Abstract

Bioethanol is an organic compound resulted from the fermentation of sugar substrates by microorganisms which is used as alternative energy sources. During bioethanol fermentation yeast are exposed to various fermentation stresses, including temperature, osmotic, and oxidative stresess. Such conditions may decrease ethanol production. We previously isolated fermentation-stress tolerance yeast isolates from traditional Balinese beverages, identified as Wickerhamomyces anomalus BT2, BT5, and BT6. However no data available regarding the bioethanol production of those isolates. Our study indicates that these strains could utilize various sugar substrates (glucose, xylose, maltose, sucrose) in oxidative fermentative media. The highest value of substrate utilization efficiency following 48 hours fermentation was shown by BT6 on glucose (61.02%), BT 2 on xylose (55.44%) and maltose (60.90%). Measurement of ethanol production by Gas Chromatography showed that the strains were able to produce higher ethanol on the glucose substrate than other substrates. For instance, BT6 could produce the highest ethanol production (5.00 g/L) amongst strains tested by using glucose as substrate. Yet, the particular strains could only produce 0.30 g/L and 0.65 g/L by using xylose and maltose, respectively. For further genetic engineering purposes, we detected ADH1 gene from all three isolates, with high homology to the alcohol dehydrogenase from Saccharomyces cerevisiae, Geobacillus stearothermophilus and Pseudomonas aeruginosa. Further strain development can be carried out targeting the ADH1 gene, important for ethanol fermentation.

Abstract

Sugarcane (Saccharum officinarum) is a plant with high economic value because it can produce sugar. Shoot borer Scircophaga excerptalis (Lepidoptera: Pyralidae) and stem borer Chilo sacchariphagus (Lepidoptera: Pyralidae) attacks are one of the issues that limit sugarcane productivity. Establishing transgenic sugarcane is one of the most efficient ways to prevent borer damage. Previously, Cry1Ab and Cry1Ac genes were successfully used to create transgenic sugarcane plants. This study aimed to detect the presence of transgenes and analyze the expression level of the Cry1Ab-Cry1Ac gene fusion in transgenic sugarcane using RT-PCR. The methods used in this study are transgene detection with PCR and gene expression analysis in normalized expression (2-ΔΔCq) with RT-PCR. The Cry1Ab-Cry1Ac gene has been integrated into all lines with varied expression levels. In 311 GV and 333 GV lines, the Cry1Ab-Cry1Ac gene was expressed in the leaf but not in the stem. Shoot and stem borer attack percentage values showed that all lines were lower than the control, with 222 EH as the lowest and 311 GV as the highest. Leaf and stem borer attack levels were compared to gene expression values of Cry1Ab-Cry1Ac. Results may indicate that the 222 EH line was resistant, but the 311 GV and 333 GV lines were not.

Abstract

Fermentation of Lactic Acid Bacteria (LAB) Pediococcus pentosaceus can improve the quality of food and its shelf life. Using commercial LAB specific media, de Man Rogosa and Sharpe (MRS) for growth on industrial scale application is not efficient. Tapioca wastewater (TW) still contains some of the nutrients needed for the growth of P. pentosaceus, but needs the enrichment of carbon sources (5% of glucose) and nitrogen sources (ammonium sulfate). This study aimed to use tapioca industrial wastewater with the addition of glucose and ammonium sulfate as an alternative growth media for lactic acid bacteria P. pentosaceus E.1222. The results showed that glucose and nitrogen had no significant effect on the number of bacterial colonies. The highest substrate efficiency was tapioca wastewater (86.81%), MRS broth (53.73%), and TW with 5% of glucose and 1% of ammonium sulfate (43.53%) respectively. Maximum growth rate (μmaks) was found in TW with 5% of glucose and 1% of ammonium sulfate (0,52 hours-1). Increasing the starter volume until 1000 mL in TW with 5% of glucose and 1% of ammonium sulfate showed a slight decrease in the log number of bacteria from 8,836 (50 mL), 8,401 (500 mL), to 8,063 (1000 mL).

Abstract

This research aims is to find starter for sorghum fermentation and to observe its influence in fermentation process and tannin level in sorghum flour. Four isolates of Lactic Acid Bacteria were tested as starter for Numbu and Kawali sorghum grain fermentation. The criteria was the ability to produce acid and cell viability. The fermentation used four treatments in duplicates. The selected isolate was inoculated into dehulled and non-dehulled sorghum grain. The fermentation time were 0, 24, and 48 h. Analysis were pH, total acidity, total sugar, viable cell, tannin level, structure of sorghum flour, the yield of flour passed 80 mesh, and flour moisture. The results showed that E1222 was selected as a starter for Numbu sorghum grain fermentation while E5 was selected for Kawali. The starter can decrease pH value, increasing total acidity, decreasing total sugar, however there was no significant difference in viable cell in fermentation process. It decreased tannin level approximately 90% in Numbu fermented flour and 77% in Kawali. There was corrosion in starch structure of fermented flour, while the yield of flour passing through 80 mesh was increased. The best treatment was fermentation of dehulled and inoculated sorghum grain (So-St) in both of sorghum varieties.

Abstract

Probiotics are living microorganisms if consumed in sufficient quantities can be beneficial for health. One of the potential bacteria as probiotics is Lactobacillus plantarum NHC6 is a lactic acid bacteria isolated from pineapple juice. This study aimed to test the ability of L. plantarum NHC6 as a probiotic in pineapple juice. Probiotics were stored until four weeks under 4°C and 10°C which weekly is monitored data on the amount of microbes, pH, percentage of lactic acid, and total dissolved solids in fermented pineapple juice. The result shows, the shelf time for pineapple juice under 4°C has a lower microbial reduction time rather than the juice storage under 10°C. Probiotics shelf time predicted for 4°C storage was 28.32 weeks and for 10°C storage was 8.67 weeks after production. The changes in physical properties (pH, percentage of lactic acid, and total soluble solids) in 10°C storage is much faster than that off in 4°C, this happens because there is still metabolic activity by bacteria at that temperature.

Abstract

The national demand for carbohydrates in flour continues to increase significantly due to the emergence of contemporary food products that use flour as the primary ingredient. One of the products used in producing food is sourdough, which is fermented flour. Lactic acid bacteria (LAB) have a role in food fermentation by producing lactic acid. Amylolytic LAB can produce amylase enzyme, a biocatalyst in the starch hydrolysis process. One of the products made from the fermentation of wheat flour is sourdough. This study aims to determine the character of amylolytic LAB in wheat and wheat flour fermentation. The research methods include rejuvenation of LAB, selection of isolates, fermentation of starchy materials, and characterization of fermented liquid. The selection of bacteria was based on cell viability and pH, which showed that isolated Pediococcus pentasaceus E1222 could be used as a starter for starchy fermentation. The highest total acid and cell viability were produced in 24-hour wheat and wheat flour fermentation. Fermented wheat flour and flour had the most favorable aroma and texture, and fermented flour had the most favorable taste. Isolate Pediococcus pentasaceus E1222 is an isolate that can be used in making sourdough.

Abstract

This study aimed to select a proteolytic LAB with probiotic properties that can be applied to manufacture protein hydrolysate from fish heads. Tests on 20 isolates of LAB showed that nine isolates were proteolytic and non-pathogenic. A total of 5 isolates could grow well at 0.5% bile salt stress, and 3 of them could grow at pH 3. These three isolates had antagonistic ability against Salmonella bacteria, and one isolate was sensitive to the antibiotic tested. Molecular identification of the selected LAB isolates showed a 100% sequence similarity with Pediococcus pentosaceus with accession number MT515895.1. The LAB isolate has high proteolytic activity since it can increase the soluble fraction of fish meal powder from 32.10% to 88.38% in 48 hr. Production of protein hydrolysate using tuna waste was carried out for 30 days. Tuna waste protein hydrolysate had a medium antioxidant activity of 25.57 ± 0.93%. The hydrolyzed protein comprised 17 amino acids, including nine non-essential and eight essential amino acids, and is dominated by glutamic acid. Selected LAB isolate is potentially used in protein hydrolysate production, especially for flavor enhancers. Keywords: antagonistic, amino acid, Pediococcus pentosaceus

Abstract

ABSTRACT This study aimed to select lactic acid bacteria (LAB) as a potential probiotic that producing anti‒microbial compounds in order to treat motile aeromonads septicemia diseases caused by Aeromonas hydrophila on catfish Clarias sp. and evaluated its performance on gnotobiotic catfish. The in vitro assay was done to select several LAB isolates based on antagonistic activity against pathogenic bacteria. The selected isolate was tested in vivo to observe their ability to improve growth performances of catfish. The study was conducted with five treatments consists of K‒ (normal catfish without addition probiotic, without challenge test), K+ (normal catfish without addition of probiotic, with challenge test), Np (normal catfish with addition of probiotic and challenge test), G (gnoto catfish without addition of probiotic, with challenge test), and Gp (gnoto catfish with addition of probiotic and challenge test). The results showed that the addition of Pediococcus pentosaceus E2211 as selected probiotic could increase survival rate, specific growth rate, and immune response towards infection of A. hydrophila. The best survival rate after challenge test was obtained in Np and Gp treatments (88.46%), followed by G treatment (65.38%), while the K+ was only 53.84%. The conclusion of this study was P. pentosaceus E2211 potentially used as a probiotic candidate for normal and gnotobiotic catfish. The presence of normal microflora with P. pentosaceus E2211 in Np treatment showed the best probiotic performance with daily growth rate 3.28%, feed conversion ratio 1.79, and total intestinal bacteria reached 108 CFU/mL significantly different from other treatments (P<0.05).Keywords: Aeromonas hydrophila, catfish, LAB, probiotic, screening  ABSTRAKTujuan penelitian ini adalah menyeleksi bakteri asam laktat (BAL) sebagai probiotik potensial penghasil senyawa antimikrob guna menanggulangi penyakit motile aeromonad septicemia akibat Aeromonas hydrophila pada ikan lele Clarias sp. dan evaluasi kinerjanya pada ikan lele gnotobiotik. Pengujian in vitro dilakukan untuk menyeleksi beberapa isolat BAL sebagai kandidat probiotik berdasarkan aktivitas antagonis terhadap bakteri patogen. Isolat terpilih kemudian diuji in vivo untuk mengetahui kemampuannya dalam meningkatkan performa tumbuh ikan lele. Penelitian ini menggunakan rancangan acak lengkap dengan lima perlakuan, yaitu: K‒ (lele normal tanpa probiotik dan tanpa tanpa diuji tantang), K+ (lele normal tanpa probiotik dan diuji tantang), Np (lele normal diberi probiotik dan diuji tantang), G (lele gnoto tanpa probiotik dan diuji tantang), dan Gp (lele gnoto diberi probiotik dan diuji tantang). Hasil penelitian menunjukkan pemberian probiotik terpilih BAL Pediococcus pentosaceus E2211 mampu meningkatkan sintasan, laju pertumbuhan, dan respons imun ikan lele terhadap infeksi A. hydrophila. Sintasan terbaik pascauji tantang diperoleh pada perlakuan Np dan Gp yaitu sebesar 88,46%, diikuti perlakuan G sebesar 65,38%, sementara pada K+ hanya mencapai 53,84%. Kesimpulan dari penelitian ini ialah isolat BAL terpilih P. pentosaceus E2211 berpotensi sebagai kandidat probiotik untuk ikan lele normal dan lele gnotobiotik Clarias sp. Keberadaan mikroflora normal yang berasosiasi dengan P. pentosaceus E2211 pada perlakuan Np menunjukkan kinerja probiotik terbaik dengan nilai laju pertubuhan harian 3,28%, rasio konversi pakan 1,79 dan total bakteri usus mencapai 108 CFU/mL yang berbeda signifikan dibanding perlakuan lainnya (P<0,05).Kata kunci: A. hydrophila, BAL, ikan lele, probiotik, seleksi

Abstract

ABSTRACT Motile aeromonad septicaemia (MAS) in catfish can be done by improving the immune responses through probiotic administration. Co-administering probiotics producing digestive enzymes are expected to have an impact on fish growth. This study was aimed to evaluate the effectivity of probiotics B. megaterium PTB 1.4 and P. pentosaceus E2211 to improve the growth performance and immune response of catfish infected by Aeromonas hydrophila. Catfish with the initial body weight of 7.36 ± 0.21 g were reared in a pond. This study was conducted with five treatments, i. e. K- (without probiotic addition and A. hydrophila injection), K+ (no probiotic addition with A. hydrophila injection), Bm (B. megaterium PTB 1.4 addition and A. hydrophila injection), Pp (P. pentosaceus E2211 addition and A. hydrophila injection), and Bm+Pp (B. megaterium PTB 1.4 addition and P. pentosaceus E2211 and A. hydrophila injection). The study results showed that Bm, Pp, and Bm+Pp treatments were able to improve the growth performance of catfish including digestive enzyme activity, daily growth rate, feed conversion ratio, and final biomass with the best results was Bm+Pp treatment. The immune response of catfish before and after the challenge test showed better survival rate, higher total leukocytes, phagocytic activity, respiratory burst, and lysozyme activity on Bm, Pp, and Bm+Pp treatment (P<0.05) than the control treatment. In conclusion, the combination of probiotics B. megaterium PTB 1.4 and P. pentosaceus E2211 in feed synergistically improved the growth performance and immune response on catfish against A. hydrophila infection. Pemberian bersama probiotik penghasil enzim pencernaan diharapkan dapat berdampak pada pertumbuhan ikan. Penelitian ini bertujuan mengevaluasi efektivitas pemberian probiotik B. megaterium PTB 1.4 dan P. pentosaceus E2211 terhadap kinerja pertumbuhan dan respons imun ikan lele terhadap infeksi A. hydrophila. Ikan lele dengan bobot 7.36 ± 0.21 g dipelihara pada kolam beton yang diberi waring. Penelitian ini menggunakan rancangan acak lengkap dengan lima perlakuan, yaitu: K- (tanpa probiotik dan tanpa diinjeksi A. hydrophila), K+ (tanpa probiotik dan diinjeksi A. hydrophila), Bm (diberi B. megaterium PTB 1.4 dan diinjeksi A. hydrophila), Pp (diberi P. pentosaceus E2211 dan diinjeksi A. hydrophila), dan Bm+Pp (diberi B. megaterium PTB 1.4 dan P. pentosaceus E2211 dan diinjeksi A. hydrophila). Hasil penelitian menunjukkan perlakuan Bm, Pp, dan Bm+Pp mampu meningkatkan pertumbuhan ikan lele. Aktivitas enzim pencernaan, laju pertumbuhan harian, rasio konversi pakan, dan biomassa panen dengan hasil terbaik yaitu perlakuan Bm+Pp. Respons imun ikan lele dengan kombinasi probiotik B. megaterium PTB 1.4 dan P. pentosaceus E2211 mampu meningkatkan sintasan, total leukosit, aktivitas fagositosis, respiratory burst, dan aktivitas lisozim, baik pada sebelum dan setelah uji tantang. Pemberian kombinasi probiotik B. megaterium PTB 1.4 dan P. pentosaceus E2211 pada pakan mampu bekerja sinergis dalam meningkatkan kinerja pertumbuhan dan respons imun ikan lele terhadap infeksi A. hydrophila. Abstract

The diversity of bacterial communities in aquatic sediment of rainforest and oil palm plantation at Sumatra was studied using pyrosequencing of 16S rRNA gene and common biodiversity indices. Phylogenetic approach was used for revealing the community shift of bacterial phyla and genera in both areas. Ecological approach used soil pH, total Carbon (TC), total Nitrogen (TN), available Phosphorus (AP) measurement and bacterial diversity with Shanon and Simpson indices, and bacterial richness with Chao1-ACE indices and OTUs. Bacterial diversity and richness in aquatic sediment of forest area was higher than that ones in oil palm plantation area, although their pH, TC, and TN in both areas were not different significantly. The majority of sequences related to Proteobacteria (34.85%), Acidobacteria (32.67%), Nitrospirae (6.86%), Chloroflexi (4.31%), and Actinobacteria (4.02%) were from forest; whereas those related to Acidobacteria (46.10%), Proteobacteria (25.86%), Nitrospirae (9.20%), Chloroflexi (4.99%), and Actinobacteria (2.34%) invented from oil palm plantation. The genera of Alphaproteobacteria and Betaproteobacteria dominated in genera phylotype of bacterial 16S rRNA phylogenetic revealed both aquatic sediment of forest and oil palm plantation. The most genera in the phylogenetic tree from aquatic sediment of both areas was Burkholderia. The bacterial community shift in aquatic sediment of forest transformation indicated higher bacterial diversity index, richness index, some of phyla and genera in aquatic sediment from forest than from oil palm plantation.
