Abstract

Piper Crocatum Ruiz & Pav leaves often be used to treat various diseases, including cancer, empirically. This study aimed to analyze the anticancer activities of Piper Crocatum bioactive compounds via In Silico analysis. The methods were biological activity analysis, cell line cytotoxicity activity, SwissADME, STITCH, molecular docking, and molecular dynamics simulation. The investigated bioactive compounds were β-asarone, methyl piperonylketone, and coumaric acid. The results showed the biological activities of the compounds related to anticancer were anti-mutagenic, TNF expression inhibitive, and MMP9 expression inhibitive. The prediction of cytotoxicity analysis results showed that investigated bioactive compounds were toxic on various tumor cell lines. Based on swissADME results, almost all compounds have good pharmacological properties, except coumaric acid. Docking analysis demonstrated the presence of bioactive compounds inhibited TNFa, HER2, and MMP9 as the target protein. The molecular dynamic result was confirmed using molecular dynamic and it shows that β-asarone interaction was stable against MMP9 and TNF protein showed by the low RMSD value. The study found the investigated bioactive compounds of Piper crocatum have the activity of anti-cancer via the inhibition of TNFa and MMP9 protein. However, further research still needs to be done to confirm the prediction results of this In Silico study. Keywords: anticancer, In Silico, molecular docking, Piper crocatum.

Abstract

Lake is one of the important natural resources in north Sulawesi and many lakes ecosystem in this province has been used and involved in tourism industry. The aims of the research is to describes the recent status of lakes in North Sulawesi and describes its opportunities for tourism development. The analysis was based on the secondary data and direct field observation. Four lakes, namely Tondano, Pangolombian, Linow and Tampusu have been used as tourism attraction in various degreed. The most intensive used was Lake Tondano, while Lake Linow recently grows as one of the favorites object. Lake Pangolombian and Tampusu receive few tourist visitations. The development of four lakes as tourism object will increase the attractiveness of North Sulawesi as ecotourism development. In such a case, the conservation of lakes becomes crucial. Abstract

Indonesia categorized as a mega-biodiversity country, with 25% of the species in the world. One of the uses of natural resources is in the tourism sector. Lately, in Indonesia, research about Social Media and Ecotourism is still not much done. Some studies in Indonesia mostly still view social media as marketing advice. In fact, social media also plays an important role in regulating the perceptions of the people who come to the ecotourism site both from the content or the ease of access. Practically this research can be used as a strategic reference by stakeholders, to be able to use Social Media as media for promotion and education. Then academically, this research can be a reference for further research on the strategy of managing ecotourism with social media. This research is a descriptive type with quantitative and qualitative approaches, with research locations in Malang. Research using an online questionnaire with Google Form. The results are, Instagram is the most used social media by students in Malang. And for the management strategy with Instagram, it can be done with an aggressive strategy that is utilizing Strength and Opportunities from Instagram such as fast, cheap, environmentally friendly and widely used by students and can be more optimized with the AISAS strategy.Keywords: ecotourism, facebook, Instagram, Malang, social media, strategy, student.

Abstract

Tujuan penelitian adalah mengetahui ultrastruktur oosit pasca vitrifikasi menggunakan transmisi elektron mikroskop. Ovarium dikumpulkan dari rumah potong hewan dan oosit dikoleksi dengan cara aspirasi dari folikel menggunakan jarum ukuran 18 G. Setelah dilakukan klasifikasi kualitas  oosit,  maka  oosit  yang  berkualitas  A  dimaturasi  secara  in  vitro. Oosit   hasil  maturasi  dipaparkan  ke  dalam  larutan  vitrifikasi  yang mengandung 30%  EG  + 0,5 M  sukrosa  dengan  lama  waktu paparan  3 menit,  oosit  dimasukkan  ke dalam  ministraw  0,25 cc  (French  straw), kemudian dipaparkan pada uap nitrogen selama 10 detik dan dimasukkan dalam kontainer nitrogen cair. Setelah 3 minggu ministraw diambil dan dilakukan penghangatan (warming) di udara selama 10 detik, dimasukkan dalam waterbath suhu 35 C selama 30 detik. Isi ministraw dituangkan ke dalam cawan petri dan oosit dibilas dua kali dengan  sukrosa 0,5 M.  Selanjutnya dilakukan pengamatan ultrastruktur oosit dengan transmisi elektron mikroskop. Hasil penelitian menunjukkan oosit hasil vitrifikasi mempunyai zona pelusida yang abnormal (fraktur), mem bran plasma mengalami lisis, beberapa butir korteks mengalami degenerasi dan perpindahan butir korteks di ruang perivitelin. Kesimpulan dari penelitian ini adalah terdapat perubahan ultrastruktur pada oosit setelah vitrifikasi.

Abstract

Tujuan penelitian ini untuk mengetahui pengaruh induksi EGF terhadap lokalisasi Cx43 dan gap junction selama ekspansi sel kumulus. Hasil penelitian ini menunjukkan bahwa ekspansi sel kumulus mempunyai signifikansi dengan meningkatnya konsentrasi EGF dan waktu kultur. Ekspansi sel kumulus diindikasikan dengan perubahan bentuk sel menjadi memanjang dan menyebar. Ekspresi Cx43 meningkat pada kultur jam ke-5 dan semakin meningkat pada kultur jam ke-10 dengan semakin padatnya protein pada badan sel. Pada kultur jam ke-15 terjadi penurunan ekspresi Cx43 pada badan sel. EGF berpengaruh terhadap peningkatan ekspansi sel kumulus tetapi tidak pada ekspresi Cx43 dengan bertambahnya konsentrasi dan waktu kultur. Hal ini menunjukkan bahwa komunikasi antar sel kumulus menurun, karena jarak antar sel yang semakin jauh.

Abstract

Pregnancy has a high risk and is more susceptible to infectious diseases. E. scaber and S. androgynus contains saponins and flavonoids as an immunomodulatory that can increase the body's immunity. The purpose of this study is to determine the immunomodulatory effects of E. scaber and S. androgynus leaf extract against cellular immune response in pregnant mice infected S. typhimurium. This research used seven treatment groups including negative control (K-) mice without injected S. typhimurium, positive control (K+) mice were injected S. typhimurium and extract treatment with 5 variant doses and dissected on the 12th and 18th days. Lymphocytes was isolated from the blood, then relative number of CD4+ and CD8+ were analyzed with flow cytometry and data continued with ANOVA. Statistical analysis showed that different extract dose and duration of treatment had a significant effect on the number of CD4+ and CD8+ T cells. E. scaber and S. androgynus act as immunomudulatory effect and treatment with combination of extracts E. scaber and S. androgynus 50 mg.kg-1 BW + 112.5 mg.kg-1 BW respectively and treatment with extract of S. androgynus 150 mg.kg-1 BW are the optimum treatment which can restore immune system conditions such as normal pregnancy without infection.Keywords: Cellular Immune, E. scaber, Immunomodulatory, S. androgynus, S. typhimurium.

Abstract

Tapak liman (Elephantopus scaber L) is one of the traditional medicinal plants, which are containing several active compounds that potentially affecting hematopoietic. The aims of this study are to elucidate the effect of leaf extract of Tapak Liman on hematopoietic in mice BALB/c, by observation on relative number of cells expressing CD4/CD8, CD4/CD62L, and TER119/B220 in the spleen; TER119/B220, TER119/VLA-4 and TER119/CD34 in bone marrow, after given the leaf extract for 2 weeks. This experiment was used 12 female mice, which is divided on 3group treated with leaf extract of Tapak Liman (P1= 0.5 g. g bw-1.day-1, P2= 1.0 g. g bw-1.day-1 and P3 = 2.0 g. g bw-1.day-1) and control. The relative numbers of cells expressing surface molecules were analyzed by flowcytometry and quantitative data were tested using one way ANOVA. The result showed the leaf extract of Tapak Liman has no significant effect on erythrocyte proliferation, on the other hand it have significant effect on both proliferation and differentiation of B lymphocytes (B220+) in the bone marrow (p=0.044) and increase the expression of CD4+, CD8+molecule in B cells (p=0.026) and erytroid (TER119+VLA-4+) in spleen and bone marrow. The administration of 1 g. g bw-1.day-1 of the extract (P2) stimulate proliferation of lymphocytes and erythrocytes lineage (TER119+VLA-4+), in spleen and bone marrow.Keywords: CD4+, CD8+, Elephantopus scaber, erythrocyte, hematopoiesis.

Abstract

This study aimed to develop an epitope-based vaccine of SARS-CoV-2 S protein through an immuno-informatics study. The whole genome of SARS-CoV-2 sequences was obtained from the GISAID database and then trimmed to obtain the S protein sequences. The alignment was done by Clustal-W of MEGA software. Epitope prediction and modeling were performed by Discotope BepiPred and the PepFold3 web server. The allergic responses and physicochemical characteristics of predicted epitopes were analyzed using the AlgPred and ProtParam from ExPASy. Molecular docking and dynamic stimulation were performed using AutoDock Vina and YASARA. Biovia Discovery Studio 2019 was used to visualize the molecular docking results. The study predicted 3 potential epitopes, including ‘GDEVRQIAPGQTGKIADYNYKLP’ (epitope 1), ‘YTMSLGAENSVAYSNN’ (epitope 2), and ‘VNNSYECDIPI’ (epitope 3) located in the spike head specifically RBD region. The epitopes did not show an allergen reaction based on IgE epitope mapping. The suitable overexpression for the host of epitopes was mammalian cells. Only epitopes 1 and 2 were stable (instability index above 40). Epitopes 1, 2, and 3 interacted with BCR with binding affinity values -6.6, -7.8, and -7.5 kcal/mol. Epitope 2 wasere stable when interacting with the BCR. Therefore, three epitopes were predicted to have high potency as the SARS-CoV-2 epitope-based vaccine.
