Abstract

Salah satu faktor yang dapat menurunkan produksi kedelai di Indonesia adalah adalah serangan penyakit pustul bakteri pada tanaman kedelai yang disebabkan bakteri Xanthomonas axonopodis pv. glycines. Tujuan penelitian ini adalah untuk mengetahui potensi bakteri antagonis Corynebacterium sp. dan Bacillus sp. dalam mengendalikan penyakit pustul bakteri pada tanaman kedelai. Penelitian ini dilakukan melalui beberapa tahapan: uji antagonis secara in-vitro dalam cawan Petri menggunakan Rancangan Acak Lengkap (RAL) dengan 8 perlakuan dan 4 ulangan, dan uji penekanan penyakit pustul bakteri di rumah kaca menggunakan Rancangan Acak Kelompok (RAK) dengan 8 perlakuan dan 4 ulangan. Aplikasi Corynebacterium sp. dan Bacillus sp. sebagai bakteri antagonis, dengan konsentrasi 106 - 108 CFU/ml mampu menekan pertumbuhan patogen X. axonopodis pv. glycine secara in vitro. Aplikasi bakteri antagonis Corynebacterium sp. dan Bacillus sp. dengan konsentrasi 106 - 108 CFU/ml  dapat menekan perkembangan penyakit pustul pada tanaman kedelai. Tingkat penekanan penyakit pustul bakteri setara dengan tingkat penekanan oleh bakterisida. Dengan demikian kedua isolat bakteri antagonis tersebut (Corynebacterium sp. dan Bacillus sp) sangat berpotensi sebagai agens pengendali hayati penyakit pustul bakteri pada tanaman kedelai.

Abstract

Mikroorganisme Lokal (MOL) merupakan produk cairan yang terbuat dari bahan-bahan alami, sebagai media hidup dan berkembangnya mikroorganisme bermanfaat. Penelitian ini bertujuan untuk mempelajari potensi bakteri dari MOL bonggol pisang sebagai antagonis terhadap Blood Disease Bacterium (BDB = Ralstonia syzygii subsp. syzygii) penyebab penyakit darah pada tanaman pisang. Isolasi bakteri dari MOL bonggol pisang dilakukan dengan metode spread plate. Pengujian penghambatan terhadap BDB menggunakan Rancangan Acak Lengkap dengan 10 perlakuan pada cawan Petri dan 11 perlakuan pada percobaan menggunakam buah pisang. Sebanyak 43 isolat bakteri berhasil didapatkan dari MOL, tetapi hanya 8 isolat yang mampu menekan pertumbuhan BDB, yaitu isolat BMOL 1, BMOL 8, BMOL 14, BMOL 21, BMOL 22, BMOL 24, BMOL 27, dan BMOL 29. Isolat BMOL1 dan BMOL 24 menunjukkan kemampuan lebih tinggi dalam menghambat pertumbuhan BDB dibanding isolat yang lain.

Abstract

Ralstonia solanacearum merupakan bakteri penyebab penyakit layu yang dapat menurunkan produksi tanaman tomat. Bakteri penghuni akar (rizobakteri) diketahui dapat berperan sebagai antagonis terhadap patogen tanaman. Penelitian ini bertujuan untuk medapatkan rizobakteri dari gulma dari famili solanaceae yaitu leunca dan ciplukan yang berpotensi sebagai antagonis terhadap R. solanacearum. Telah diperoleh delapan isolat rizobakteri yang berpotensi menekan pertumbuhan R. solanacearum. Semua isolat rizobakteri yang diuji memiliki potensi yang sama dengan kontrol (bakterisida Streptomisin sulfat) dalam menekan perkembangan penyakit layu bakteri. Berdasarkan karakterisasi fisiologi dan biokimia diketahui bahwa masing-masing isolat rizobakteri memiliki karakter yang berbeda.

Abstract

Penelitian kelimpahan bakteri bermanfaat pada petak lahan padi yang dibudidayakan menggunakan pendekatan Pengelolaan Hama Terpadu (PHT) dan konvensional telah dilakukan di Desa Bayem, Kecamatan Kasembon, Malang. Penelitian bertujuan untuk mengetahui kelimpahan bakteri (Pseudomonas sp., Bacillus sp., penambat nitrogen dan pelarut fosfat) pada lahan padi sawah yang menerapkan PHT dan cara konvensional. Penelitian dilakukan dengan pengambilan sampel tanah secara sistematik, dibiakkan pada media selektif  lalu dilakukan perhitungan kelimpahan bakteri. Hasil perhitungan kelimpahan bakteri dianalisis uji T untuk membandingkan kelimpahan bakteri Pseudomonas sp., Bacillus sp., penambat nitrogen dan pelarut fosfat antara lahan PHT dan konvensional. Hasil penelitian menunjukkan bahwa tidak ada perbedaan yang signifikan kelimpahan bakteri pelarut fosfat dan bakteri penambat nitrogen di lahan PHT dan lahan konvensional baik pada fase vegetatif maupun generatif. Ada perbedaan yang signifikan kelimpahan bakteri Bacillus sp. di lahan PHT dan lahan konvensional baik pada fase secara vegetatif maupun generatif. Tidak dapat diketahui adanya perbedaan kelimpahan bakteri Pseudomonas sp. pada lahan PHT dan lahan konvensional karena data tidak diperoleh pada plot lahan PHT. Tidak ada perbedaan kelimpahan bakteri bermanfaat pada lahan PHT dan konvensional kecuali kelimpahan Bacillus sp pada fase generatif diduga karena aplikasi pada lahan PHT masih dilakukan pada musim pertama dan baru berjalan dalam waktu beberapa minggu.

Abstract

Sugarcane disease disturbance is one of the limiting factors in achieving high productivity. Xylaria sp. has become endemic to sugarcane plantations in Lampung and South Sumatra. The intensity of disease attacks in Indonesia by 25% and 26% can reduce sugar production 12.3 and 15.4%, respectively. Losses due to disease attacks are greater in ratoons because plant cane sugarcane which is attacked by this disease, in ratoon stage, there will be a decrease in the number of shoots. The aim of this study was to obtain isolates of Trichoderma spp. which have antagonistic abilities and have chitinase enzymes so that they are effectively used as biological agents against Xylaria sp. The research method of antagonist test used dual culture test and qualitative test of chitin substrate was used to assess the chitinolytic index of four isolates of Trichoderma spp. The parameters were the inhibition of Trichoderma spp. isolates and the chitinolytic index of the four isolates moreover the value of the chitinase enzyme activity of the selected isolates. Mechanism of antagonist T10 are competition, antibiosis and mycoparasitism. The results showed that the Indonesian Sugar Research Institute (ISRI) T10 isolate could be used as a biological agent against Xylaria sp. The isolate had an inhibitory ability 73.33% with a chitinolytic index value 1.15 and the activity value of the chitinase enzyme crude extraction at a dilution 10-1 was 10.99 units/mL.

Abstract

We have studied the arthropods biodiversity in two paddy field ecosystems, namely, paddy field ecosystem using Integrated Pest Management (IPM) system and non-IPM paddy field ecosystem. This study was conducted from April 2011 - November 2011 in three locations, that is, Pasar Kamis village and Sungai Rangas village in Banjar regency, and Guntung Payung village in Banjarbaru city, South Borneo Province. In this study, we used insect nets, yellow sticky traps, light trap and pitfall trap to get the sample or catch the arthropods in one period of planting season. The arthropods caught were then classified into some classes: pest (herbivore), natural enemy (parasitoid and predator), and other arthropods. After that, the Species Diversity Index was determined using its Shannon-Wiener Index (H), Evenness (e), Species Richness (R), and Species Similarity Index (IS). The sum of arthropods which have the characteristic of pest and parasitoid were higher in the IPM paddy fields than in the non-IPM paddy fields, and the sum of other arthropods were the same. The highest H and e values were in the IPM paddy field in Pasar Kamis village. The IS value for each three locations were 77.5% in Pasar Kamis village, 93.42% in Guntung Payung village, and 78.76% in Sungai Rangas village.Keywords: diversity, arthropods, ecosystem, and SLPHT

Abstract

The cultivation of tobacco is one of users of agro-chemical substances such as insecticides, herbi-cides, defoliants, and fertilizers among other food crops and plants with high economical value. The use of these chemicals may bring negative effects regarding the richness and abundance of arthropods. The study of arthropod community in the Virginia tobacco ecosystem was carried out in Central Lombok, West Nusa Tenggara Province, during the 2010 plantation year. It was aimed at finding the composition, structure, and dynamic of the arthropod diversity around the tobacco field. Samples were obtained by using trapping techniques (pitfall traps, yellow-pan traps, and sweep net). The number of arthropods found in Virginia tobacco field are 69, consisting of 65 species of insects (belonging to 46 families and 8 orders) and 4 species of spiders (belonging to 4 families). The majority of insects found was Hymenoptera, dominated by bees. Based on the ecological functions, the major group of arthro-pods documented was phytophagous (20 species), mostly Coleoptera and Orthoptera. Yet, the number of predators was relatively more abundant than that of the phytophagous. The number of kinds of ar-thropods commonly interacting around the field fluctuated during the growing period, while in the cultivation period the number decreased. The diversity of the species (H) and the ratio of abundance of the natural enemies and phytophagous in the field was high.

Abstract

The study aimed to identify the content of the triterpenoid saponin compound from Barringtonia asiatica seeds collected from coastal of Malalayang beach, Manado, North Sulawesi. The method of extraction was the maceration with methanol solvent. The identification of the isolate was by thin layer chromatography, column chromatography, and GCMS. The phytochemistry test result of B. asiatica seed extract contained the compounds of alkaloid, saponin, and tannin. Then, the compound was separated by a thin layer chromatography method with a solvent system of methanol:chloroform:water. It produced three nodes that were spread around polar (Rf 0.24), semipolar (Rf 0.6) and non-polar (Rf 0.78) areas. Meanwhile, the process of column chromatography could only separate two chemical components namely semipolar (Rf 0.6) and polar (Rf 0.76). The identification with GCMS resulted in three compounds of Triterpenoid saponins, namely 2.4-bis-(1.1-dimethyl ethyl); 4-Dodecylphenol; and 2.6 bis-(1.1-dimethyl ethyl-4-methyl).

Abstract

This research aims to assess the environmental factors that affect the abundance of scales populations on the apple crop. The study was conducted in July 2012 to January 2013. The study was conducted at two sites with different altitude. Bumiaji village's altitude is ±900 m and Tulungrejo village's altitude is ±1,515 m asl. Multiple linear regression analysis was conducted to determine the environmental factors that affect the abundance of scales populations on the apple crop. Based on the results of the regression analysis, air temperature and humidity affect the abundance of scales populations in Binangun while rainfall and long solar radiation have no effect. Scales population abundance of the apple crop in Binangun was influenced by air temperature and humidity one week before. Based on the results of the regression analysis, air temperature, air humidity and rainfall affect the abun dance of scales population in Tulungrejo while long solar radiation has no effect. Scales population abundance ofthe apple crop in Tulungrejo was affected by air temperature two weeks before, while humidity and rainfall were affected one week before. The higher air temperature scales, flea population level will increase. And the higherrelative humidity, scales population levels will increase.

Abstract

Environmentally friendly and conventional cultivation systems that are selected and used by farmers can directly or indirectly affect microbes in the soil. Microbial population diversity can be used as a sensitive parameter to soil quality. Bacterial populations are a determining factor in ecosystems that are important because of biological and biogeochemical cycles, and heterotrophic activities. This study suggests looking at environmentally friendly (O1) and conventional (K1, K2, K3) soil bacteria on soil planted with sweet maize and function of bacteria in ecosystems. The results of this study that diversity on environmentally friendly was higher than conventional soil. The value was 1,775; while those of the conventional lands 1, 2 and 3 were 1,587; 1,245 and 1,320. In O1 soil, the most common genus was Agrobacterium and bacteria which were only found in environmentally friendly soils were Bacillus and Clostridium. Bacteria found such as O1F and O1G which were related to B. paramycoides could be used as biological agents against Cercospora leaf spot disease, B. megaterium could bind nitrate to the rhizosphere and dissolve phosphate in the soil, and B. aryabhattai which was potential as biological fertilizers and bioremediation
