Abstract

Nature serves as an excellent inspiration for researchers in the fields of chemistry and medicine. Terrestrial or marine organisms produce billions of natural products with unique chemical and physical properties. Some of them have been used in traditional therapy for specific diseases since ancient times. Although their exact chemical structures have yet to be elucidated clearly in that time, investigations in medicinal chemistry have been well documented. Furthermore, utilizing natural products in drug design and development offers advantages such as high biocompatibility, low toxicity, fewer side effects, wide bioactivities, and large structure diversity. Nowadays, rational drug research using computer-aided drug design is well established to cut the long way of drug discovery and overcome the resistance cases and the increment in the number of active patients. This review will highlight some natural products to comprehensively understand their journey from unknown natural products, isolation, purification, characterization, in silico evaluation, bioactivity screening assay, drug modifications, in vitro investigation, in vivo examination, and clinical trial.

Abstract

Various agricultural products are known to have anti-microbial, anti-cancer, and anti-inflammatory effects. As we can mention, Walnut (Juglans regia) husk, orange (Citrus sinensis) peel, Broad bean (Vicia faba) peel, and Nettle (Urtica urens) are proven to have antimicrobial and anticancer actions. Also, plant diseases such a Pectobacterium carotovorum, Ralstonia solanacearum, Dickeya chrysanthemi, and Pseudomonoas syringae are known to cause annual damage to plant products. Therefore, in the current study, the researchers evaluated their antibacterial activity on the mentioned bacteria under in vitro conditions. Extracts of J. regia husk, C. sinensis peel, V. faba outer peel and U. urens were prepared by maceration method and their anti-bacterial activity on P. carotovorum, D. chrysanthemi, P. syringae, R. solancearum was evaluated using blank disk and well diffusion to obtain minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) values. J. regia (husk) and V. faba (outer peel) extracts revealed an anti-bacterial effect on all 4 studied bacteria, while C. sinensis (peel) only inhibited the growth of P. syringae and U. urens only had this effect on P. syringae and R. Solancearum. The inhibition zones varied from 8 to 14 mm, while almost all MIC and MBC rates were 6.25 and 12.5 mg/mL, respectively. Based on the antimicrobial results, the extracts that showed suitable antibacterial effects on certain bacteria can be further studied to be used as natural pesticides.

Abstract

In this study, edible films were made from glucomannan from local porang tubers, carrageenan, and sorbitol as the plasticizer. This research method begins with the making of modified porang flour with three immersion treatments, namely control (A), 10% lime solution (B), and modified lime and mocaf starter fermentation (C) to reduce oxalate levels. Plasticizers were added with variations of carrageenan 2;3;4%, and sorbitol 1;2;3;4; and 5 g. The results showed that the modified lime immersion and fermentation treatment reduced calcium oxalate levels, and the highest yields of glucomannan were 2.58% and 10.07 g. The FTIR analysis results of the isolated glucomannan showed that the spectrum formed by the presence of O-H functional groups, CH2 groups, C-O-C groups, absorption areas of 805 cm-1 and 872 cm-1 indicated the presence of the main constituent of glucomannan, namely β-pyranose. In comparison, the characteristics of the edible film obtained an average thickness between 0.32–1.50 mm. The highest thickness was obtained in the composition of the addition of 3% carrageenan and 4 grams of sorbitol variation C. The percentage of water resistance of edible film variations A and C ranged from 10.04% - 22.02%. Variations A and C with a composition of 2% carrageenan and 1 gram of sorbitol, the edible film is completely degraded. The surface morphology of the edible film obtained from variation A with a concentration of 2% carrageenan and 5 grams of sorbitol showed a slightly porous surface.

Abstract

Diabetes mellitus is a chronic non-communicable disease that occurs due to insulin deficiency or reduced function in the body. Therefore, the scientific investigation of herbal plants and their effectiveness is essential. It has been shown that gallic acid has hypoglycemic properties. In this study, the anti-diabetic and anti-lipid peroxidation properties of gallic acid were investigated. For this purpose, Streptozotocin was injected intraperitoneally into male rats at a dose of 60 mg/kg to induce type I diabetes. Seven days after Streptozotocin injection, blood samples were taken and rats with serum glucose levels higher than 250 mg/dL were considered diabetic. The rats were divided into 4 groups. Two groups of diabetic rats received either water or gallic acid at a dose of 40 mg/kg, and two groups of normal rats received either water or gallic acid at 40 mg/kg dose. The treatment period for all groups was 8 weeks. After the end of the period, blood samples were taken from the rats under fasting conditions. The collected blood samples were analyzed for serum biochemical factors (glucose, cholesterol, triglycerides, High-density lipoprotein, and creatinine) using spectrophotometry with the respective kits, and the level of lipid peroxidation in red blood cells was measured. The administration of gallic acid at a dose of 40 mg/kg (8 weeks) significantly reduced serum levels of glucose, triglycerides, cholesterol, alanine aminotransferase, aspartate aminotransferase, very low-density lipoprotein, low-density lipoprotein, and creatinine, while significantly increasing serum levels of high-density lipoprotein. Additionally, the level of Malondialdehyde in red blood cells was decreased in the diabetic group receiving gallic acid compared to the diabetic control group, and in normal rats receiving gallic acid at a dose of 40 mg/kg (8 weeks), it had no significant effect on serum factors and had no toxicity.
