Abstract

Rhizome with high productivity and content of bioactive ingredients is the hope to be achieved from the cultivation of galanga (Kaempferia galanga L.). Galanga is used traditionally as a medicine for stomach ulcers, colds, headaches, coughs, diarrhea, facilitating menstruation, skin diseases, and rheumatism. Production of secondary metabolites is influenced by environmental factors and the place of growth. This study aims to determine the effect of the growing environment from two different locations on the essential oil content and its constituent components from the rhizome of the galanga accessions. The samples observed were nine accessions of galanga harvested at 6 months which were planted in the Bogor, namely Darmaga (L1) and Cisarua (L2) with a altitudes of 214 m asl and 780 m asl with different agro-climatic conditions. The accessions tested included Purbalingga (PBG), Cilacap (CLP), Purworejo (PWJ), Karanganyar (KRA), Pacitan (PCT), Madiun (MAD), Galesia 1 (GAL 1), Galesia 2 (GAL 2), and Galesia 3 (GAL 3). GAL 1, GAL 2, and GAL 3 are high yielding varieties of galanga that have been released by the Minister of Agriculture of the Republic of Indonesia and used as comparisons based on their essential oil content. Observations were made on the levels of essential oils and their constituent compounds using GC-MS (Gas Chomatography-Mass Spectroscopy). Differences in growing places affect the content of essential oils and their constituent components from the rhizome of the galanga accession. Environmental factors that influence are sunlight and water availability. The highest essential oil content (3.78%) was obtained from the MAD accession planted in Cisarua (L2) and the highest EPMS compound (34.74%) was obtained from the PCT accession planted in Darmaga (L1). There were 73 compounds detected with four main components, namely delta-3-carene, pentadecane, ethyl cinnamate, and ethyl p-methoxy cinnamate (EPMS).

Abstract

Inflammatory diseases are diseases characterized by inflammatory symptoms. Acute inflammatory disease can cause dysregulation of the inflammatory immune response, thereby inhibiting the development of protective immunity against infection. Among the acute inflammatory disease is COVID-19. The initial viral infection causes the antigen-presenting cells to detect the virus through a phagocytosis mechanism in the form of macrophage and dendritic cells. Lactobacillus fermentum and L. plantarum are gram-positive bacteria potentially serving as immunomodulators caused by inflammation and immune system response. Short-chain fatty acids (SCFA) produced by Lactobacillus can induce immune response through tolerogenic dendritic cells. This probiotic bacterium can induce the production of different cytokines or chemokines. Following the results of in vitro and in vivo tests, L. fermentum and L. plantarum can induce IL-10 release to activate regulatory T-cell and inhibit tumor necrosis factor-α (TNF-α) binding activity of nuclear factor kappa B (NF-κB). Literature review showed that dysregulation of inflammatory immune response disorders due to inflammatory disease could be treated using probiotic bacteria L. fermentum and L. plantarum. Therefore, it is necessary to conduct further studies on the potential of indigenous Indonesian strains of these two bacteria as anti-inflammatory and immunostimulants.

Abstract

Hypertension is a structural or functional change in the arteries or the organs it supplies caused by increased blood pressure. Angiotensin-converting enzyme (ACE) can increase blood pressure by converting inactive angiotensin I to active (angiotensin II). Captopril is a hypertension drug that can inhibit ACE activity. Sembung leaf (Blumea balsamifera DC) is a plant that can potentially have antihypertensive activity. This study aims to identify the interaction of active compounds in sembung leaves against ACE as antihypertensive drug candidates through an in silico test based on pharmacodynamic and pharmacokinetic parameters using two docking software, Autodock Vina and PyRx. The results showed that Luteolin was the best test ligand besides having lower ΔG and Ki than control ligands and higher Kd than control ligands. Luteolin was identified to interact with Zn and hydrogen bond interactions at the active site and met the criteria in Lipinski analysis, ADME, and toxicity, so this compound is relatively safe to be used as a drug candidate for the treatment of hypertension.

Abstract

Penelitian ini dilakukan untuk mengetahui konsentrasi optimum dan model isotherm bovine serum albumin (BSA) dalam matriks silika gel dengan pemanfaatan silika yang bersumber dari pasir alam Takari. Penelitian ini dilakukan melalui tahapan preparasi sampel, ekstraksi silika (SiO2), penentuan konsentrasi optimum, dan model isotherm BSA pada matriks silika gel yang dianalisis menggunakan spektrofotometer UV-Vis. Penentuan konsentasi optimum BSA dalam matriks silika gel dibuat dengan variasi konsentrasi 10, 20, 30, 40, dan 50 mg/L pada pH dan waktu kontak optimum. Hasil penelitian menunjukkan bahwa konsentrasi optimum adsorpsi BSA pada konsentasi 40 mg/L dengan efisiensi adsorpsi sebesar 99,06%. Adsorpsi BSA mengikuti model isotherm Langmuir tipe 2 dengan nilai R2 = 0,9492 dan kapasitas adsorpsinya sebesar 9,5057 mg/g. Proses adsorpsi BSA oleh silika dalam matriks silika gel terjadi secara kimia dan monolayer.

Abstract

A problematic bone remodeling cycle is known to produce more osteoclasts than osteoblasts, making bones more fragile and this condition shows osteoporosis. In this context, estrogen deficiency in the human body is associated with the regulation of osteoporosis. Therefore, this study aimed to investigate the knowledge about basic concepts of bone, osteoporosis, phytoestrogens, and Indonesian medicinal plants for osteoporosis treatment. Data were obtained from literature on various databases including Science Direct, Wiley Online Library, Scopus, Pubmed, and Google Scholar. Adequate therapy is needed to increase estrogen content and an effective approach is to consume medicinal plants that contain phytoestrogens, which have identical structure and activity to human estrogen (17β-estradiol). The results showed that there were observations comprising in vitro, in vivo, and in silico studies on 18 Indonesian medicinal plants as antiosteoporosis treatments. The Leguminosae or Fabaceae family, which has a significant amount of isoflavones (the primary group of phytoestrogens) was found to dominate as an antiosteoporosis agent. Therefore, the development of phytoestrogens therapy from Indonesian medicinal plants must be implemented for the future treatment of osteoporosis.

Abstract

Sponge-associated marine fungi are potential source for secondary metabolite compounds. The aim of this research was to investigate sponge-associated marine fungus as secondary metabolite producers against Escherichia coli. The fungus was isolated from Indonesian marine sponge Stylissa sp. and identified as Aspergillus hortai through a combination of morphological and molecular characteristics of ITS DNA and β-tubulin genes. The fungus was tested against E. coli using fungal broth and mycelial extracts. The optimized condition was achieved by fungal broth grown in corn meal broth at 6-days of shaking incubation. Fungal extract was produced using three liters of filtered fungal broth and extracted in ethyl acetate. The antibiotic activity of the extract is vulnerable to 45°C heat and basic or acidic conditions. Therefore, the extraction was done at pH 7 with evaporation at 40°C. The extract shows 7 major bands on TLC with 1 band shows activity against E. coli (Rf 0.81) on bioautogram. The band was observed as a yellow color and turned black in short-wave UV and did not show any fluorescence in long-wave UV. This research shows that sponge-associated marine fungi obtained from Indonesia has the potential as anti E. coli worth to be explored for searching new antibiotics.

Abstract

Temulawak (Curcuma xanthorrhiza) is known to be used as ingredient in Jamu and traditionally utilized to cure a range of illness. The global market demand for the rhizome of the temulawak affecting the urgency of sustainable rizhome production. The aim of this research was to analyze the effects of root endophytic and arbuscular mycorrhizal fungi on growth and rhizome curcumin content of the temulawak. There were five fungal inoculation treatments on C. xanthorrhiza seedling grown in sterilized and unsterilized growth media. They were root endophytic fungi A. niger (A), Glomus sp. (G), combination of A. niger and AMF Glomus sp. applied at the same time (GA) and at different time (G-A) and control. The experiment was conducted under greenhouse condition. The plant growth parameters, fungal colonization, and rhizomes curcumin content were measured. The result showed that there was a significant interaction between fungal inoculation and control treatments. In general, fungal inoculation on both sterilized and unsterilized growth media increased the plant growth and rhizomes curcumin content. On the sterilized growth media, C. xanthorrhiza inoculated by A. niger showed the best growth parameter. On the other hand, the best growth parameter on the unsterilized media was showed by C. xanthorrhiza inoculated by mixed fungal inoculums. The highest amount of curcumin content was showed by C. xanthorrhiza inoculated by Glomus sp. as a single inoculation.

Abstract

Used cooking oil (UCO) is abundant in Indonesia because of lifestyle of people. Especially UCO from street vendors had very low quality based on its Free Fatty Acid (FFA) content that reached 5.074%. When UCO was deposed into drainage, it would pollute the environment. Low grade UCO from street vendors contained Unsaturated-Fatty-Acid, Saturated-Fatty-Acid, and other hydrocarbons. Recycling UCO to replace edible oil considered more desirable rather than biodiesel since higher added value. FFA was the most undesired compound affecting cooking oil quality. FFA reducing with diluted 0.1 M NaOH 10% v/v could reduce FFA from 5.07% to 0.53%. Bleaching earth (BE) content mostly Si, Nb, Al, Ca and Na was safe to treat UCO for edible cooking oil. BE played cleaning UCO but absorbed the oil 17%. ANOVA analysis for BE and Moringa oleifera leaves (MOL) treatment fits with Reduced Quadratic model. Based on the ANOVA, the model showed that the decrease of FFA was determined significantly by BE amount and temperature with P-values less than 0.05. Optimization for BE treatment was done using Response Surface Method to find out the lowest FFA. And based on the simulation, it was obtained 0.2854% while the experimental results showed 0.2908%. Furthermore, BE combined with Moringa oleifera leaves (MOL) that known anti-oxidant source was used to treat UCO. UCO originally content stronger anti-oxidant activity rather fresh palm oil with IC50 62.12 and 83.03 mg/kg, respectively. Stronger anti-oxidant in UCO allegedly derived from fried foods. Treatment 10% BE and 5% MOL considered optimal for good appearance edible oil, little yellow greenish. Moreover, GC/MS analysis showed improvement active compounds in treated UCO with 5, 7.5 and 10 g MOL in 150 ml UCO with 24.36%, 30.32% and 37.76% active compounds, respectively. Moreover, anti-oxidant measured with IC50 the treated UCO were 43.18, 42.33 and 41.78 mg/kg, respectively. Increasing the number of MOL did not significantly increase the anti-oxidant activity. Anti-oxidant detected in treated UCO identified such as ethyl acridine, indolizine derivatives, cyclotrisiloxane-hexamethyl, benzimidazoles, and Fumaric acid. Based on the results, UCO recycling is applicable for a community, save expenses and strengthen food security and food sovereign. Further thought for its implementation is to design an integrated system from collecting UCO in a reservoir tank equipped with filter, recycling process and finally product tank at mini plant scale. Abstract

This research aims to develop a sensitive and selective nonenzymatic electrochemical sensor for glucose detection using a glassy carbon electrode modified with platinum nanoparticles (PtNPs) decorated on reduced graphene oxide (RGO). The structural properties and surface morphology of PtNPs/RGO composite were characterized using Raman spectroscopy and scanning electron microscopy (SEM). In addition, cyclic voltammetry (CV) and differential pulse voltammetry (DPV) techniques were employed to investigate glucose measurements in human urine samples. The developed sensor shows an increasing anodic peak of glucose with a linear response at a concentration range from 10 to 1000 µM, with a detection limit of 5 µM. The proposed sensor also demonstrated good reproducibility, indicated by the value of relative standard deviation (%RSD) of 3.9%, and maintained its current response over seven consecutive measurements. Moreover, the proposed sensor exhibited high selectivity for glucose detection against several potential interferences, such as potassium (K+), chloride (Cl−), magnesium (Mg2+), ascorbic acid, dopamine, and urea, with recovery values of 96-102%, which are acceptable within the analytical range. Furthermore, this proposed sensor successfully detected glucose in human urine samples, and their concentrations were not significantly different when measured with a commercial glucose sensor.

Abstract

A simple, portable visible reflectance spectrophotometer was developed for classifying the quality of Curcuma xanthorrhiza Roxb. The device was assembled using consumer electronic components: an LED as the light source, a DVD optical layer as the diffraction grating, and a webcam as the detector, with web-based software (Spectral Workbench) as the signal processor. The spectrophotometer's performance was evaluated using standard Sudan III samples and powdered C. xanthorrhiza samples from two quality classes (A and B). Spectral data were analysed using principal component analysis (PCA) and partial least squares (PLS) regression.  PCA successfully grouped Sudan III samples by concentration and classified C. xanthorrhiza by curcuminoid content. A PLS regression model was developed for quantifying Sudan III, yielding R² values of 0.9909 for both calibration and cross-validation, with RMSEs of 0.3821% and 0.4152%, respectively. The results closely matched those from a commercial spectrophotometer. A PLS-discriminant analysis (PLS-DA) classification model for C. xanthorrhiza was also developed, achieving sensitivity and specificity values of 1. Additionally, semi-quantitative parameters such as decision limit (26.15% B), detection capability (41.06% B), and unreliability range (26.15–41.06% B) were calculated. The classification model showed strong sensitivity, specificity, and detection capabilities within acceptable limits. These findings suggest that this low-cost reflectance spectrophotometer, combined with chemometric methods, holds promise as a practical tool for the quality control of raw herbal materials.
