Abstract

Imunomodulator merupakan bahan yang dapat mengembalikan ketidakseimbangan sistem imun. Ekstrak rumput Kebar (Biophytum petersianum Klotzsch) mempunyai senyawa-senyawa aktif yang berperan sebagai agen imunomodulator, salah satunya adalah flavonoid. Penelitian ini bertujuan untuk mengetahui pengaruh dosis ekstrak rumput Kebar terhadap peningkatan jumlah sel B220 pada mencit betina. Rumput Kebar diekstrak dengan air yaitu dengan cara merebus (masak) semua bagian tanamannya (akar, batang, daun) dari masing-masing jenis dalam aquabides dengan perbandingan 100 gram dalam 3 liter aquabides) sambil diaduk dan dibiarkan mendidih , kemudian larutan tersebut disaring dan dibiarkan dingin lalu dibekukan dalam freezer selama 1 – 2 hari untuk selanjutnya dijadikan bubuk dengan metode pengering bekuan (freeze drying). Bubuk / jeli yang terbentuk kemudian digunakan sebagai bahan penelitian dengan cara melarutkan kembali dengan aquades sesuai dosis pada perlakuan. Selanjutnya ekstrak rumput Kebar diberikan pada 4 kelompok mencit (masing-masing 5 ulangan) secara oral dengan dosis 0, 0,48; 0,96 dan 1,92 mg/gr BB setiap hari selama 2 minggu. Setelah perlakuan, dilakukan dislokasi pada mencit, kemudian sel-sel limfositnya diisolasi dari limfa dan dilakukan perhitungan jumlah sel dengan flow cytometry. Hasil penelitian menunjukkan bahwa pemberian ekstrak rumput Kebar menunjukkan pengaruh signifikan terhadap peningkatan jumlah sel dosis 0,48 mg/g BB terjadi peningkatan sebesar 7,860% menjadi 29,870%; dosis 0,96 mg/g BB terjadi peningkatan sebesar 6,800% menjadi 28,810%, dan dosis 1,92 mg/g BB terjadi peningkatan sebesar 0,6225% menjadi 22,6625%. Ekstrak rumput Kebar aktivitas sebagai imunomodulator yakni dosis 0,48 mg/gr BB lebih bersifat imunostimulator, sedangkan dosis 0,96 mg/g BB dan dosis 1,92 mg/g BB lebih lebih bersifat imunosupresor.

Abstract

Lake is one of the important natural resources in north Sulawesi and many lakes ecosystem in this province has been used and involved in tourism industry. The aims of the research is to describes the recent status of lakes in North Sulawesi and describes its opportunities for tourism development. The analysis was based on the secondary data and direct field observation. Four lakes, namely Tondano, Pangolombian, Linow and Tampusu have been used as tourism attraction in various degreed. The most intensive used was Lake Tondano, while Lake Linow recently grows as one of the favorites object. Lake Pangolombian and Tampusu receive few tourist visitations. The development of four lakes as tourism object will increase the attractiveness of North Sulawesi as ecotourism development. In such a case, the conservation of lakes becomes crucial. Keywords: economy development, Lake tourism, North Sulawesi.

Abstract

Indonesia categorized as a mega-biodiversity country, with 25% of the species in the world. One of the uses of natural resources is in the tourism sector. Lately, in Indonesia, research about Social Media and Ecotourism is still not much done. Some studies in Indonesia mostly still view social media as marketing advice. In fact, social media also plays an important role in regulating the perceptions of the people who come to the ecotourism site both from the content or the ease of access. Practically this research can be used as a strategic reference by stakeholders, to be able to use Social Media as media for promotion and education. Then academically, this research can be a reference for further research on the strategy of managing ecotourism with social media. This research is a descriptive type with quantitative and qualitative approaches, with research locations in Malang. Research using an online questionnaire with Google Form. The results are, Instagram is the most used social media by students in Malang. And for the management strategy with Instagram, it can be done with an aggressive strategy that is utilizing Strength and Opportunities from Instagram such as fast, cheap, environmentally friendly and widely used by students and can be more optimized with the AISAS strategy.Keywords: ecotourism, facebook, Instagram, Malang, social media, strategy, student.

Abstract

Kidney stones are one of the urologic diseases that have plagued mankind for centuries. The main constituents of stones in the kidney are calcium oxalate monohydrate (COM) crystals. Nowadays, there are varieties of drugs and treatments that can be made to minimize the grievances due to kidney stone disease. The treatment can be done either by using chemicals or traditional medicine. Calcusolâ„¢ is one of the popular herbal products that have been used by Indonesian people in curing the kidney stone disease. The main constituent that was contained in Calcusolâ„¢ is an extract of the tempuyung leaves (Sonchus arvensis L.), which is expected could cure the kidney stone disease. This study used primary cultured renal epithelial cells of mice to determine the optimal dose of Calcusol and the optimal dose of COM. The primary Kidney epithelial cell were treated with Calcusolâ„¢ and COM at various doses. The analysis of the cell death either by necrosis or apoptosis pathways was analyzed by flow cytometric analysis. The results that were obtained is the percentage of cell death that is then analyzed by using a complete randomized design (CRD) One Way Anova. Based on the results that were obtained, it is known that the optimal dose of Calcusolâ„¢ in vitro were ranging from 75 ppm to 100 ppm, whereas the optimal dose of COM suggested for 500 ppm.

Abstract

Diabetes mellitus is a metabolic disease that is caused either by the decrease of insulin secretion frompancreatic β cells or the insensitivity of target cells against insulin. High glucose levels (hyperglycemia condition)can trigger the formation of free radicals, the main cause of diabetes micro and macrovascular complications. Theformation of free radicals and AGE (advanced glycation end-products) is assumed to became the key factor in thedecline of granulocyte cell production as well as the disruption of these cells functional activity. The purpose ofthis research was to determine the role of VipAlbumin® in inhibiting the adverse effects of increased blood glucoselevels, which highly influence the production of granulocyte. This study was divided into in vitro and in vivostage. BALB/C mice were used as experimental animals at in vivo stage and induced to undergo diabetes through100 mg/kg BW streptozotocin (STZ) injection at the age of 5 days. VipAlbumin® administered orally for 14 days,which began when mice reached the age of 14 weeks. The administration of VipAlbumin® divided into 3 dosesi.e. 0,01664 mg/gr BW (1st dose), 0,416 mg/gr BW (2nd dose), and 10,4 mg/gr BW (3rd dose). The further step wasa flowcytometric analysis to see the development of granulocyte cells relative amount, which were isolated fromthe bone marrow. The result of this analysis shows that VipAlbumin® administration, particularly at the 2nd and3rd dose, were able to modulate granulocyte cells development in the bone marrow.

Abstract

CD4+CD25+FOXP3+ is a regulatory T cell that plays a role in the body's tolerant system to control activated effector cells. T regulatory plays an important role as a trigger for tumor progressiveness, where an increase in the amount of T regulatory FOXP3 in the tumor state correlates with a poor prognosis. Breast cancer is a type of cancer that tends to increase every year. The purpose of this study was to study the influence of a combination of Phyllanthus urinaria and Catharanthus roseus plant extracts on CD4+CD25+FOXP3+ modulation. Mice was injected subcutaneously with the carcinogen compound DMBA for six weeks at 0.015 mg/kg weight. The study design was conducted using a complete random design (RAL) which was divided into six groups namely healthy (normal) control, cancer control (DMBA injection), Cisplatin and a combination of extracts with three different doses (dose 1 (Phyllanthus urinaria 500 weight and Catharanthus roseus 75mg/kg weight) and dose 3 (Phyllanthus urinary 2000mg/kg weight and Catharanthus roseus 375mg/kg weight). The relative number of T regulatory cells is measured using flow cytometry analysis. The measurements were made using the Cell Quest ProTM program. Statistics analyze using one way ANOVA (p<0.05). Tukey HSD analysis with SPSS 26 software used to analyze real differences between treatments. A relative increase in CD4+CD25+FOXP3+ occurs in cancer control. While the decrease occurred in every treatment except in the dose group 1. The optimal dose to reduce CD4+CD25+FOXP3+ amounts significantly occurred in the dose group 3. This indicates the influence of a combination of Phyllanthus urinaria and Catharanthus roseus on CD4+CD25+FOXP3+ differentiation. Abstract

Tujuan penelitian adalah mengetahui ultrastruktur oosit pasca vitrifikasi menggunakan transmisi elektron mikroskop. Ovarium dikumpulkan dari rumah potong hewan dan oosit dikoleksi dengan cara aspirasi dari folikel menggunakan jarum ukuran 18 G. Setelah dilakukan klasifikasi kualitas  oosit,  maka  oosit  yang  berkualitas  A  dimaturasi  secara  in  vitro. Oosit   hasil  maturasi  dipaparkan  ke  dalam  larutan  vitrifikasi  yang mengandung 30%  EG  + 0,5 M  sukrosa  dengan  lama  waktu paparan  3 menit,  oosit  dimasukkan  ke dalam  ministraw  0,25 cc  (French  straw), kemudian dipaparkan pada uap nitrogen selama 10 detik dan dimasukkan dalam kontainer nitrogen cair. Setelah 3 minggu ministraw diambil dan dilakukan penghangatan (warming) di udara selama 10 detik, dimasukkan dalam waterbath suhu 35 C selama 30 detik. Isi ministraw dituangkan ke dalam cawan petri dan oosit dibilas dua kali dengan  sukrosa 0,5 M.  Selanjutnya dilakukan pengamatan ultrastruktur oosit dengan transmisi elektron mikroskop. Hasil penelitian menunjukkan oosit hasil vitrifikasi mempunyai zona pelusida yang abnormal (fraktur), mem bran plasma mengalami lisis, beberapa butir korteks mengalami degenerasi dan perpindahan butir korteks di ruang perivitelin. Kesimpulan dari penelitian ini adalah terdapat perubahan ultrastruktur pada oosit setelah vitrifikasi.

Abstract

Tujuan penelitian ini untuk mengetahui pengaruh induksi EGF terhadap lokalisasi Cx43 dan gap junction selama ekspansi sel kumulus. Hasil penelitian ini menunjukkan bahwa ekspansi sel kumulus mempunyai signifikansi dengan meningkatnya konsentrasi EGF dan waktu kultur. Ekspansi sel kumulus diindikasikan dengan perubahan bentuk sel menjadi memanjang dan menyebar. Ekspresi Cx43 meningkat pada kultur jam ke-5 dan semakin meningkat pada kultur jam ke-10 dengan semakin padatnya protein pada badan sel. Pada kultur jam ke-15 terjadi penurunan ekspresi Cx43 pada badan sel. EGF berpengaruh terhadap peningkatan ekspansi sel kumulus tetapi tidak pada ekspresi Cx43 dengan bertambahnya konsentrasi dan waktu kultur. Hal ini menunjukkan bahwa komunikasi antar sel kumulus menurun, karena jarak antar sel yang semakin jauh.

Abstract

This study aimed to develop an epitope-based vaccine of SARS-CoV-2 S protein through an immuno-informatics study. The whole genome of SARS-CoV-2 sequences was obtained from the GISAID database and then trimmed to obtain the S protein sequences. The alignment was done by Clustal-W of MEGA software. Epitope prediction and modeling were performed by Discotope BepiPred and the PepFold3 web server. The allergic responses and physicochemical characteristics of predicted epitopes were analyzed using the AlgPred and ProtParam from ExPASy. Molecular docking and dynamic stimulation were performed using AutoDock Vina and YASARA. Biovia Discovery Studio 2019 was used to visualize the molecular docking results. The study predicted 3 potential epitopes, including ‘GDEVRQIAPGQTGKIADYNYKLP’ (epitope 1), ‘YTMSLGAENSVAYSNN’ (epitope 2), and ‘VNNSYECDIPI’ (epitope 3) located in the spike head specifically RBD region. The epitopes did not show an allergen reaction based on IgE epitope mapping. The suitable overexpression for the host of epitopes was mammalian cells. Only epitopes 1 and 2 were stable (instability index above 40). Epitopes 1, 2, and 3 interacted with BCR with binding affinity values -6.6, -7.8, and -7.5 kcal/mol. Epitope 2 wasere stable when interacting with the BCR. Therefore, three epitopes were predicted to have high potency as the SARS-CoV-2 epitope-based vaccine.

Abstract

Kegiatan pengabdian masyarakat ini dilakukan di Pesantren Al Fatih di Desa Klampar Kecamatan Proppo, Kabupaten Pamekasan Madura. Tujuan kegiatan pengabdian kepada masyarakat ini adalah untuk meningkatkan kualitas pengelolaan usaha peternakan sapi pedaging, dan untuk meningkatkan peran dan fungsi pondok pesantren sebagai pendidikan non formal guna mendukung ketahanan pangan serta penguatan penerapan biosecurity dalam upaya pencegahan PMK. Metode yang digunakan dalam kegiatan ini melalui observasi lokasi, diskusi dan wawancara, Participatory Rural, Participatory Technology Development, Educative, Forum Group Discussion (FGD). Hasil kegiatan menunjukkan bahwa peserta sangat antusias saat pelaksanaan. Edukasi yang diberikan pada peserta memberikan peningkatan pengetahuan tentang biosecurity yang signifikan, dari nilai pre-test 68.86 point, sedangkan nilai post-test 78.16 point, sehingga terjadi peningkatan 14%.  Pengetahuan yang diaedukasikan meliputi; biosecurity PMK, sanitasi kandang, biosecurity keluar masuk wilayah farm/peternakan dan isolasi sapi yang sakit. Kesimpulan dalam kaegiatan ini adalah, peserta mampu memahami tentang materi peningkatan kualitas ternak melalui biosafety dan biosecurity PMK dan menerapkan teknologi tepat guna tentang formulasi pakan, dan sanitasi kandang untuk pencegahan dan reinfeksi PMK.
