Abstract

Operational implementation, such as laboratory staff and equipment, differ for each frozen semen producer, resulting in variability in the quality assessment of the frozen semen produced. Conventional assessments subject to subjectivity, human error, and high variability. Computer-assisted semen analysis (CASA) is considered more objective. This study aimed was to conduct a comparative analysis of motility and concentration assessment of frozen semen sperm using conventional and the portable CASA AndroScope. Twenty-one laboratory assistants from 21 national and regional frozen semen producers participated in this study. Sperm motility and concentration were assessed conventionally (using a Neubauer chamber) and AndroScope was used with five replicates. The results of the comparison of sperm motility scores revealed significant differences (p<0.05) between conventional examination and using AndroScope. The results of the comparison of the sperm concentration calculations revealed no significant difference (p>0.05) between the calculations performed using the Neubauer chamber and AndroScope. Assessment with the AndroScope is considered reliable and can replace conventional assessment in assessing motility and sperm concentration, thereby improving and standardizing the assessment performed by frozen semen producers.

Abstract

This study aims to find out the characteristic of gayo horse and to identify the population of gayo horse. Data collection was conducted through selection process over 100 local horse distributed in Central Aceh Sub-districts, Gayo Lues and Bener Meriah. The result was 30 gayo horses with varying age distribution. Detail observation was performed on six gayo horses of male (n=3) and female (n=3). The observation consisted of morphological observation and morphometry. Data collection of gayo horse population was based on annual report from local Animal Husbandry and Fishery Official over the last 5 years. Result showed that gayo horse has straight cranial shape similar to thoroughbred with smaller size, thick and stiff mane, and ears akin to donkey. Gayo horse body height ranges between 113-120 with body weight ranging around 215-280 kg. Gayo horse is agile despite living in mountain and able to carry heavy weight. There has been drastic decline of population in year 2010-2014. In conclusion, gayo horse is included as large pony category, and without any rescue effort, gayo horse is predicted to be extinct in 2037.

Abstract

Sheep farming plays a crucial role in Indonesia's livestock industry; however, the effectiveness of artificial insemination (AI) is hindered by the less-than-ideal quality of cryopreserved semen. This research assessed the longevity and progressive motility recovery rate (RR) of frozen semen from local Indonesian rams, using a homemade Tris egg yolk (TEY) extender and three commercial extenders (AndroMed®, Steridyl®, BoviFree®). Semen from three rams (Batur, Garut, and Sakub) was processed, frozen in 0.25 mL straws (50 × 10⁶ sperm per straw), and its quality was evaluated using CASA. Longevity was monitored hourly from 0 to 4 h at 37°C. The data were analyzed using a Randomized Block Design with ANOVA, followed by Duncan’s Multiple Range Test. Sperm motility decreased over incubation time for all extenders, with notable differences at 4 h (total motility, p = 0.017; progressive motility, p = 0.023). Commercial extenders preserved higher total and progressive motility than TEY, with Steridyl® showing the highest mean RR (64.11%; mean all rams; RR analysis, n = 18), significantly outperforming Andromed®, BoviFree®, and TEY (p = 0.007). The superior results of Steridyl® align with its low-density lipoprotein content, which stabilizes the sperm membranes during cryopreservation. In summary, Steridyl® was the most effective extender for maintaining motility and RR of frozen semen from local Indonesian rams, potentially enhancing AI success in small ruminant programs.
