Abstract

An study was carried out to determine the physiological data on plasma protein characteristics of Bal cattle. A total of 24 plasma samples of male and female bali cattle were characterized by using sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS PAGE). The data were presented descriptively. The results showed that on the basis of their estimated molecular weights, at least 14 protein bands (1st to band 14th) were identified with the molecular weights of 963,50 kDa, 530 kDa, kDa 346,82, 124,84 kDa, 89,85 kDa, 68,67 kDa, 54,71 kDa, 37,77 kDa, 20,78 kDa, 16,95 kDa, 16,18 kDa, 15,46 kDa, 12,56 kDa and 10,46 kDa, respectively from the highest to the lowest. Furthermore, plasma protein bands of 14 bali cattle were grouped into five fractions, namely albumin, globulin a1, a2, b, and g. Albumin fraction shown by the band 6th to 14th with a molecular weight of 68,67 to 10,46 kDa respectively. Globulin fraction a1 and a2 shown by the band 5th and 4th with a molecular weight of 89,85 kDa and 124,84 kDa respectively. b- globulin fraction shown by the band 3rd with a molecular weight of 346,82 kDa. g-globulin fraction was shown by the band 1st and 2nd with a molecular weight of 963,50 kDa and 530 kDa respectively. The percentage of area band Bali cattle plasma protein fraction consists of 92% albumin, globulin fraction a2 of 3%, g-globulin of 2%, globulin a1 and b of 1%. It can be concluded that the plasma protein band Bali cattle male and female calves (aged 0-1.5 years), puberty (ages 2-2.5 years) and adults (aged 3-5 years) amounted to 14 protein bands with varying thickness comprises of the five fractions are albumin, globulin a1, a2, b, and g. ABSTRAK Penelitian ini bertujuan untuk mengetahui karakteristik protein plasma sapi bali. Sebanyak 24 sampel plasma sapi bali jantan dan betina dikarakterisasi menggunakan metode sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS PAGE). Data hasil penelitian dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa berdasarkan perhitungan bobot molekul pita protein ke-1 sampai pita ke-14 secara berurutan memiliki bobot molekul yaitu 963,50 kDa, 530 kDa, 346,82 kDa, 124,84 kDa, 89,85 kDa, 68,67 kDa, 54,71 kDa, 37,77 kDa, 20,78 kDa, 16,95 kDa, 16,18 kDa, 15,46 kDa, 12,56 kDa, dan 10,46 kDa. Selanjutnya, 14 pita protein plasma sapi bali dikelompokan menjadi lima fraksi yaitu albumin, globulin a1, a2, b, dan g. Fraksi albumin ditunjukan oleh pita ke-6 sampai pita ke-14 dengan bobot molekul 68,67-10,46 kDa. Fraksi globulin a1 dan a2 ditunjukan oleh pita ke-5 dan ke-4 dengan bobot molekul 89,85 kDa dan 124,84 kDa. Fraksi globulin b ditunjukan oleh pita ke-3 dengan bobot molekul 346,82 kDa. Fraksi globulin g ditunjukan oleh pita ke-1 dan ke-2 dengan bobot molekul 963,50 kDa dan 530 kDa. Persentase luas pita protein plasma sapi bali terdiri dari fraksi albumin 92%, fraksi globulin a2 3%, globulin g 2%, globulin a1 dan b 1%. Dapat disimpulkan bahwa pita protein plasma sapi bali jantan dan betina pedet (umur 0-1,5 tahun), pubertas (umur 2-2,5 tahun), dan dewasa (umur 3-5 tahun) berjumlah 14 pita protein dengan ketebalan bervariasi terdiri dari lima fraksi yaitu albumin, globulin a1, a2, a, dan g.

Abstract

Study to determine the potential of tempe flour in improving bone quality of ovariectomized rats (ovx)based on calcium (Ca) and phosphorus(P) levels in the plasma and bones; osteoblast and osteoclastprofiles have been conducted for 4 months. A total of 20 female Sprague Dawley albino rats, age of 3months and average body weight of 200 g were used in this study. The rats were divided into 4 groups,i.e.normal rats + diets (K0); normal rats +tempe flour (K1); ovx + diets (OV0); and ovx + tempe flour (OV1),respectively. The Ca and P levels in plasma and bones were analyzed using Atomic AbsorptionSpectrophotometer (AAS) and spectrophotometric methods. In addition, the profiles of osteoblast andosteoclast were also observed. The results showed that Ca and P levels in plasma and bones of normal ratswere 11.08 mg/dL and 7.06 mg/dL; 30.88 mg/dL and 18.63 mg/dL, respectively. The highest Ca and P levelsin both plasma and bones were observed in normal rats which were given tempe flour (K1), 11.25 mg/dLand 7.42 mg/dL; and 31.60 mg/dL and 19.78 mg/dL, respectively. Similarly, Ca and P levels in plasma andbones of ovxwhich were given tempe flour were slightly higher (10.91 mg/dL and 6.96 mg/dL; and 29.89 mg/dL and 17.87 mg/dL) compared to ovx without tempe flour diets (9.38 mg/dL and 6.46mg/dL; and 26.58 mg/dL and 16.75 mg/dL). The numbers of osteblast was found slightly higher both in normal rats and ovxwhich were given tempe flour; whereas slightly decreased of osteoclast was observed only in ovx which weregiven tempe flour. The administration of tempe flour in ovx diets could improved bone quality as indicatedby the Ca and P levels in plasma and bones and decreased numbers of osteoclast.

Abstract

Isoflavone compounds have been known to have biological effects in improving health. The purpose ofthis study was to determine the bioavailability of isoflavone in plasma and its role as lowering bloodglucose levels in hyperglycemia rats. Bioavailability of isoflavones in plasma was performed in 18 ratsstrains Spraque Dawley age 3 months with 200 g of body weight on average and were given 300 ppm oralof isoflavone single dose. The analysis of isoflavon as done at 0, 1, 6, 12, 18 and 24 hours using HPLCmethod. Study effect on blood biochemical values used 25 male rats, which was divided into 5 treatmentgroups. One group as a negative control (K1) was given aquades; positive group hyperglycemia (K2), and 3other groups (K3, K4 and K5) were a positive groups of hyperglycemia and given isoflavone different doses,i.e. 1.0, 1.5 and 2.0 mg/200g bw per oral. Blood glucose were analyzed by using glucose oxidase biosensor,while cholesterol, HDL (Hight-density lipoprotein) and triglycerides were analyzed enzymatically using aspectrophotometer. The results showed the analyzes isoflavone genistein was more efficiently absorbedthan daidzein and glisitein. Isoflavone as found at highest concentration in plasma at 6 hours after oraladministration i.e: genistein of 11,17 ppm, daidzein of 2,86 ppm and glisitein of 2,47 ppm. Theadministration isoflavones at dose 2 mg/200g bw could decrease blood glucose levels in hyperglycemiarats, and also has hipocholesterolemia effect.

Abstract

Bali cattle (Bos (Bibos) banteng) often experience fractures (fractures) of metacarpale, femur bone or tibia-fibular bone. This study was aim to reveal the strength and density of female Bali cattle bone which determines the occurrence of fractures. In order to reveal the strength of bali cattle bones, the elements that need to be explored include: bone density, cortical thickness of tibia bone, bone diameter, and bone resistance to pressure. To determine bone density performed by measuring bone specific graavity, cortical thickness of bone, carried out by splitting the bone by sawing the tibia bone extensively, then measuring the thickness of the wall of the bone cylinder using a caliper. Measurement of bone resistance to pressure was done by weighing the bone with a certain weight until the bone breaks or cracks and then measured by checking in which load the bones breaks or cracks. The result shown that the average density of Bali cow os tibia is 1.95 g/mL, average os tibia diameter is 26.45 mm, os tibia cortical thickness from top, middle, and bottom in sequence are 5.50 mm, 6.25 mm, and 5.35 mm. The result of os tibia compressive strength test is 9.26 Pascal with average load can be hold is 56.75 Newton and average wide of os tibia cross section cut is 6.05 cm2. This study result can be used as representation of Bali cow os tibia strength that has not been widely reported and can be used as recommendation for improvement of female Bali cattle livestock management.

Abstract

Research to confirm the abnormalities of blood corpuscles in Bali ducks based on blood smear examination has been conducted. The research samples consisted of 105 ducks from various regencies in Bali. The blood smear examination was conducted by the method described in this literature and the data was collected and tabulated by means of the descriptive method. Erythrocyte abnormalities were: polychromasia (61.9%), anisocytosis (17.1%), poikilocytosis (21.0%). Abnormalities associated with leukocyte composition were: Lymphopenic (46.7%); Heterophylic (44.8%); Heteropenic (3.8%), also Thrombocyto penia (2.9%) was present in 105 ducks. In conclusion 61,9% ducks examined deal with regenerative anaemia. They also have some blood abnormalities such as basophylia, eosinophylia, heterophylia, heteropenia, lymphopenia, and monocytosis.

Abstract

Kintamani dog as one of germ plasm owned by Bali province has been widely accepted as dog of Indonesian origin which need to be preserved. Report have shown that puppies of Kintamani dogs sold in Denpasar animal market often die due to Canine parvovirus (CPV) infection. The mortality of CPV infection in puppies can reach as high as 91% espescially in unvaccinated dogs. As the mortality of CPV in dogs is very high, it is important to find out the seroprevalence of CPV infection in Kintamani dogs in Sukawana village. Up to now, the seroprevalence of CPV infection in Sukawana, the natural habitate of Kintamani dog has never been reported. In this study the sample collection and area selection was conducted by haemaggutination inhibition (HI) test. Sera sample were concluded positive if the HI titers of sera were > 64 HI units. Seroprevalence of CPV infection was calculated by dividing the number of positive sera with the total sera samples. The seroprevalence of CPV among dogs was determined using non parametric analysis (Chi-Square). From 70 sera samples collected 67.1% (47/70) were antibody positive against CPV. The highest seroprevalence was found in Banjar Sukawana 22.8% (16/70). A higher seroprevalence was found in female dogs 45.7% (32/70) compare to male dogs 21.4% (15/70). Kintamani dogs aged between 724 month have the highest seroprevalence 27.1% (19/70). Based on the distribution of antibody titers, the seroprevalence antibody >64 HI was 65.7%. The result showed that the high titer (> 64 HI) of antibody against CPV, it was shown that CPV infection has occurs naturally in kintamani dog at Sukawana village.

Abstract

Glycogen is found at all of body tissue, especially mostly in liver and muscle. The objectives of thisresearch was to evaluate the content of glycogen in liver and muscle of diabetic rats that were treated withextract of tempe. A total of twenty male Spraque Dawley rats of 2 months old were used in this study. Therats were divided into four groups: (1) negative control group (K-), that were not treated extract of tempeand nondiabetic, (2) positif extract of tempe group (ET), that were treated with extract of tempe andnondiabetic (3) positif diabetic group (DM), and (4) diabetic and extract of tempe group (DM+ET). Extractof tempe was orally administered with dose 300 mg/kg bw/day. The treatment was conducted for 28 days.Effect of extract of tempe on body weight of all rats was determined at various time interval at 0, 4, 7, 14,21, and 28 days. At the end of the experiment, all rats were sacrificed by cervical dislocation. Liver andmuscle gastrocnemius were collected for analysis of glycogen level. The result of this study showed thatadministration methanol extract of tempe of 300 mg/kg bw/day can increase body weight, glycogen synthesisin the liver and muscle in normal rats (rat of ET group) and also diabetic rats (rat of DM+ET group). At theend of research, diabetic rats (rat of DM group) were decrease of body weight up to 5.7%. On the rat ofDM+ET group, rat of K(-) group and rat of ET group were increase of body weight of 5.7%, 19.3% and 20.3%,respectively. Glycogen level both liver, and muscle at rat of ET group and rat of DM+ET group wereincrease each of 9.29%, 2.2% in liver and 18.27% and 4.02% in muscle. Glycogen level at rat of DM groupwere decrease up to 42.5% in liver and 31.6% in muscle.

Abstract

Catharanthus roseus is one plant recognized as medical potential which can decrease cholesterol. Thepresent study was carried out to evaluate the effect of aqueous leaf extracts of C. roseus on plasma lipidprofil levels in rats cholesterol rich diet and atherogenic index. This study was carried out on 15 SpraqueDawley male rats randomly distributed into five groups (n=3). Rats hypercholesterolemic wereadministrated cholesterol rich diet containing 1% (w/w). Normal control group with normal diet whitoutextracts (K1), positive control hypercholesterolemic group (K2) with cholesterol rich diet whitout extracts,and others three groups (K3-K5) were feed high cholesterol and 1 mL of aqueous leaf extracts of C. roseuswith a dose of 20% (w/v), 40% (w/v) and 80% (w/v) respectively, administered twice daily 1 mL orally.Treatment was given for 28 days. After treatment, the plasma lipids profile such cholesterol total, highdensity lipoprotein (HDL), low density lipoprotein (LDL), triglyceride (TGA) level were measured. Theresults showed that during the period of treatment with cholesterol rich diet have produced under condition hypercholesterolemic of rats with average cholesterol levels of 124.33 ± 4.04 mg/dL (K2 group). Treatmentwith extract dose of 80% showed the best results among the other doses in reducing levels of total cholesterol,triglycerides, LDL and raise HDL levels. Extract treatment dose of 80% up to four weeks resulted incholesterol total (79.33±3.51 mg/dL), TGA (72.33±6.65 mg/dL), HDL (55.00±3.60 mg/dL), and LDL(9.87±5.34mg/dL). In addition, value ratio of cholesterol:HDL was 1.4: 1 and atherogenic index value was0.44. These results suggest that extracts of leaf C. roseus optimum doses 80% (w/v) has hipolipidemicactivity in hypercholesterolemic rats and it has low atherogenic index value.

Abstract

This research aimed to study the effect of isoflavon-riched tempe flour on calcium (Ca, phosphate(P), and estrogen levels in plasma et normal female rats during their growth period. A level oftwenty-five 2 months old female Sprague Dawley rats with an avarage body weight of 200 g wasrandomly divided into into 5 groups: one with control group (KO: without treatment) and fourtreatment groups (K1, K2, K3, K4 : animals were given tempe flour with isoflavon at 1; 2; 4; and 6 mg/200 g/bw, respectively). The treatment was conducted for two months, following this blood plasmawas collected to analyse the level of calcium, phosphor, and estrogen, respectively. The resultshowed that although the plasma level of Ca, P, and estrogen was higher in the treatment groupcompare to the control group, this was not significantly different (P>0,05). The highest plasmalevel of Ca, P, and estrogen was seen in anmal receiving tempe flour with 4 mg/ 200 g bw/dayisoflavon.
