Abstract

Brokoli (Brassica oleracea L. var. italica Plenck) merupakan sayuran yang memiliki nilai gizi tinggi dan popular di Indonesia. Produksi brokoli di Indonesia rendah karena umumnya kultivar yang ditanam di Indonesia adalah kultivar introduksi. Kultivar introduksi tidak cocok di lingkungan tropis Indonesia sehingga memerlukan upaya pemulian tanaman untuk perbaikan kultivar. Salah satu upaya perbaikan kultivar yang popular saat ini adalah kultur mikrospora. Kultur mikrospora dapat memproduksi tanaman homozigot atau galur murni sehingga nantinya dapat dijadikan tetua dalam program pemuliaan tanaman. Penentuan Jumlah kromosom sangat berguna dalam pengembangan kultur mikrospora dan pemuliaan tanaman hibrida. Kultur mikrospora menghasilkan variasi jumlah kromosom yang beragam sehingga pengetahuan jumlah kromosom penting dilakukan dalam kultur mikrospora. Penelitian ini menggunakan 3 kultivar brokoli Green Magic, Royal Green dan BL 10001. Penentuan jumlah kromosom menggunakan metode Squash dengan pewarna aceto-orcein pada ujung akar. Hasil penelitian menunjukkan jumlah kromosom pada kultivar BL 10001 adalah 18 kromosom, pada kultivar Royal Green adalah 9 kromosom dan pada kultivar Green Magic adalah 14 kromosom.

Abstract

Sugarcane is an economically valuable plant that continues to be developed to produce superior varieties, the technique to get new varietis used is microspore culture biotechnology. There are many obstacles in the implementation of the microspore cultivar method. Therefore, in this research, a study of sugarcane anthers was carried out. This research used sugarcane panicles for anther selection and then observed under a microscope. Data is analyze with descriptivey method. The results show that white and yellow anthers are the right materials for microspore culture donors

Abstract

Tanaman lili (Lilium longiflorum Thunb.) merupakan tanaman hias dan bunga potong. Permintaan bunga potong lili sangat menjanjikan tetapi ketersediaan bibit masih terbatas. Penelitian ini terdiri dari perkecambahan biji lili secara in vitro pada media ½ Murashige & Skoog (MS), multiplikasi bagian eksplan pangkal dan ujung tunas pada berbagai perlakuan media MS kombinasi ZPT BAP dan NAA. Tujuannya untuk mendapatkan kombinasi media yang tepat dan mengetahui persentase tertinggi bagian eksplan yang mampu tumbuh tunas, akar dan kalus. Kombinasi media MS dengan BAP dan NAA yaitu (a) kontrol 0:0 mg L-1, (b) 0:0.1 mg L-1, (c) 0:1 mg L-1, (d) 0.1:0 mg L-1, (e) 0.1:0.1 mg L-1, (f) 0.1:1 mg L-1, (g) 1:0 mg L-1, (h) 0.1:1 mg L-1, (i) 1;1 mg L-1. Penelitian ini disusun dalam RAL faktorial dua faktor yaitu kombinasi pemberian BAP dan NAA, multiplikasi eksplan bagian pangkal dan ujung tunas. Biji lili mampu berkecambah sebanyak 58%. Media c dan i yang mampu menginduksi eksplan untuk tumbuh tunas, akar dan kalus. Media c mampu menginduksi tunas, akar dan kalus, media i mampu menginduksi akar dan kalus. Persentase respon pertumbuhan eksplan bagian pangkal tunas yaitu pertumbuhan tunas 11.1% dan akar 13.3%, sedangkan eksplan bagian ujung tunas menunjukkan pertumbuhan akar 6.7% tanpa pertumbuhan tunas. Pertumbuhan kalus menunjukkan hasil yang sama pada eksplan pangkal dan ujung tunas yaitu 4.4%.

Abstract

Kentang (Solanum tuberosum L.) adalah salah satu komoditi pangan penting di dunia. Penyediaan benih kentang generasi awal yang bebas penyakit sistemik virus merupakan masalah utama dalam pembudidayaan kentang. Produksi benih kentang generasi awal berupa umbi mini di rumah kaca masih memiliki resiko kontaminasi oleh penyakit virus. Teknik produksi umbi mikro secara in vitro dapat menjadi metode alternatif untuk produksi benih sumber. Tujuan penelitian ini adalah untuk menentukan media yang optimum dalam produksi umbi mikro pada empat genotipe kentang, yaitu AOTX98202-1RU, ATX9202-3RU, ATTX98468-5R/Y dan ATTX98518-5P/Y. Keempat genotipe dikulturkan pada media Murashige and Skoog (MS) dengan tambahan 6% sukrosa, dengan atau tanpa 2 g L-1 phytagel, dan dengan atau tanpa 10 mg L-1 kinetin. Stek dua buku dikulturkan pada tiap botol kultur, masing – masing dengan lima ulangan. Kultur diinkubasi selama dua minggu pada suhu 23°C dengan fotoperiode 16 jam terang/hari, lalu suhu 16 °C selama 2 minggu dengan fotoperiode 16 jam terang/hari, dan suhu 16 °C dalam ruang gelap selama 6 minggu, dan diamati selama 10 minggu. Hasil penelitian menunjukkan bahwa tiap genotipe memiliki respon yang berbeda terhadap media. Perlakuan 2 g L-1 phytagel + 10 mg L-1 kinetin (T4) merupakan media yang efektif untuk mendorong pertumbuhan umbi mikro pada genotipe ATTX98468-5R/Y dan ATTX98518-5P/Y. Genotipe ATTX98468-5R/Y pada media T4 menghasilkan umbi mikro terbanyak (4.2 umbi mikro/botol), sedangkan ATTX98518-5P/Y menghasilkan rata-rata berat umbi tertinggi (363.6 mg). Hasil penelitian menunjukkan pentingnya untuk mengembangkan protokol khusus untuk tiap genotipe agar mendapatkan produksi umbi mikro yang optimal.

Abstract

This study produced biodiesel from Malapari seed oil (Pongamia pinnata L.) by optimizing the esterification and transesterification processes. The objectives of this study were to determine the sulfuric acid (H2SO4) catalyst concentration in the esterification reaction that can reduc the free fatty acid (FFA) content of the seed oil to ≤2%, analyze the oil to methanol molar ratio (i.e.,1:6, 1:9, and 1:12) and KOH concentration (i.e.,2%, 4%, and 6%) that will produce the highest biodiesel yield, and evaluate the quality of the produced biodiesel according to the Indonesian National Standar (SNI) 04-7182-2015. The research was conducted from October to December 2023 at the Plant Physiology Laboratory and the Physical Chemistry Laboratory, FMIPA, UNUD. The research applied Completely randomized design. Quantitative biodiesel data were analyzed using Analysis of Variance. If significant differences (α < 0.05) were detected, then the quantitative biodiesel data were further analyzed using Duncan's Post Hoc Test. Results showed that an H2SO4 concentration of 7% reduced the FFA content to 1.915%. A molar ratio of 1:12 with a 2% KOH catalyst produced the highest biodiesel yield of 65.35%. The produced met the SNI for the saponification number (i.e., 52.82–133.69 mg-KOH/g), iodine number (i.e.,7.19–22.42 g_I2/100_g), and cetane number (i.e., 86.48–132.68). However, the water content (i.e., 0.721%–2.407%), viscosity (i.e., 1.73–6.18 mm²/s), acid number (i.e., 0.224–4.685 mg_KOH/g), density (i.e., 874–917 kg/m³), FFA content (i.e., 0.116 -% to 2.229%), and methyl ester content (48,13% to–132.12%_mass) partially did not meet the standards. This study highlights the potential of Malapari seed oil as a biodiesel feedstock, with further required needed to meet all quality standards.

Abstract

Background: Biodiesel production from malapari seeds generates waste in the form of seed cake that can still be utilized. This seed cake can be processed into compost because it contains various essential minerals for plant growth. This study analyzes the quality of compost made from malapari seed cake based on the SNI 19-7030-2004 standard. Methods: The compost fertilizer was fermented for 31 days using four treatments. (P1) malapari cake 1kg + Orgadec 2g; (P2) malapari cake 334g + goat manure 333g + burnt rice husk 333g + Orgadec 2g; (P3) Malapari cake 200g + goat manure 400g + burnt rice husk 400g + Orgadec 2g; (P4) Malapari cake 400g + goat manure 200g + burnt rice husk 200g + Orgadec 2g. Results: Water, phosphorus, and potassium content tests in treatments P1, P2, P3, P4 met specifications. pH tests in treatments P1, P2, P4 met specifications, P3 did not meet specifications. Nitrogen tests in P1 and P2 met specifications, but P3 and P4 did not. The organic C test in P1 did not meet specifications, while P2, P3, and P4 met specifications. The electrical conductivity test in P1 met specifications, while P2, P3, and P4 did not. Color and odor in P1 did not meet specifications, while P2, P3, and P4 met specifications. Conclusions: Compost from malapari seed cake has met the compost quality specifications according to SNI 19-7030-2004 for P2, so it is suitable for plant application.

Abstract

Indonesia’s energy sector continues to face persistent challenges in the context of rapid economic growth, increasing energy demand, and mounting environmental concerns. Despite the constitutional mandate requiring the state to ensure equitable access to energy and sustainability, existing regulatory frameworks frequently fall short of achieving these objectives. This research aims to examine the legal and policy frameworks governing the energy sector in Indonesia, where energy serves as a critical component of economic development and public welfare.The urgency of this study lies in addressing the disconnect between the normative ideals (das Sollen) of equitable and sustainable energy governance and their actual implementation (das Sein) in current policy practices. The research employs a normative legal method, relying on a literature review of both primary and secondary legal sources, coupled with conceptual and statutory interpretation.The findings indicate that current policies often fail to ensure equitable distribution, meaningful public participation, and adequate environmental protection. Accordingly, legal reforms are necessary to advance justice in energy access and to establish enforceable emission limits and environmental standards. The conclusion drawn from this study underscores the importance of bridging the gap between normative aspirations and practical realities in order to achieve inclusive, sustainable, and just energy governance.
