Abstract

Lily (Lilium longiflorum Thunb.) was known as ornamental plants, especially as cut flower. Lily varieties in Indonesia are still lacking and the price is quite expensive but demand of lily is increased steadily. The first step for successful plant breeding is understanding phenophysiology of the plants, in particular flowering phenology. The aim of this research is to provide information on flowering lily phenology, to be a basic knowledge in improving for planning plant breeding programs. In this research, plants were observed from bud initiation until fruit production (phase F0-F4). Additional morphologies observed such as: high plant, leaf shape and color, shape and type of root or bulb. Result shows that period of phase F0-F3 of sampled lily is 52 days. The plant height of lily is about 50-150 cm, has elongated single lancet leaf shape, green leaf, bulbs, and has fiberous root.

Abstract

This research was aimed to determine adaptability of  eight introduced  broccoli cultivars in the tropical climate, the effect of fertilizers, as well as determine the content of vitamin A, C and fiber owned by the cultivar. This research was using randomized complete block design (RCBD) with two factors. The first factorsare different broccoli cultivars namely Broccoli 'Lucky', Broccoli 'B-106', Broccoli 'B-116', Broccoli 'B-112', Broccoli 'Green 2109' F1, Broccoli 'Green calabrase' and Broccoli 'Green Magic 'and Broccoli' Mujur'as a control. The second factors fertilizer of ZA, manure and urea. This research was performed in Kopang, Central Lombok, West Nusa Tenggara, for seven months from December 2014 to Juni 2015.Analysis of the nutrient content was done at the Laboratory of Analytical Chemistry, University of Mataram, Lombok. Parameters observed in this research were plant height, number and weight of leaves, and quality of curd. Data were analyzed by analysis of variance (ANOVA) if significantly different then tested further by Duncan's Multiple Range Test (DMRT) at 5% level. The results showed all the cultivars able to adapt. Broccoli 'B-106' is a cultivar that has the quality and quantity of curd most excellent and able to adapt to high temperatures. ZA fertilizer application will produce curd weight and diameter greater than that ofmanure or urea. The research also showed that cultivar B-112 has the highest content of vitamin A, B-116 has the highest vitamin C and Green 2109F1 has the highest fibers. Abstract

In vitro propagation study was carried out at Plant Tissue Culture Laboratory, Faculty of Agriculture, Udayana University. It was conducted using Completely Randomized Design with two factors.  The first factor was plant growth regulator (PGR), consist of nine concentration mixture of IBA (0, 0.5, 1 mg/L) and BAP (0, 1, 2 mg/L). The second factor was grape varieties, consisted of Prabu Bestari and Jestro Ag 86.  There were five replicates for each treatment combination. Variable observed included presence and absence of callus, percentage of callus growth, percentage of shoot growth, number of shoot per explants, emergence of shoots, percentage of root growth and number of roots per explants.  Data were statistically analyzed using Analysis of Variance (ANOVA).  When treatment showed a significant different (P<0.05) or highly significant (P<0.01) mean separation was conducted following Tukey Test. Data were analyzed using MINITAB Statistical Program. Results showed that PGR and varieties was highly significant for percent of callus growth.  PGR also showed significant effect on shoot growth percentage, while there was significant difference found on root growth percentage.  It can be concluded that combination of 1 mg/L IBA and 2 mg/L BAP was able to produce the highest percentage of callus growth. Addition of 2 mg/L BAP without IBA was able to produce the highest percentage of shoot growth. Percentage of root growth of Jestro Ag 86 was higher than Prabu Bestari.

Abstract

Plant breeding is an effort to develop plants by manipulating the genetic system of plants to get new superior cultivars. Plant breeding often produces incompatible seeds, so a rescue process is needed to reduce the risk of embryo death. The addition of gibberellic acid could break the seed dormancy for seed that hard to germinate. The purpose of this study was to find out the best concentration of gibberellic acid for germination of citrus globular embryos from protoplast fusion. Research was conducted at the Biology and Cell Biology Research Group In Vitro Culture Laboratory, Indonesian Center for Agricultural Biotechnology and Genetic Resources Research and Development (BB BIOGEN), Agricultural Research and Development Agency, Ministry of Agriculture, Bogor. The study used Completely Randomized Design (CRD) with concentration 0, 1, 2 and 3 ppm of gibberellic acid as treatment, each consists of 7 replicates. This study used globular embryos measuring ± 3 mm of inbred FS 29 and FS 89 from protoplast fusion between Siam Simadu and Mandarin Satsuma citrus. Observations were made on the percentage of germinated embryos, time of germination, the number of the complete and incomplete shoot (abnormal, only part of the roots or buds that developed). Data were analyzed using Crosstab analysis test with IBM SPSS Statistics 22 software. Result showed that addition of gibberellic acid induce faster germination time and increase seedling percentage. The best germination medium for citrus FS 29 was VMW with 2 ppm and 3 ppm gibberellic acid with the seedling percentage around 85,7% in 7 days, while the best germination medium for citrus FS 89 was VMW with 1 ppm gibberellic acid with 100% seedling embryos in 8,5 days. VMW medium added with 1 ppm, 2 ppm and 3 ppm gibberellic acid for citrus FS29 and VMW medium added with 2 ppm and 3 ppm for citrus FS89 were good to get the complete sprouts with a succes rate of 100%. Abstract

Dendrobium heterocarpum Lindl. is one of the orchid species which is often seeked. It is because this species has a mystique appearance and special fragrance. The research aimed to observe the growth and developing responses of protocorm like bodies (PLBs) of D. heterocarpum after 12 weeks in three different medium, i.e. Murashige and Skoog (MS), Kursor C and Western 3 (W3). The D. heterocarpum seeds were planted in MS, Kursor C and W3 medium. Qualitatively the orchid growth were observe including growth score and colour score. The result showed that growing time of MS was 4 weeks, a week faster than the growing time on Kursor C and W3 medium. The growing phase on MS medium and Kursor C medium reached phase 1 whereas on W3 medium reached phase 2. PLB’s colour after 12 weeks on MS medium reached phase E, strong yellowish green phase (142A). Meanwhile, on Kursor C and W3 was on phase D, moderate yellowish green (143D).

Abstract

Dendrobium heterocarpum Lindl. is a native orchid in Indonesia. Its population continue to decline due to unsustainable exploitation. One solution to increase its population is via in vitro technique propagation. This research aimed to determine effect of hormone addition (Zeatin and NAA) to induce shoot multiplication of D. heterocarpum Lindl. grown in Murashige and Skoog (MS) basic medium. The research was conducted in Tissue Culture Laboratory, Department of Biology, Faculty of Mathematics and Science, Udayana University from November 2015 until March 2016. The experiment employed Completely Randomized Design (CRD) with 20 combinations of treatments, with three replicates each treatment. The treatment werer addition of Zeatin and NAA, with five concentrations of Zeatin (0, 0.1, 0,5, sweet corn and hybrids corn as the natural Zeatin); four NAA concentrations (0, 0.05, 0.1 and 0.5). Variable recorded include such as the number of shoots, roots and explant’s vigor. The quantitative data which include number of leaves and shoots after 12 weeks, were analyzed using Analysis of Variance (ANOVA); and if significantly different, tested further using Duncan Multiple Range Test (DMRT) at 5% level. Z5N3 medium revealed significantly different on amount of leaves and shoot height and also the best medium on amount of roots. Best hormone combination that can induce multiplication of new shoots are medium Z4N3 and Z5N1.

Abstract

This study aims to determine the type of Hemerocallis fulva L. pollen, pollen viability and growth after storage at 280C, 40C, -200C, -800C temperature for 0-8 weeks. The study was conducted from January - March 2010. Pollen samples were taken from UPT. Botanical Gardens Plant Conservation 'Eka Karya’ Bali. Pollen storage and observations was carried out at the Laboratory of Plant Biotechnology and Development Structure, Biology Department, Faculty of Science, Udayana University. Viability test were conducted using hanging drop media and agar media with GA3. The results showed that type of Hemerocallis fulva L. pollen were: monosulkat, Prolate spheroiodal, (54.25 X 47.47), P/E: 1.14, Zono, colpate, reticulate. Fresh pollen viability on hanging drop media was 8.33%, decreased to 4% after storage at 4°C for one week and -20°C for 5 weeks. Whereas on agar media, control pollen has viability of 3.17% and increased at -20°C to become 11.67%. Mature pollen can be observed at the level of uninukleat, binukleat, and trinukleat. The development of pollen with the media hanging drop on the control has a long reed 115.94 ?m and length increased at 4 ° C with storage duration ie 256.39 weeks to 1 ?m, whereas on an agar medium + GA3 to control reed 149.82 ?m in length and at 28 ° C, 4 ° C, -20 ° C and -80 ° C respectively 94.73 ?m; 142.1 ?m; 95.53 ?m; 118.67 ?m at 1 week storage time.

Abstract

Begonia bimaensis Undaharta & Ardaka adalah salah satu Begonia yang dikembangkan di Kebun Raya ‘Eka Karya’ Bali. Jenis ini merupakan spesies endemik yang ditemukan di Gunung Muria di Kabupaten Bima, Pulau Sumbawa. Begonia bimaensis memiliki rambut putih yang tumbuh pada permukaan atas daunnya. Perbanyakan tanaman B. bimaensis perlu dilakukan dengan teknik kultur jaringan karena terbatasnya tanaman dan untuk menjaga kelestarianya. Tujuan penelitian ini adalah mengetahui teknik sterilisasi yang tepat untuk inisiasi eksplan daun B. bimaensis dan mengetahui kombinasi ZPT kinetin dan 2,4-D yang paling efektif serta pengaruhnya terhadap pertumbuhan kultur B .bimaensis. Penelitian ini menggunakan media dasar MS (Murashige dan Skoog) dengan penambahan kombinasi ZPT kinetin (0 ppm, 1 ppm, dan 2 ppm) dan 2,4- D (0 ppm, 0,5 ppm, dan 1 ppm). Percobaan terdiri dari 9 perlakuan yang setiap perlakuan terdiri dari 3 ulangan dengan tiap ulangan terdiri dari 1 botol. Hasil penelitian menunjukkan bahwa teknik sterilisasi yang tepat yaitu menggunakan kombinasi sterilan yang berbeda (sabun Sunlight, fungisida, pemutih Bayclin, dan alkohol) dan waktu perendaman menggunakan fungisida selama 30 menit. Pemberian ZPT kinetin dan 2,4-D terhadap B. bimaensis mengakibatkan pertumbuhan yang lebih cepat. Pengaruh paling tinggi terhadap pertubuhan kultur B. bimaensis diperoleh pada hari ke-12 setelah tanam dengan pemberian kinetin 2 ppm dan 2,4-D 0,5 ppm. Kata kunci: in vitro, Begonia, 2,4-D, kinetin

Abstract

The existence of development can change the composition, vegetation structure of mangrove forest. The development can also reduce the absorption of carbon gas in mangrove forests. This study aims to: (1) determine the composition, structure of mangrove vegetation in the area of ??mangrove forests of Ngurah Rai Denpasar forest park, (2) determine the condition of biomass and the potential of mangrove carbon reserves in Tahura Ngurah Rai Denpasar. This research was conducted using the Line Transect Plot method. Along the transect line plots of 10mx10m, 5mx5m and 2mx2m are made. The Purposive Sampling method was used to determine the carbon absorption in the study. Potential analysis of carbon biomass deposits is carried out based on allometric numbers. The percentage analysis of the CO2 absorption is done based on the results of analytical laboratory test. From the analysis of vegetation there are 8 types of mangroves. The highest of important value index in Station I is Rhizopora apiculata with a value of 128.10%, while the highest value in Station II is Rhizopora mucronata with a value of 130.83% and the highest value in Station III is Rhizopora apiculata with a value of 199.99%. The highest sapling level of INP was Rhizopora mucronata 171.70%, Station II Rhizopora mucronata 205.85%, Station III Rhizopora apiculata 164.57%. Seedling rate of Rhizopora mucronata in station I was 233.33%, station II Rhizopora mucronata 134.72%, station III Rhizopora apiculata 80.28%. The greatest aboveground biomass is found in station II with 325,848 tons / ha, the highest carbon stock is found in station III with 325,296 tons / ha and the highest of carbon absorptions potential is found in station III with 1,193.80 ppm. Abstract

Broccoli (Brassica oleracea L. var. italica Plenck.) is a vegetable crop belongs to Brassicaceae family.  Broccoli has high nutrition, high in fiber and contains isotiacyanate that has anticancer activity.  Broccoli market in Indonesia, particularly in modern supermarkets, increases 15-20% per year, however, productivity was still low, therefore effort to increase broccoli production in Indonesia need to be investigated. Field trial was conducted at Batur Village, Kintamani District, Bangli Regency, Bali, to find out cultivars that were adapted in tropical region. Introduced cultivars being trialed were ‘Alborada’,‘Belstar’, ‘Fiesta’, ‘Sarasota’, ‘Bay Meadows’, ‘Castle Dome’, ‘Liutenant’, ‘Iron Man’, ‘Marathon’, ‘Green Gold’, ‘Imperial’, ‘Green Magic’ and ‘Lucky’ as control.  Variable observed were vegetative growth, curding period, and productivity of each cultivar.  This study employed Randomized Completely Block Design with 3 replicates (plots) and 8 plants each plot.  Data were analyzed using ANOVA (Analysis of variance) on Costat program, followed by Duncan’s Multiple Range Test (DMRT) on 1% level.  Results show that each cultivar varied in adaptability in tropical region. ‘Castle Dome’ has the highest productivity, while ‘Fiesta’ was the lowest.
