Abstract

Background: Indonesia is a major producer of essential oils, with patchouli and peppermint oils being widely used in aromatherapy. Aromatherapy products, such as wind oil, provide therapeutic benefits but are often underused by younger consumers. Developing formulations that appeal to a broader audience may increase their use. Objective: This study aimed to formulate an aromatherapy oil using patchouli and peppermint essential oils, evaluate its physical properties, and test consumer preferences. Method: Four formulations (F1–F4) were prepared with varying concentrations of patchouli and peppermint oils. Physical tests, including organoleptic, homogeneity, clarity, pH, and spreadability, were conducted over four weeks. A hedonic test with 20 respondents assessed sensory preferences. Results: Formulations remained stable in terms of homogeneity and clarity throughout the study. pH values were within the acceptable range for skin application, though they increased slightly during storage. Spreadability improved over time, with F2, F3, and F4 meeting the ideal range by the fourth week. F4, containing 6% patchouli and 15% peppermint oils, was the most preferred in the hedonic test, particularly for color, smell, and clarity. Conclusion: Patchouli and peppermint essential oils can be effectively combined to create a stable, consumer-preferred aromatherapy oil. F4 showed the best physical and sensory qualities, making it a promising formulation for future development.

Abstract

Diabetic ulcers are known to be one of the most feared complications and account for 80% of hospital admissions in patients with Diabetes Mellitus (DM). Diabetic ulcers occur due to bacterial infections, exacerbated by the formation of biofilm in the wounds. Nanotechnology presents a new alternative strategy for inhibiting biofilm formation, one of which is nanogel. The richness of marine biota that has the potential as an anti-biofilm candidate includes Petrosia sp. This study aims to extract and test the anti-biofilm activity of the Petrosia sp. formulation as a nanogel for healing diabetic ulcers caused by biofilm infection of Staphylococcus aureus. The extraction method used is maceration with 96% ethanol as a solvent, and biofilm activity is determined using the microtiter broth method. The procedure for creating incisional wounds on male mice induced with diabetes by alloxan is performed after shaving the fur in the area to be incised, followed by the application of 2% lidocaine cream as a local anesthetic. Subsequently, a preclinical trial on male mice is conducted by monitoring the stages of S. aureus biofilm wounds in both the test group and the positive control group. The results show that the nanogel formula from Petrosia sp. can inhibit up to 78.04% ± 0.01 on diabetic ulcer wounds with S. aureus biofilm over a period of 10 days and is highly potential to be developed as a new anti-biofilm candidate for diabetic ulcers caused by S. aureus biofilm.

Abstract

Red pidada fruit peel extract gel preparation can increase the efficiency and comfort of topical use because of its activity in the healing of open wounds. The aim of this research was to determine how variations in HPMC levels affect the physical quality and stability of red pidada fruit peel extract gel preparations, using the maceration method to obtain thick extracts and formulations with different HPMC concentrations (1% HPMC, 3% and HPMC 5% formula were tested for physical quality and stability, including the Storage Stability Test for 4 weeks. Effectiveness testing was also carried out in white mice using an incision wound method with a depth of 1.5 mm and macroscopic observation. The effectiveness test concentration was set to 5%. The results of data analysis using the Spss One Way ANOVA test showed that higher HPMC levels resulted in a thicker gel, viscosity, stickiness, spreadability, and higher pH. It can be concluded that the gel formulation that meets the requirements is that formula 2 provides organoleptic test results with a thick green color typical of red pidada, good homogeneity test, spreadability test 5.52 cm, adhesion test 2.61 minutes, viscosity test 2700cCps, pH test 4.92. During storage, the gel exhibited good consistency. The longer the storage time, the closer the pH changes to alkaline values. With the Anova value, there is a significant difference in the three formulas with a value of p=0.005 Keywords: red pidada (Sonneratia caseolaris) fruit peel, wound healing, HPMC, gel

Abstract

Catheter-associated urinary tract infections (CAUTI) have increased significantly every year. Approximately 70-80% of these infections involve biofilms. This study aims to determine the effectiveness of nano spray formula of kelubut leaf extract in inhibiting and eradicating biofilm formation on catheters. Kelubut (Passiflora foetida L.) leaf extract was obtained through maceration process to obtain useful compounds. Antibiofilm activity was assessed using microbroth dilution, with scanning electron microscopy for visualization. The nano spray formulation activity of kelubut (Passiflora foetida L.) leaf extract was able to inhibit biofilm growth by (77.74% ± 0.01), while ciprofloxacin was slightly more effective with a value of (80.22% ± 0.01). SEM analysis confirmed the ability of the kelubut leaf extract nanospray formulation to inhibit and damage Escherichia coli biofilms. The nano spray formulation of kelubut leaf extract (Passiflora foetida L.) coded NS1 is able to inhibit biofilm formation activity on catheters and has the potential to be developed as an antibiofilm candidate against bacteria that cause urinary tract infections.

Abstract

Microbial biofilm-associated infections, particularly those caused by Pseudomonas aeruginosa and Candida albicans, have emerged as global challenges due to resistance to conventional antimicrobials. This study investigated the antibiofilm activity of ethanol extract from the sponge Petrosia sp., collected from the waters of Maratua Island, against both pathogens. Extraction was performed using 96% ethanol maceration, followed by in vitro biofilm inhibition and eradication assays using the broth microdilution method in microtiter plates. Biofilm density was quantified through crystal violet staining and absorbance measurement (OD₅₉₅ nm). The results demonstrated that the extract inhibited both intermediate-stage (24 hours) and mature-stage (48 hours) biofilm formation, with an MBIC₅₀ of 0.125% for both microorganisms. At a concentration of 1%, the inhibition percentages reached 73.86% for P. aeruginosa and 84.70% for C. albicans. The highest mature biofilm eradication activity was observed at 1% concentration, with MBEC₅₀ values of 51.98% for P. aeruginosa and 80.53% for C. albicans, approaching the effectiveness of positive controls (nystatin 82.04% and chloramphenicol 61.72%). The extract exhibited a dose-dependent pattern, with greater efficacy against C. albicans, possibly due to differences in biofilm matrix composition. These findings confirm the potential of Petrosia sp. as a source of multitarget antibiofilm compounds, particularly for fungal infections, and underscore the significance of Indonesia’s marine biodiversity as a basis for innovative therapeutic development.

Abstract

Candida albicans can cause two infections in humans: superficial and systemic. The ability of C. albicans to infect the host is influenced by virulence factors and character changes so that it can fool the immune system. From the character change factor, C. albicans can form a biofilm. This study aims to determine the good concentration in inhibiting and determine the antifungal and antibiofilm activity of nanoemulsion mouthwash formulation of bajakah tampala (Spatholobus littoralis Hassk) skin extract against C. albicans. This research was conducted with an experimental method. The formulation used a spontaneous magnetic stirrer technique to make nanoemulsion preparations. Antifungal and antibiofilm tests were carried out by dilution method using a 96-well plate and a microplate reader with a wavelength of 620 nm to determine the percentage inhibition against C. albicans and determine MIC50 and MBIC50. The results showed that the nanoemulsion mouthwash formulation of S. littoralis inhibited the planktonic and biofilm of C. albicans. The concentration of 1% is expressed as MIC50 and MBIC50. Therefore, the nanoemulsion formulation of S. littoralis extract could inhibit the growth of C. albicans in the oral cavity.
