Abstract

Kandungan bahan bioaktif pada tanaman memiliki beragam potensi aktivitas biologis dan dimanfaatkan dalam budidaya ikan sebagai alternatif untuk pencegahan dan pengobatan penyakit ikan. Serapoh (Chromolaena odorata) diketahui memiliki bahan bioaktif namun penerapan untuk pencegahan dan pengobatan penyakit pada kakap putih belum pernah diteliti. Penelitian ini bertujuan untuk mengevaluasi potensi antioksidan dan antibakteri daun serapoh secara in vitro terhadap Vibrio harveyi penyebab penyakit black body syndrome pada benih kakap putih. Penelitian secara in vitro melingkupi analisis fitokimia, uji antioksidan dan antibakteri. Hasil uji menunjukkan bahwa ekstrak daun serapoh mengandung flavonoid, tannin, saponin, dan steroid. Rendemen dari hasil maserasi dengan pelarut akuades, etanol, etil asetat, dan n-heksan berturut-turut adalah 11,34%; 9,13%; 4,21%; dan 1,48%. Ekstrak etil asetat memiliki kandungan total fenol yang tertinggi (212,8 mg/g) dibanding ekstrak yang lain. Kandungan total flavonoid yang tertinggi terdapat pada ekstrak etanol (195,5 mg/g) diikuti dengan ekstrak etil asetat (20,2 mg/g), n-heksan (10,6 mg/g), dan akuades (8,1 mg/g). Nilai potensi antioksidan ekstrak etanol lebih tinggi (86,59%) dibanding ekstrak yang lain namun potensi antioksidan ekstrak etanol, etil asetat, dan akuades tidak berbeda nyata dengan asam askorbat sebagai pembanding. Ekstrak etanol, etil asetat, dan n-heksan dapat menghambat pertumbuhan V. harveyi. Ekstrak etanol bersifat bakteriostatik (1,25 mg/mL) dan bakterisidal (5 mg/mL), serta menyebabkan kerusakan sel sehingga metabolit seluler seperti asam nukleat dan protein dapat keluar dari sel. Hasil penelitian menunjukkan bahwa ekstrak daun serapoh memiliki potensi antioksidan dan antibakteri terhadap V. harveyi sehingga dapat digunakan untuk pencegahan dan pengobatan penyakit black body syndrome pada benih kakap putih.Bioactive compounds in plants have various potential biological activities and are commonly used in fish farming as alternatives to prevent and treat fish diseases. Serapoh (Chromolaena odorata) is known to have bioactive compounds, yet its application to prevent disease in Asian seabass has not been studied. This study aimed to evaluate the antioxidant and antibacterial potential of serapoh leaves in vitro against Vibrio harveyi, causing black body syndrome disease in Asian seabass. The performed tests in this study consisted of phytochemical analysis, antioxidant, and antibacterial tests. The results showed that serapoh leaf extract contains flavonoids, tannins, saponins, and steroids. The yields obtained from maceration with aquadest, ethanol, ethyl acetate, and n-hexane solvents were 11.34%; 9.13%; 4.21%; and 1.48%, respectively. Ethyl acetate extract had the highest total phenol content (212.8 mg/g) compared to the other extracts. Ethanol extract has the highest total flavonoid content (195.5 mg/g) followed by ethyl acetate (20.2 mg/g), n-hexane (10.6 mg/g), and aquadest (8.1 mg/g). The highest antioxidant potential value was shown by ethanol extract (85.59%), but the antioxidant potentials of ethanol, ethyl acetate, and aquadest extracts were not significantly different from ascorbic acid. Ethanol, ethyl acetate, and n-hexane extracts can inhibit the growth of V. harveyi. Ethanol extract has bacteriostatic (1.25 mg/mL) and bactericidal (5 mg/mL) properties. The exposure of V. harveyi to ethanol extract resulted in cellular damage that can release cellular metabolites such as nucleic acids and proteins. In conclusion, serapoh leaf extract had antioxidant and antibacterial potential against V. harveyi and could be used to prevent or treat black body syndrome in Asian seabass.

Abstract

Snakehead fish Channa striata is a high commercial freshwater fish commodity. It has the potential as a pharmaceutical ingredient. Intensive snakehead fish cultivation starts to experience a problem, namely Motile Aeromonas Septicemia (MAS) caused by Aeromonas hydrophila. The purpose of this study is to evaluate the effect of the addition of commercial yeast in feed to improve snakehead fish immunity. This study was designed with a completely randomized design (CRD) with 5 treatments with 3 replications (reared in the net) and 6 treatments with 3 replications (reared in the aquarium). The treatments were K (feed without yeast), F5 (feed with the addition of 5 g/kg of cake yeast), R3 (feed with the addition of  3 g/kg of tempeh yeast, R5(feed with the addition of 5 g/kgof tempeh yeast), and R7 (feed with the addition of 7 g/kg of tempeh yeast). The results show that the survival rate after 30 days is 88.89- 92.22%. The best treatment after A. hydrophila infection was found in fish fed with 3 g/kg oftempehyeast with a survival rate of 56.67%, total erythrocytes of 4.07x 106 mm-3 cells, hemoglobin of 7.40 g% of total leukocytes 4.97x 104 mm-3 cells and phagocytic activity of 33.67. In conclusion, the addition of tempeh yeast at a dose of 3 g/kg could be used as an alternative to prevent the effect of A. hydrophila pathogen infection in snakehead fish.

Abstract

Pacific whiteleg shrimp (Litopenaeus vannamei) is a very important commodity, which accounts for almost 70% of the total world shrimp production. However, this production is still threatened by disease attacks, one of which is caused by white spot disease (WSD). Herbs are widely used as immunostimulants as an effort to prevent diseases. Black cumin (Nigella sativa) and stone breaker (Phyllanthus niruri) have long been known as excellent immunostimulants in human. This study aimed to evaluate the potential of N. sativa and its combination with P. niruri as an immunostimulant in L. vannamei for preventing WSD. Tested shrimp (±3.0 g) were treated by N. sativa (N), P. niruri (P) and the combination of both (1N:1P, 2/3N:1/3P, 3/4N:1/4P, 1/3N:2/3, 3/5N:2/5P, 1/4N: 3/4P, 2/5N:3/5P) through feed for 28 days. They were then infected by white spot syndrome virus (WSSV) and observed for 7 days. The parameters observed included immune response and production performance.  N. sativa was able to reduce the mortality of test shrimp infected with WSSV, with an RPS value of 71%. N. sativa can be combined with P. niruri in the right composition (1/2N:1/2P and 2/3N:1/3P), while the other combinations are antagonistic. The administration of the best treatments in this study, relatively did not affect the value of total hemocyte count, phenoloxidase activity, respiratory burst activity, hemolymph clotting time and production performance of tested shrimp.

Abstract

Bakteri Streptococcus agalactiae merupakan patogen utama yang menyerang ikan nila mulai fase benih hingga dewasa. Tujuan penelitian ini yaitu untuk menguji efikasi vaksin booster S. agalactiae di induk ikan nila sebelum pemijahan kedua terhadap imunitas maternal untuk pencegahan streptococcosis. Vaksin gabungan sediaan sel utuh dan produk ekstraselular (ECP) S. agalactiae diinjeksi sebanyak 0,4 mL/kg induk ikan dengan perbandingan 50:50% (v/v) dari dosis penyuntikan, sedangkan kontrol diinjeksi dengan phosphate buffered saline (PBS). Vaksin diberikan ke induk ikan pada fase tingkat kematangan gonad dua (TKG 2). Perlakuan penelitian yaitu induk diinjeksi PBS (K), induk diinjeksi vaksin satu kali (A), dan induk diinjeksi vaksin booster setelah pemijahan pertama (B). Uji tantang benih dari setiap induk perlakuan dilakukan melalui perendaman S. agalactiae 107 CFU/mL selama 30 menit pada umur benih 5, 10, 15, dan 20 hari setelah menetas. Hasil penelitian menunjukkan terdapat perbedaan yang signifikan (P<0,05) pada total leukosit dan aktivitas fagositik induk ikan. Level antibodi dan lisozim induk, telur, dan benih dari perlakuan B signifikan lebih tinggi (P<0,05) dibandingkan dengan perlakuan induk lainnya. Daya tetas telur dari perlakuan induk B (94,52%) signifikan lebih tinggi (P<0,05) dari pada perlakuan induk lainnya. Nilai Relative percent Survival benih dari perlakuan induk B tidak berbeda signifikan (P>0,05) dengan perlakuan benih dari induk A pada hari ke-5 tetapi, signifikan lebih tinggi (P<0,05) pada hari ke-10 hingga 20 pascatetas (96,61-54,17%). Pemberian vaksin booster di induk ikan nila sebelum pemijahan kedua dapat menstimulasi peningkatan imunitas dan transfer imunitas maternal ke anaknya untuk pencegahan streptococcosis.

Abstract

The most common problem in barramundi Lates calcarifer seedling production is the high mortality (> 90%) caused by nervous necrosis virus (NNV) infection. This research aims to evaluate the resistance and gene expression of barramundi challenged by NNV. Two populations were used in this study, i.e., Australian, and Situbondo-originated barramundi populations. The immune-related gene expression levels in the liver, head of kidney, and spleen were observed at 48 and 96 hours after post-infection (hpi). Barramundi’s survival and blood parameters were evaluated post-NNV infection. The results showed that the highest survival was revealed in Situbondo’s barramundi (42.0±4.47%) compared to Australian barramundi (20.0±7.07%) and no mortality was observed in the control without NNV infection. The higher survival rate in barramundi from Situbondo was in line with the blood profile. The number of red blood cell from Situbondo barramundi post-NNV infection (ST) at 96 hpi was higher (P<0.05) than Australian barramundi post-NNV infection (AT). The number of white blood cell of ST at 48 hpi was higher (P<0.05) than AT, but started to decrease at 96 hpi in ST barramundi. The total white blood cell in AT barramundi increased from 48 to 192 hpi. TNFα and IL1-β gene expression levels were significantly higher in the liver, head kidney, and spleen of Situbondo compared to Australian barramundi at 48 hpi, while MHCIIα gene expression in Situbondo’s was significantly higher compared to Australian barramundi at 96 hpi. These results indicate the important roles of all the genes in the barramundi’s immune responses against viral infection. Based on the results of the research, Situbondo’s barramundi has the potential to be used as a candidate for generating broodstock of disease-resistant strain.

Highlight Research A molecular marker for shrimp selection in disease resistance. Single nucleotide polymorphisms (SNP) in the ALF gene strongly correlate with shrimp resistance to WSSV infection. Higher ALF gene expression in survivor shrimp. SNP as molecular marker inherited in the first generation (G1) shrimp. ARMS-PCR method successfully detect SNP in the shrimp ALF gene. Abstract The Pacific white shrimp (Litopenaeus vannamei) is Indonesia's main export commodity, but its production is constrained by the white spot syndrome virus (WSSV). Selective breeding of disease-resistant broodstock based on single nucleotide polymorphism (SNP) in the anti-lipopolysaccharide factor (ALF) gene is an alternative strategy for solving the disease problem. This study aimed to detect the SNP g.455 A>G in the anti-lipopolysaccharide factor (ALF) shrimp gene, evaluate the correlation of SNP with WSSV-resistance trait, analyze the expression level of immunity genes and genotype frequencies of the WSSV-resistance population shrimp and analyze the SNP inheritance in the first generation of selected shrimp. A total of 120 individuals from 4 families were used to detect the SNP marker using tetra-primer amplification refractory mutation system-polymerase chain reaction (ARMS-PCR). The correlation of the SNP marker with survival rate (SR) was analyzed using a general linear model (GLM) between genotype frequencies and SR. Genotypic similarities between broodstock and pedigree were analyzed using Chi-square. SNP g.455 A>G was successfully detected using the ARMS-PCR method and had a strong correlation between the marker and SR (p-value of AA = 0.012; AG = 0.359, and GG = 0.001). The resistant population has significantly higher ALF and SOD gene expression levels and AA genotype frequency. The SNP marker was inherited, so the broodstock and pedigree have the same genotype frequencies according to chi-square analysis (χ2 = 0.46 and p-value = 0.497). These results suggested that the g.455 genotype AA could be selected to produce WSSV-resistant Pacific white shrimp.

Abstract

Pariwisata adalah sektor yang berkembang pesat dan signifikan di Indonesia, seiring dengan hal tersebut peningkatan sumber daya manusia pelaku pariwisata adalah sebuah keniscayaan, termasuk sumber daya manusia di bidang perhotelan. Tujuan dari penulisan ini adalah untuk mengetahui pengaruh pengembangan karir, kepemimpinan terhadap kepuasan kerja melalui motivasi kerja di hotel. Penelitian ini adalah penelitian kuantitatif dengan populasi penelitian karyawan salah satu grup hotel di Jakarta, dengan jumlah sampel sebanyak 215 pegawai. Menggunakan metode analisis jalur, hasil penelitian mengungkapkan bahwa terdapat pengaruh langsung yang signifikan antara pengembangan karir dengan motivasi, kepemimpinan dengan motivasi, pengembangan karir dengan kepuasan kerja, kepemimpinan dengan kepuasan kerja dan pengaruh tidak langsung antara pengembangan karir terhadap kepuasan kerja melalui motivasi kerja serta kepemimpinan terhadap kepuasan kerja melalui motivasi kerja.

Abstract

Viral inhibitor protein (VIP) contributes to shrimp resistance against virus infection. VIP can be used as a new strategy to control white spot syndrome virus (WSSV) infection in shrimp culture by inhibiting its action. This study aimed to evaluate the application of VIP on the whiteleg shrimp (Litopenaeus vannamei) to inhibit the WSSV infection. Shrimp at an average body weight of 6.66±0.08 g was injected by recombinant VIP with two different concentrations (1 μg/g and 10 μg/g) and challenged with WSSV. As a positive control was the shrimp not injected with recombinant VIP and challenged with WSSV, and as a negative control was the shrimp not injected with recombinant VIP and not challenged with WSSV. The results showed that the survival rate of shrimp that were injected with recombinant VIP was significantly increased after being challenged with WSSV (P<0.05). The survival and immune response of shrimp that were injected with recombinant VIP were higher than the positive controls (P<0.05). In this study, the application of recombinant VIP can significantly increase the shrimp resistance against WSSV up to 95.65% higher than the positive control.

Abstract

ABSTRACT  Based on caries history and caries lesion white spot is known as the best indicator in determining caries development. However, these indicators cannot reach the target accuracy of around 80%.  Caries or cavities are problems that are often found in the oral cavity. According to WHO global data (2000) shows 60-90% of school children in industrialized countries have cavities. Based on the results of the 2007 Riskesdas in the province of South Kalimantan, the percentage of the population with dental and oral problems was 29.2%.This activity aims to identify def-t, DMF-T, carry out promotive, preventive and curative, namely by counseling, mass toothbrushes and filling applications with ART fillings on permanent first molars for students at SDN Kurau, Tanah Laut Regency, South Kalimantan Province . The community service target is 100 students. However, only 44 students and 12 parents attended. The results of the def-t examination obtained d = decay or dental caries of 165, e = extraction or teeth that could not be treated 97 teeth and f = fillings was 0, so the number of def-t was 350 teeth with an average of 7.98. Whereas the DMF-T had 80 teeth with an average of 1.82. The caries rate for primary and permanent teeth was high, that is, each child had 7-8 cavities for primary teeth while permanent teeth had cavities 2. Restoration with ART materials that can performed as many as 72 teeth. And 2 months later an evaluation was carried out, the results obtained were that 68 teeth were still holding on with the ART fillings, while 4 teeth had fallen out. The fact found in the field is that dental caries in primary teeth and permanent teeth is still very high, namely 143 teeth and 80 teeth in students of SDN Kurau, Tanah Laut Regency and what can be done with ART restoration on permanent first molars and referrals to the puskesmas or The hospital performed pulpal treatment and tooth extraction. It is recommended to treat teeth that can still be treated, namely the collaboration of the teacher (school), parents and the health center. Abstract

Dental caries is one of the most common chronic and endemic diseases in school-aged children. Untreated caries can affect weight, growth, quality of life and impact the health of permanent teeth. External factors that influence the occurrence of dental caries are the level of knowledge, attitudes and behavior of children in oral health. By empowering children's independent ability to obtain their oral health status, it is hoped that the Caries Free Program can be realized. The expected form of independence is that school children are able to gain knowledge, attitudes and positive behaviors independently. This can be realized if there is an active change of agent in their peer environment. The formation of the Little Doctor Team in this school environment is expected to make them a Promoter of Dental & Oral Health in their peer environment. The purpose of the Community Service is to form a Little Doctor Team in Elementary School, equip them with knowledge about how to brush their teeth properly, as well as the introduction of Basic Dental & Oral Health Tools at SD Islam Al Madani Banjarbaru. The target of Community Service is selected students of SD Islam Al Madani Banjarbaru who are formed into a Little Doctor Team. The problem can be solved by forming a Little Doctor Team, as well as providing briefings on how to brush teeth properly and simple Dentistry Tools.
