Abstract

IMPACT OF AGRICULTURAL ACTIVITIES ON EUTROFICATION LEVELS AND TYPES OF FITOPLANKTON IN LAKE BUYAN BULELENG REGENCY PROVINCE OF BALIThis research was conducted to find out the impact of agricultural activities in the various levels of eutrophication and diversity of phytoplankton in Buyan Lake, Buleleng, Bali Province. Purposive sampling method was organized to determining research station, where the sampling stations were determined based on various considerations, such as conditions of study area, and predominant use of agricultural land in the study area. Sampling method of phytoplankton and the lake water were done by setting the 4 stations that represent agricultural activities around Buyan Lake. Phytoplankton samples were taken by filtering the water on the lake surface as much as 100 liters, using the plankton net with a mesh size of 25µm. Phytoplankton parameters was analyzed in laboratory. Data was analyzed using analysis of phytoplankton abundance, Equity index and dominance index. Results showed that abundance of phytoplankton at each stations in Buyan Lake was ranged between 1150 – 1791.67 cells / l. Average abundance of phytoplankton in all stations was 1504.17 cells / l. Based on the abundance of phytoplankton, waters of Buyan Lake was classified into water that have low fertility rate (oligotrophic). Agricultural activities around Buyan Lake resulted a moderate eutrophication level (mesotrofik).

Abstract

Artificial reefs are made to act as temporary substitutes for small areas of damaged original reef. The structures attract fish and other marine organisms, which can eventually be exploited (e.g. for human food), so it is expected that they will form the basis around which natural reefs can grow and recover. The purpose of this study is to document recruitment of organisms onto the artificial structures, evaluate the effectiveness of the structures in terms of shape and recruitment potential, and use the recruitment information to show which species occur in the surrounding reefs. This research was conducted from May-July 2010 in north Bali, in the Village District Gerokgak Patas, Kampung Baru Village in Buleleng District, and Penuktukan Village, Kecematan Tejakula. The research method used was a visual census method to determine the number of fish species and fish abundance on artificial reefs at three different locations. Three replications were performed at each location. 2481 individuals from 18 families (38 species) of reef fish were observed. The area with the greatest abundance of fish and the highest number of fish species was in Kampung Baru, with 1170 individuals from 21 species of fish. Dominant reef fish species were Plotosus lineatus (Cat Fish) - 510 individuals, Pomacentrus auriventris (Goldbelly damsel)- 256 individuals, Dascyllus trimaculatus (Three spot damsel) - 335 individuals, Pseudanthias squamipinnis (Lyretail Anthias) - 320 individuals. A Sorensen analysis showed that reef fish communities on the artificial reefs in the form of 'hexadomes' were not derived from the surrounding reefs.

Abstract

The Effect of Ethyl Methane Sulphonate (EMS) on Growth and Variations of Marigold (Tagetes sp.) The aims of this research are to determine the variation of marigold (Tagetes sp) derived from seed treated with EMS and to recommend the EMS concentrations that are able to induce varietion. Seeds of marigold cv Narai Orange were soaked in water for 6 hours, followed by soaking in EMS at concentration of 0%, 0.3%, 0.6% and 0.9% for 4 hours. This study employed Randomized Complete Blok Design with 10 replicates and each replicate consisted of 10 plants. Six plants were randomly chosen for measurements. The total number of samples observed were 240 plants. Observations were made on the percentage of the growth, plant height, number of leaves, number of branches, diameter and weight of flowers. Data obtained from the observations were analyzed using Analysis of Variance (ANOVA), followed by DMRT (Duncan’s Multiple Range Test) if there is a significant difference between treatments. The EMS treatment reduced all characters observed. The EMS concentration of 0.6% showed plant that had yellow flowers. The 0.9% EMS treatment resulted in one plant with chimera, 6 dwarf plants, 2 plants with thin stems, and 1 short plant with many branches. Untreated plants did not show any variation.

Abstract

The aim of this research was to obtain a method for pollen storage of Citrus species in Bali. Pollen from 12 Citrus genotypes was collected from five different regions; Petang, Plaga, Kintamani, Besakih, and Payangan Village.  Pollen from intact anthers were stored at three different storage temperatures, i.e., 25oC (room temperature), 4oC (refrigerator), and -20oC (freezer), with or without oven dried at 40oC for 20 hours before storage.  Pollen viability was determined using in vitro germination test.  Germination media contained 20 g/l sucrose and 8 g/l agar.  Result shows that -20oC was the best storage temperature.  At this temperature, only slight reduction of pollen viability after 2 months storage.  Storage at room temperature (25oC – 30oC) was not effective; pollen was infected with fungus within a week of storage. Oven dried pre-treatment before storage made better performance on pollen stored at 4oC.  Pollen moisture content decreased after oven dried, which also decreased fungus infection.  Storage of pollen is an important aspect to plant breeding programs, when individuals to be hybridized do not flower at the same time or are geographically separated.  This is the first report on pollen storage and viability of Citrus genotypes grown in Bali.

Abstract

Tanaman tebu adalah keluarga poace yang berpotensi menghasilkan nilai ekonomi tinggi karena manfaatnya sebagai produk unggulan dalam menghasilkan gula serta bioetanol yang banyak digunakan oleh masyarakat luas. Untuk itu perlu upaya pemuliaan tanaman yang cepat untuk memenuhi kebutuhan pasar. Teknik kultur mikrospora merupakan aplikasi bioteknologi yang efisien dalam menghasilkan tanaman haploid. Teknik ini belum berhasil diaplikasikan pada tanaman tebu sehingga dalam penelitian ini bertujuan untuk memicu embriogenesis mikrospora tebu dengan aplikasi kultur mikrospora pada varietas bululawang. Metode yang digunakan yaitu dengan stress perlakukan perendaman mannitol 0.3M sebelum isolasi kultur pada lama perendaman yang berbeda. Analisis data menggunakan uji anova dan uji duncan. Hasil yang diperoleh yaitu Perlakuan stress yang diberikan berhasil memicu embriogenesis mikrospora dengan lama perendaman anther optimum pada 7 hari pada mannitol 0.3M

Abstract

Indonesia is the biggest tuna exporter in Southeast Asia. With a high number of tuna catch, it is worried that the catch will decrease tuna population, specifically longtail tuna. To anticipate the decrease, there needs to be a conservation program to protect longtail tunas from scarcity. One method used to protect longtail tuna is by genetic conservation. The aim of this research is to understand the genetic and phylogenetic variety of the longtail tuna in Pabean Surabaya Fish Market. The polymerase chain reaction was used to amplify segment of the mitochondrial control region gene from members of these sample, used forward primer CRK 5’-AGCTC AGCGC CAGAG CGCCG GTCTT GTAAA-3’ and reverse primer CRE 5’-CCTGA AGTAG GAACC AGATG-3’. Based on the sequencing process, 28 out of 29 samples longtail tuna were analyzed successfully. The results of the 28 sample analysis of longtail tuna based on its genetic variety and phylogenetic tree reconstruction showed a haplotype variety (Hd) score of 1,00000, and nucleotide (π) variety of 0,1939. Genetic variety value showed that longtail tuna has great adaption capabilities toward environmental changes time to time. Phylogenetic tree reconstruction results showed 7 clades with a genetic range of 0,005 – 0,035, which shows that all samples are closely related. The results of this study can be used as basic information in forming regulations on longtail tuna sustainable management and genetic conservation.

Abstract

Mining and nickel processing activities in Pomalaa, Kolaka Regency have an impact on the condition of coastal waters. Several researchers who have conducted research in the area stated that the Pomalaa coast has been polluted by the presence of nickel metal so that there is an initiative to reduce the impact of Ni in the waters by bioremediation of the microalgae Chlorella vulgaris and Spirulina platensis. This study aims to analyze the growth of microalgae types C. vulgaris and S. platensis cultured on media containing nickel with different concentrations and also to determine the ability of microalgae to reduce nickel heavy metals at different concentrations. This research was carried out at the Fisheries Laboratory of Haluoleo University , while the AAS (Atomic Absorption Spectrophotometric) analysis was carried out at the Kendari Environmental Laboratory UPTD Laboratory on January – May 2021. The results of the analysis showed that there was a significant difference in the growth density of microalgae cells (C. vulgaris and S. platensis) which were given the treatment/concentration. Ni different. The absorption capacity of C. vulgaris ranged from 0.73 to 4.97 mg/l, while s. platensis ranged from 0.082 – 2.11 mg/l. C. vulgaris is superior in reducing nickel because C. vulgaris can live in polluted water conditions. Keywords: Bioremediation; Nickel; C. vulgaris; S. platensis.

Abstract

Leucaena is a plant that produces biomass productivity in the form of hardwood for fuel with low humidity and high calorific value. However, Leucaena is also classified as an invasive plant which can cause the urgency of native plant species and ecosystems in Indonesia. Therefore, the formation of sterile Leucaena needs to be done, one of which is through genetic transformation using Agrobacterium tumefaciens. Callus is used as a target for transformants in the genetic transformation process, so it is necessary to use appropriate media and PGR. This study aimed to determine the type of media and the concentration of 2,4-D on callus induction. This research is an experimental study with a completely randomized design (CRD) method with two factors. The first factor is the type of media (MS and WPM) and the second factor is the concentration of 2,4-D (0; 0.25; 0.50; 0.75; 1.00; 1.25 and 1.50 mgL-1 ). Each treatment was repeated three times so that 42 experimental units were obtained. Parameters observed were callus initiation, callus fresh weight (gram), callus texture and color. Quantitative data is analyzed by analysis of variance (ANOVA). The results showed that the use of media had a significant effect (P<0.05) on callus fresh weight. The use of 2,4- D concentration had a significant effect (P>0.05) on callus texture. The use of WPM media resulted in the fastest callus emergence time (6.67±0.57), the best callus texture (crumb callus type 2) and the best callus color (green). Meanwhile, the highest fresh weight (2,48±0.83) was in the use of MS media. The fastest callus emergence time occurred in the control (without the addition of 2,4-D) (7.33±0.57 and 6.67±0.57), the highest average fresh callus weight (2,48±0.83 and 2.35±0.32) occurred in the treatment with the addition of 1.00 mgL-1 2,4-D with a crumb callus texture of type 2 and callus green color only appeared in the treatment with a concentration of 0.25 mgL-1.

Abstract

Longtail Tuna (Thunnus tonggol) is one of the neritic species of tuna from the Scrombridae family. T. tonggol is oceanicromus and ussually found in tropical and subtropical waters in the Indo-Pacific region. Although known as one of the tuna species, the information of this species is very lacking, especially in Indonesia. Therefore, this study aims to determine the genetic diversity and kinship relationships of longtail tuna (T. tonggol) collected at Sagulung fish market, Batam, Riau Islands. Molecular analysis were carried out using genetic markers namely mitochondrial D-loop (control regian) with the stages of DNA extraction, PCR (Polymerase Chain Reaction), electrophoresis, sequencing, and data analysis. A total of 21 longtail tuna samples were identified using molecularly and confirmed as T. tonggol species, which have a base length ranging from 482 - 523 bp (base pairs). The value of haplotype diversity (Hd) of longtail tuna was 1,00000 and the value of nucleotide diversity (?) was 0,01654. The results of phylogenetic tree reconstruction showed that all samples collected were in the same group (clade) with the average genetic distance in one T. tonggol clade obtained a value of 0.010 which has a close relationship between samples. This result can be use as additional information for longtail tuna species in Indonesia.

Abstract

Nusa Penida MPA was established with the aim to conserving natural resources and ensuring the sustainability of livelihoods. The Government of Indonesia uses the EVIKA tools to assess the effectiveness of MPA management. This study examines the effectiveness of MPA management by looking at indicators of coral reef ecosystem health and resource utilization to develop an effective management strategy using the force filed analysis method. The results showed that the condition of coral and fish biomass in the Nusa Penida MPA experienced a significant increase. Hard coral cover was in the medium category with a cover of 43% and fish biomass of 816.80 kg/ha. The coral reef health index of the Nusa Penida MPA is 5 which indicates that coral cover is in the medium category with high recovery potential and the coral reef fish category is low. Nusa Penida marine resources are used for fisheries, tourism as well as for trade. This study recommends a strategy to increase the effectiveness of Nusa Penida MPA management by investing in increasing human resource capacity in the Bali MPA management unit in competence on monitoring ecosystem and preparing a management budget plan along with efforts to fulfill program budgets.
