Abstract

The current study aimed to investigate the effectiveness of Elephantopus scaber and Sauropus androgynus as anti-inflammatory agents in pregnant mice infected with Escherichia coli. This study used seven treatments group (n=3): K- (healthy pregnant mice), K+ (pregnant mice infected with E. coli), P1 (pregnant mice infected with E. coli + E. scaber 100%), P2 (pregnant mice infected with E. coli + E. scaber 75% and S. androgynus 25%), P3 (pregnant mice infected with E. coli + E. scaber 50% and S. androgynus 50%), P4 (pregnant mice infected with E. coli + E. scaber 25% and S. androgynus 75%), P5 (pregnant mice infected with E. coli + S. androgynus 100%). Flow cytometry analysis was used to analyze cell populations expressing CD4+TNFα+, CD4+IFNγ+ and regulatory T cells (CD4+CD25+CD62L+). All treatment groups significantly (p<0.05) decreased TNFα and IFNγ levels, while the P2 group was more effective in increasing regulatory T cells at the 1st and 2nd trimesters of the pregnancy. This study showed that E. scaber and S. androgynus combination alleviated inflammation by reducing inflammatory cytokines (TNFα and IFNγ) and increasing T-regulatory cells. Therefore, E. scaber and S. androgynus combination could suppress the inflammation during pregnancy and infection. Abstract

Pulmonary fibrosis is a chronic disease characterized by progressive connective tissue deposition that replaces healthy lung tissue. This study aimed to investigate the effect of Elephantopus scaber L. Ethanol Extract (ESEE) treatment on the relative number of IL-2 cytokine and lymphocyte activation in bleomycin (BLM)-induced pulmonary fibrosis mice model. Fifty-six male BALB/c mice were divided into seven treatment groups: N (normal); V or vehicle (corn oil); PF or Pulmonary Fibrosis (BLM 2 mg.kg-1); Dex (Dexamethasone 3 mg.kg-1 + BLM); D1-D3 (ESEE at doses of 0.0504, 0.1008, and 0.2016 mg.kg-1 BW + BLM). ESEE, dexamethasone, and corn oil were administered orally, followed by intraperitoneal bleomycin injection daily for 14 days. Mice were dissected on days 7 and 14, and spleens were isolated to analyze cell populations expressing CD4+IL-2+, CD8+IL-2+, CD4+CD62L+, and CD8+CD62L+. The results showed that bleomycin injection could increase the relative number of IL-2 and decrease the relative number of naive T cells compared to normal mice. ESEE treatment significantly reduced the relative number of IL-2, thus decreasing naive T cell activation after one week of bleomycin injection compared to the mice model. In contrast, the increased IL-2 production led to the increasing naive T cell activation after two weeks of bleomycin injection. Therefore, ESEE treatment has the potential to maintain homeostasis through modulation of IL-2 production and T-lymphocyte activation in the pulmonary fibrosis mice model. Abstract

IL-6 and IL-17 are pro-inflammatory and pro-fibrotic cytokines that increase pulmonary fibrosis due to lung alveolar epithelial cell damage. Tapak liman leaves (Elephantopus scaber L.) have anti-inflammatory and anti-asthmatic properties. This study aimed to investigate the effects of the Elephantopus scaber L. ethanol extract (ESEE) on IL-6 and IL-17 produced by CD4+ and CD8+ in the bleomycin-induced pulmonary fibrosis mice model. Fifty-six male BALB/c mice will be divided into seven groups consisting of healthy mice (N), vehicle mice (V), pulmonary fibrosis (PF), Dexamethasone (DEX) as a drug control, and three doses of ESEE (0.0504, 0.1008, and 0.2016 mg.kg-1 BW). ESEE will be administered orally, followed by intraperitoneal bleomycin injection for 14 days. Mice are then dissected on days 7 and 14, and the spleen will be isolated for analysis of the expression of IL-6 and IL-17. The results showed that ESEE effectively reduced levels of IL-6 and IL-17 cytokines produced by CD4+ and CD8+ T cells, and doses three of ESEE (0.2016 mg.kg-1 BW)  (0.2016 mg.kg-1 BW) showed the most effective reduction activity than the Dexamethasone group. The treatment was proven to reduce the expression of IL-6 and IL-17 in mice with a model of pulmonary fibrosis. Keywords: bleomycin, Elephantopus scaber L., IL-6, IL-17, pulmonary fibrosis.

Abstract

Calcium carbonate (CaCO3) has many benefits for medicine, manufacturing, and nanotechnology due to its antibacterial and anti-inflammatory properties. Interlocking bricks (i-bricks) are widely used in sustainable housing for efficient air circulation and reducing humidity inside the house. If CaCO3 is mixed with interlocking brick, it can inhibit bacterial growth, leading to a healthier indoor environment. Therefore, this study aimed to investigate the physical characterization and antibacterial activity of CaCO3-reformulated i-brick, and its anti-inflammatory potential in mice infected with Salmonella typhimurium. The physical characterization of CaCO3-reformulated i-brick (F1-F10 formulas), including its density, compression, and absorption, was analyzed. The inhibitory activity of CaCO3-reformulated i-brick on S. typhimurium growth was determined using a linear regression equation between culture age and cell density values. BALB/c male mice were randomly divided into four groups (n=4): healthy mice (N), S. typhimurium-infected mice (S), S. typhimurium-infected mice treated with F6 formula (F6) and F7 formula (F7) for 7 days. After 7 days of treatment, lymphocytes were isolated to determine the relative number of TGF-beta, interleukin (IL)-10, and TNF-alpha using flow cytometry analysis. The results showed that formulas 6 and 7 of CaCO3-reformulated i-bricks exhibited optimal physical properties, including density, compression, and water absorption. These formulas also inhibited the growth of S. typhimurium. Furthermore, formulas 6 and 7 possessed anti-inflammatory effects in S. typhimurium-infected mice indicated by the high increase in IL-10 and TGF-beta production and low levels of TNF-alpha. In conclusion, CaCO3 brick formulation can inhibit the growth of S. typhimurium and exert an anti-inflammatory effect. Further investigation is needed to reveal the effect of CaCO3-reformulated i-bricks on another inflammatory marker to further elucidate its anti-inflammatory mechanism.

Abstract

Phenolic and flavonoid compounds have been found to have positive benefits due to their antioxidant activity. Alpinia purpurata (Viell.) K. Schum, or lengkuas merah, is known to have aromatic rhizomes and is rich in active compounds such as flavonoids and phenolics. This research aimed to investigate the total phenolic and flavonoid content of A. purpurata ethanol extract and its antioxidant activity. A. purpurata was extracted using Microwave-Assisted Extraction (MAE) with 97% ethanol solvent. The ethanol extract of A. purpurata was then analyzed for the total phenolic and flavonoid content. The antioxidant activity and nitric oxide (NO) levels were determined by DPPH and NO scavenging assay, respectively. The results showed that A. purpurata ethanol extract had a high phenolic content (148.76 ± 1.03 mg GAE.g-1extract) but weak DPPH scavenging activity (IC50 of 219.06 ± 8.15 µg.mL-1), indicating that high phenolic content does not necessarily correlate with strong antioxidant properties. In contrast, the extract exhibited strong NO scavenging activity with an IC50 of 22.73 ± 7.57 µg.mL-1. Despite its high phenolic content, the weak DPPH activity indicates that total phenolic concentration alone is not always a reliable indicator of antioxidant strength. Further studies should include additional antioxidant assays to comprehensively evaluate the extract’s antioxidant potential.

Abstract

The carcinogenic compounds such as DMBA known for having the ability to promote the DNA damage that affects the growth regulator genes. Therefore, uncontrolled cell growth will occur and change the immune system. Elephantopus scaber extract is considered as an alternative way of handling breast cancer since it capable to affect the work of the immune system. Hence, this study aims to determine the effect of E. scaber extract on quantitative changes of CD4+, CD8+ T cells and proinflammatory cytokines CD4+IFNγ+ and CD4+TNF-α+ after induction of DMBA and estrogen. In vivo experiments were carried out by using 5-6 weeks old of female mice BALB/c strain. The extract of E. scaber leaf powder was obtained from Material Medica Batu, Malang. The experimental animals were divided into five groups such as negative control group (K-), positive control group (K+), mice treated with E. scaber extract for a week (group 1), mice treated with E. scaber extract for 2 weeks (group 2), and mice treated with E. scaber extract for 3 weeks (group 3). Induction of DMBA and estrogen was conducted by injection of DMBA (7.12 dimetylbenz (α) anthracene) 0.56 mg.kgBW-1 and estradiol hormone 0.0504 mg.kgBW-1. Mice were treated with E. scaber extract with a dose of 50 mg.kgBW-1. Lymphocytes were isolated from spleen and observed the CD4+, CD8+ T cells and proinflammatory cytokines CD4+IFN-γ+ and CD4+TNFα+ T cells through flow cytometry analysis. One-way ANOVA (p <0.05) and SPSS were used to analyze this data. The results showed that the induction of DMBA and estradiol hormone affected mice fur loss. It also decreased the immune system by lowering the number of CD4 and CD8 T cells. However, E. scaber extracts increased the number of pro-inflammatory cytokines TNF-α. Furthermore, the DMBA and estradiol induction also promoted the number of CD4+IFN-γ+ in the first week, but decreased the number of CD4+IFN-γ+ at weeks 2 and 3, then increase the number of CD4+IFN-γ+. So, the treatment of E. scaber extracts demonstrated the ability to restore the homeostasis of the immune system in the first and second weeks.  Keywords: CD4+, CD8+, DMBA, estrogen, E.scaber, IFN-γ+, TNF-α+

Abstract

The spike protein of SARS-CoV-2 is crucial for initiating infections by binding to host cells and mediating membrane fusion. In this study, HEK-293T cells were transfected with plasmids encoding three structural proteins of SARS-CoV-2, i.e., Spike (S), Membrane (M), and Envelope (E). This transfection enabled the formation of SARS-CoV-2 Virus-Like Particles (VLPs), which allows for safer studies of the virus and its proteins. To serve as a marker for expression, an enhanced green fluorescent protein (EGFP) was fused to the spike protein, resulting in a Spike-EGFP (S-EGFP) fusion protein. The characteristics of SARS-CoV-2 spike protein expression in the transfected HEK-293T cells were then investigated using polymerase chain reaction (PCR) and flow cytometry. The PCR analysis revealed non-specific DNA band smearing, which did not provide conclusive confirmation of spike protein expression. However, flow cytometry analysis demonstrated that approximately 30% of the transfected cells exhibited green fluorescence, indicating the expression of the Spike-EGFP fusion protein. These findings, obtained through flow cytometry, confirmed the successful spike protein expression in transfected HEK-293T cells.

Abstract

Tujuan penelitian adalah mengetahui ultrastruktur oosit pasca vitrifikasi menggunakan transmisi elektron mikroskop. Ovarium dikumpulkan dari rumah potong hewan dan oosit dikoleksi dengan cara aspirasi dari folikel menggunakan jarum ukuran 18 G. Setelah dilakukan klasifikasi kualitas oosit, maka oosit yang berkualitas A dimaturasi secara in vitro. Oosit hasil maturasi dipaparkan ke dalam larutan vitrifikasi yang mengandung 30% EG + 0,5 M sukrosa dengan lama waktu paparan 3 menit, oosit dimasukkan ke dalam ministraw 0,25 cc (French straw), kemudian dipaparkan pada uap nitrogen selama 10 detik dan dimasukkan dalam kontainer nitrogen cair. Setelah 3 minggu ministraw diambil dan dilakukan penghangatan (warming) di udara selama 10 detik, dimasukkan dalam waterbath suhu 35 C selama 30 detik. Isi ministraw dituangkan ke dalam cawan petri dan oosit dibilas dua kali dengan sukrosa 0,5 M. Selanjutnya dilakukan pengamatan ultrastruktur oosit dengan transmisi elektron mikroskop. Hasil penelitian menunjukkan oosit hasil vitrifikasi mempunyai zona pelusida yang abnormal (fraktur), mem bran plasma mengalami lisis, beberapa butir korteks mengalami degenerasi dan perpindahan butir korteks di ruang perivitelin. Kesimpulan dari penelitian ini adalah terdapat perubahan ultrastruktur pada oosit setelah vitrifikasi.

Abstract

Tujuan penelitian ini untuk mengetahui pengaruh induksi EGF terhadap lokalisasi Cx43 dan gap junction selama ekspansi sel kumulus. Hasilpenelitian ini menunjukkan bahwa ekspansi sel kumulus mempunyai signifikansi dengan meningkatnya konsentrasi EGF dan waktu kultur. Ekspansi sel kumulus diindikasikan dengan perubahan bentuk sel menjadi memanjang dan menyebar. Ekspresi Cx43 meningkat pada kultur jam ke-5 dan semakin meningkat pada kultur jam ke-10 dengan semakin padatnya protein pada badan sel. Pada kultur jam ke-15 terjadi penurunan ekspresi Cx43 pada badan sel. EGF berpengaruh terhadap peningkatan ekspansi sel kumulus tetapi tidak pada ekspresi Cx43 denganbertambahnya konsentrasi dan waktu kultur. Hal ini menunjukkan bahwa komunikasi antar sel kumulus menurun, karena jarak antar sel yang semakin jauh.

Abstract

Lake is one of the important natural resources in north Sulawesi and many lakes ecosystem in this province has been used and involved in tourism industry. The aims of the research is to describes the recent status of lakes in North Sulawesi and describes its opportunities for tourism development. The analysis was based on the secondary data and direct field observation. Four lakes, namely Tondano, Pangolombian, Linow and Tampusu have been used as tourism attraction in various degreed. The most intensive used was Lake Tondano, while Lake Linow recently grows as one of the favorites object. Lake Pangolombian and Tampusu receive few tourist visitations. The development of four lakes as tourism object will increase the attractiveness of North Sulawesi as ecotourism development. In such a case, the conservation of lakes becomes crucial.
