spesies rujukan di situs IUCN (International Union for the Conservation of Nature) Red list. Hasil identifikasi molekuler menunjukan bahwa sampel sirip hiu dari  kota Manado  memiliki kemiripan yang tinggi dengan spesies : Carcharhinus falciformis  (HM1 dan HM2) dan C. melanopterus (HM3).  Menurut data IUCN Red list, C. falciformis dan C. Abstract

Sea cucumbers are one of the marine biotas that contain bioactive compounds that have the potential as antibacterial ingredients. The purpose of this study was to perform antibacterial testing on fractions of the extract of the sea cucumber H. atra meat and to conduct a zoochemical analysis to determine the content of bioactive compounds that can inhibit bacterial growth. Antibacterial testing using disc diffusion method against Staphylococcus aureus and Escherichia coli bacteria. The results showed that the methanol extract of sea cucumber flesh could inhibit the growth of both types of test bacteria. In antibacterial testing for S. aureus, the ethyl acetate fraction was 11.8 mm, the n-hexane fraction was 7 mm, and the methanol fraction was 8.6 mm, while for E. coli the ethyl acetate fraction was 10.88 mm, the n-hexane fraction was 7 mm, and 8.6 mm methanol fraction. The compounds contained in the ethyl acetate fraction of sea cucumber H. atra flesh extract are alkaloids, flavonoids, steroids, and saponins which are compounds that can inhibit bacterial growth with their respective working mechanisms.Keywords: Disc diffusion, Extraction, Fractionation, Sea cucumber (H.atra), Zoo-chemical, AbstrakTeripang merupakan salah satu biota laut yang memiliki kandungan senyawa bioaktif yang berpotensi sebagai bahan antibakteri. Tujuan penelitian ini melakukan pengujian antibakteri pada fraksi-fraksi dari ekstrak daging teripang H. atra dan melakukan analisis zookimia untuk mengetahui kandungan senyawa bioaktif yang dapat menghambat pertumbuhan bakteri. Pengujian antibakteri menggunakan metode difusi cakram terhadap bakteri Staphylococcus aureus dan Escherichia coli. Hasil penelitian menunjukkan bahwa ekstrak metanol daging teripang memiliki dapat menghambat pertumbuhan kedua jenis bakteri uji. Pengujian antibakteri terhadap S. aureus fraksi etil asetat sebesar 11,8 mm, fraksi n-heksan 7 mm, dan fraksi metanol 8,6 mm sedangkan untuk bakteri E. coli fraksi etil asetat sebesar 10,88 mm, fraksi n heksana 7 mm, dan fraksi metanol 8,6 mm. Abstract

Taman Nasional Bunaken (TNB) is located in North Sulawesi Province, Republic of Indonesia. TNB offers an exploration experience of marine tourism such as snorkeling and diving. This study was conducted to determine the statistics and correlation of weather parameters on hydrodynamics in the waters of the small island of TNB. This information can be used by marine tourism users. The weather data studied are wind direction and speed. The hydrodynamic conditions studied were wave height and ocean currents. Respondent surveys were also conducted on tourist users at the research sites. The wind direction for the period January - April is dominant from the Northeast, May - September is dominant from the East to the South, October tends to be evenly distributed from the East to the West, and November - December is dominant from the West. Significant wind speeds that occur in January, February, and March range from 18 – 36 knots. The average wave height shows a significant value in January – March, tends to be lower in April – June, fluctuates in July – November, and increases again in December. The maximum current velocity at a sea depth of 16.5 m is stronger than the maximum speed at a depth of 0.5 m and 8.0 m in January – April, and May August. The correlation value of wind speed and sea wave height is significant at 0.96 and the correlation between wind speed and the current speed is 0.74. These results indicate that there is a strong -very strong relationship between parameters. The survey results show that wind speed, wave height, and ocean currents have the same pattern. January, February, November, and December are the months of extreme or bad weather and hydrodynamic conditions.Keywords: Taman Nasional Bunaken, Weather, Hydrodynamics, CorrelationAbstrakTaman Nasional Bunaken (TNB) terletak di Provinsi Sulawesi Utara, Negara Republik Indonesia. TNB  menawarkan pengalaman eksplorasi suasana wisata bahari seperti snorkling dan diving. Penelitian ini dilakukan untuk mengetahui statistik dan korelasi parameter cuaca terhadap hidrodinamika di perairan pulau kecil TNB. Informasi tersebut dapat digunakan oleh pengguna wisata bahari. Data cuaca yang diteliti adalah arah dan kecepatan angin. Kondisi hidrodinamika yang diteliti ialah tinggi gelombang dan arus laut. Survei responden juga dilakukan kepada pengguna wisata di lokasi penelitian. Arah angin periode Bulan Januari - April dominan dari arah Timur Laut, Bulan Mei – September dominan dari arah Timur hingga Selatan, Bulan Oktober cenderung merata dari arah Timur hingga Barat dan Bulan November – Desember dominan dari arah Barat. Kecepatan angin signifikan terjadi pada bulan Januari, Februari, dan Maret berkisar antara 18 – 36 Knot. Rata-rata tinggi gelombang menunjukan nilai signifikan pada bulan Januari – Maret, cenderung merendah pada bulan April – Juni, berfluktuasi pada bulan Juli – November dan meningkat kembali di bulan Desember. Kecepatan maksimum arus laut di kedalaman 16.5 m lebih kuat jika dibandingkan kecepatan maksimum di kedalaman 0.5 m dan 8.0 m pada bulan Januari – April serta bulan Mei – Agustus. Nilai korelasi kecepatan angin dan tinggi gelombang laut signifikan sebesar 0.96 serta korelasi kecepatan angin dan kecepatan arus sebesar 0.74. Hasil tersebut menunjukkan antar parameter memiliki hubungan yang kuat - sangat kuat. Hasil survei menunjukan kecepatan angin, tinggi gelombang dan arus laut mempunyai pola yang sama. Bulan Januari, Februari, November, dan Desember merupakan bulan-bulan terjadinya kondisi cuaca serta hidrodinamika ekstrem atau buruk.Kata kunci: Taman Nasional Bunaken, Cuaca, Hidrodinamika, Korelasi

Abstract

In the last few decades, coral reef ecosystem has been under significant pressure due to anthropogenic impacts and naturally driven factors. The coral reef ecosystem is unique because it has very high biodiversity and many organisms depend on its existence. Every organism in a coral reef ecosystem uniquely contributes to how coral reef communities survive and function. However, coral reef ecosystems are also vulnerable to pressures that cause degradation, interfering with the ecosystem balance. This study aims to monitor the ecological condition of coral reefs and the abundance of reef fish to see how the MPA program in Bahoi village helps the coral reef ecosystem from degradation. The data was collected using Line Intercept Transect (LIT) method, which was drawn 70 meters transect line horizontally at a depth of 3-5 meters. The same transect line was also used to collect data about reef fish by using a Visual Census. The observation distance length is 2.5 meters to the left and right of the transect line. Results of the study showed that the coral cover of MPA Bahoi Village is in the Fair category and the abundance of reef fish in the MPA area is in the Moderate category.  Keywords: Coral Cover; Coral Reef; Marine Protected Area (MPA); Monitoring; Reef Fish
