Abstract

Penelitian ini bertujuan mengetahui kemampuan perkembangan awal embrio sapi in vitro menggunakan media tunggal untuk maturasi, fertilisasi, dan kultur berbahan dasar tissue culture medium (TCM) 199. Oosit sapi dikumpulkan dari rumah potong hewan dengan teknik aspirasi dan diklasifikasikan berdasarkan kekompakan sel kumulus dan sitoplasma yang homogen. Oosit dimaturasi pada medium TCM 199 yang disuplementasi dengan 10 IU/ml pregnant mare’s serum gonadotropin (PMSG), 10 IU/ml human chorionic gonadotropin (hCG), dan 10% fetal bovine serum (FBS), dilakukan selama 24 jam pada inkubator 5% CO2, 39 C. Fertilisasi dilakukan pada dua media yang berbeda yaitu media rutin fertilisasi dan media berbahan dasar TCM 199 dengan suplemen bovine serum albumin (BSA) dan heparin. Setelah fertilisasi, kumulus sel dihilangkan (denudasi), kemudian dikultur pada media TCM 199 yang disuplementasi dengan asam amino esensial dan non-esensial serta 10% FBS selama 3 hari. Hasil penelitian menunjukkan tingkat maturasi oosit pada sistem yang digunakan mampu mendukung 81,5% oosit mencapai tahap metafase II (M-II). Tingkat pembelahan embrio lebih tinggi pada media rutin dibandingkan dengan media TCM 199 yakni masing-masing 44,4 dan 23,2%. Jumlah embrio tahap 4-8 sel pada kedua perlakuan tidak berbeda nyata. Dapat disimpulkan media tunggal berbasis TCM dapat digunakan untuk produksi embrio in vitro.

Abstract

The purpose of this study was to evaluate the effectiveness of cysteamine supplementation on the maturation medium and/or in vitro fertilization medium with regards to improving the normal fertilization rate of sheep oocytes, which are characterized by the formation of two pronuclei. Grade A and B oocytes were matured in medium-199 with 0.3% bovine serum albumin (BSA), 10 IU/mL follicle stimulating hormone (FSH), 10 IU/mL human chorionic gonadotrophin (hCG), and 50 μg/mL gentamicin added for 24 hours in a 5% CO2 incubator at 39° C. The treatment group was divided into the following groups: a control group with no cysteamine (P1), a group with 100 μM cysteamine in the maturation medium (P2), a group with 100 μM cysteamine in the fertilization medium (P3), and a group with 100 μM cysteamine both in the maturation and fertilization medium (P4). The fertilization was carried out by incubating sperm-oocytes for 12 hours and then staining them with 1% aceto-orcein to observe the formation of a pronucleus. Normal fertilization rates obtained by each treatment group were 56.5% (P1), 57.1% (P2), 57.8% (P3), and 59.9% (P4) with no significant difference (P0.05) between groups. It was concluded that 100 μM cysteamine supplementation in both the maturation medium and fertilization medium was not able to increase the normal fertilization rate of sheep oocytes.

Abstract

Endometritis merupakan gangguan reproduksi akibat infeksi uterus yang menyebabkan penurunan efisiensi reproduksi dan kerugian ekonomi yang cukup besar pada peternakan sapi perah. Tujuan penelitian untuk mengetahui dan menganalisis secara spesifik tentang prevalensi, faktor kejadian dan pengaruh endometritis terhadap efisiensi reproduksi pada sapi perah. Penelitian ini menggunakan data sekunder 580 ekor sapi perah dari 98 peternak yang dikoleksi dari data recording reproduksi sapi perah di KPBS Pangalengan periode Januari sampai dengan Juni 2020. Data prevalensi, faktor kejadian endometritis, service per conception, conception rate dan pregnancy rate dianalisis secara deskriptif menggunkan Microsoft Excel, sedangkan jarak lahir ke IB pertama dan days open dianalisis mengunakan independent-sample t test. Hasil penelitian ditemukan data 120 ekor sapi perah yang mengalami endometritis dan 460 ekor tanpa endometritis dengan tingkat prevalensi sebesar 20,69%, prevalensi tertinggi pada laktasi ≥2, faktor kejadian berturut-turut adalah infeksi post partus pada partus normal (41,67%), kesulitan melahirkan (41,67%), retensio plasenta (9,17%), abortus (4,17), hipokalsemia/milk fever (2,50%) dan metritis (0,53%). Sapi dengan endometritis mengalami penundaan jarak lahir ke IB pertama dibandingkan tanpa endometritis (123,57±52,77 vs 90,94±41,18 hari) dan perpanjangan days open (146,98±63,01 vs 104,31±46,13 hari). Dapat disimpulkan bahwa kejadian endometritis dapat menurunkan efisiensi reproduksi pada sapi perah dengan perpanjangan days open sekitar 2 siklus estrus.

Abstract

Perguruan tinggi sebagai tempat insan akademik berkiprah saat ini mengalami berbagai tantangan dan ancaman yang dapat mengganggu fungsinya sebagai pusat pengembangan ilmu pengetahuan dan teknologi (iptek), serta menjadi penjaga keberlanjutan peradaban dan moralitas insan akademik agar tidak terdistorsi dan tererosi. Kemajuan teknologi (khususnya teknologi informasi), dekadensi moral global, tekanan ekonomi, orientasi praktis dan fokus pada pencapaian jangka pendek, perubahan paradigma pendidikan serta iklim ketidakpastian, menjadikan nilai-nilai tradisional perguruan tinggi terancam tergeser menjadi mesin penghasil tenaga kerja semata. Perguruan tinggi, yang seharusnya berperan sebagai benteng terakhir penjaga nilai-nilai akademik, serta sebagai agen perubahan (agent of change) yang berorientasi pada nilai kebaikan dan penjaga nilai moral (guardian of value), perlu jati diri insan akademik yang unggul, sehingga peran dan fungsi perguruan tinggi dapat tetap dipertahankan. Untuk memberikan pertimbangan dalam upaya menjaga tersebut, Policy Brief ini disusun melalui serangkaian proses diskusi anggota tim PAH Jati Diri Insan Akademik dan proses FGD yang melibatkan narasumber internal dan eksternal IPB. Policy Brief ini diakhiri dengan rekomendasi yang dapat diusulkan kepada penyelenggara pendidikan tinggi Indonesia agar fungsi perguruan tinggi dan insan akademik dapat tetap terjaga

Abstract

This study aimed to evaluate the effect of sorghum-Indigofera forage-based feed on the pre-mating performance of Garut ewes. A total of 28 Garut ewes was divided into two groups of treatment so that each treatment consisted of 14 heads. The treatment of field grass (FG) as a control and the feed composition in the formula of 50% sorghum forage silage + 40% Indigofera hay + 10% rice bran (SI). The animals were distributed under a completely randomized design, and the data were analyzed using a t-test. The results showed that the dry matter intake (DMI) and nutrient digestibility values of FG were higher (p<0.05) than SI. However, the impact on body weight gain, blood urea nitrogen (BUN) and blood cholesterol level did not differ (p>0.05). The average pre-mating bodyweight of Garut ewes for all treatments was the same number relatively, were 35 kg/head. BUN and blood cholesterol levels in the FG and SI treatments were 36.18±3.47 vs 35.27±4.23, and 66.13±5.56 vs 69.25±3.95 mg/dl, respectively. It can be concluded that SI feed had higher efficiency values and better readiness for the reproductive phase. Therefore, SI feed can be used as a substitute for field grass for the reproductive stage of Garut ewes.

Abstract

This study aimed to determine the effect of sorghum-indigofera (SI) mixed feed and pregnant mare serum gonadotropin (PMSG) hormone stimulation on ewe’s reproductive performance and Garut lamb’s pre-weaning growth. Twenty-eight multiparous Garut ewes were included into four treatment groups (2 ´ 2) with a factorial-Completely Randomized Design (CRD). The first factors were the types of feed local forage (LF) and SI, while the second factor was PMSG hormone stimulations (with and without PMSG stimulation). Synchronized estrous used twice injections with PGF2α, 11 days apart, and 500 IU PMSG was adminstrated at the second PGF2α injection intramuscularly. The observed variables included ewes' performance after parturition and post-natal lamb's growth. The obtained data were analyzed by multivarians of analysis (MANOVA) with α = 0.05 and continued with Tukey's Test. The result showed that feeding ewes with SI without PMSG stimulation resulted in the highest number of pregnancies, litter size, and lactation compared to other treatments. Feed type and PMSG stimulation factors has no any interaction effect (p > 0.05) on birth weight, weaning weight, and milk production. However, birth weight and milk production were affected (p < 0.05) by the feed, while weaning weight was affected (p < 0.05) by the PMSG stimulation. The fastest lamb growth rate was found in the groups of ewes fed by SI without PMSG stimulation (120 g/head/day). It can be concluded that SI could improve the ewe’s reproductive performance and the post-natal growth of Garut lambs. Meanwhile, the PMSG hormone could beneficially with sorghum-indigofera mixed feed.

Abstract

Penelitian ini bertujuan untuk mengevaluasi pengaruh suplementasi melatonin dalam media pengencer terhadap kualitas spermatozoa domba pasca-ekuibrasi. Melatonin dikenal sebagai antioksidan kuat yang mampu menangkal stres oksidatif, terutama yang terjadi selama proses kriopreservasi dan pencairan semen. Parameter yang diamati dalam penelitian ini meliputi motilitas, integritas membran plasma, keutuhan tudung akrosom, dan viabilitas spermatozoa, baik sebelum maupun setelah proses ekuibrasi. Hasil penelitian menunjukkan bahwa penambahan melatonin pada konsentrasi 1 mM memberikan peningkatan yang signifikan terhadap motilitas spermatozoa (83,33 ± 2,8%) dan integritas membran plasma (81,63 ± 0,62%) dibandingkan dengan kelompok kontrol. Selain itu, viabilitas spermatozoa pasca-ekuibrasi juga meningkat secara signifikan pada kelompok perlakuan (76,41 ± 1,4%) dibandingkan dengan kelompok kontrol (71,67 ± 2,2%). Penambahan melatonin dalam media pengencer terbukti mampu mempertahankan kualitas spermatozoa selama proses kriopreservasi. Temuan ini menunjukkan bahwa melatonin memiliki potensi besar sebagai aditif dalam pengencer semen guna meningkatkan efektivitas dan keberhasilan program inseminasi buatan pada ternak, khususnya pada domba.   Kata kunci: Ekuiblirasi, Melatonin, Spermatozoa

Abstract

This study aims to observe the changes in the characteristics of vaginal mucus to diagnose early pregnancy in dairy cattle. The characteristics of vaginal mucus include pH value, consistency, volume, and Vaginal Electrical Resistance (VER) value to diagnose early pregnancy using a Draminski Estrous Detector. The sample used that collected in this research was 52 dairy cattle. The research method consists of taking samples and observing the characteristics of the mucus. Data analysis includes normality test, T-test and Pearson correlation. The results showed that the mucus volume of pregnant cattle was less (1.280.356 mL) than that of non-pregnant cattle (2.210.437 mL), the pH value in pregnant cattle tended to be acidic, while non-pregnant cattle were classified as neutral, and there were differences in the consistency of mucus in pregnant cattle (4.00.0), while non-pregnant cows tended to be liquid (2.60.204). The volume and consistency of vaginal mucus in pregnant and non-pregnant cattle were significantly different (P0.05). The VER value of pregnant cattle was higher (36563.2 ohms) than non-pregnant cattle (19055.3 ohms). The VER value was highly correlated (r= 0.819) with the pregnancy status of dairy cattle. Thus, VER can be used as an accurate indicator to determine the status of early pregnancy in dairy cattle.

Abstract

This study to do examine the effectiveness of using heparin and hypotaurine to improve the in vitro fertilization rate. In first stage, oocytes were matured in the maturation medium supplemented with 0.3 mg/mL L-carnitine to improve nuclear maturation. In second stage, matured oocytes were fertilized with semen incubated in fertilization medium supplemented with 0.1 IU/mL heparin and 10 M hypotaurine. The final concentration of sperm used was 5x106 spermatozoa/mL, and its interaction with oocytes was carried out for 12 hours in 5% CO2 incubator at 38.5 C. The results showed that percentage of oocytes that reached metaphase II was significantly higher (P0.05) in media supplemented with 0.3 mg/mL L-carnitine compared to control (88.24.9% vs 79.85.1%). Furthermore, fertilization rate was characterized by formation of 2 PN was significantly higher (P0.05) in media supplemented with heparin and hypotaurine than control (82.62.0% vs 73.32.4%). In conclusion, L-carnitine supplementation at dose of 0.3 mg/mL on maturation media was effective in increasing percentage of matured oocytes. The combination of 0.1 IU/mL heparin and 10 M hypotaurine in fertilization medium was also successful in elevating the percentage of fertilized oocytes.

Abstract

This study aimed to determine the biopotency of pregnant mare serum gonadotropin (PMSG) in stimulating the formation of dominant follicles and the biopotency of human chorionic gonadotropin (hCG) in triggering an increase in the ovulation rate of the natural-estrus donor group and the prostaglandin F2 (PGF2) induced group using female Aceh cattle (n= 20) which were divided into two treatment groups: the group with natural estrus (A) and the group with PGF2 -induced estrus (B). Each donor group was superovulated using the injection of PMSGhCG (A1, B1) and only PMSG (A2, B2). The observations on total follicles and CL were carried out at D10, D14/D+0 and D21/D+7 using ultrasonography (USG). The results showed that PMSG gave a better superovulatory response to the development of dominant follicles (P0.01) at A1, A2, B1, and B2. The administration of hCG hormone 2000 IU i.m. was not able to increase the rate of ovulation. Total follicles after the addition of hCG were also not significantly different between A1:A2 and B1:B2. Statistically, the number of CL in all donor groups in D21 was not significantly different (P0.05). It concluded, superovulation using PMSG gave the same results to the donor cows with natural estrus and the donor cows with PGF2-induced estrus. The hCG hormone with a dose of 2000 IU injected i.m. was also not able to increase the ovulation rate of Aceh cattle superovulated with PMSG 2500 IU i.m. ____________________________________________________
