Abstract

One of the long-term losses in homeostatic balance for any organism is a sign of aging skin. The state of hyperpigmentation caused by the activity of the tyrosinase enzyme is an indicator of aging. The solution that can be recommended to prevent premature signs of aging is by caring for the skin using the most affordable pharmaceutical preparation, namely Hand Sanitizer Gel. Hand Sanitizer Gel which has anti-aging properties can be produced from bioactive compounds from horticultural commodity plants because it has been proven and is quite popular. Apart from being able to be used as an active ingredient in functional food, it can also be used as a pharmaceutical, namely the preparation of active cosmetic ingredients. This is due to the content of vitamins, antioxidants, essential oils, proteins, even phenolic compounds and other environmentally friendly active ingredients found in horticultural commodity plants, especially the fruit group used in this research. Red dragon fruit has been proven to contain organic compounds such as gallic acid, elagic acid and ferulic acid which can inhibit the melanogenesis process both in vitro and in vivo from the results of previous findings. Lemons contain saponins, flavonoids, alkaloids and tannins so they can be designed to inhibit the tyrosinase enzyme. This research aims to determine the inhibitory power of the tyrosinase enzyme from extracts of horticultural commodity plants, both red dragon fruit and lemon peels in the form of hand sanitizer gel formulations and preparations. The results of this research have proven that based on the IC50 value, horticultural commodity plant extracts from lemon peel have better inhibitory power for the tyrosinase enzyme compared to red dragon fruit, namely 3,509 µg/mL. The conclusion in this research is that horticultural commodity plants in the form of lemon peel extract are more recommended as an active substance in inhibiting the tyrosinase enzyme when compared with red dragon fruit extract based on the IC50 value even though the hand sanitizer gel dosage form has the potential to inhibit the tyrosinase enzyme.

Abstract

This study aimed to isolate and characterize endophytic bacteria from black turmeric rhizomes (Curcuma aeruginosa Roxb) and test their potential as antibacterial agents against Streptococcus pneumoniae. This experimental research began with the isolation of endophytic bacteria from surface-sterilized black turmeric rhizomes. The obtained pure isolates were characterized macroscopically (colony shape, margin, elevation, pigment) and microscopically (cell shape, Gram staining). The growth curve of potential isolates was determined, and antibacterial activity was tested using the disc diffusion method against S. pneumoniae, with ciprofloxacin as a positive control. Six endophytic bacterial isolates (PS1, PS2, PS3, PS4, PS5, PS6) were successfully isolated, showing variations in colony morphology, and all were classified as Gram-negative bacilli. All isolates exhibited strong antibacterial activity against Streptococus pneumoniae, with inhibition zone diameters ranging from 15.3 mm to 20.6 mm. Black turmeric rhizomes (Curcuma aeruginosa Roxb) harbor diverse endophytic bacteria with significant potential as antibacterial agents against the pneumonia pathogen, Streptococcus pneumoniae.

Abstract

Staphylococcus epidermidis is a coagulase-negative Staphylococcus (CoNS) that causes a spectrum of superficial skin infections including wound infections, folliculitis, and burn-associated skin sepsis, particularly through its capacity to form highly resistant biofilms. The emergence of methicillin-resistant S. epidermidis (MRSE) with polysaccharide intercellular adhesin (PIA)-mediated biofilm has severely limited available therapeutic options. This study assessed the concentration-dependent bacteriostatic activity and minimum inhibitory concentration (MIC) of Citrus aurantifolia lime peel ethanolic extract against S. epidermidis ATCC 12228 using the Kirby-Bauer disc diffusion method on Muller Hinton Agar (MHA). Extract from lime peels sourced in Tulungagung, East Java was prepared by ethanol maceration. Fourteen concentrations (0.5–150 mg/mL) were tested in triplicate. The MIC was 1 mg/mL (inhibition zone 6.73 ± 0.06 mm). Inhibition zones increased progressively from 6.73 mm (MIC, 1 mg/mL) to 11.60 ± 0.35 mm (150 mg/mL), with the very strong threshold (>10 mm) first crossed at 70 mg/mL. Concentration-response linearity was confirmed (R² > 0.97). The positive control (tetracycline 30 µg) produced 28.17 ± 0.59 mm. These findings establish a pharmacological basis for incorporating lime peel extract into wound care and topical antiseptic formulations targeting S. epidermidis.

Abstract

Diarrhea remains a major global health challenge caused by pathogenic bacteria including Escherichia coli, Salmonella typhi, Staphylococcus aureus, and Bacillus cereus. African leaf (Vernonia amygdalina Delile.) contains bioactive compounds such as flavonoids, saponins, tannins, and alkaloids with recognized antibacterial properties. This study determined the Minimum Inhibitory Concentration (MIC) of ethanol extract of V. amygdalina prepared by Ultrasonic-Assisted Extraction (UAE) at three extraction times (5, 15, and 30 minutes) against four diarrhea-causing bacteria using the disc diffusion method. Extraction yields were 8.8%, 10.45%, and 11.9% for UAE at 5, 15, and 30 minutes, respectively. Antibacterial testing at concentrations ranging from 300 to 1.56 mg/mL demonstrated predominantly moderate inhibition zones (5–10 mm), with some concentrations reaching the strong category (10–20 mm). The MIC for B. cereus and S. aureus was 1.56 mg/mL across all extraction times. For S. typhi, the MIC was 1.56 mg/mL (5 and 30 min UAE) and 12.5 mg/mL (15 min UAE). For E. coli, the MIC was 6.25 mg/mL (5 min UAE) and 1.56 mg/mL (15 and 30 min UAE). One-way ANOVA confirmed statistically significant differences among concentrations (p < 0.05). The 30-minute UAE produced the highest yield and generally superior antibacterial activity, supporting its selection as the optimal extraction condition. These findings highlight the potential of V. amygdalina ethanol extract as a natural antibacterial agent for diarrheal disease management.

Abstract

Pigeon pea (Cajanus cajan L. (Huth.)) contains flavonoids that exhibit potential antidiabetic activity through the inhibition of the α-glucosidase enzyme. The main limitation in the application of flavonoid compounds is their low solubility and stability. Nanoencapsulation using chitosan–TPP via a simple ionic gelation method can enhance the stability and bioactivity of active compounds. This study aimed to develop a nanoencapsulation formulation of pigeon pea extract and to evaluate its physical characteristics, stability, and α-glucosidase inhibitory activity. The research was conducted using variations in nanoencapsulation extract concentrations: F1 (0.2%), F2 (0.4%), and F3 (1%). Physical characterization included particle size, polydispersity index (PDI), and zeta potential, along with a freeze–thaw stability test for five cycles. Antidiabetic activity was assessed by measuring α-glucosidase inhibition and determining the IC₅₀ value. Data were analyzed using one-way ANOVA followed by Tukey’s post hoc test (p < 0.05). The results showed that particle size decreased from 290.40 nm (F1) to 198.30 nm (F3), with polydispersity index values below 0.3 for F2 and F3. The zeta potential ranged from −28.10 to −36.10 mV. F3 demonstrated the highest stability and the strongest antidiabetic activity with an IC₅₀ value of 68 ppm (p < 0.05). In conclusion, the nanoencapsulation formulation of pigeon pea extract produced small-sized, stable particles with effective α-glucosidase inhibitory activity. The optimal formulation was F3, which shows potential for further development as a natural-based antidiabetic nanoencapsulation system.
