Abstract

Kulit kacang tanah mengandung luteolin yang berpotensi sebagai antiinflamasi secara in vitro dan in vivo. Penelitian ini bertujuan untuk mengetahui efek antiinflamasi ekstrak etanol kulit kacang tanah (Arachis hypogaea L.) pada tikus jantan galur Wistar yang diinduksi karagenin Penelitian menggunakan rancangan acak lengkap pola searah. Dua puluh lima ekor tikus putih jantan galur Wistar dibagi menjadi 5 kelompok. Kontrol negatif menggunakan CMC-Na 0,5% 2,5 mL/200 gBB, kontrol positif menggunakan Na-diklofenak 6,75 mg/kgBB. Ekstrak etanol kulit kacang tanah yang diujikan yaitu dosis 50, 100 dan 200 mg/kgBB. Larutan uji diberikan secara peroral dengan volume pemberian 2,5 mL/200 gBB 1 jam sebelum kaki hewan uji diradangkan dengan karagenin 1% secara subplantar. Pengukuran volume kaki tikus dilakukan tiap 0,5 jam selama 6 jam. Data volume udem dihitung nilai AUC (Area Under the Curve) rata-rata volume udem terhadap waktu dan persen daya antiinflamasi. Data dianalisis dengan anava satu jalan dan dilanjutkan uji LSD (Least Significant Difference) dengan taraf kepercayaan 95%.  Hasil penelitian menunjukkan bahwa ekstrak etanol etanol kulit kacang tanah (Arachis hypogae L.) dengan dosis 50, 100 dan 200 mg/kgBB mempunyai persen daya antiinflamasi berturut-turut sebesar 16,33%; 26,39% dan 31,70%, sedangkan Na-diklofenak 6,75 mg/kgBB sebesar 32,24%.Kata kunci: kulit kacang tanah (Arachis hypogaea L.), ekstrak etanol, antiinflamasi

Abstract

Previous research has shown that some compounds in leaves and seeds of sugar apple have a cytotoxic activity. The aim of this research was to determine the cytotoxicity of semipolar fraction of ethanolic extract from sugar apple stem bark (Annona squamosa L.) on T47D cancer cells. The semipolar fraction of ethanolic extract from sugar apple stem bark was collected by fractionation using Vacuum Liquid Chromatography (VLC) with hexane:ethyl acetic (9:1, 8:2, 7:3, and  6:4)  as  mobile  phase.  Cytotoxicity from the fractions of five different concentration namely; 25, 50, 100, 150, and 250, µg/mL was measured by MTT assay. The potency of the cytotoxicity was defined by the ability of the fraction to inhibit the growth of T47D cells indicated by the value of IC50. Qualitative analysis of contained compounds in the fraction was done by Thin Layer Chromatography (TLC) method using silica gel F 254 as a stationary phase and hexane:ethyl acetic (7:3) as a mobile phase. UV 254 and 366 nm lamp also Dragendorff, citroboric, and FeCl3 spray reagents were used to visualize the spots of the secondary metabolites. The result proved that the semipolar fraction of ethanolic extract from sugar apple stem bark showed potential cytotoxicity on T47D cancer cells with IC50value of 70,77 µg/mL. Qualitative analysis showed that the fraction contained flavonoids and alkaloids which is presumably responsible for its cytotoxic activity.

Abstract

Mangrove plants as traditional medicine have long been used by the society for the therapy of anticancer diseases. So far, the potency of mangrove plants as anticancer has not been studied intensively. The methanol extract of R. mucronata stem bark has cytotoxic activity on myeloma cells. This study aims to determine the isolates of the ethyl acetate fraction of the methanol extract of the stem bark of R. mucronata and R. apiculata, and to examine the potency of cytotoxic activity against MCF-7 and T47D cells. The powder of the stem bark of R. mucronata and R. apiculata were extracted with methanol by maceration, and then the methanol extract was fractionated consecutively using n-hexane, ethyl acetate, and methanol. Ethyl acetate fraction was then purified by column chromatography with a diameter of 2 cm and a height of 30 cm with a static phase in the form of silica gel 60 (0.063-0.2 mm) Merck 7734 and eluent n-hexane:ethyl acetate (6: 4, 5: 5, 6: 4, 7: 3, 8: 2, 9: 1) and n-hexane: methanol (9,5: 0,5) 10 ml each. The eluates that produce the same number and pattern of stains were combined into one isolate. Then the eluate was evaporated at room temperature. Then the purification results of the extract R. mucronata were isolated using preparative TLC with 0.25 mm GF254 silica gel as static phase and n-hexane mobile phase: ethyl acetate (6: 4), while the apiculata extract with n-hexane mobile phase: ethyl acetate (5: 5). The results of the stain separation of the compound were scraped off and separated from the static phase using a solvent. The purity of isolates compound was examined using TLC in the static phase of silica gel GF254 0.25 mm. The qualitative test results of the ethyl acetate fraction of R. apiculata stem bark with an NMR spectrophotometer showed a composition of cis-3- (3,5-dihydroxyphenyl) acrylic acid, while the ethyl acetate fraction of R. mucronata stem bark contained n-hexan-3-ol compounds. The cytotoxicity test of ethanol extract, n-hexane fraction, ethyl acetate fraction and ethanol extract was carried out on MCF-7 and T47D cells using the MTT method. The results proved that the ethanol extract, n-hexane fraction, ethyl acetate fraction and ethanol fraction from the ethanol extract of R. mucronata and R. apiculata stem bark, based on cytotoxic activity, were inactive against MCF-7 and T47D cells.

Abstract

Mareme plant (Glochidion arborescens Blume.) is a plant that has the potential to treat cancer. In a study on the leaves and bark of the Mareme (Glocidion arborescens Blume.) plant, tests for antioxidant, antidiabetic, and anti-inflammatory activities were previously carried out. Secondary metabolites that play a role in cytotoxic activity in Mareme plants are flavonoids, and these compounds can specifically kill the progression of cancer cells. This study aims to determine the content of compounds, examine cytotoxic activity, and determine the percentage of living cells in the leaves and bark of Mareme (Glochidion arborescens Blume.). The leaves and bark of Mareme (Glochidion arborescens Blume.) were extracted by maceration using a 96% ethanol solvent, then identification of the compounds was carried out to determine the content contained in the ethanol extract of the leaves and stem bark of Mareme, and then a cytotoxic test was carried out using the MTT assay method. The compounds contained in Mareme leaves are flavonoids, tannins, steroids, saponins, and polyphenolics, while the chemical compounds in Mareme stem bark are flavonoids, tannins, saponins, and polyphenolics. Tests were carried out at concentrations of 500, 600, and 800 µg/mL. The results of the cytotoxic test showed that the value of live cells in the ethanol extract of Mareme leaves (Glochidion arborescens Blume.) was 114.21, 106.21, and 78.91%, respectively. While the value of live cells in the ethanol extract of Mareme stem bark (Glochidion arborescens Blume.) was 59.56, 56.37, and 42.85%, respectively. The higher the concentration, the smaller the living cells produced.

Abstract

Diabetes mellitus is a metabolic disorder indicated by high blood glucose levels. Red castor is a plant that has potential in the treatment of diabetes mellitus. This study aims to measure the impact of ethanol extract from the roots of the red castor plant (Jatropha gossypiifolia L.) in reducing blood glucose levels in male white Wistar rats that have been induced with alloxan at a dose of 150 mg/kgBW. The study used 25 male mice divided into 5 treatment groups. The negative control group was given distilled water as a substitute for the substance being tested, while the positive control group was given a dose of glibenclamide of 5 mg/kgBW. Apart from that, there were 3 treatment groups that were given extract from red castor root at doses of 25, 50 and 100 mg/kgBW respectively. Treatment was given for 14 days. Alloxan 150 mg/kgBW was used so that mice experienced an increase in blood glucose levels exceeding 200 mg/dL. The results of decreasing blood glucose levels respectively at doses of 25, 50, and 100 mg/kgBW were 101.0 mg/dL, 81.6 mg/dL, and 79.4 mg/dL. The percentage reduction in blood glucose levels obtained at doses of 25, 50, and 100 mg/kgBB was 73.63%, 77.89%, and 80.10%. Based on the results of statistical tests, the effect of using ethanol extract of red castor root at doses of 25 mg/kgBB, 50 mg/kgBB, and 100 mg/kgBB is comparable to using 5 mg/kgBB glibenclamide
