Abstract

The coastal area of Sarawet Village, East Likupang District, North Minahasa Regency, North Sulawesi Province has a mangrove forest covering an area of 379 hectares. Apart from being a living habitat for marine biota, this large area of mangrove is also an ecotourism destination. The purpose of this research is to determine the suitability of mangrove lands as ecotourism areas; Determine the ecological carrying capacity of the area for mangrove ecotourism activities; Assess public perceptions regarding the benefits of mangrove ecosystems and their potential to be developed as ecotourism destinations. The research method is a survey describing ecological, socioeconomic, institutional, and infrastructure conditions. Data collection is grouped into primary data and secondary data. Primary data collection was carried out through direct observation in the field by measuring the ecological potential of mangroves, visual observation of biota, and information from the community obtained directly at the research location through structured interviews with respondents. Based on the results of the study, the Tourism Suitability Index (IKW) value for the Sarawet Village mangroves was 2.73, which indicates the Very Suitable category for being developed as a mangrove river ecotourism area, as well as the public's perception of the benefits of the mangrove ecosystem and its potential to be developed as an ecotourism destination is very good. so it is hoped that this can improve the welfare of the existing community. Keywords: Mangroves, Ecotourism, Sarawet Abstrak Kawasan pesisir Desa Sarawet Kecamatan Likupang Timur Kabupaten Minahasa Utara Provinsi Sulawesi Utara memiliki hutan mangrove seluas 379 hektar, keberadaan mangrove yang luas ini selain menjadi habitat hidup bagi biota laut juga sebagai tujuan ekowisata. Tujuan dan manfaat penelitian ini adalah menentukan kesesuaian lahan mangrove sebagai kawasan ekowisata; Menentukan daya dukung ekologis kawasan untuk kegiatan wisata mangrove ekowisata; Mengkaji persepsi masyarakat tentang manfaat ekosistem mangrove dan potensinya untuk dikembangkan sebagai tujuan ekowisata. Metode penelitian yang digunakan yaitu melakukan survei untuk mendeskripsikan kondisi ekologi, sosial ekonomi, kelembagaan dan infrastruktur. Pengumpulan data dikelompokkan menjadi data primer dan data sekunder. Pengumpulan data primer dilaksanakan melalui pengamatan langsung di lapangan dengan melakukan pengukuran potensi ekologi mangrove, pengamatan biota secara visual dan informasi dari masyarakat diperoleh langsung di lokasi penelitian melalui wawancara secara terstruktur dengan responden. Berdasarkan hasil kajian diperoleh nilai Indeks Kesesuaian Wisata (IKW) mangrove Desa Sarawet yakni sebesar 2,73 yang menunjukkan kategori Sangat Sesuai untuk dapat dikembangkan sebagai Kawasan ekowisata sungai mangrove, serta persepsi masyarakat tentang manfaat ekosistem mangrove dan potensinya untuk dikembangkan sebagai tujuan ekowisata adalah sangat baik sehingga diharapkan hal tersebut dapat meningkatkan kesejahteraan masyarakat yang ada. Abstract

Indonesia is a tropical country with high marine biodiversity, including sea cucumber. The high demand for sea cucumber has led fishermen to catch sea cucumber regardless of the type or size suitable for harvesting. The lack of cultivation and regulations governing the protection of sea cucumbers has caused some species to be endangered. Sea cucumber species with high selling prices are increasingly difficult to find in West Likupang District. This study aims to identify strategies for managing sea cucumber fisheries sustainably and provide policy recommendations for their sustainable management in West Likupang District. This study used a survey method and then analyzed it with a SWOT analysis (strengths, weaknesses, opportunities, threats). The SWOT analysis reveals several strategies for achieving sustainable sea cucumber fisheries: legislation, conservation efforts, aquaculture development, regulated fishing, proper post-harvest handling, and educating fishermen on regulations. Keywords: Management, regulation, conservation, sea cucumber, SWOT Abstrak Indonesia merupakan negara tropis yang mempunyai keanekaragaman hayati laut yang tinggi, salah satunya ialah sumber daya trepang (Holothuridea). Permintaan pasar yang tinggi terhadap trepang, menyebabkan nelayan menangkap trepang tanpa mengindahkan jenis dan ukuran yang layak panen. Belum adanya budi daya dan regulasi yang mengatur perlindungan terhadap trepang menyebabkan beberapa spesies sudah terancam punah. Spesies trepang dengan harga jual yang tinggi makin sulit ditemukan terutama di Kecamatan Likupang Barat. Penelitian ini bertujuan untuk menentukan strategi pengelolaan sumber daya perikanan trepang, untuk mencapai perikanan trepang yang berkelanjutan, dan membuat rujukan atau rekomendasi kebijakan untuk pengelolaan sumber daya perikanan trepang (Holothuroidea) yang berkelanjutan secara umum Di Provinsi Sulawesi Utara, khususnya di Likupang Kabupaten Minahasa Utara. Penelitian ini menggunakan metode survei kemudian dianalisis dengan analisis SWOT (kekuatan, kelemahan, peluang, dan ancaman). Berdasarkan hasil analisis, terdapat beberapa strategi yang dapat dikembangkan untuk mencapai perikanan trepang yang berkelanjutan, yakni: regulasi berupa peraturan perundangan, upaya konservasi, pengembangan usaha budidaya, penangkapan yang terukur, penanganan pasca panen/penangkapan dari alam, dan sosialisasi kepada nelayan terkait peraturan Kata kunci: Pengelolaan, regulasi, konservasi, trepang, SWOT

Abstract

We used the Nansen Bottle Sampler to collect water samples in the deepsea area, ranging from 150 meters to 175 meters in six water points around Manado Bay, to test the quality of e-DNA water samples to detect target species in the fishing area. Therefore, the basis of the case study method with a sampling technique was carried out on July 29 2023 using Power Water Sterivex Kits, water samples were then stored at -250C and were then taken to TBRC, University of the Ryukyus for further laboratory works, such as; eDNA extract, eDNA quality testing, 1st and 2nd PCR and Electrophoresis eDNA analysis processes using MiFish-U primers with a target of 163–185bp and 375 bp, following the MiFish protocol.  Based on the results of the eDNA extract solution, it is known that the quality of eDNA from the 6 sampling sites locations ranged between 2.8 µg/mL – 4.4 µ g/mL, which means a good quality of eDNA. Moreover, it showed that the presence of DNA fragments at Kappa 60◦C Gelelectrophoresis 1st-PCR, 12S rRNA gene product (163–185bp), and Kappa 60◦C and 65◦C Geklelectrophoresis 2nd-PCR Products according to the target amplicon 375 bp. This means we can conduct the next step, the PCR sequence analysis.  Then, eDNA quality testing, 1st and 2nd PCR, and Electrophoresis of e-DNA analysis process were using MiFish-U F/R primers with a target of 375 bp, it is known that the concentration of Nanodrop from the 6 sampling locations ranges between 2.8 µg/mL – 4.4 µg/mL while the core or quality eDNA ranged from 1.56 µg/mL – 2.50 µg/mL. Based on identification results, five types of species were detected; Myctophum lychnobium, Selar crumenophthalmus, Photonectes sp., Oreochromis sp. Thunnus obesus and Homo sapiens were generated using eDNA metabarcoding on the mitochondria genome database MitoFish. Selanjutnya dasar metode studi kasus dengan teknik pengambilan sampel secara sampling dilakukan pada tanggal 29 Juli 2023 menggunakan Power Water Sterivex Kits, sapel air disimpan pada -250C yang selanjutnya dibawa ke TBRC, University of the Ryukyus untuk pnelitian laboratorium lanjutan seperti ekstrak eDNA, Pengujian kualitas eDNA, 1st and 2nd PCR dan Elektrophoresis proses analisis eDNA menggunakan primer MiFish-U dengan target 375 bp, mengikuti MiFish protokol. Berdasarkan hasil pengujian larutan ekstrak eDNA diketahui bahwa kualitas eDNA dari 6 titik lokasi sampling berkisar antara 2.8 ng/mL – 4.4 ng/mL dan menunjukkan adanya fragment DNA pada Kappa 60◦C Geklelectrophoresis 1st-PCR Produk 12S rRNA gene (163–185bp, dan Kappa 60◦C dan 65◦C Geklelectrophoresis 2nd-PCR Produk sesuai amplikon target 375 bp. Hal ini berarti dapat dilanjutkan pada tahap analisis sekuens PCR. Pengujian kualitas eDNA, 1st and 2nd PCR dan Elektrophoresis proses analisis eDNA menggunakan primer MiFish-U F/R dengan target 375 bp, diketahui bahwa konsentrasi Nanodrop dari 6 titik lokasi sampling berkisar antara 2.8 µg/mL – 4.4 µg/mL sedangkan kemurnian atau kualitas eDNA berkisar antara 1.56 µg/mL – 2.50 µg/mL. Hasil identifikasi menyatakan lima jenis spesies terdeteksi; Myctophum lychnobium, Selar crumenophthalmus, Photonectes sp., Oreochromis sp. Thunnus obesus, Homo sapiens dihasilkan dengan menggunakan eDNA metabarcoding pada MitoFish database genom mitokondria. Abstract

The conservation of tawes fish (Barbonymus gonionotus) is carried out through aquaculture and restocking, where seed transportation is a crucial factor affecting survival rates. Mishandling during transport can increase stress, metabolism, and mortality risk. Reducing stress during transport can be achieved by using natural anesthetic agents such as clove oil (Eugenia aromatica) and low-temperature regulation. This study aims to analyze the effects of clove oil dosage and temperature on the induction time and recovery of tawes fish seeds. The experiment was designed using a completely randomized design (CRD) with two factors: clove oil dosage (0 ppm, 5 ppm, 10 ppm) and temperature (16–19°C and 24–27°C), with 18 experimental units (3 replications). Data were analyzed using ANOVA (JMP-SAS). The results showed that combining clove oil and temperature significantly affected induction and recovery times. The 10 ppm clove oil treatment at 16–19°C resulted in the fastest induction time, while the 0 ppm treatment at 24–27°C led to the fastest recovery. The interaction between these factors demonstrated that clove oil significantly influenced the fish's response to temperature changes. Keywords: tawes fish seed, transportation, temperature, clove oil, induction, sedative.   Abstrak   Pelestarian ikan tawes (Barbonymus gonionotus) dilakukan melalui budidaya dan restocking, di mana transportasi benih menjadi faktor krusial yang memengaruhi tingkat kelangsungan hidupnya. Kesalahan dalam penanganan dapat meningkatkan stres, metabolisme, dan risiko kematian benih. Upaya menekan stres selama transportasi dilakukan dengan penggunaan agen pembius alami, seperti minyak cengkih (Eugenia aromatica) dan pengaturan suhu rendah.Penelitian ini bertujuan menganalisis pengaruh dan dosis minyak cengkih serta suhu terhadap waktu induksi dan pemulihan (recovery) benih ikan tawes. Percobaan menggunakan rancangan acak lengkap (RAL) dengan dua faktor: dosis minyak cengkih (0 ppm, 5 ppm, 10 ppm) dan suhu (16–19°C dan 24–27°C), dengan 18 unit percobaan (3 ulangan). Data dianalisis menggunakan ANOVA (JMP-SAS).Hasil penelitian menunjukkan bahwa kombinasi minyak cengkih dan suhu berpengaruh signifikan terhadap waktu induksi dan pemulihan benih. Perlakuan 10 ppm minyak cengkih pada suhu 16–19°C menghasilkan waktu induksi tercepat, sedangkan tanpa minyak cengkih pada suhu 24–27°C mempercepat pemulihan. Interaksi kedua faktor ini menunjukkan bahwa minyak cengkih secara nyata mempengaruhi respons ikan terhadap perubahan suhu. Abstract

This thesis aims to study the tsunami hazards in the Maluku Sea region, particularly in the coastal area of Bitung, Indonesia. The research focuses on modeling and analyzing the potential tsunamis triggered by earthquake events in that region. The data used includes bathymetry, topography, coastline, and river data obtained from the Geospatial Information Agency (Badan Informasi Geospatial-BIG). The tsunami modeling process utilizes the software COMCOT (Cornell Multi-grid Coupled Tsunami).  The earthquake source parameters (PusGen) used in the modeling, such as magnitude, focal depth, length of the fault plane, the width of the fault plane, dislocation of the fault plane, strike, dip, slip/rake, and modeling area, were obtained from worst-case scenarios based on PusGen's research results. The research consists of two main stages: the pre-verification stage, where data processing and tsunami modeling are conducted, and the verification stage, where field surveys are carried out to determine the coordinates for Temporary Evacuation Sites (TEP) and Final Evacuation Sites (FEP).  Various equipment, such as GPS, altimeters, stopwatches, cameras, and drones, are used during the field verification. The research procedures include a literature review, data collection of topography, bathymetry, earthquake parameters, and fault mechanism. Subsequently, tsunami modeling is conducted, and hazard analysis is performed based on the model results. Tsunami hazard maps are generated to highlight high-risk areas in the coastal region of Bitung, and evacuation routes are identified to prepare the coastal community of Bitung to face potential tsunami threats.  The research offers important information to assist in the development of warning systems and efficient evacuation strategies to protect the coastal community of Bitung from tsunamis' devastating impacts. Abstract

This study aims to analyze the effect of commercial papain enzyme with different concentrations and duration of soaking on increasing the degree of hatching of pearl catfish (Clarias gariepinus) eggs. The study used a Randomised Group Design (RAK) with variations in papain enzyme concentration (0, 2, 4, 6, and 8 ppm) and soaking duration (5, 10, and 15 minutes). The results showed that papain enzyme concentration and soaking duration significantly affected the degree of hatching (p < 0,001). The optimal concentration of papain enzyme was 6 ppm with a soaking duration of 10 minutes, resulting in the highest hatching rate of 70,52%±2,92. The lowest egg adhesion was produced at a concentration of 8 ppm with a duration of 15 minutes (32%±4,00), while the highest degree of fertilization was directed at a concentration of 6 ppm with a duration of 10 minutes (79%±3,61). The measurement results of water quality parameters are: temperature 25,6-26,4°C, pH 7,12-7,47, and dissolved oxygen 4,06-4,41 ppm. These results indicate that the papain enzyme effectively reduces egg adhesion and increases the hatching rate of pearl catfish eggs.  Keywords: mutiara catfish; papain enzyme; hatching rate; concentration; immersion duration.   Abstrak Penelitian ini bertujuan untuk menganalisis pengaruh enzim papain komersial dengan konsentrasi dan durasi perendaman yang berbeda terhadap peningkatan derajat penetasan telur ikan lele mutiara (Clarias gariepinus). Penelitian menggunakan Rancangan Acak Kelompok (RAK) dengan variasi konsentrasi enzim papain (0, 2, 4, 6 dan 8 ppm) dan durasi perendaman (5, 10 dan 15 menit). Hasil penelitian menunjukkan bahwa konsentrasi enzim papain dan durasi perendaman berpengaruh signifikan (p <0,001) terhadap derajat penetasan. Konsentrasi optimal enzim papain adalah 6 ppm dengan durasi perendaman 10 menit, menghasilkan derajat penetasan tertinggi sebesar 70,52%±2,92. Daya rekat telur terendah dihasilkan pada konsentrasi 8 ppm dengan durasi 15 menit (32%±4,00), sedangkan derajat pembuahan tertinggi dihasilkan pada konsentrasi 6 ppm dengan durasi 10 menit (79%±3,61). Hasil pengukuran parameter kualitas air yaitu: suhu 25,6–26,4°C, pH 7,12–7,47 dan oksigen terlarut 4,06–4,41 ppm. Hasil ini menunjukkan bahwa enzim papain efektif mengurangi daya rekat telur dan meningkatkan derajat penetasan telur ikan lele mutiara.  Kata kunci: ikan lele mutiara; enzim papain; derajat penetasan; konsentrasi; durasi perendaman

Abstract

Abstrak Penelitian ini bertujuan untuk menganalisis tingkat kepuasan tenaga medis terhadap implementasi Rekam Medis Elektronik (RME) di Rumah Sakit Manembo-Nembo Bitung. Implementasi RME bertujuan untuk meningkatkan efisiensi dan kualitas pelayanan kesehatan. Metode yang digunakan adalah penelitian kuantitatif dengan pendekatan cross-sectional, di mana data dikumpulkan melalui kuesioner yang diberikan kepada 208 tenaga medis yang bekerja di rumah sakit tersebut. Hasil penelitian menunjukkan bahwa sebagian besar tenaga medis merasa puas dengan sistem RME, terutama dalam hal timeliness dan ease of use, meskipun masih ada kendala dalam aspek content dan accuracy. Variabel content, accuracy, format, ease of use, dan timeliness memiliki hubungan signifikan dengan kepuasan tenaga medis. Penelitian ini memberikan rekomendasi untuk perbaikan lebih lanjut pada sistem RME, terutama pada peningkatan pelatihan dan penyajian informasi dalam sistem. Kata Kunci: kepuasan tenaga medis, Rekam Medis Elektronik, content, accuracy, timeliness Abstract This study aims to analyze the level of satisfaction of medical staff regarding the implementation of Electronic Medical Records (EMR) at Manembo-Nembo Bitung Hospital. The implementation of EMR aims to improve the efficiency and quality of healthcare services. The method used in this study is a quantitative research design with a cross-sectional approach, where data were collected through a questionnaire distributed to 208 medical staff working at the hospital. The results of the study show that most medical staff are satisfied with the EMR system, especially in terms of timeliness and ease of use, although there are still challenges in the content and accuracy aspects. The variables content, accuracy, format, ease of use, and timeliness have a significant relationship with medical staff satisfaction. This study provides recommendations for further improvements in the EMR system, particularly in enhancing training and information presentation. Abstract

Pengangkutan benih ikan sering kali menyebabkan stres fisiologis yang berdampak negatif terhadap kesehatan dan kelangsungan hidup ikan, terutama akibat perubahan lingkungan seperti suhu dan perlakuan anestetik. Penelitian ini dilakukan untuk mengkaji pengaruh dosis minyak cengkeh dan suhu sebagai anestetik terhadap glukosa darah dan sintasan benih ikan Tawes. Rancangan penelitian yang digunakan adalah rancangan acak lengkap (RAL) Faktorial, dengan dua faktor yakni faktor dosis minyak cengkih (A) dengan 3 taraf yaitu A1 (0 ppm), A2 (5 ppm), dan A3 (10 ppm), dan faktor suhu (B) terdiri dari 2 taraf yaitu B1 (16-19 °C) dan B2 sebagai kontrol (24-27 °C), dengan tiga kali ulangan. Penelitian ini terdiri atas 18 unit percobaan, di mana setiap unit diisi dengan 50 ekor benih ikan Tawes sehingga total ikan yang digunakan sebanyak 900 ekor. Benih dimasukkan ke dalam kantong berisi 5 L air (1/3 dari volume kantong), ditambahkan oksigen sebanyak 2/3 volume kantong. Selanjutnya dilakukan uji transportasi selama 12 jam, setelah proses pengangkutan, ikan dipelihara di akuarium berukuran 60 x 40 x 40 cm, dengan volume 96 L. Data dianalisis menggunakan analisis sidik ragam (ANOVA) pada program statistik JMP Pro 16 (SAS 9.4). Perlakuan terbaik ditemui pada perlakuan dosis minyak cengkih 10 ppm + suhu 24-27°C dengan kadar glukosa darah setelah transportasi dan setelah pemeliharaan masing-masing sebesar 119,9±6,80 mg/dL dan 86,56±5,01 mg/dL. Sintasan setelah transportasi dan setelah pemeliharaan masing-masing sebesar 98,7±1,16 % dan 96,7±1,16 %. Interaksi faktor minyak cengkih dan suhu menunjukkan bahwa faktor minyak cengkih tidak memberikan pengaruh secara nyata terhadap glukosa darah dan sintasan.

Abstract

Sea Surface Temperature (SST) is the temperature of the water measured at sea level. SST is a crucial marine parameter that influences the weather and climate in Indonesia. The value of sea surface temperature will be very closely related to factors such as the depth of measurement from sea level, the intensity of sunlight received, season, weather, water depth, air circulation, and cloud cover. This study aims to design a prototype tool to measure wind speed and sea surface temperature parameters. The components used are the DS18B20 sensor, JL-FS2 sensor, Arduino Mega 2560, and the SIM900a GSM Module. The system is displayed in real-time via the Thingspeak website, accessed via a PC or smartphone. The DS18B20 sensor, as a sea surface temperature sensor, can measure these parameters by looking at the trend of changing day and night temperatures. The JL-FS2, as a wind speed sensor, has been able to measure wind speed parameters during field tests at FADs.

Abstract

Produksi perikanan budidaya dapat ditingkatkan melalui penyediaan pakan yang sesuai dengan kebutuhan ikan, mencakup aspek jenis, ukuran, maupun jumlahnya. Pakan mikrokapsul adalah jenis pakan buatan berukuran sangat kecil (mikron) yang dirancang khusus untuk memenuhi kebutuhan nutrisi ikan pada tahap larva dan benih.Tujuan penelitian ini adalah untuk menganalisis karakteristik morfometrik dan pola pertumbuhan ikan yang diberi pakan mikrokapsul inklusi berbeda. Penelitian ini menggunakan 1.080 ekor benih ikan nila merah berukuran 1-3 cm pada wadah akuarium. Akuarium yang digunakan berukuran 60x40x40 cm yang kemudian diisi air sebanyak 72 L dan ditebar sebanyak 1 ekor L-1. Penelitian ini didesain dengan menggunakan rancangan acak lengkap dengan lima perlakuan dengan masing-masing tiga ulangan. Perlakuan satu yaitu pemberian pakan komersil (kontrol), perlakuan dua yaitu pemberian pakan mikrokapsul tepung Tubifex sp., perlakuan tiga yaitu pemberian pakan mikrokapsul tepung Maggot, perlakuan empat yaitu pemberian pakan mikrokapsul tepung Spirulina sp., dan perlakuan lima yaitu pemberian pakan mikrokapsul tepung Chlorella sp.. Hasil penelitian menunjukkan bahwa karakteristik morfometrik ikan nila berbeda pada setiap perlakuan. Hasil uji regresi linear sederhana terhadap parameter pola pertumbuhan ikan nila pada semua perlakuan bersifat alometrik negatif, yakni pertumbuhan panjang lebih cepat daripada pertumbuhan beratnya. Hasil pengukuran kualitas air pada semua perlakuan menunjukkan hasil yang sesuai dengan baku mutu untuk pemeliharaan ikan nila dengan kisaran nilai suhu berkisar 25,1-29,6 o C, oksigen terlarut berkisar 5,1-6,8 mg L-1, pH berkisar 7,42-8,14, dan amonia berkisar 0,01-0,17 mg L-1.Aquaculture production can be increased by providing feed that is appropriate to the need of fish, both in terms of type, size, and quantity. Microcapsule feed is a type of artificial feed with very small sizes (microns) specifically designed to fulfil the nutritional need at the larval and fry fish stages. This study aimed to analyze the morphometric characteristics and growth patterns of fed microcapsules with different inclusions. The study was conducted using 1.080 tilapia with size of 1-3 cm in aquariums. The aquarium used was 60x40x40 cm and was then filled with 72 L of water with a stocking density of 1 fish L-1. This study used a completely randomized design with five treatments and three replications each. Treatment one is the provision of commercial feed (control), treatment two is the provision of microcapsule feed with Tubifex sp flour, treatment three is the provision of microcapsule feed with Maggot flour, treatment four the provision of microcapsule feed with Spirulina sp flour, and treatment five is the provision of microcapsule feed with Chlorella sp flour. The results of the study showed that the morphometric characteristics of tilapia were different in each treatment. The results of the simple linear regression test on the parameters of tilapia growth patterns in all treatments were negative allometric, namely the growth in length was faster than the growth in weight. The results of water quality measurements in all treatments showed results that were in accordance with quality standards for tilapia with a temperature range of 25,1-29,6 oC, dissolved oxygen range of 5,1-6,8 mg L-1, pH range of 7,42-8,14, and ammonia range of 0,01-0,17 mg L-1.
