Abstract

Brine shrimp (Artemia salina) cytotoxicity assays (BSLT) are one of the commonly used cytotoxic test methods to assess a plant extract's pharmacological activity and toxicity. Thus, this study aimed to examine the toxic levels of Kecubung (Datura metel) leaves extracted using different solvents, namely ethanol and ethyl acetate. The results showed that the LC50value of flower ethanol, flower ethyl acetate, leaf ethanol, and stem ethyl acetate extracts had an LC50value 1000, which was included in the toxic category. The LC50value of flower ethanol is 121.044 ppm, flower ethyl acetate 105.89 ppm, leaf ethanol 639.589 ppm, and stem ethyl acetate 635.276 ppm. Ethanol leaf extract at a concentration of 1000 ppm showed the highest mortality with a percentage of 60% of the total number ofA. salina. The flower ethanol extract showed the highest mortality at a concentration of 250 ppm with a percentage of 67% and at a concentration of 1000 ppm with a percentage of 70% of the total number ofA. salina. Meanwhile, flower ethyl acetate and stem ethyl acetate extract at a concentration of 1000 ppm obtained 100% mortality ofA. salinain the first 6 hours. The result shows that the flower ethyl acetate and stem ethyl acetate extract at a concentration of 1000 ppm is very toxic compared to other concentrations.Keywords:ToxicityArtemia salinaLC50Ethyl acetateEthanol

Abstract

Lokop is one of the villages in Serbajadi District, East Aceh Regency, Aceh Province. The village has a watershed named the river Lokop. This river is located in the Leuser Ecosystem Area which is inhabited by various aquatic biota, especially fish, with the level of diversity and distribution patterns that need to be studied and analyzed conoftened. This study used the purposive sampling method, with the parameters being the level of diversity and distribution patterns of fish in the Lokop river. Data collection was carried out at 5 observation stations based on the typology of river habitat, each station was divided into three substations representing parts of the basin, river bank and river basin approximately 50 m apart. Fish sampling is carried out at each station using a plot of 10 x 10 m2. The number of plots at each station is 9 plots with a total of 45 plots. The results showed that the diversity index (H') value of fish in the Lokop river was 2.39 in the moderate category and the distribution pattern of fish in the Lokop river has a uniform pattern and clusters. Out of a total of 17 species obtained, 8 species (47%) were clustred, 8 species (47%) were uniformly distributed and 1 species (6%) could not be analyzed because only one individual was found.Keywords:FishRiverDiversityDistribution

Abstract

Penelitian ini bertujuan untuk mengidentifikasi bentuk, warna, dan kelimpahan mikroplastik pada lima spesies ikan (Rastrelliger kanagurta, Nemipterus japonicus, Sardinella fimbriata, Megalaspis cordyla, Eleutheronema tetradactylum). Penelitian ini dilakukan dari Juni hingga Agustus 2025, dimana sampel diperoleh dari TPI Pantai Labu. Analisis mikroplastik dilakukan di laboratorium Biologi, FMIPA Universitas Negeri Medan. Proses ekstraksi mikroplastik dimulai dengan memasukkan organ ikan target kedalam larutan H2O2 30% sebanyak 20 ml dan FeSO4  0,05M sebanyak 5 tetes kemudian diinkubasi selama 24 jam pada suhu 40ºC. Selanjutnya organ ikan yang telah terurai disaring menggunakan Whatman, dibilas menggunakan larutan NaCl jenuh, dan hasil saringan ditampung ke dalam cawan petri. Sampel yang telah dikeringkan selanjutnya diamati menggunakan mikroskop stereo. Hasil analisis mikroplastik pada lima spesies ikan menunjukkan kontaminasi, dengan nilai kelimpahan total tertinggi pada spesies Rastrelliger kanagurta (58,7 partikel/individu), diikuti Megalaspis cordyla (44,3), Sardinella fimbriata (31,5), Eleutheronema tetradactylum (29,6), dan Nemipterus japonicus (29,3 partikel/individu). Hasil analisis mikroplastik menunjukkan bentuk mikroplastik yang ditemukan terdiri atas tiga bentuk, yaitu fiber, fragmen, dan film dengan dominasi bentuk fiber dan warna hitam pada seluruh spesies ikan. Selanjutnya hasil uji one way ANOVA menunjukkan bahwa jenis ikan berpengaruh nyata terhadap kelimpahan mikroplastik (p 0,05), di mana R. kanagurta berbeda signifikan dengan N. japonicus, S. fimbriata, dan E. tetradactylum, sedangkan ketiga spesies terakhir tidak berbeda nyata satu sama lain

Abstract

The threat of decreasing populations of Tor species continues to increase due to intensive fishing, habitat destruction and infrequent cultivation efforts. Monitoring of this group of fish must be carried out routinely to reveal the population conditions of fish. This research aims to reveal the population conditions of fish through an analytical approach to the length-weight relationship and fish otolith size. All fish samples were then taken for otoliths to measure their size and analyzed further using a scanning electron microscope (SEM). The research was conducted in the Tangkahan River, North Sumatra, Indonesia from January-March, 2023. The results showed that 43 samples of T. douronensis, 34 samples of T. tambra, and 24 samples of T. tambroides had b values of 2.853, 2.757, and 2.353, respectively. The b value in this study indicated that the fish had a negative allometric growth pattern. The size of the otoliths of the three Tor species shows that T. tambra has the highest length (the distance measured from the anterior to the posterior of the of sagitta otolith) on average reaching 9.31 mm, while T. tambroides and T. douronensis only reach 1.53 mm and 1.83 mm. The results were confirmed by ANOVA showing significant differences (P 0.05) in the mean value of otolith size based on Tor species, where the otolith size of T. tambra was significantly different compared to the other two species. The condition of Tor species in the Tangkahan River has experienced ecological pressure. This indication was concluded from the results of the analysis of the length-weight relationship of fish which showed b value 3 (negative allometric). The results show that the characteristics and size of the otoliths can differentiate Tor species and facilitate identification efforts related to management and conservation.Keywords:Habitat DestructionNegative AllometricAnterior of Sagitta OtolithPosterior of Sagitta Otolith

Abstract

Analisis isi lambung ikan red devil (Amphilophus labiatus) di Danau Toba bertujuan untuk mengetahui jenis makanan dan dampak yang ditimbulkannya sehingga diketahui risiko invasif yang ditimbulkan di Danau Toba. Penelitian dilaksakan pada bulan Februari April 2025. Metode yang digunakan yaitu metode survey ditiga lokasi penelitian secara purposive sampling (Kecamatan Porsea Kab. Toba, Kecamatan Muara Kab. Tapanuli Utara, Kecamatan Onan Runggu Kab. Samosir) Sebanyak 25 sampel red devil dikoleksi dari masing-masing lokasi. Hasil penelitian yang diperoleh menunjukkan bahwa morfologi ikan red devil terdiri dari 2 warna yaitu merah atau oranye, merah kehitaman. Identifikasi panjang relatif usus = 1,40-1,61 menunjukkan ikan red devil omnivora. Makanan utama fitoplankton dengan nilai IP 79,48%, di lokasi I dan II zooplankton sebagai makanan pelengkap dan lokasi III zooplankton potongan akar dan lumut berperan sebagai makanan pelengkap, untuk makanan tambahan di lokasi I dan II yaitu potongan akar, potongan daun dan lumut sedangkan lokasi III hanya potonganlumut sebagai makanan tambahan. Nilai ISC = 0,011-0,014%. Risiko invasif dari ikan red devil di Danau Toba yaitu sebagai menyebabkan kompetisi makanan tinggi serta menyebabkan ikan red devil cepat mendominasi wilayah perairan dan menyebabkan ikan lain berkurang populasinya serta sulitdikendalikan populasinya.
