Abstract

Abstract

Abstract

Abstract. Pandawani NP, Listihani L, Widnyana IK, Ariati PEP, Selangga DGW. 2022. High impact of Clerodendrum paniculatum leaf extract to suppress zucchini yellow mosaic virus infection in zucchini plants. Biodiversitas 23: 2914-2919. Zucchini yellow mosaic virus (ZYMV) is an important virus in zucchini plants (Cucurbita pepo L.). ZYMV infection is prevalent in C. pepo in Bali and is extremely difficult to eradicate. This study aimed to determine the efficacy of pagoda leaf extract (Clerodendrum paniculatum L.) in reducing yield loss in C. pepo. A factorial randomized block design with two variables was employed in this study. The application time of the C. paniculatum extract was the first variable, and concentration was the second. Plant extract was applied during seed immersion, one day before and after inoculation, and on the same day of virus inoculation. The concentrations of C. paniculatum extract employed were 10%, 30%, and 50%. The crude extract was sprayed onto the C. pepo leaves. Plants treated with extract one day after virus inoculation at 10%, 30%, and 50% concentrations exhibited yellowing, yellowing with cupping, mosaic, mosaic with cupping, and stunting. While application of extract one day before virus inoculation at 30% and 50% concentrations resulted in yellowing and yellowing with cupping signs. Plants infected with ZYMV first showed flowering at 37 DAP, whereas plants treated with C. paniculatum leaf extract one day before virus inoculation started flowering at 32 DAP. Application of C. paniculatum leaf extract one day before ZYMV inoculation on C. pepo effectively suppressed 40% yield loss. The findings of present investigation revealed information about the potential of C. paniculatum leaf extract as a plant virus control agent through the mechanism of induction in C. pepo.

Abstract

Abstract. Prihatiningsih N, Rahayuniati RF, Djatmiko HA, Lestari P, Wulansari NK, Widnyana IK, Sutanto KD. 2024. Endophytic bacterial isolate diversity in suboptimal field rice and their potential in sheath blight control. Biodiversitas 25: 3359-3367. Rice root endophytic bacteria from suboptimal fields rice are beneficial for controlling plant pathogens. The present research aimed to characterize morphology and biochemical properties, identify secondary metabolites of rice root endophytic bacteria and evaluate their potential as biocontrol for sheath blight caused by Rhizoctonia solani. The present study focused on five endophytic bacteria, specifically Bacillus sp., with white colonies, rod-shaped cells, Gram-positive characteristics, and catalase-positive. These bacteria were selected from rice root samples due to their potential as antagonists against the pathogenic fungus R. solani. The inhibition potential of endophytes against R. solani was determined by measuring the radius of the fungal colony in the presence of the endophytic bacteria isolates. Among them, NP KR4 showed the highest inhibition percentage of 46.00 ± 6.25%, which was attributed to the antibiosis mechanism causing hyphal swelling. A molecular test using the 16S rRNA gene was conducted to determine the isolates' genetic identity. Comparing the gene sequences with those in the genetic database, it was found that NP A5, NP KR4, and NP KR7 isolates were similar to B. subtilis strain 2JKP676166, although with a few base pair differences. NP A6 exhibited a close relation to B. subtilis strain YT2 HQ143571, while NP SB3 was identified as B. subtilis subsp. stercoris strain EG1265MN704551. Further analysis of the endophytic bacteria's secondary metabolites through Gas Chromatography and Mass Spectroscopy revealed the presence of alkaloids, phenols, alcohols, and fatty acids. Among the isolates, NP KR4 demonstrated the highest biocontrol effectiveness against sheath blight, with an efficacy of 58.74%. Overall, the results indicated that the rice root endophytic bacteria isolated from suboptimal fields possess multiple contemporary traits, including antifungal activity and strain-specific differences as Bacillus subtilis. These findings suggest promising prospects for developing bio-fungicide formulations to promote sustainable rice production.

Abstract

Abstract. Suanda IW, Widnyana IK, Dharmadewi AAIM. 2025. Isolation and identification of pathogenic Fusarium oxysporum isolated from soybean leaves. Biodiversitas 26: 2261-2268. Not all fungi isolated from soybeans are pathogenic. Understanding the identity and pathogenicity of the fungi found is crucial for providing appropriate control recommendations, which can significantly impact soybean farming. This research aimed to compare the pathogenicity of various fungi isolates obtained from soybean plants with withering symptoms in fields in Bali, along with morphological and molecular identification. Five fungi isolates were chosen for the pathogenicity test, and the most aggressive isolate was further tested for morphological and molecular detection with PCR by using ITS1 and ITS4 primers. The pathogenicity test was performed by inoculating fungi into healthy soybeans from Anjasmoro, Argomulyo, and Argopuro cultivars, and the symptoms were assessed five days after inoculation. Five days after inoculation, the soybean leaves turned yellowish brown, the roots were few and short on all tested fungi isolates, while the uninoculated control seed remained asymptomatic. The five isolates were different in aggressiveness; the disease area percentage ranged between 81% and 98%, depending on the isolate. The most aggressive isolates inoculated in soybeans are the JKP2 and JKP3 isolates; the highest plant proportion inoculated by these isolates showed the most severe symptoms (the diseased area was 98% or complete plant rot). The JKP2 culture grown in PDA media is generally characterized by white to greenish purple, reverse white, and fluffy. At the same time, the mycelium of JKP3 isolate is white, reverse purple in the center, and white at the margins. Phylogenetic analysis confirmed the two isolates observed to be Fusarium oxysporum. To our knowledge, this is the first report of F. oxysporum isolated from soybeans in Bali.
