Abstract

Identification of Fusarium solani Fungi Associated with Stem Rot Disease in Dragon Fruit Plant (Hylocereus sp.) at Bangorejo District, Banyuwangi Regency Dragon fruit plants at Banyuwangi in the maintenance process always experience disorder such as pathogenic attacks. The part of dragon fruit plant whichis often attacked by pathogens is the stem which is caused the stem to rot. One type of photogenic fungus in dragon fruit plants is Fusarium solani. This fungus attacks the stem of the dragon fruit plant, so that it can cause the stem of the plant looks wilted and brown rot.The cause of stem rot disease of dragon fruit plant in Banyuwangi is not yet known exactly, thus identification in macroscopic, microscopic, and molecular need to be done. Recently, the use of molecular methods in fungi identification has increased to solve this problem. One of the molecular approaches to identify fungi is by using a device called Polymerase Chain Reaction (PCR).The result of this research showed mycelium that grown on PDA media (Potato Dextrose Agar) shaped like a thick cotton expands like a cloud, the shape of the edge of the colony is uneven, the surface color of the colony is white, the color of the bottom of the colony is white, the direction of the growth of the mycelium is to the side and the structure of the mycelium is smooth. In microscopic identification there found a long macroconidia, crescent shaped with blunt ends, 1-5 septa, microconidia shaped in round to oval, long hyphae and bersepta. The molecular identification with PCR to determine the presence of Fusarium solani mushroom by using universal primary pairs of ITS4 and ITS5 successfully amplified and produced DNA fragments measuring 560 bp.

Abstract

Detection of Ocurrence Citrus Vein Phloem Degeneration (CVPD) Disease on Citrus Plant with Comprehensive Symptoms using Polymerase Chain Reaction (PCR) Technique Citrus Vein Phloem Degeneration (CVPD) is one of very serious disease of citrus plant. The disease is caused by Liberobacter asiaticum bacteria and transmitted by Diaphorina citri Kuw insect. The outspread of disease can also caused by the use of citrus seedlings which have been infected by CVPD. Preliminary visual observations CVPD disease symptoms, there are two types of plants showing symptoms of partial and systemic. Partial symptoms are not all the leaves of plants showing symptoms CVPD, while the systemic symptom is on the whole leaves of plants showing symptoms CVPD. This research aimed to determine the outspread of L. asiaticum bacteria in every parts of the plant that made detection by PCR. The research has been conducted from in Batukaang, Bangli and Plaga, Badung then continued at Laboratory of Genetic Resources and Biology Molecular, Udayana University. Results of PCR visualization using 1% agarose gel showed that there were a DNA band at 1160 bp on the leaves, twigs, branches and trunks. Because the size of 1160 bp DNA bands is expression by L. asiaticum, then the citrus leaf samples were detected positive for the L. asiaticum and it can be ascertained that L. asiaticum haas been distributed to all parts of the plant.

Abstract

Effectility of some Extract to Larvae of Plutella xylostella Development in the LaboratoryThe research was conducted in November 2017 to Februari 2018 in Udayana University Plant Pest and Disease Laboratory. The aim of this research is to know the ability of some plant leaf extract to larvae of Diamonback Moth (Plutella xylostella) development in the laboratory. Plant extract materials used in this experiment were leaf of Lantana camara, Chromolaena odorata, Tithonia diversifolia, and Nicotiana tabacum. Process of extracting was done in Laboratory of Genetic Resources and Molecular Biology Faculty of Agriculture Udayana University. This research uses Completely Randomized Design (CRD) with 4 treatment of leaf extract at 10% concentration and 1 without treatment (control), each treatment was repeated 10 times. Observations were made until the larvae does not do activity or until become adult. Testing of four types of plant leaf extracts showed the following results: Each of C. odorata leaf extract, L. camara, T. diversifolia and N.tabacum used as research material had different potential in suppressing P.xylostella development. Application of L. camara, T. diversifolia and N. tabacum on larvae can develop through 4 instars before becoming pupa, treatment with plant leaf extract is most effective to suppress the development of larvae biology P.xylostella is a leaf extract of C. odorata that result were 100% mortality on larvae stase and the lowest is on the treatment of N. tabacum extract were 20%.

Abstract

ABSTRACT Vesicular Arbuscular Mycorrhizae (VAM) Identification of Red Onion (Allium cepa L.) and Taro (Colocasean esculenta (L.)Schott) Rhizosphere and Its Spore Multiplication in Zeolite Media Efforts to increase agricultural production can be done with a variety of technologies. Technology is widely applied to farmers in the cultivation process is the use of inorganic fertilizers with high doses exceeding the dose of balanced fertilizer. In the production of onion crops in the field, also using inorganic fertilizer application on an ongoing basis which may result in damage to the ground both in terms of texture and structure. Red onions and taro roots are shallow causing both of these plants are unable to absorb nutrients to the maximum if exercised enough chemical fertilizer. This study was aimed to determine genus and species of VAM in red onion and taro rhizosphere, its colonization in root tissue, and to examine zeolite media compatibility with corn as a symbiont. Based on the results of the study, it was found three species of VAM spores of red red onion plant rhizosphere, namely: Acaulospora denticulata, A. laevis, Glomus ambisporum, and five species of VAM spores recovered from the rhizosphere of taro plants namely: Acaulospora foveata, A. koskei, A. capsicula, Scutellospora calospora and Glomus ambisporum. Infections were found in plants onion, taro and corn plants form symbiotic arbuscular, vesicles and inner spore. VAM from the rhizosphere of plants onion and taro can be reproduced using zeolite media and symbionts plant corn. Keywords: MVA, rhizosphere, Acaulospora, zeolite, inner spore

Abstract

AbstractThe existence of Hinduism in Bali and generally inIndonesia gained recognition by the state as the official religion through a complicated process. This article will answer the question in the title above to see pemaskaraan (polishing) process and replicate the Balinese religion. The purpose of pemaskaraan (polishing) in order to enhance his form of religion and culture in order to lookmore beautiful than the original. Pemaskaraan (polishing)process is detected when the organic intellectuals manage their fear of the religions of non-Hindu in an effort to maintain the authenticity of Bali. Pemaskaraan (polishing) Balinese religion, emerged as organic intellectuals reaction against the widespread threats of religious conversion in Bali. Pemaskaraan (polishing) realized by smoothing the basis of the Hindu religion.And replication is the impersonation of religion of the majority, so the Balinese religion is not alienated in the society of nation and state.

Abstract

AbstractPuri Kesiman had has a strategic position that functioning asthe headquarters of the struggle in the early twentieth century,since Kesiman involvement in “Puputan of Badung”, andthe Indonesian independence struggle on Bali. As the onlyremaining palace of the King of Badung until now, it will beinteresting to study how the Puri of Kesiman can survive longenough and its action in the present. Especially in relation tothe position and role of the palace (puri) in Balinese society,especially in the increasingly pluralistic Village Kesiman whereit is located. This study discusses the history of the CastleKesiman, Kesiman Puri’s role in the history of Bali general andhistory of Badung particular; and traces the position of PuriKesiman in early independence era till today.

Abstract

This study is an intellectual exercise applying genealogical history writing. Its scope is neither to write factual truths of the past nor to seek the origin of an event, but to raise an issue of a cultural practice of the present. In the second decade of the XXI century, there are still some people who think, talk, and act on their belief in kawitan, glorified ancestors, and make it an entry point for their political interests. The result of this study is some knowledge which is not only valuable to the historiography of Indonesian history but also to the study of oral traditions. Through this study, it is expected that they both can complement each other, and that the science of history does not need to be awkward to use oral tradition as a source of information and researchers studying oral traditions can use genealogical methods in their research

Abstract

AbstractThe purpose of this study is to discuss the relations of power in the history of the contemporary Hindu religion in Bali.  The concept of power relations refers to the concept of Power and Discourse of Michel Foucault, while the term “invented religion” is a development of Invented Tradition by Eric Hobsbawm. “Invented religion” in this study is defined as the practice of Hindu religion, which was invented, constructed, and officially manifested; and the religious practice which emerged in a relatively short time; and in a few years  it has been regarded as an established religion. Based on the concept of Power and Discourse of Foucault, I argue that there are relations of power that shape and sustain the practice of “invented religion.”  I will try to find the power relations in a God worshipping discourse in Samuantiga and Jagatnatha temples. There are two research questions that I raise for the argument, namely how and why these two discourses emerge and thrive in contemporary’s practice of Hinduism?  The answer to both questions will be sought at the present Balinese-Hindu intellectual thought. Then, it is assessed using the Genealogy Method of  Foucault. My conclusion is that the presence of the two discourses is only to harmonize Hinduism with  Islam as the majority religion in Indonesia.

Abstract

Damage and Product Losses by Fruit Flies Bactrocera dorsalis Complex (Diptera: Tephritidae) at Citrus Cultivation. This research knows the damage and nominal loss caused by fruit flies attack on citrus cultivation. The study took place from May to September 2016, from fruit to harvest, fruit fly observations were performed using relative methods. The results showed the damage of fruit at citrus cultivation in place of research have category relative moderate until heavy. Of the 10 plant samples observed 3 damaged plants reached 51%. Then 7 other plant samples suffered damage in the medium category. But the average damage reached 44%, this is a fruit fly is categorized as a very harmful pest. Nominal losses in 10 sample plants observed with the number of fruit fell as much as 943 fruit and fruit weight reached 157.17 kg obtained nominal loss of Rp. 471,500. Based on the data obtained hectar loss of Rp.29.468.750. Fruit fly attacks have increased along with the condition of citrus fruits that begin to experience physiological development so that the loss of the number of fruit fall that resulted in economic losses.

Abstract

Diveristy and Inhibition of Air-Borne Spores That Contaminate Substrat of OysterMushroom (Pleurotus ostreatus (Jacq. Ex Fr) Kummer). The aim of research to know thediversity, prevalence and inhibition of air-borne spores that could potentially contaminatesubtsrat baglog of oyster mushrooms. In this study using the method: (1) the arrest of sporescarried out during the hours of 7 am to 13 pm by placing three petri dish that already containsmedia PDA 3 pieces, and repeated 3 times, (2) inhibition ability of air-borne spores againstoyster mushroom. The place and time of the study conducted at Jl. Siulan Gang Zella No. 7Denpasar, and implemented in October 2014 to February 2015. The results showed that 13genera, including Aspergillus spp, as many as 10 with a prevalence of 13.51%, Aspergillusniger 7 (9.45%), Brachysporium sp., 1 (1.35%) , Cunninghamella sp. 1 (1.35 %), Fusariumspp. as many as 19 (25.6 %), Giotrichum sp. 1 (1.35%), Mucor spp. , a total of 18 (24.32%),Neurospora spp., as many as 8 (10.81%), Penicillium spp. 2 (2.70%), Phytophthora spp. 4(5.40%), Stachybotrys sp. 1 (1.35%), Trichoderma sp. 1 (1.35%) and Umbelopsis sp. 1 (1.35%). The highest prevalence achieved by Fusarium spp . amounting to 25.67 %, followed byMucor spp. 24.32%. Diversity index (H') was obtained from each replicate of 0.6438, 0.7048,0.5611 with a 0.6366 average. Dominance index (C) obtained by 0.6331. 0.8646, 0.6146, andaverage 0.7041. Diversity including < 1, marked by low diversity and dominance index closeto 1, meaning there is dominance. The dominance held by Fusarium spp . and Mucor spp.Inhibitory effects of air -borne fungi on the growth Oyster mushroom in vitro; The highestachieved by Fusarium spp, with inhibition of 94.00 ± 1.2%, followed by Aspergillus spp.amounting to 92.15 ± 1.5%, and the lowest was achieved by Penicillium spp . amounting to70.37 ± 2.5%.
