Abstract

Pencegahan kerusakan jaringan pankreas akibat radikal bebas pada kondisi hiperglikemia kronik sangat penting dan vitamin E dapat bertindak sebagai pemusnah radikal bebas dalam mencegah perkembangan diabetes melitus. Tujuan penelitian ini untuk mengamati pengaruh vitamin E pada kondisi hiperglikemia pada aktivititas enzim superoxide dismutase (SOD), glutathione peroxidase (GPx), dan kadar malondialdehyde (MDA) pada jaringan pankreas tikus. Penelitian dilakukan di Laboratorium Biokimia Fakultas Kedokteran Hewan Universitas Udayana, tahun 2010. Sebanyak 15 ekor tikus putih jantan strain Spraque Dawley umur 3 bulan digunakan dalam penelitian ini. Tikus percobaan dibagi menjadi tiga kelompok perlakuan. Kelompok kontrol negatif (K0), kelompok tikus hiperglikemia (K1), serta kelompok tikus hiperglikemia dan diberi vitamin E dosis 7 mg/200 g bb/hari (K2). Kelompok hiperglikemia diberi larutan 50% glukosa, dosis 1,5 cc/ekor per oral dua kali sehari. Perlakuan diberikan selama 1 bulan. Kadar glukosa darah diperiksa menggunakan metode biosensor glukosa oksidase. Enzim SOD, GPx, dan MDA dianalisis menggunakan metode spektrofotometri. Kadar glukosa darah perlakuan K2 sebesar 153,5±8,2 mg/dL tidak berbeda (p>0,05) bila dibandingkan dengan perlakuan K1 sebesar 154,1±5,7 mg/dL. Aktivitas SOD perlakuan K2 sebesar 43,21±3,32 U/g lebih tinggi dibandingkan dengan perlakuan K1 sebesar 31,99±3,55 U/g (p<0,05). Aktivitas GPx perlakuan K2 sebesar 10,44±0,54 U/g tidak berbeda bermakna (p>0,05) bila dibandingkan dengan perlakuan K1 sebesar 9,39±0,59 U/g. Kadar MDA perlakuan K2 sebesar 20,27±0,87 pmol/g lebih rendah dibandingkan dengan perlakuan K1 sebesar 25,24±0,82 pmol/g (p<0,05). Simpulan, kadar SOD dan GPx rendah, sedangkan kadar MDA tinggi pada pankreas tikus dalam kondisi hiperglikemia. Pada tikus hiperglikemia yang diberi vitamin E, aktivitas SOD dan GPx tinggi, sedangkan kadar MDA rendah. [MKB. 2011;43(2):72–6].Kata kunci: Enzim antioksidan, hiperglikemia, pankreas tikus, vitamin EHiperglycemic and Vitamin E Effect on Malondialdehyde and Antioxidant Intracellular Enzyme in Rat Pancreatic TissuePrevention of pancreatic tissue damage by free radicals under conditions of hyperglycemia chronic would be important and vitamin E can act as scavengers of these oxygen radicals in preventing the development of diabetes mellitus. This study aims was to evaluate the effect of vitamin E on the hyperglycemia conditions on the blood glucose, enzyme superoxide dismutase (SOD), glutathione peroxidase (GPx), and malondialdehyde (MDA) levels in rat pancreatic tissue. Research conducted at the Laboratory of Biochemistry Faculty of Veterinary Medicine Udayana University, in 2010. A total of 15 male rats white of strains Spraque Dawley old 3 months used in this study. All of rats were divided into three groups. Negative control group (K0), group of rats hyperglycemia (K1), and group of hyperglycemia and given vitamin E dose of 7 mg/200 g bw/day (K2). Hyperglycemia group was given 50% glucose solution doses of 1.5 cc/head orally twice daily. Treatment was conducted for 1 month. Blood glucose level was measured by using glucose oxidase biosensor method. Pancreatic SOD, GPx activities, and MDA levels were measured by using spectrophotometric methods. Blood glucose levels in the K2 treatment of 153.5±8.2 mg/dL was not significantly different (p>0.05) when compared with the K1 treatment of 154.1±5.7 mg/dL. Activities of SOD enzymes in the K2 treatment of 43.21±3.32 U/g higher than the K1 treatment of 31.99±3.55 U/g (p<0.05). Activities of GPx enzymes in the K2 treatment of 10.44±0.54 U/g was not significantly different (p>0.05) when compared with K1 treatment of 9.39±0.59 U/g. MDA level in the K2 treatment of 20.27±0.87 pmol/g lower than the K1 treatment of 25.24±0.82 pmol/g (p <0.05). In conclusion, SOD and GPx enzymes levels are low whereas MDA level is high in the rat pancreatic under conditions of hyperglycemia. The hyperglycemic rat by treatment with vitamin E shows high SOD and GPx levels and low MDA levels. Abstract

Bakso merupakan produk olahan daging asal hewan yang sering ditemui di kalangan masyarakat karena rasanya yang enak, relatif murah, mudah disajikan, dan mudah ditemui. Pada tahap produksi, bakso seringkali ditambahkan bahan pengawet dengan maksud untuk memperpanjang masa simpan produk dalam 12-24 jam. Salah satu masalah yang dihadapi pedagang bakso yaitu produk bakso harus habis terjual sebelum mengalami pembusukan sehingga beberapa oknum ditemui mengawetkan produk dagangnya menggunakan bahan berbahaya seperti formalin dan boraks. Penggunaan formalin dan boraks dengan dosis terkecil pada makanan telah dilarang di dalam Peraturan Menteri Kesehatan No. 33 tahun 2012, termasuk pada produk bakso. Penelitian ini bertujuan untuk mengetahui tingkat pemahaman penjual bakso sekaligus mendeteksi secara kualitatif adanya senyawa formalin dan boraks yang dijual di sekitar pasar tradisional di Kota Denpasar. Sampel yang diidentifikasi diambil dari sembilan pedagang menetap, lima pedagang kaki lima, dan enam pedagang keliling selama tiga minggu berturut-turut. Tingkat pemahaman penjual diambil berdasarkan sejumlah pertanyaan kuisioner. Pemeriksaan kualitatif senyawa formalin dilakukan menggunakan reagen NaOH, natrium nitroprusida, dan fenilhidrazin, sedangkan, pemeriksaan kualitatif senyawa boraks dilakukan menggunakan uji kertas kurkumin. Hasil studi menunjukkan persepsi pedagang terhadap penggunaan bahan pengawet berbahaya dinilai sudah baik. Hasil analisis kualitatif terhadap formalin dan boraks pada bakso menunjukkan hasil negatif di semua sampel, dari minggu pertama hingga ketiga.

Abstract

Diabetes is marked by elevated levels of blood glucose, and progressive changes of the structure of pancreatic islet histopathology. The objective of this research was to analyse the glucose level and histophatological feature in pancreatic islet in alloxan compound induced rats. A total of ten male Spraque Dawley rats of 2 months old were used in this study. The rats were divided into two groups: (1) negative control group (K-), and (2) positif induced alloxan group (diabetic group =DM). The rats were induced by a single dose intraperitonial injection of alloxan compound 120 mg/kg of body weight. The treatment was conducted for 28 days. Blood glucose levels of rats were analysed at 0, 4, 7, 14, 21, and 28 days following treatment. At the end of the experiment, rats were sacrificed by cervical dislocation. Pancreas was collected for analysis of histopathological study by Immunohistochemical technique, and ultrastructural study using transmission electron microscope (TEM). The result showed that Langerhans islet of diabetic rat (rat of DM group) showed a marked reduction of size, number of Langerhans islet of diabetic rat decrease, and characterized by hyperglycemic condition. By using TEM, beta cells of DM group showed the rupture of mitochondrial membrane, the lost of cisternal structure of inner membrane of mitocondria, reduction of insulin secretory granules, linkage between cells acinar with free Langerhans islet, and the caryopicnotic of nucleus. Abstract

Oxidative stress is defined as over-production of free radicals which lead to cells damage, pathological condition and cell death. The objective of this study was to analyze respond of oxidative stress and isoflavone treatment on superoxide dismutase (SOD) enzyme activities and lipid peroxidation in rat liver. A total of fifteen male Spraque Dawley rats were used in this study. They were sub-divided into three groups; (1) a negative control group, (2) a stress oxidative group, and (3) treatment by stress condition followed by treatment with isoflavone. Stress condition was achieved by five days fasting together with swimming for 5 mins/day and only drinking water ad libitum. Isoflavone was orally administrated on a dose of 1 mg/200g bw/day for five days. At the end of the experiment, rats were sacrificed by anesthesia. Liver was collected for analysis of SOD enzyme activities, SOD immunohistochemical analysis, and malondialdehyde (MDA) level. Result showed that stress condition increase free radicals that showed by decreased SOD activity, and increased MDA level. Isoflavone treatment could get over reduction of SOD and prevented increase of MDA level in the liver of rats under stress conditions. Abstract

ABSTRAK: Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan secara in vitro dan mengidentifikasi golongan senyawa flavonoid yang terkandung dalam fraksi n-butanol ekstrak kulit terong belanda. Uji aktivitas antioksidan dilakukan dengan metode DPPH. Pemisahan dan pemurnian fraksi aktif n-butanol dilakukan dengan kromatografi kolom menggunakan silika gel yang dielusi dengan n-butanol-etil asetat-asam asetat 10% (2:7:1) dan diidentifikasi dengan spektrofotometer UV-Vis dan FTIR. Fraksi n-butanol memiliki aktivitas antioksidan yang kuat dengan nilai IC50 sebesar 69,89 mg/L. Empat senyawa flavonoid (isolat A, C, H, dan J) berhasil diisolasi dan diidentifikasi sebagai golongan flavon, flavonol dan isoflavon. Senyawa tersebut dipercaya memiliki aktivitas antioksidan.     ABSTRACT: The present study was conducted to determine in vitro antioxidant activity and ctivity and to identify flavonoid class of compounds present in n-butanol active fraction of Terong Belanda (Solanum betaceum cav.) peel extract. Antioxidant activity test was performed by DPPH (1,1-diphenyl-2-pycrylhidrazyl) method. Separation and purification of n-butanol active fraction were done using silica gel column chromatography eluted with n-butanol-ethyl acetate-acetic acid 10% (2:7:1) and identified by UV-Vis and FTIR spectrophotometer. n-Butanol fraction had strong antioxidant activity with IC50 value by 69.89 mg/L. Four flavonoids (A, C, H, and J isolates) were isolated and identified as flavone, flavonol and isoflavone. This compounds are believed to be responsible for their antioxidant activity.

Abstract

The classification between religion and believer by the apparatus of the Indonesia state and zendeling/missionaries creates persuasive discrimination to Marapu believer in Tarung Village. Formal education was made formalization religious instrument to Marapu believer because in the practice, students must profess which is recognized juridically by the apparatus of the Indonesian state. Formulation of the problem in this research involve (1) how the process of formalization religious to Marapu believer through formal education? and (2) how implication of formalization religious to Marapu religiousity in Tarung Village?. The formalization Marapu religious through formal education to Tarung’s people can be studied by applying the theory of hegemony, religious conversion theory, and theological inculturation theory. While the concepts used as a reference in this research are religious formalization, Marapu, formal education, and implication. This qualitative research uses etnography research methods including the technique of collecting data through the observation, interview, literatur study and data analysis in order to process data findings.The research findings show that Marapu believer must be Christianity or Catholic in order to get formal education. Marapu as teachings without scripture, make a different interpretation in Tarung’s people, it can be an obstacle for reach of common identity. For Tarung’s people “half Marapu and half Christian/Catholic” is two principle that workable, behind the succumb strategy between Marapu leaders and Christian/Catholic leaders for the sake of certain interests. Marapu of Tarung’s people have inculturation with basic Christianity/Catholicism.

Abstract

The phenomenon of dating applications is increasingly growing in society. This is evidenced by the rise of dating applications. Tinder is one of the most talked about dating apps. Tinder brings people a change in the way it is social. Since Tinder was launched, now humans can easily make new friends virtually. Dating applications are believed to be able to provide a solution when a match comes for its users. This is what attracts researchers to dig deeper into the phenomenon of matchmaking through the Tinder application as a matchmaking platform in the digital era. The theory used by the researcher is social action theory and theory of love by Wheel theory. This research also uses a qualitative descriptive method and a qualitative approach. The results show that the resource person uses the Tinder application to find new friends, girlfriends and even husbands / wives. The researcher concluded that the actions taken by the informants on average had the same answer. This action is done for the purpose of finding a new socials circle.

Abstract

Radikal bebas berasal dari proses metabolisme dan faktor eksternal yang dapat menyebabkan tidak stabilnya sel darah. Antioksidan dapat melindungi sel darah dari serangan radikal bebas, yang bersumber dari kulit pisang. Tujuan dari penelitian ini untuk mengetahui pengaruh pemberian ekstrak kulit pisang kepok terhadap profil darah tikus putih yang diberikan latihan intensif. Rancangan Penelitian menggunakan metoda rancangan acak lengkap (RAL) dengan sampel 27 ekor tikus putih jantan (200-225 gram/ekor) dibagi menjadi 3 perlakuan yang masing-masing terdiri atas 9 ekor tikus, yaitu kelompok kontrol (P0), kelompok renang (P1), dan kelompok renang dan ekstrak pisang (P2) selama 28 hari. Spesimen darah diambil melalui vena orbitalis, selanjutnya dilakukan uji hematologi lengkap untuk mengentahui profil darah. Hasil penelitian menunjukkan bahwa kadar hemoglobin dan total eritrosit P1 nyata lebih rendah (P<0.05) dari kontrol (P0), sedangkan P2 tidak berbeda nyata dengan kontrol (P0). Total leukosit P1 dan P2 tidak berbeda nyata, akan tetapi nyata lebih tinggi (P<0,05) dari P0. Netrofil dan limfosit pada P1 dan P2 nyata lebih tinggi (P<0,05) dari P0, akan tetapi eosinofil dan monosit tidak berbeda dengan P0. Kadar HCT dan MCHC perlakuan P1 nyata lebih rendah (P<0,05) dari kontrol, sedangkan P2 tidak berbeda dengan kontrol (P1). Kadar MCV baik P1 maupun P2 tidak berbeda dengan kontrol (P0). Dari hasil penelitian ini dapat disimpulkan, ekstrak pisang kepok dapat mempertahankan kadar hemoglobin dan eritrosit namun belum dapat mempertahankan kadar leukosit dan nilai deferensial leukosit.
