Abstract

Tuberculosis in children is often a systemic complication because of their imperfect cellular immune system. ESAT-6, CFP-10, and MPT-64 are small molecular dominant antigens secreted by Mtb into the blood and filtered by the kidneys into urine. The high proportion of children with TB cases according to the Ministry of Health of the Republic of Indonesia in 2013 was 7.9%-12%. Diagnis is difficult to establish with sputum and blood samples. Therefore, it is necessary to support the diagnosis using urine samples with ICT rapid test. The purpose of this study was to determine the examination of Mtb ESAT-6, CFP-10, and MPT-64 antigens in the urine of pediatric TB patients with ICT to support the diagnosis. The method used was analytic observation with case control design. The results of the study showed that 32 of the urine of children with TB who were examined with ICT, 22 showed ESAT-6, CFP-10, and MPT-64 positive and 10 negative. Whereas, from 32 urine of healthy children, 25 were negative and 7 were positive. Analysis results showed 95% confidence (CI), p = 0.0002 (p <0.05), 78% specificity and 68.8% sensitivity compared to TB diagnosis from pediatricians. As a conclusion, Mtb ESAT-6, CFP-10, and MPT-64 antigens can be examined in the urine of pediatric TB patients with ICT to support the diagnosis.

Abstract

Currently, therapy with Platelet Rich Plasma (PRP) has been widely used and continues to grow for various clinical applications. Along with its development, there are various options in the method of obtaining PRP, either automatic or manual, while one of the most reliable according to the literature is a double centrifugation method. The purpose of this research is to produce an optimization of the double centrifugation method. This study uses experimental data obtained by researchers at the Clinical Pathology Laboratory of Dr. Soetomo Hospital, Surabaya. Experiments generally were conducted on stored blood obtained from the blood bag from PMI with CPD anticoagulant. Optimization of double centrifugation method to produce PRP with optimum platelet count could be obtained by using an "I"-shaped tube as well as other specialized equipment which are currently not available in standard clinical pathology laboratory, and also fresh blood. The resulting increase amounted to 4.11 times with the platelet count of 1.152 million.

Abstract

Sepsis is a major problem causing high morbidity and mortality in patients who hospitalized in intensive care. In sepsis, there is a change in the distribution of albumin in the intravascular and extravascular compartment, so hipoalbumin is occured. Using albumin in septic patients from several studies that have been done are still controversial. Analyzing albumin infusion with changes in levels of albumin, cytokines and SOFA score in septic patients in intensive care dr. Soetomo. The design of this study is a prospective longitudinal observational. The study was conducted in the intensive care unit or ICU dr. Soetomo in February 2015. The inclusion and exclusion criteria were determined that obtained 15 research subjects. They were examined with serum albumin and proinflammatory cytokines as well as evaluating the SOFA score. Then, it measure with ANOVA test methods. Infusion of albumin positively correlated with increased levels of serum albumin and a negative correlation with the levels of IL6, TNF-a and SOFA score. It also found a correlation between changes in levels of albumin with changing levels of IL6 (p <0:01) and changes in SOFA score (p <0.05).

Abstract

The skin, the largest organ in the human body, serves as a protective barrier against external factors. Exposure to UV-B radiation accounts for approximately 90% of skin damage, leading to aging characterized by dryness and wrinkles. Proper nutrition has been associated with skin repair and delayed aging. Astaxanthin, known for its pharmacological properties including anti-cancer, anti-diabetic, anti-inflammatory, and antioxidant activities has garnered attention for its ability to improve damaged skin when administered as a daily supplement. This study investigated the roles of TNF-α and IL-10 as inflammatory markers in skin damage and repair, which remain underexplored. A true experimental randomized posttest-only control group design was employed using 24 male white rats divided into four groups: (1) normal group (N), untreated rats; (2) control group (C), rats administered with olive oil (5 ml/kg BW); (3) treatment group 1 (P1), rats exposed to UV-B and administered with olive oil (5 ml/kg BW); and (4) treatment group 2 (P2), rats exposed to UV-B and administered with astaxanthin (10 mg/kg BW) combined with olive oil (5 ml/kg BW). ANOVA and Games-Howell post-hoc tests revealed significant differences (p < 0.05) among groups, except between the control (C) and normal (N) groups. Astaxanthin (10 mg/kg BW) demonstrated both curative and protective effects by reducing TNF-α expression (pro-inflammatory) and increasing IL-10 expression (anti-inflammatory) in UV-B-induced rats. These findings highlight astaxanthin's potential as a therapeutic agent for UV-B-induced skin damage.
